식생가뭄반응지수(VegDRI) 국내 적용방안 기초연구 (Preliminary Research on Domestic Application of Vegetation Drought Response Index (VegDRI))

  • 최근 가뭄 모니터링을 위해 과거에 비하여 고해상도의, 물리적으로 기반을 두는 정보가 요구되고 있다. 기존에 주로 활용하고 있는 통계적 방법론 기반의 가뭄지수들은 지니고 있는 한계에 대해 여러 개선과정을 거치고 있으나, 기상변수로부터 지표상의 식생 관련 변수로의 전파 과정에 대한 개별 통계적 가뭄지수 간의 관계 설명이 매우 어렵다. 이와 같은 관계로, 국내 유역에서의 물리적 기반을 둔 고해상도 가뭄 판단방법에 대한 시도가 필요한 시점이다. Brown et al. (2008)은 위성기반 식생정보, 기상학적 가뭄지수, 지형학적 조건을 고려한 식생가뭄반응지수(Vegetation Drought Response Index; 이하 VegDRI)를 개발하였다. 학습자료에 대해 CART 기반의 경험적 모델을 구축하여, 격자마다 근-실시간 자료를 적용한 VegDRI를 산출하여 고해상도의 지도를 산출하는 방식을 제시하였다. VegDRI는 NCDC의 U.S. Drought Monitoring에 활용되고 있으며, NOAA의 Drought Task Force Assessment Protocol에서는 가뭄 모니터링의 기준으로 설정되어 있다. 본 연구에서는 국내에 VegDRI를 적용하고자 필요한 자료수집 및 전처리 과정을 거쳐 결과를 도출하였다. 기상청 ASOS 기상관측소에서 얻은 기상변수, MODIS 위성으로부터 추출된 정규식생지수(Normalized Difference Vegetation Index; NDVI), 지형학적 정보와 기상학적 가뭄지수(SPI, PDSI)를 기계학습으로 모델링하여 VegDRI를 산출하였다. 산출된 VegDRI 공간분포도에 대하여 기존에 활용되던 유관기관의 가뭄 판단방법과의 유사성과 차이점을 비교 검토하여 적용성을 평가하였다.

이변량 빈도해석을 이용한 낙동강 유역의 지역 가뭄 위험도 평가 기준 산정 (Estimation of Regional Drought Risk Assessment Criteria Using Bivariate Frequency Analysis in Nakdong River Basin)

  • 가뭄으로 인해 발생할 수 있는 가장 심각한 피해는 용수 부족으로 인한 수자원 시스템의 용수공급 실패이며, 따라서 가뭄 위험도는 사용 가능한 용수의 부족과 관련하여 정량화되어야 한다. 이러한 맥락에서 수자원 시스템의 가뭄 위험도를 평가하기 위해 주로 신뢰도(reliability), 회복도(resiliency) 및 취약도(vulnerability)와 같은 세 가지 이수안전도 평가지표가 사용된다. 이러한 평가지표는 각각 용수공급 실패가 평균적으로 얼마나 자주 발생하는지, 얼마나 오래 지속되는지, 또한 어느 정도의 규모로 발생하는지를 위험도를 정량화하는 것으로, 용수공급 실패사상의 빈도, 지속기간 및 심도를 나타낸다. 본 연구에서 DRI(Drought Risk Index)는 신뢰도, 평가도 및 회복도의 가중평균값으로 정의되며, 이는 지속기간과 심도를 변수로 하는 이변량 가뭄빈도해석과 같은 변수를 공유한다. 본 연구에서는 두 가지 형태의 DRI 를 이용하여 지역 가뭄 위험도 평가 기준 산정 방안을 제시하였다. DRI_O(observed DRI)는 용수부족 시계열을 통해 산정된 공급실패 사상으로부터 산정되며, DRI_D(designed DRI)는 이변량 빈도해석을 통해 산정된 특정 지속기간을 갖는 확률가뭄심도로부터 계산된다. 기후변화 시나리오를 이용해 DRI_O 를 산정함으로써 미래의 이수안전도를 예측할 수 있으며, 이를 DRI_D 와 비교하여 지역의 용수부족으로 인한 가뭄 위험도를 산정하는 방법을 제안하였다. 또한 기존에는 주로 과거 최대 가뭄사상을 목표안전도로 설정하였으나 DRI_D 를 이용하여 보다 현실적인 목표안전도를 설정할 수 있다. 낙동강 권역의 10 개 중권역의 10 개 기후변화 시나리오를 대상으로 분석을 수행한 결과 병성천 유역과 형산강 유역이 각각 최저 및 최고 위험도를 갖는 것으로 분석되었으며, 지역 안전도 기준은 평균적으로 재현기간 5-20 년 사이의 범위를 갖는 것으로 나타났다.

식생가뭄반응지수 (VegDRI)를 활용한 위성영상 기반 가뭄 평가 (Satellite-based Hybrid Drought Assessment using Vegetation Drought Response Index in South Korea (VegDRI-SKorea))

  • The development of drought index that provides detailed-spatial-resolution drought information is essential for improving drought planning and preparedness. The objective of this study was to develop the concept of using satellite-based hybrid drought index called the Vegetation Drought Response Index in South Korea (VegDRI-SKorea) that could improve spatial resolution for monitoring local and regional drought. The VegDRI-SKorea was developed using the Classification And Regression Trees (CART) algorithm based on remote sensing data such as Normalized Difference Vegetation Index (NDVI) from MODIS satellite images, climate drought indices such as Self Calibrating Palmer Drought Severity Index (SC-PDSI) and Standardized Precipitation Index (SPI), and the biophysical data such as land cover, eco region, and soil available water capacity. A case study has been done for the 2012 drought to evaluate the VegDRI-SKorea model for South Korea. The VegDRI-SKorea represented the drought areas from the end of May and to the severe drought at the end of June. Results show that the integration of satellite imageries and various associated data allows us to get improved both spatially and temporally drought information using a data mining technique and get better understanding of drought condition. In addition, VegDRI-SKorea is expected to contribute to monitor the current drought condition for evaluating local and regional drought risk assessment and assisting drought-related decision making.

  • The biomedical applications of chitosan have been widely researched. FN mediates its biological effects through binding to the hetero-dimeric transmembrane glycoproteins, integrins, which physically couple the cytoskeleton to the ECM. FN binds to the integrin through a consensus site including the Arg-Gly-Asp (RGD) sequence within tenth type III module (Ruoslahti & Pierschbacher 1987). A short sequence Pro-His-Ser-Arg-Asn (PHSRN) has also been identified as a synergistic motif within ninth type III module for binding to ${\alpha}$5${\beta}$1 integrin (Aota et al. 1994). (omitted)

Extracellular S100A4 negatively regulates osteoblast function by activating the NF-κB pathway

  • Patients with inflammatory bone disease or cancer exhibit an increased risk of fractures and delayed bone healing. The S100A4 protein is a member of the calcium-binding S100 protein family, which is abundantly expressed in inflammatory diseases and cancers. We investigated the effects of extracellular S100A4 on osteoblasts, which are cells responsible for bone formation. Treating primary calvarial osteoblasts with recombinant S100A4 resulted in matrix mineralization reductions. The expression of osteoblast marker genes including osteocalcin and osterix was also suppressed. Interestingly, S100A4 stimulated the nuclear factor-kappaB (NF-${\kappa}B$) signaling pathway in osteoblasts. More importantly, the ex vivo organ culture of mouse calvariae with recombinant S100A4 decreased the expression levels of osteocalcin, supporting the results of our in vitro experiments. This suggests that extracellular S100A4 is important for the regulation of bone formation by activating the NF-${\kappa}B$ signaling pathway in osteoblasts.

ST5 Positively Regulates Osteoclastogenesis via Src/Syk/Calcium Signaling Pathways

  • For physiological or pathological understanding of bone disease caused by abnormal behavior of osteoclasts (OCs), functional studies of molecules that regulate the generation and action of OCs are required. In a microarray approach, we found the suppression of tumorigenicity 5 (ST5) gene is upregulated by receptor activator of nuclear $factor-{\kappa}B$ ligand (RANKL), the OC differentiation factor. Although the roles of ST5 in cancer and ${\beta}-cells$ have been reported, the function of ST5 in bone cells has not yet been investigated. Knockdown of ST5 by siRNA reduced OC differentiation from primary precursors. Moreover, ST5 downregulation decreased expression of NFATc1, a key transcription factor for osteoclastogenesis. In contrast, overexpression of ST5 resulted in the opposite phenotype of ST5 knockdown. In immunocytochemistry experiments, the ST5 protein is colocalized with Src in RANKL-committed cells. In addition, ST5 enhanced activation of Src and Syk, a Src substrate, in response to RANKL. ST5 reduction caused a decrease in RANKL-evoked calcium oscillation and inhibited translocation of NFATc1 into the nucleus. Taken together, these findings provide the first evidence of ST5 involvement in positive regulation of osteoclastogenesis via Src/Syk/calcium signaling.

Validity of the dietary reference intakes for determining energy requirements in older adults

  • BACKGROUND/OBJECTIVES: The objectives of this study were to evaluate the accuracy of the Dietary Reference Intakes (DRI) for estimating the energy requirements of older adults, and to develop and validate new equations for predicting the energy requirements of this population group. MATERIALS/METHODS: The study subjects were 25 men and 23 women with a mean age of $72.2{\pm}3.9\;years$ and $70.0{\pm}3.3\;years$, and mean BMI of $24.0{\pm}2.1$ and $23.9{\pm}2.7$, respectively. The total energy expenditure (TEE) was measured by using the doubly labeled water (DLW) method, and used to validate the DRI predictive equations for estimated energy requirements (EER) and to develop new EER predictive equations. These developed equations were cross-validated by using the leave-one-out technique. RESULTS: In men, the DRI equation had a -7.2% bias and accurately predicted the EER (meaning EER values within ${\pm}10%$ of the measured TEE) for 64% of the subjects, whereas our developed equation had a bias of -0.1% and an accuracy rate of 84%. In women, the bias was -6.6% for the DRI equation and 0.2% for our developed equation, and the accuracy rate was 74% and 83%, respectively. The predicted EER was strongly correlated with the measured TEE, for both the DRI equations and our developed equations (Pearson's r = 0.915 and 0.908, respectively). CONCLUSIONS: The DRI equations provided an acceptable prediction of EER in older adults and these study results therefore support the use of these equations in this population group. Our developed equations had a better predictive accuracy than the DRI equations, but more studies need to be performed to assess the performance of these new equations when applied to an independent sample of older adults.

PARK2 Induces Osteoclastogenesis through Activation of the NF-κB Pathway

  • Osteoclast generation from monocyte/macrophage lineage precursor cells needs to be tightly regulated to maintain bone homeostasis and is frequently over-activated in inflammatory conditions. PARK2, a protein associated with Parkinson's disease, plays an important role in mitophagy via its ubiquitin ligase function. In this study, we investigated whether PARK2 is involved in osteoclastogenesis. PARK2 expression was found to be increased during the receptor activator of nuclear factor-κB ligand (RANKL)-induced osteoclast differentiation. PARK2 gene silencing with siRNA significantly reduced osteoclastogenesis induced by RANKL, LPS (lipopolysaccharide), TNFα (tumor necrosis factor α), and IL-1β (interleukin-1β). On the other hand, overexpression of PARK2 promoted osteoclastogenesis. This regulation of osteoclastogenesis by PARK2 was mediated by IKK (inhibitory κB kinase) and NF-κB activation while MAPK (mitogen-activated protein kinases) activation was not involved. Additionally, administration of PARK2 siRNA significantly reduced osteoclastogenesis and bone loss in an in vivo model of inflammatory bone erosion. Taken together, this study establishes a novel role for PARK2 as a positive regulator in osteoclast differentiation and inflammatory bone destruction.

식사구성안의 생애주기별 권장 식단의 영양 적정성 분석 -2015 한국인 영양소 섭취기준- (Nutritional Adequacy Analysis of Recommended Menu in Dietary Reference Intakes for Koreans 2015)

  • Objectives: Recommended Menu (RM) prepared according to the Target Pattern expected to meet the dietary reference intake (DRI) of nutrients. Nutritional adequacy of RM in 'DRI for Koreans 2015' were analyzed to verify whether such expectation was fulfilled. Methods: Dishes in RM are categorized by 5 food groups, and number and types of dishes for main meal and between-meals were analyzed. The energy and 12 nutrients (protein, dietary fiber, Ca, P, Fe, Na, K, vitamin A, thiamin, riboflavin, niacin and vitamin C) contents in 10 RMs were calculated using the food composition table (CD) in 'DRI for Koreans'. Energy, energy contribution ratio, and nutrient contents in 10 RMs for 18 age groups were evaluated based on the 'DRI for Koreans 2015'. Results: Number of dishes per meal were 4.83, and representative table setting consisted of cooked rice + soup (or stew) +3 side dishes. Energy contents of RM were 75~109% of estimated energy requirement (EER). None of the RM met the DRI of all 12 nutrients examined. Calcium was the most insufficient nutrient. Only 1-2 years' RM met the DRI, all the other RMs did not meet the calcium DRI. Dietary fiber and potassium contents were also insufficient in most RM. In adult male's RM, only 1 nutrient, i.e. calcium did not meet the DRI, but in female adult's RM, 5~6 nutrients did not meet the DRI. Energy contribution ratio of carbohydrate, protein, and fat in RM were 59.0~70.4%, 15.7~17.5%, 12.1~23.5%, respectively. And 4 RMs out of 10 exceeded the upper limit of acceptable macro-nutrient distribution range (AMDR) of carbohydrate and 3 RMs out of 10 RM were below the lower limit of fat AMDR. Contribution ratio of nutrients were ${\geq}40%$ by food groups were as follows: grain group in energy and carbohydrate; meat fish egg legume group in fat, protein, and niacin; vegetables group in dietary fiber, vitamin A, and vitamin C; milk dairy products group in calcium. Conclusions: RM prepared according to the Target Pattern did not meet nutritional adequacy as expected. Especially calcium, potassium, and dietary fiber contents are needed to be increased in many RMs. Further, energy content in RM needs to be adjusted for fat sugar food group assigned in Target Pattern and condiment.

가뭄 위험도 평가를 위한 종합적 지수 개발에 관한 연구 (Development of comprehensive drought risk index for drought risk assessment)

  • 가뭄은 다른 자연재해들과 달리 복잡한 자연의 순환 시스템에 의해 발생하므로 시기가 명확하지 않으며, 사회 전반에 걸친 큰 규모의 피해가 발생한다. 이러한 피해를 저감하기 위해 가뭄에 대한 다양한 연구들이 진행되고 있다. 가뭄에 대한 기존 연구들을 살펴보면, 강수량과 같은 가뭄의 원인을 중점적으로 다룬 가뭄지수 연구가 대다수이다. 한편, 실제 가뭄 피해에 대한 연구들은 위험 혹은 취약성 분석으로 지수를 산정하는 방법으로 연구되고 있으며 농업 가뭄에 집중한 결과를 제시하고 있다. 본 연구에서는 가뭄 현상의 원인을 제공하는 기상학적 요인뿐 아니라 피해 대상과 대상의 내부시스템 능력에 따른 영향을 고려한 가뭄위험지수(Drought Risk Index, DRI)을 연구하였다. 또한, 생활용수, 농업용수, 공업용수를 고려하여 농업가뭄과 더불어 사회 경제학적 가뭄의 피해 평가가 가능한 통합 DRI를 산정하였다. 크론바흐 알파 계수를 통하여 지수 구성 요소들 간의 신뢰성과 DRI와 실제 가뭄 피해간의 스피어만 상관 계수를 사용하여 국내 유역에 대한 DRI의 타당성을 확인하였고 이를 통해 지수의 적합성을 분석하였다. 결과적으로 기후변화 시나리오를 사용하여 한반도의 2030, 2050, 2080년대 통합 DRI를 제시하였다.
