Purpose: Fruit and vegetable juices are known to be rich sources of antioxidants, which have beneficial effects on diseases caused by oxidative stress. The purpose of this study was to directly compare the antioxidant activities of fruit and vegetable juices marketed in Korea. Methods: We analyzed four fruit juices, two vegetable juices, two yellow-green juices, and six mixed vegetable juices. Antioxidant activities were analyzed using 2,2-diphenyl-1-picrylhydrazyl (DPPH) test, 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonate) (ABTS) test, and oxygen radical absorbance capacity (ORAC) assay. Protective effects against DNA damage were determined using an ex vivo comet assay with human lymphocytes. Results: DPPH radical scavenging activities were in the following order: blueberry juice > mixed vegetable C juice > kale juice > mixed vegetable P juice > grape juice. ABTS radical scavenging activities were in the following order: blueberry juice > mixed vegetable C juice > grape juice > mixed vegetable P juice > kale juice. Peroxyl radical scavenging activities as assessed by ORAC assay were in the following order: blueberry juice > kale juice > mixed vegetable C juice > grape juice. Grape or blueberry juice showed strong abilities to prevent DNA damage in lymphocytes, and the difference between them was not significant according to the GSTM1/GSTT1 genotype. Conclusion: Antioxidant activities of fruit and vegetable juices and ex vivo DNA protective activity increased in the order of blueberry juice, grape juice, and kale juice, although the rankings were slightly different. Therefore, these juices rich in polyphenols and flavonoids deserve more attention for their high antioxidant capacity.
This study evaluated the antioxidant activity of the extract and fractions of Alnus firma. Alnus firma had the highest total phenolic content ($452.80{\pm}7.01{\mu}g$ gallic acid equivalents/mg) in a methanol (MeOH) fraction and the highest total flavonoid content ($112.29{\pm}11.14{\mu}g$ rutin equivalents/mg) and antioxidant capacity ($936.23{\pm}0.07{\mu}g$${\alpha}$-tocopherol equivalents/mg) in an ethylacetate (EA) fraction. The antioxidant activities of various solvent extract fractions of Alnus firma were evaluated using various antioxidant assays, including ${\beta}$-carotene-linoleate assay, reducing power assay, 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assay, metal chelating activity assay, superoxide anion radical scavenging assay, and lipid peroxidation inhibition assay using the ferric thiocyanate method. These activities were compared with those of ascorbic acid, butylated hydroxyanisole (BHA), gallic acid (GA), butylated hydroxytoluene (BHT), and ${\alpha}$-tocopherol. First, at a $250{\mu}g/ml$ concentration, the EA and MeOH fractions of A. firma showed 92.43% and 89.20% DPPH radical scavenging activity, respectively. Second, $50{\mu}g/ml$ of the EA fraction exhibited 72.49% superoxide anion radical scavenging activity, a little greater than the same dose of GA (60.88%). Finally, 0.5 and 1 mg/ml of the EA fraction showed 73.45% and 73.29% inhibition of peroxidation in the ${\beta}$-carotene-linoleic acid system, respectively. The decreasing order of reducing power was EA fraction > n-butanol (BuOH) fraction > dichloromethane (DCM) fraction > n-hexane (HX) fraction. The results obtained in the present study indicated that Alnus firma can be used as an easily accessible potential source of natural antioxidants.
Vincamine, one of the major indole alkaloids in vincaminor (Vinca minor L.) is commonly used for treating cerebrovascular diseases. The antioxidant activity of vincaminor extracts and vincamine were measured by 1.1-diphenyl-2-picrylhydrazyl (DPPH) and lipid malonaldehyde (MA) assay. Vincaminor leaves were pulverized and extracted with various solvents such as water, methanol, and ethanol. The antioxidant activities of the extracts varied in accordance with solvents and assays. In DPPH assay, the water extract showed the highest antioxidant activity. In lipid MA assay, However, the ethanol extract inhibited MA formation from cod liver oil by 82% at the level of 5,000 ${\mu}g/mL$. Vincamine in the extract was analyzed by high-performance liquid chromatogram and the concentration of vincamine was 0.419$\pm$0.005 ${\mu}g/mL$.

In this study, the antioxidant and anti-inflammatory properties of Colpomenia sinuosa extracts were identified. Antioxidant experiments included DPPH, ABTS, nitrite scavenging experiments, and FRAP, polyphenol concentration measurements, flavonoid concentration measurements. DPPH assay results showed an antioxidant function of 2.821 mg ascorbic acid/g extract. ABTS assay results showed an antioxidant function of 3.923 mg ascorbic acid/g extract. nitrite assay results showed an antioxidant function of 17.89 mg ascorbic acid/g extract. In FRAP, 1 mg of the Colpomenia sinuosa extract showed a reduction of 2.062±0.177 ㎍ of ascorbic acid. For polyphenols, 62.85±3.18 mg/g was shown. Flavonoids showed 10.01±0.24 mg/g. In the meantime, cell experiments showed cytotoxicity and anti-inflammatory functions. In cytotoxicity experiments, Colpomenia sinuosa extracts showed cytotoxicity of less than 20% and an inflammatory inhibition of 30.93±2.93% at a concentration of 100 ㎍/mL. These results indicate that Colpomenia sinuosa extract is available as functional cosmetic material.

This study attempted to investigate the antioxidant capacity and whitening effects of the extract from the mixture of 7 different vegan cosmetic ingredients. To examine radical scavenging capacity, DPPH, ABTS and FRAP assays were performed. In addition, polyphenol and flavonoid concentrations were measured to check antioxidant substances. Specifically, in a DPPH test, 105.74 mg ascorbic acid/g antioxidant capacity was observed. In an ABTS assay, 85.31 mg ascorbic acid/g antioxidant capacity was found. Lastly, in an FRAP assay, 1mg of the extract revealed ascorbic acid 198.01±5.50 ㎍ reducing power. Polyphenol and flavonoid concentrations were 30.19±0.75 mg/g and 9.12±0.36 mg/g respectively. In a cell-based assay, cytotoxicity and whitening activity were examined. In terms of cytotoxicity, '20% or less' was observed. Furthermore, the inhibition of melanin synthesis was '34.70±2.97%' at 100 ㎍/mL, confirming the possibility of the extract from the mixture of 7 different substances as a cosmetic ingredient.
Ganoderma lucidum has been traditionally used as a medicine for treatment of bronchitis, arthritis, and high blood pressure, and it has been reported to display many biological activities including anticancer and immune activities. Since mushroom mycelium is known to have excellent biological activities together with mushroom fruiting body, studies on biological activities of mushroom mycelium have been actively conducted. Thus, the present study compared the biological activities before and after the cultivation of Ganoderma lucidum mycelium on Atractylodes rhizoma. When the radical scavenging activity was assessed by the DPPH assay, ARGL (ethanol extract of Atractylodes rhizoma mycelium fermented with Ganoderma lucidum) showed radical scavenging activity of 5.58~82.56% at concentrations of $10{\sim}500{\mu}g/assay$, while AR (ethanol extract of Atractylodes rhizoma) showed radical scavenging activity of 5.27~72.08% at the same concentrations. When measured by using the ABTS assay, ARGL showed higher radical scavenging activity than AR, which was consistent with the result obtained by the DPPH assay. In the MTT assay, the cytotoxicity of ARGL against all cell lines was higher than that of AR. In particular, the cytotoxicities of AR and ARGL against Hep3B at a concentration of $400{\mu}g/assay$ were 71.81% and 86.40%, respectively. In addition, the result obtained by the SRB assay was consistent with the result obtained by the MTT assay. According to the results mentioned above, there is a high probability that medicinal herb cultures using mycelium can be used as sources of functional foods since the cytotoxicities against cancer cells and antioxidant activities increased when the mycelium was fermented with Atractylodes rhizoma.
Objectives : Oxidative stress has a critical role in neurodegenerative diseases. In this study, we investigated the antioxidant and neuroprotective effects of the ethanolic extract of Banryong-hwan (BRHE) in SH-SY5Y cells and primary rat mesencephalic dopaminergic neurons. Methods : To assess the antioxidant effects, we carried out 1,1-diphenyl-2-picrylhydrazyl(DPPH) free radical scavenging assay, 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulphonic acid)(ABTS) radical cation decolorization assay, and determination of total polyphenolic content. We evaluated the effect of BRHE treatment on neuroprotection against 6-hydroxydopamine(6-OHDA) toxicity using thiazolyl blue tetrazolium bromide(MTT) assay, nitric oxide(NO) assay, reactive oxygen species(ROS) assay in SH-SY5Y cells and tyrosine hydroxylase(TH) immunocytochemistry in primary rat mesencephalic dopaminergic neurons. Results : BRHE showed IC50 values of 328.10 ${\mu}g/mL$ and 43.12 ${\mu}g/mL$ in DPPH assay and in ABTS assay, respectively. Total polyphenolic content was 180.76 ${\mu}g/mL$. In SH-SY5Y cells, BRHE significantly attenuated the toxicity induced by 6-OHDA at the concentrations of 25-100 ${\mu}g/mL$ pre- and post- treatment in MTT assay. While 6-OHDA increased the NO and ROS contents, BRHE decreased them in a dose dependent manner. Moreover, in primary dopaminergic neuron culture, BRHE significantly protect-ed the dopaminergic cell loss against 6-OHDA toxicity up to 136% at the concentration of 75 ${\mu}g/mL$. Conclusions : These results demonstrate that BRHE has neuroprotective effect against 6-OHDA induced neurotoxicity through decreasing NO and ROS generation.

The purpose of this study was to research for anti-oxidation and anti-wrinkle effects of Acar mono Sap (AM). To cosmetic effect of AM, safety effect (MTT assay), anti-wrinkle effect (elastase, MMP-1 inhibition assay) and anti-oxidant effect (DPPH assay) were measured. When water extract of AM was used for cell viability, it was over 100% at 6% (6 ml/100 ml in phosphate buffer) concentration. AM showed 45.7% elastase inhibition and 23.7% MMP-1 inhibition at 50% (50 ml/100 ml in phosphate buffer) concentration so that it had good anti-wrinkle characteristic. And AM showed 68.9% antioxidation capacity at 50% concentration by using a DPPH assay. Consequently, AM can be used as natural materials or additives for human skin owing to their beneficial biologic functions, including the anti-wrinkle effect, for cosmetic compositions.

The aim of this study was to evaluate the anti-Helicobacter pylori activity of fractions and major aglycon compounds (baicalein, chrysin, oroxylin A, wogonin) of Scutellariae Radix. Minimum inhibitory concentration (MIC) measurement, DPPH radical-scavenging assay, DNA protection assay, and urease inhibition analysis were performed. The ethyl acetate (EtOAc) fraction showed the potent anti-Helicobacter activity, and therefore, compounds in the EtOAc fraction were subjected to further assay. The MICs of chrysin, oroxylin A, and wogonin against Helicobacter pylori 26695 were 6.25, 12.5 and $25{\mu}g/mL$, respectively. Baicalein exhibited the most effective DPPH radical-scavenging activity. DNA protection using Fenton reaction, chrysin, oroxylin A, and wogonin showed effective DNA protective effect. This result was also confirmed by quantitative real-time polymerase chain reaction (qRT-PCR). Regarding Jack bean urease (0.5 mg/mL, 50 unit/mg) inhibition, 20 mM ofbaicalein and chrysin inhibited urease activity by 88.2% and 72.5%, respectively.
Antioxidative and free radical scavenging properties of different stem extracts of Euphorbia trigona were evaluated and correlated with its total phenolic content. Aqueous, acetone and methanolic extracts of shade dried stem were obtained and were concentrated in vacuo. The antioxidant and free radical scavenging activities of stem extracts was determined by 2,2-Diphenyl-1-picrylhydrazyl (DPPH) scavenging assay, reducing power assay, deoxyribose degradation assay and $Fe^{2+}$ chelating assay. Total phenolic contents (TPC) were evaluated using Folin-Ciocalteu reagent. The results confirmed that the plant is a rich source of polyphenolic compounds which are invariably higher compared to other herbs. All extracts showed TPC in the range of 146.6 - 168.6 mg/g gallic acid equivalents at $300{\mu}g/ml$ of extract. Among the three extracts ME showed highest scavenging activity as evidenced by maximum scavenging of DPPH (83.2%), $OH{\bullet}$ radicals (94.81%), $Fe^{2+}$ chelating activity (88.59%) and a high reducing power 0.623 at $300{\mu}g/ml$. Our results demonstrate that Euphorbia trigona, an unexplored xerophytic plant could be potential source of natural antioxidants and phytotherapeutic agents. The plant possess invariably high amount of polyphenolic compounds with a broad spectrum of antioxidant properties and could be further used for food, feed and pharmaceutical applications.
