Hizikia fusiformis is well known edible brown seaweed both in Korea and Japan. It has been intensively studied due to its pronounced health benefits. In this study, the radical scavenging (antioxidative) activities of its hydrophilic phlorotannin constituents were studied. An aqueous extract/original extract (OE) of H. fusiformis was initially prepared with heat, enzymes and pH control treatments. Then the original extract was further fractionated (with methylene chloride and methanol) and crude hydrophilic phlorotannin extract (CHPE) was prepared. The radical scavenging activities of both OE and CHPE were determined by using electron spin resonance (ESR) spectrophotometric assays such as 1,1-diphenyl-2-picrylhydrazyl (DPPH), hydroxyl and alkyl radical assays. The CHPE reported significantly (p < 0.05) higher total phenolic (phlorotannins) content (1.23 mg ${\cdot}ml^{-1}$) than that of the OE (0.21 mg ${\cdot}ml^{-1}$). Both OE and CHPE have reported good radical scavenging activities and those activities were dose-dependent. The CHPE have demonstrated significantly higher radical scavenging activities than that of the OE. In comparison, the DPPH radical (6 $\times$ 10$^{-5}$ M) scavenging activities of all the CHPE concentrations (0.25, 0.5 and 1 mg ${\cdot}ml^{-1}$) tested were significantly higher (37.3, 78.2 and 91.6%, respectively) than that of the OE counterparts (11.4, 34.6 and 61.7%, respectively). Alkyl radical scavenging percentages of CHPE at 0.0625, 0.125, 0.25 and 0.5 mg${\cdot}ml^{-1}$ were significantly higher (34.3, 69.2, 80.4 and 88.7%, respectively) than that of the OE (16.6, 41.4, 62.3 and 77.4%, respectively). The percentages of hydroxyl radical scavenging activities of CHPE at the concentrations of 0.25, 0.5 and 1 mg ${\cdot}ml^{-1}$ were 32.5, 59.4 and 84.2 % respectively. Moreover, the hydroxyl radical scavenging activity of OE was quite lower than that of the CHPE. Therefore, these results suggest that the hydrophilic phlorotannins of H. fusiformis are potential radical scavengers thus, a great source of antioxidative nutraceuticles.
This study was carried out in order to determine the biological activities such as antioxidant activities in nine cultivars of Momordica charantia L. (Korea, China, Japan and Philippine native cultivars). Their antioxidant activities were measured using DPPH free radical scavenging, ABTS cation radical scavenging, and Nitrite scavenging ability. The highest total polyphenol content (16.82 ${\mu}g/m{\ell}$) measured in the native Korea cultivar, and this value was 4.0 ${\mu}g/m{\ell}$ higher than that of the 'Verde Beuhas' cultivar (12.82 ${\mu}g/m{\ell}$). The 'Peacock' cultivar had the highest total flavonoid amount which was 4.38 ${\mu}g/m{\ell}$. The free radical scavenging activity using DPPH method was the highest in the native China cultivar ($RC_{50}$ = 102.6 ${\mu}g/m{\ell}$). ABTS cation radical scavenging activity according to cultivars was significantly higher in 'Peacock'. Nitrite scavenging ability showed the most remarkable effect at the pH 1.2, exhibited to 81.5${\sim}$86.9% by addition of ethanol extract 1${\mu}g/m{\ell}$ from Momordica charantia L. These results suggest that Momordica charantia L. had the potent biological activities, and that their activities exhibited differently depending on cultivars.
Levels of quercetin in different parts of onion were investigated using high performance liquid chromatography (HPLC) and liquid chromatography mass spectrometry (LC/MS) suitable for use with functional food material. Two main peaks were observed on HPLC chromatograms from the extracts of the skin, and the outer, middle, and core parts of onion. Using LC/MS, peak 1 was tentatively identified as quercetin monoglucoside at m/z 466.4, and peak 2 as quercetin with [M]+ at m/z 303.3. The levels of quercetin in the skin, and the outer, middle and core parts of the plant were 16.83,2.67,0.95, and 0.35 mg/g, respectively. In the study of 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, skin, the nonedible part, contained the highest amount of quercetin, compared to the other edible parts, and showed the highest DPPH radical scavenging activity. Levels of quercetin and DPPH radical scavenging activity increased from core to skin. All parts of onion exhibited the strongest antibacterial activity only against Staphylococcus aureus and Vibro parahaemolyticus. Antibacterial activities of onion exhibited that S. aureus was more sensitive than V. parahaemolyticus. Among the four onion extracts, the middle part showed the strongest inhibitory activity against S. aureus but all onion extracts showed similar antibacterial activities against V. parahaemolyticus.

In this study, the comparison of antioxidative effect of the methanol (MeOH) extracts from grains were investigated in vitro radical scavenging system. Ten grains (black rice, rice, barley, wheat, millet, sorghum, glutinous millet, buckwheat, phellines linteus rice and brown rice) were extracted with MeOH. Among the MeOH extracts of grains, sorghum and black rice showed effective scavenging activities of 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical. The $IC_{50}$ values of sorghum and black rice were $47.4{\mu}g/mL$ and $50.6{\mu}g/mL$, respectively. In addition, black rice also exerted the strongest activities on hydroxyl radical (${\cdot}OH$) scavenging activity. Furthermore, the MeOH extracts of black rice showed effective and dose dependant scavenging activities of DPPH radical and ${\cdot}OH$. These results indicated that black rice showed strong free radical scavenging activity. It suggested that black rice could be a promising natural antioxidant against free radical-induced oxidative damage.
The antioxidative and free radical scavenging activity of water extracts of dandelion were investigated. Antioxidative and radical scavenging activity were assessed by means of different tests; inhibition of peroxidation on linoleic acid model system, scavenging DPPH radical, scavenging of hydroxyl radical by chemiluminescence assay, scavenging of superoxide anion radical by EPR spectroscopy and scavenging of hydrogen peroxide. The leaf extract showed strong antioxidant activity in linoleic acid system. The antioxidant activity of water extracts of dandelion increased with increasing concentrations of extracts. The scavenging activity of the dandelion extracts, on inhibition of the DPPH radical, was related to the reaction time. Hydroxyl radical were generated by lenten reaction and dandelion extract was found to scavenge OH˙in a concentration-dependent manner. The water extract of leaf had effective scavenging activities on hydrogen peroxide and superoxide anion radical. From the these data, it is evident that water extract of dandelion leaf is an effective scavenger for OH˙, O$_2$¨, DPPH˙, hydrogen peroxide. And, the antioxidative effect observed is believed to be partly due to this radical scavenger activity.
Antioxidative activities of major pentacyclic terpenoids from the semi-fermented tea of Camellia sinensis L. were investigated. The free radical scavenging activities of triterpenoids $1{\sim}3$ were examined with of DPPH and superoxide anion radical scavenging activity. The $IC_{50}$ of compounds 1 and 2 for DPPH radical scavenging activities were 23.1 and $37.2{\mu}g/mL$ respectively, and for superoxide anion radical scavenging activities were 37.2 and $35.2{\mu}g/mL$, respectively. According to this result, compounds 1 or 2 in semi-fermented tea could be the candidates for bioactive material having antioxidant activity.

Objective : This study was designed to investigate effect of immature fruits from Citrus unshiu Markovich to mature them on antioxidative and antiaging-related enzyme activities in vitro. Methods : Fruits from Open field-cultivated C. unshiu Markovich (ripe and unripe fruits of Citri) made a purchase in Jeju island. We measured total polyphenol and flavonoid contents in mature C. unshiu Markovich (MC) and immature C. unshiu Markovich (IMC) 70% ethanol extract. DPPH free radical, ABTS radical, and hydroxyl radical scavenging activities in both extracts were determined. Also, reducing power, SOD-like activities, elastase inhibition, and collagenase inhibition activities of these extract were assayed at 5, 1, 0.5, and 0.1 mg/㎖. Results : The total polyphenol contents in the ethanol extract of IMC were higher than those of MC (26.99 ± 0.89 mg/TAEg). And total flavonoid contents in them were very similar to both extract (MC; 7.25 ± 0.4 mg/RUEg and IMC; 7.44 ± 0.18 mg/RUEg). DPPH free radical scavenging activities, ABTS radical scavenging capacity, reducing power, and SOD-like activities of IMC-treated group showed significant increase compared to those of MC group in all treated concentrations. Particularly, DPPH free radical scavenging and elastase inhibition activities in IMC-treated group at 5 mg/㎖ showed similar with positive control group. Besides, IMC extract at 5 mg/㎖ concentration surpassed positive control (BHA or Vit. C) in SOD-like activities and ABTS radical scavenging capacity. Conclusion : These results suggest that IMC ethanol extract may be used as a natural antioxidant and a antiaging material for development of functional foods and cosmeceuticals.

The study was aimed at evaluating the antioxidant and antimicrobial activities of methanol extract of Caesalpinia bonducella leaves (MECB) (Family: Caesalpiniaceae). The effect of MECB on antioxidant activity, reducing power, free radical scavenging (DPPH radical, nitric oxide radical, superoxide anion radical, hydroxyl radical and hydrogen peroxide radical scavenging), total phenolic content and antimicrobial activities were studied. The antioxidant activity of MECB increased in a dose dependent manner. About 50, 100, 250 and 500 g of MECB showed 53.4, 61.2, 69.1 and 76.2 % inhibition respectively on peroxidation of linoleic acid emulsion. Like antioxidant activity, the effect of MECB on reducing power increased in a dose dependent manner. The free radical scavenging activity of MECB was determined by DPPH radical scavenging method. The potency of this activity was increased with increased amount of extract. MECB was found to inhibit the nitric oxide radicals generated from sodium nitroprusside $(IC_{50}\;=\;102.8\;g/ml)$ whereas the $IC_{50}$ value of curcumin was 20.4 g/ml. Moreover, the MECB was found to scavenge the superoxide generated by photoreduction of Riboflavin. MECB was also found to inhibit the hydroxyl radical generated by Fenton reaction, where the $IC_{50}$ value is 104.17 g/ml compared with catechin 5 g/ml, which indicates the antioxidant activity of MECB. The MECB capable of scavenging hydrogen peroxide in a concentration-dependent manner. The amounts of total phenolic compounds were also determined. Antimicrobial activities of MECB were carried out using disc diffusion methods with five Gram positive, four Gram negative and four fungal species. The results obtained in the present study indicate that MECB leaves are potential source of natural antioxidant and antimicrobial agents.
The aim of this study is to examine the radical scavenging activity of perilla and sesame oil that Koreans traditionally consume. For DPPH radical scavenging activity, oil and its hexane/70% methanol extracts (ME) are used and for superoxide and hydroxyl radical scavenging activities, ME are used. Unrefined perilla oil, sesame oil, and refined sunflower oil are used. The yields for ME of perilla, sesame and sunflower oil are 0.57, 0.61, and 0.30%, respectively, and the amounts of phenolic compounds in ME of corresponding oil are 18.77, 88.64 and $0.05\;{\mu}g$ tannic acid/mg, respectively. $IC_{50}$ for DPPH scavenging activity of perilla, sesame and sunflower oil are 2.12, 1.91, and 3.35 mg/mL, respectively and those for ME of corresponding oils are 0.42, 0.07, and 43.11 mg/mL, respectively. In DPPH assay, the solvent used for oil sample is iso-octane and that for ME is methanol. Superoxide anion scavenging activity of ME of perilla, sesame and sunflower oil tested at 1 mg/mL concentration are 21.10, 13.25, and 3.14%, respectively. Hydroxyl radical scavenging activities of those samples tested at 1 mg/mL concentration are 86.08, 93.30, and 93.17%, respectively. In summary, the refining process seems to remove the phenolic compound during oil processing. Antiradical substances in perilla and sesame oils responsible for scavenging DPPH radicals are present in the methanol fraction, while the antiradical substances in the sunflower oil are in the lipid fraction. DPPH scavenging activity of ME of sesame oil is significantly higher than that of perilla oil (p<0.05). However, superoxide anion scavenging capacity of ME of perilla oils was found to be greater than that of both sesame and sunflower oils (p<0.05).
As a part of our search for novel antioxidants from the seaweeds, we have investigated radical scavenging effect for their crude extracts using 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, authentic peroxynitrite, and 3-morpholinsydnonimine (SIN-1), a peroxynitrite-generating species in vitro. Thirty-four seaweeds were screened for $ONOO^-$ and DPPH radical scavenging activities. A potent inhibitory effect against peroxynitrite generated by SIN-1 at $5{\mu}g/ml$ of methanol extracts was observed in order of Ishige okamurae(95.3%), Sargassum hemiphyllum(90.2%), Symphyocladia latiuscula(89.6%), Porphyra suborbiculata(86.7%), and Gelidium amamsii(85.9%), Also, a significant scavenging effect against direct authentic peroxynitrite was revekaled for methanol extracts of Ishige okamurae(66.2%) and Sargassum hemiphyllum(55.2%) and the acetone/methylene chloride(1:1) extract of Gigatina tenella (61.0%). In our measurement for evaluating the capacity to scavenge the stable free radical of DPPH, acetone/methylene chloride(1:1) extracts of Symphyocladia latiuscula, Gloiopeltis furcata, and Sargassum thunbergii and the methanol extract of Sargassum sp. showed an inhibitory potency of 85.8%, 82.8%, 74.1%, and 64.0%, respectively.
