The Production of mutant protein by a transcription-based mechanism and in vivo technique for determining transcriptional mutagenesis

  • When an elongating RNA polymerase encounters DNA damage on the template strand of a transcribed gene it can either be arrested by or be transcribed through the lesion. Lesions that arrest RNA polymerases are thought to be subject to transcription-coupled repair, whereas that damage that is bypassed can cause miscoding, resulting in mutations in the transcript (transcriptional mutagenesis). We have developed a technique using a plasmid-based luciferase reporter assay to determine the extent to which a particular type of DNA base modification is capable of causing transcriptional mutagenesis in vivo. The system uses Escherichia coli strains with different DNA repair backgrounds and is designed to detect phenotypic changes caused by transcriptional mutageneis under nongrowth conditions. In addition, this method is capable of indicating the extent to which a particular DNA repair enzyme (or pathway) suppresses the occurrence of transcriptional mutagenesis. Thus, this technique provides a tool with which the effects of various genes on non-replication-dependent pathways resulting in the generation of mutant proteins can be gauged.

  • In the existed study, a fire outbreak in a reinforced concrete structure looses the organism by the different contraction and expansion of hardened cement pastes and aggregate, and causes cracks by thermal stress, leading to the deterioration of the durability. So accurate diagnosis of deterioration is needed based on mechanism of fire deterioration in general concrete structures. Fundamental information and data on the properties of concrete exposed to high temperature are necessary for accurate diagnosis of deterioration. Therefore, this study is willing to propose fundamental data for accurate diagnosis of deteriorated concrete structure by fire damage with experiment according to the curing conditions.

  • The nano imprint process is one of the next generation lithography has been mentioned as one of major nanoreplication techniques because it is simple process, low cost, high replication fidelity and relatively high throughput. This process requires a surface contact between a template with patterns and a wafer on a stage. After contact, the vertical moving the template to the wafer causes some directional motions of the stage. Thus the stage must move according to the motions of the template to avoid the damage of the transferred patterns on the wafer. This study is to develop the wafer stage with a passive compliance to overcome the damage. This stage is designed with the concept like that it has a monolithic, symmetry and planar 6-DOF mechanism.

  • Acoustic Emission was monitored during tensile test and loading-holding-unloa-ding cycle test for two types (notched and unnotched) of CFRP specimens. AE activities showed that the fiber breakage during tensile tests depended upon the specimen geometry. We obtained new AE parameter such as the ration (damage ratio= AE events during unloading test / AE events during loading test) and the felicity ratio from which we investigated dynamic fracture process of CFRP specimens. The damage ratio of AE events was shown to be a good indicator to distinguish the generated fracture mechanism, such as fiber breakage and delamination. Also, ultrasonic testing results after loading-holding-unloading cycle test were good agreement with AE test results to detect defects or fiber breakage.

Regulation of Nrf2 Mediated Phase II Enzymes by Luteolin in human Hepatocyte

  • This study attempted to confirm the antioxidative potential of luteolin against tert-butyl hydroperoxide (t-BHP) induced oxidative damage and to investigate its molecular mechanism related to glutathione (GSH)-dependent enzymes in HepG2 cells. Treatment with luteolin resulted in attenuation of t-BHP induced generation of reactive oxygen species (ROS) and oxidative stress-mediated cell death. In addition, accelerated expression of GSH-dependent antioxidative enzymes, glutathione peroxidase (GPx) and glutathione reductase (GR), and heme oxygenase (HO)-1, as well as strengthened GSH content was induced by treatment with luteolin, which was in accordance with increased nuclear translocation of nuclear factor-erythroid 2 p45-related factor 2 (Nrf2), a transcription factor for phase 2 enzymes, in a dose-dependent manner. These results suggest that the cytoprotective potential of luteolin against oxidative damage can be attributed to fortified GSH-mediated antioxidative pathway and HO-1 expression through regulation of Nrf2 in HepG2 cells.

Ar 중성빔과 $BCl_3$를 이용한 $ZrO_2$의 원자층 식각에 관한 연구

  • 본 연구에서는 중성빔을 이용한 Atomic Layer Etching(ALET) system을 이용하여 $ZrO_2$의 atomic layer etching mechanism에 대하여 연구하였다. Ar neutral beam irradiation dose와 $BCl_3$ gas pressure의 변화에 따라 $ZrO_2$ etch rate와 RMS roughness를 관찰했을 때, Ar neutral beam irradiation dose이 $1.485{\times}10^{16}atoms/cm^2{\bullet}cycle$ 이상이고 $BCl_3$ gas pressure가 0.15mTorr 이상 일 때 $ZrO_2$ etch rate은 $1.07\;{\AA}/cycle$의 일정한 값에서 유지됨을 확인하였다. 그리고 ALET와 ICP Etcher을 통해 $ZnO_2$를 각각 식각하여 physically or chemically damage를 비교한 결과, ALET가 기존의 ICP Etcher system보다 $ZrO_2$ 식각공정에 대해 적은 damage를 받는 것을 ARXPS를 통해 관찰 하였다.

  • This paper describes an investigation on bearing failure due to spragging that has been continuously occurred in turbine hearings. The spragging is defined as the damage found on the leading edge of unloaded pads in the tilting pad journal bearing, In general, the damage mechanism by spragging is classified into fatifgue failure, The principle cause of spragging could be thought as the self-excited vibration by the absence of a stable static equilibrium position of upper pads with no preload. Because of serious consequences of system breakdowns due to bearing failures, determination ar the causes of failure and effective method for countermeasures are very important. This paper describes both the causes of spragging and countermeasures for prevention of such failure, which are taken place in the electric power plants.

Protective Effect of Metabolized Chungpesagan-tang on Hypoxia/Reperfusion Induced-PC12 Cell Damage (저산소/재관류로부터 청폐사간탕의 PC12 세포 보호 효과)

  • This research was performed to investigate the protective effect of Chungpesagan-tang (CST) from hypoxia/reperfusion induced-PC12 cell damage. To elucidate the mechanism of the protective effect of CST, cell viability, changes in activities of superoxide dismutase, glutathione peroxidase, catalase, caspase 3 and the production of malondialdehyde were observed after treating PC12 cells with CST which was metabolized by rat liver homogenate. Pretreatment of CST with liver homogenate appeared to increase its protective effect against hypoxia/reperfusion insult. The result showed that CST exhibited the highest protective effect against hypoxia/reperfusion at the dose of $1\;{\mu}g/ml$ in PC12 cells, probably by recovering the redox enzyme activities and MDA to control level.

  • With the use of a new method the deformation mechanism of high-temperature-brazed joints can be obtained in a very short time. For that purpose a SEM(Scanning Electron Microscope) was equipped with a high temperature tensile testing machine. By means of SEM-investigation the damage behaviour of high-temperature-brazed joints is exa mined at elevated temperature. Based on these it is possible to make a qualification of the influence over single parts on the damage beginning and behaviour in dependence of temperature. This shall be shown exemplarily for the high temperature material NiCr20TiAl (Nimonic 80A).
