Identification of binding motifs for skeletal ryanodine receptor and triadin

  • In skeletal muscle cells, depolarization of the transverse tubules (T-tubules) results in Ca$\^$2+/ release from the sarcoplasmic reticulum (SR), leading to elevated cytoplasmic Ca$\^$2+/ and muscle contraction. This process has been known as excitation-contraction coupling (E-C coupling). Several proteins, such as the ryanodine receptor (RyR), triadin, junctin, and calsequestrin (CSQ), have been identified to be involved in the Ca$\^$2+/ release process. However, the molecular interactions between the SR proteins have not been resolved. In the present study, the mechanisms of interaction between RyRl and triadin have been studied by in vitro protein binding and $\^$45/Ca$\^$2+/ overlay assays. Our data demonstrate that the intraluminal loop II of RyR1 binds to triadin in Ca$\^$2+/-independent manner. Moreover, we could not find any Ca$\^$2+/ binding sites in the loop II region. GST-pull down assay revealed that a KEKE motif of triadin, which was previously identified as a CSQ binding site (Kobayasi et al.,2000 JBC) was also a binding site for RyR1. Our results suggest that the intraluminal loop II of RyR could participate in the RyR-mediated Ca$\^$2+/ release process by offering a direct binding site to luminal triadin.

The Microencapsulated Ascorbic Acid Release in vitro and Its Effect on Iron Bioavailability

  • The present study was carried out to examine the stability of microencapsulated ascorbic acid in simulated-gastric and intestinal situation in vitro and the effect of microencapsulated ascorbic acid on iron bioavailability. Coating materials used were polyglycerol monostearate (PGMS) and medium-chain triacylglycerol (MCT), and core materials were L-ascorbic acid and ferric ammonium sulfate. When ascorbic acid was microencapsulated by MCT, the release of ascorbic acid was 6.3% at pH 5 and 1.32% at pH 2 in simulated-gastric fluids during 60 min. When ascorbic acid was microencapsulated by PGMS, the more ascorbic acid was released in the range of 9.5 to 16.0%. Comparatively, ascorbic acid release increased significantly as 94.7% and 83.8% coated by MCT and PGMS, respectively, for 60 min incubation in simulated-intestinal fluid. In the subsequent study, we tested whether ascorbic acid enhanced the iron bioavailability or not. In results, serum iron content and transferring saturation increased dramatically when subjects consumed milks containing both encapsulated iron and encapsulated ascorbic acid, compared with those when consumed uncapsulated iron or encapsulated iron without ascorbic acid. Therefore, the present data indicated that microencapsulated ascorbic acid with both PGMS and MCT were effective means for fortifying ascorbic acid into milk and for enhancing the iron bioavailability.

The clinical efficacy of thoracolumbar fascia release for shoulder pain

  • Objective: This study aimed to elucidate the effects of thoracolumbar fascia release (TLFR) on the degree of pain and disability in patients with shoulder pain. Design: Randomized control trial. Methods: Thirty subjects with shoulder pain participated in this study. They were allocated to TLFR group (n=15) and manual physical therapy (MPT) group (n=15). Shoulder pain and disability index (SPADI) and the score on the visual analogue scale (VAS) were measured before and after TLFR. Results: In the TLFR group, the degree of shoulder pain as indicated by SPADI measured after the intervention significantly differed from that before the intervention (p<0.05); moreover, in the MPT group, the degree of shoulder pain was significantly lower (p<0.05). The data of the 2 groups before the intervention significantly differed from those after the intervention (p<0.05). SPADI significantly differed within the groups (p<0.05), but not between the groups. The sum of SPADI did not differ significantly between the groups. The VAS scores of shoulder pain measured before the intervention significantly differed from those measured after the intervention (p<0.05) in the both groups. After the intervention, shoulder pain decreased significantly in the TLFR group as compared to that in the MPT group. Conclusions: TLF release was effective in reducing shoulder pain. The results of this study can be applied in clinical practice for TLFR performed to reduce shoulder pain. Further studies will need to be performed to elucidate the effects of TLFR on functional recovery.

Modeling of the Environmental Behavior of Tritium Around the Nuclear Power Plants

  • The relationship between the tritium release rate from the nuclear power plant and tritium concentration in the environment around the Kori site was modeled. The tritium concentration in the atmosphere was calculated by multiplying the release rates and $\chi$/Q values, and the d3V deposition rate at each sector according to the direction and the distance was obtained using a dry deposition velocity. The area around Kori site was divided into 6 zones according to the deposition rate. The six zones were divided into 14 compartments for the numerical simulation. Transfer coefficients between the compartments were derived using site characterization data. Source terms were calculated from the dry deposition rates. Tritium concentration in surface soil water and groundwater was calculated based upon a compartment model. The semi-analytical solution of the compartment model was obtained with a computer program, AMBER. The results showed that most of tritium deposited onto the land released into the atmosphere and the sea. Also, the estimated concentration in the top soil agreed well to that measured. Using the model, tritium concentration was predicted in the case that the tritium release rates were doubled.

Development of a Mechanistic Fission Gas Release Model for LWR $UO_2$ Fuel Under Steady-State Conditions

  • A mechanistic model has been developed to predict the release behavior of fission gas during steady-state irradiation of LWR UO$_2$ fuel. Under the assumption that UO$_2$ grain surface is composed of fourteen identical circular faces and grain edge bubble can be represented by a triangulated tube around the circumference of three circular grain faces, it introduces the concept of continuous formation of open grain edges tunnels that is proportional to grain edge swelling. In addition, it takes into account the interaction between the gas release from matrix to grain boundary and the reintroduction of gas atoms into the matrix by the irradiation-induced re-solution of grain face bubbles. It also treats analytically the behavior of intragranular, intergranular, and grain edge bubbles under the assumption that both intragranular and intergranular bubbles are uniform in both radius and number density. Comparison of the present model with experimental data shows that the model's prediction produces reasonable agreement for fuel with centerline temperatures of 1000 to 140$0^{\circ}C$, wide scatter band for fuel with centerline temperatures lower than 100$0^{\circ}C$, and underprediction for fuel with centerline temperatures higher than 140$0^{\circ}C$.

COSMOS : A Computer Code for the Analysis of LWR $UO_2$ and MOX Fuel Rod

  • A computer code COSMOS has been developed based on the CARO-D5 for the thermal analysis of LWR UO$_2$ and MOX fuel rod under steady-state and transient operating conditions. The main purpose of the COSMOS, which considers high turnup characteristics such as thermal conductivity degradation with turnup and rim formation at the outer part of fuel pellet, is to calculate temperature profile across fuel pellet and fission gas release up to high burnup. A new mechanistic fission gas release model developed based on physical processes has been incorporated into the code. In addition, the features of MOX fuel such as change in themo-mechanical properties and the effect of microscopic heterogeneity on fission gas release have been also taken into account so that it can be applied to MOX fuel. Another important feature of the COSMOS is that it can analyze fuel segment refabricated from base irradiated fuel rods in commercial reactors. This feature makes it possible to analyze database obtained from international projects such as the MALDEN and RISO, many of which were collected from refabricated fuel segments. The capacity of the COSMOS has been tested with some number of experimental results obtained from the HALDEN, RISO and FIGARO programs. Comparison with the measured data indicates that, although the COSMOS gives reasonable agreement, the current models need to be improved. Burning Characteristics of Wood-based Materials using Cone Calorimeter and Inclined Panel Tests

  • Research to discuss the fire performance of materials requires tools for measuring their burning characteristics and validated fire growth models to predict fire behavior of the materials under specific tire scenarios using the measured properties as input for the models. In this study, burning characteristics such as time to ignition, weight loss rate, flame spread, heat release rate, total heat evolved, and effective heat of combustion for four types of wood-based materials were evaluated using the cone calorimeter and inclined panel tests. Time to ignition was affected by not only surface condition and specific gravity of the tested materials but also the type and magnitude of heat source. Results of weight loss rate, measured by inclined panel tests, indicated that heat transfer from the contacted flame used as the heat source into the inner part of the specimen was inversely proportional to specific gravity of material. Flame spread was closely related with ignition time at the near part of burning zone. Under constant and severe external heat flux, there was little difference in weight loss rate and total heat evolved between four types of wood-based panels. More applied heat flux caused by longer ignition time induced a higher first peak value of heat release rate. Burning characteristics data measured in this study can be used effectively as input for fire growth models to predict the fire behavior of materials under specific fire scenarios.

Enhancing the ability of strain energy release rate criterion for fracture assessment of orthotropic materials under mixed-mode I/II loading considering the effect of crack tip damage zone

  • In this study, considering dissipated energy in fracture process zone (FPZ), a novel criterion based on maximum strain energy release rate (SER) for orthotropic materials is presented. General case of in-plane loading for cracks along the fibers is assumed. According to the experimental observations, crack propagation is supposed along the fibers and the reinforcement isotropic solid (RIS) concept is employed as a superior model for orthotropic materials. SER in crack initiation and propagation phases is investigated. Elastic properties of FPZ are extracted as a function of undamaged matrix media and micro-crack density. This criterion meaningfully links between dissipated energy due to toughening mechanisms of FPZ and the macroscopic fracture by defining stress intensity factors of the damaged zone. These coefficients are used in equations of maximum SER criterion. The effect of crack initiation angle and the damaged zone is considered simultaneously in this criterion and mode II stress intensity factor is extracted in terms of stress intensity factors of damage zone and crack initiation angle. This criterion can evaluate the effects of FPZ on the fracture behavior of orthotropic material. Good agreement between extracted fracture limit curves (FLC's) and available experimental data proves the ability of the new proposed criterion.

Comparative Evaluation on the Cost Analysis of Software Development Model Based on Weibull Lifetime Distribution (와이블 수명분포에 근거한 소프트웨어 개발모형의 비용 분석에 관한 비교 평가)

  • In this study, the finite-failure NHPP software reliability model was applied to the software development model based on the Weibull lifetime distribution (Goel-Okumoto, Rayleigh, Type-2 Gumbe), which is widely used in the software reliability field, and then the cost attributes were compared and evaluated. For this study, failure time data detected during normal operation of the software system were collected and used, the most-likelihood estimation (MLE) method was applied to the parameter estimation of the proposed model, and the calculation of the nonlinear equation was solved using the binary method. As a result, first, in the software development model, when the cost of testing per unit time and the cost of removing a single defect increased, the cost increased but the release time did not change, and when the cost of repairing failures detected during normal system operation increased, the cost increased and the release time was also delayed. Second, as a result of comprehensive comparative analysis of the proposed models, it was found that the Type-2 Gumble model was the most efficient model because the development cost was lower and the release time point was relatively faster than the Rayleigh model and the Goel-Okumoto basic model. Third, through this study, the development cost properties of the Weibull distribution model were newly evaluated, and the analyzed data is expected to be utilized as design data that enables software developers to explore the attributes of development cost and release time.

Movement of Pacific cod Gadus macrocephalus in the Korean Southeast Sea, ascertained through pop-up archival tags and conventional tags (Pop-up식 전자태그와 재래식 태그로 알게된 한국 남해동부해역 대구 Gadus macrocephalus의 이동)

  • To estimate the movement of Pacific cod Gadus macrocephalus in the Korean Southeast Sea, three type tags were used. A total of 97 Pacific cod were tagged and released with either archival tags or conventional tags. Of these releases, commercial fishermen recovered thirteen conventional tags, and five of seven pop-up tags transmitted data to Argos satellites. Pacific cod began to move towards East Sea after release, and they spent most of their time at depths of 100 to 300m, water temperatures of 0.8 to $14.0^{\circ}C$. However, geographical ranges of their movement limited to area around the southern East Sea. Pacific cod attached conventional tag were recaptured near the release site(Jinhae Bay: main spawning ground) about one or two year after release. Data obtained from tagging investigations suggest that they migrated annually from spawning ground to habtat of the Korean Southeast Sea.
