Antioxidant Compounds from the Root Bark of Hibiscus syriacus

  • A new lignan named as hibiscuside, (+) pinoresinol 4-O-[-glucopyranosyl (12) -rhamnoside] (1), and a known lignan, syringaresinol (2) were isolated from the root bark of Hibiscus syriacus together with two feruloyltyramines (3,4) and three known isoflavonoids (5,6,7). The structures of these compounds have been established on the basis of their NMR, Mass, UV spectra. Among these phenolic compounds,6-O-acetyl daidzin (5), 6-O- acetyl genistin (6), and 3-hydroxy daidzein (7) with IC50 values of 8.2, 10.6, and 4.1 M, respectively, significantly inhibited lipid peroxidation in rat liver microsomes Hibiscuside (1), E and Z-N-feruloyl tyramines (3,4) exhibited moderate antioxidant activity.

Production of a High Value-Added Soybean Containing Bioactive Mevinolins and Isoflavones

  • The production of mevinolin, a potent hypocholesterolemic drug, and the bioconversion of isoflavones were investigated in soybeans fermented with Monascus pilosus KFRI-1140. The highest yields of 2.94 mg mevinolins and 1.13 mg isoflavone aglycones per g dry weight of soybean were obtained after 20 days of fermentation. Mevinolin was present in the fermentation substrate predominantly in the hydroxycarboxylate form (open lactone, 94.8$\sim$96.7%), which is currently being used as an hypocholesterolemic agent. The significant (p<0.01) bioconversion (96.6%) of the glucoside isoflavones (daidzin, glycitin, genistin) present in the soybean to the bioactive aglycones (daidzein, glycitein, genistein), with a 15.8-fold increase of aglycones was observed. The results suggest that Monascus-fermented soybean has potential as a novel medicinal food or multifunctional food supplement.

  • Five flavonoids were isolated from the heartwoods of Maackia fauriei. On the basis of spectroscopic evidences, the structure of these compounds were established as (-)maa ckiain(3-hydroxy-8,9-methylenedioxypterocarpan), ononin(4'-methoxyisoflavone- 7-O-${\beta}$-D-glucoside), wistin(4',6-dimethoxyisoflavone-7-O-${\beta}$-D-glucoside), daidzin(4'-hydroxyisoflavone-7-O-${\beta}$-D-glucoside) and 2'-methoxy-4'- hydroxyisoflavone-6-O-${\beta}$-D-glucoside. In these compounds, 2'-methoxy-4'-hydroxyisoflavone-6-O-${\beta}$-D-glucoside was for the first time isolated from the plant source.

Dihydrodaidzein production from soybean hypocotyl extract by human intestinal bacterium MRG-1

  • Phytoestrogenic S-equol production in human gut exclusively depends on the biotransformation of daidzein to dihydrodaidzein (DHD). With a growing demand for the DHD enriched biomaterials, the commercial soybean hypocotyl extract (SHE) was chosen as a substrate for the microbial DHD production by human gut bacterium MRG-1, anaerobic DHD producer. To optimize the production of DHD, anaerobic fermentation conditions, including sterilization time, growth stage of inoculum, and growth media, were investigated. Maximum DHD production (1.2 g/L) was achieved after 48 h incubation when 1% (w/v) of SHE in the 20-min-sterilized Gifu Anaeboic Medium media was inoculated with OD600 0.3-0.4 of MRG-1. This is the first report that crude soy biomaterial, instead of pure compounds, such as daidzin and daidzein, is utilized for the production of the DHD enriched biomaterial.

Quality Properties and Isoflavone Contents of Chungkukjang Containing Isoflavone Extracted from Arrowroot (Pueraria lobata Ohwi) (칡 이소플라본을 첨가한 청국장의 품질 특성 및 이소플라본 함량)

  • This study was conducted to determine the characteristics of chungkukjang with added isoflavone extracted from arrowroot and to determine its utility as a functional food substance. Crude isoflavone was prepared from arrowroot by the ethanol extraction method and frozen dried (AI). Samples of chungkukjang each had different amounts of isoflavone extracts added to them: 0 (control), 1.76 (AC1-chungkukjang), 3.52 (AC2-chungkukjang), and 7.11 (AC3-chungkukjang) g/kg. The pH, color, slime material content, calcium, and isoflavone of each sample were measured to investigate the quality and changes in isoflavone content. As AI increased, the pH of chungkukjang decreased to 7.46~7.53 compared to the pH of control (7.63). The slime material content range increased to 4.46~6.16%. However, there was no significant difference in the general components of chungkukjang between each of the groups (Con, AC1, AC2, AC3). In colors of chungkukjang, values for $L^*$, $a^*$, and $b^*$ decreased as AI increased. Meanwhile, the calcium content of AC1, AC2 and AC3 tended to increase by 11.18~12.82% compared to the control. This may be due to the influence of Ca in the arrowroot extract powder. There was a remarkable increase in total isoflavone content, by 30.81~130.66%, with an order of AC3>AC2>AC1. In all of the groups, the content of genistein and daidzein, aglycone form, increased dramatically, by 65.00~128.34%, and 89.38~142.91%, respectively, compared to the control. In AC3, in particular the genistin and daidzin content increased by 103.47% and 188.13%. These results showed that AI can be used as a source of isoflavone supplementation in chungkukjang.

Molecular Cloning and Functional Expression of Extracellular Exo-β-(1,3)-Glucanase from Candida fermentati SI (Candida fermentati SI의 exo-β-(1,3)-glucanase유전자의 클로닝 및 그 특성)

  • An isoflavone glucosidase that catalyzes the hydrolysis of isoflavone glucosides into glucose and corresponding aglycones was purified from Candida fermentati SI. The N-terminal sequence was determined to be GLNCDYCN. We designed degenerate primers on the basis of these amino acid sequences and successfully cloned the full structural gene sequence of the isoflavone glucosidase using inverse PCR. The exo-β-(1,3)-glucanase gene consists of 1227 base-pair nucleotides, encoding a 408-amino-acid sequence that shares 41–96% amino acid homology with other yeast exo-β-(1,3)-glucanases belonging to glycoside hydrolase family 5. The recombinant exo-β-(1,3)-glucanase was expressed in Pichia pastoris X-33, using a pPICZA vector system, and further characterized. The molecular mass of the purified exo-β-(1,3)-glucanase was estimated by SDS-PAGE to be 47 kDa. The optimal pH and temperature were pH 4.5 and 40℃, respectively. The Km values of the purified exo-β-(1,3)-glucanase for daidzin and genistin were 0.12 mM and 0.14 mM, respectively. The Vmax values of the purified isoflavone glucosidase were 945.03 U/mg for daidzin and 835.92 U/mg and for genistin.

A Study on the Estrogenicity of Korean Arrowroot (Pueraria thunbergiana) (한국산 칡의 Estrogen 활성에 관한 연구)

  • To assess the estrogenicities of Korean arrowroot (Pueraria thunbergiana) the contents of nine phytochemicals which are known to present were analyzed by HPLC. Also the estrogenecities of these phytochemicals were assayed using estrogen receptor dependent transcriptional expression assay. Daidzein and puerarin are major compounds in arrowroot, which were contained in the order of: root>stem>leaf>flower>seed. Among the assayed phytochemicals daidzein, genistein, biochanin A, formononetin, puerarin and genistin were highly estrogenic. The total estrogenicities in different parts of arrowroot and those by regions in Korea were also assessed. Roots had the highest estrogenic effects. The estrogenicities also were shown in stem and leaf. The differences in the estrogenicities of arrowroots by regions in Korea were shown. These results demonstrated that the estrogenic phytochemicals content was the highest in Korean arrowroot and also present in stem and leaf.

Isoflavonoid Contents, Antibacterial Activities, and Physiological Activities of Cheonggukjang Made from Sword Bean (작두콩 첨가 청국장의 Isoflavonoids 함량과 항균력 및 생리활성)

  • This research aimed to examine the isoflavonoid contents, antibacterial activities, and physiological activities of Cheonggukjang made from sword bean (CS). The effects of adding sword bean were compared with those of raw materials (RM), steamed materials (SM), and traditional Cheonggukjang (TC). In the case of the antibacterial activity on Gram-positive bacteria, the result of CS in ethanol extract was the highest in Bacillus cereus, and the result of water extract was the highest in Staphylococcus aureus. Optimization of Iso-flavonoids Extraction Process from Kudge Using Ultrasonic Irradiation Energy (초음파에너지를 이용한 칡으로부터 이소플라보노이드의 추출공정 최적화)

  • In this study, we used the ultrasonic extraction process as a method to extract antioxidant substances from kudzu, and measured the content of iso-flavonoids puerarin, daidzein, daidzin contained in kudzu. The response surface methodology which is a statistical analysis method for optimizing the extraction amount of iso-flavonoids from the kudzu and the process condition for maximizing the yield was applied. It is the final objective of this study to effectively derive the condition of the process that matches the target response with a minimum number of experiments and analyze the effect of each process condition on the response. In the response surface methodology, the central composite design was applied and the optimum condition was analyzed, and the three independent variables were set to ultrasonic irradiation time, volume ratio of ethanol/ultrapure water, ultrasonic irradiation power. Using the response surface methodology, the optimum conditions with the maximum extraction yield and the content of iso-flavonoids were evaluated as ultrasonic irradiation time (24.75 min), ethanol / ultrapure water volume ratio (39.75 vol%), ultrasonic irradiation power (592.36 W). The overall satisfaction level appears as high as 0.8938, which is recognized at a significance level within 5%. As a result of analyzing the optimization process, it was confirmed that the ultrasonic irradiation time is the factor that most affects the responses.

Isoflavones and biotransformed dihydrodaidzein production with in vitro cultured callus of Korean wild arrowroot Pueraria lobata (한국산 야생칡 캘러스에서의 이소플라본 및 생물전환에 의한 디하이드로다이드제인 분석)

  • Pueraria lobata, a medicinally important leguminous plant produces various isoflavones including puerarin, daidzin and daidzein which are metabolized to equol via dihydrodaidzein and tetrahydrodaidzein by the bacterial fermentation of natural isoflavone sources in human intestines. In this study, we described callus proliferation and isoflavone production in callus of Korean wild arrowroot and dihydrodaidzein biosynthesis in callus extract fermented with Pediococcus pentosaceus. Proliferation was the best at callus cultured in the medium containing 1.0 mg/L TDZ and 1.0 mg/L NAA at light condition for 12 days. Puerarin was significantly more produced at callus cultured in the medium containing 2.0 mg/L kinetin and 1.0 mg/L NAA at dark condition for 16 days, but daidzin and daidzein were not significant. Callus extract was successfully fermented with P. pentosaceus and dihydrodaidzein, which is one of equol precursors formed by biotransformation, was confirmed to be produced. These results will facilitate mass production of callus and isoflavones as equol precursors from Korean wild arrowroot and can be applied for the production of equol by biotransformation in vitro.
