유산균 발효에 의한 인삼열매 추출물의 항산화 및 항노화 효과 (Antioxidant and Antiaging Effect of Ginseng Berry Extract Fermented by Lactic Acid Bacteria)

  • 본 연구에서는 유산균 발효에 의한 인삼열매 추출물의 여러 가지 생리활성을 평가하였다. 고성능액체크로마토그래피를 수행하여 인삼열매 추출물과 유산균 발효 인삼열매 추출물의 ginsenoside Re, Rc 및 Rb1의 함량분석 결과 유산균 발효에 의해 ginsenoside Re, Rc 및 Rb1의 함량이 모두 증가하였다. 인삼열매 추출물과 유산균 발효 인삼열매 추출물의 DPPH free radical 소거활성과 SOD 유사활성 측정결과 1.00 %의 농도에서 유산균 발효 인삼열매 추출물은 각각 86.34, 76.82 %의 DPPH free radical 소거활성과 SOD 유사활성을 나타내었고, 인삼열매 추출물은 각각 49.78, 40.80 %의 DPPH free radical 소거활성과 SOD 유사활성을 나타내어 유산균 발효에 의해 항산화 활성이 증가하는 것을 확인하였다. 또한 유산균 발효 인삼열매 추출물 0.50 %의 농도로 처리한 피부 섬유아세포에서 procollagen type I (COL1A1)은 828.13 % 증가하였으며, matrix metalloproteinase (MMP)-1은 87.88 %, tumor necrosis factor (TNF)-${\alpha}$는 99.92 %감소하는 효과를 보여주었다. 이처럼 항산화, 주름, 항염에 효능을 보이는 유산균 발효에 의한 인삼열매 추출물은 기능성 화장품 소재로 개발될 수 있는 가능성이 있다.

The Chemical and 1,1-Diphenyl-2-Picrylhydrazyl Radical Scavenging Activity Changes of Ginsenosides Rb1 and Rg1 by Maillard Reaction

  • The chemical and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity changes of ginsenoside $Rb_1$-glycine and ginsenoside $Rg_1$-glycine mixtures by Maillard reaction were investigated to identify the role of Maillard reaction in the increased antioxidant activity of ginseng by heat-processing. The DPPH radical scavenging activity of $Rg_1$-glycine mixture was more strongly increased by heat-processing than that of $Rb_1$-glycine mixture. From the analyses of ginsenosides, $Rb_1$ was gradually changed into 20(S)-$Rg_3$, 20(R)-$Rg_3$, $Rk_1$ and $Rg_5$ by heat-processing. $Rg_1$ was gradually changed into 20(S)-$Rh_1$, 20(R)-$Rh_1$, $Rk_3$ and $Rh_4$ by heat-processing. However, the generation of these less-polar ginsenosides was not related to the increased DPPH radical scavenging activity of $Rb_1$-glycine and $Rg_1$-glycine mixtures because their DPPH radical scavenging activities were already significantly increased when dried at $50^{\circ}C$, which temperature induce no structural changes of ginsenosides. In the comparison of browning compound levels of $Rg_1$-glycine and $Rb_1$-glycine mixtures, the extents of Maillard reaction were positively correlated with their increased free radical scavenging activities. Based on the chemical and DPPH radical scavenging activity changes of $Rg_1$-glycine and $Rb_1$-glycine mixtures by heat-processing, we clearly identified that the increased free radical scavenging activity of ginsenoside is mediated by the Maillard reaction between sugar moiety of ginsenoside and amino acid.

골쇄보 열수추출물 약침액의 항염증반응 및 DPPH 라디칼 소거능 활성에 대한 연구 (The Anti-inflammatory Effect and DPPH Free Radical Scavenging Capability of Rhizoma drynariae Aqueous Extract)

  • 목적 : 한의학에서 골절이나 신장질환에 사용되어 왔던 골쇄보열수추출물 약침액이 항염증 및 항산화활성에 대한 효능이 있는지를 연구하고자 한다. 방법 : 골쇄보열수추출물 약침액이 세포독성에 미치는 영향을 관찰하기 위하여 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT) assay를 실시하였다. 골쇄보열수추출물 약침액이 항염증효능에 미치는 영향을 관찰하기 위하여 lipopolysaccharide(LPS)로 유도된 RAW264.7 대식세포에서의 산화질소(NO) 및 프로스타글란딘($PGE_2$) 생성 억제력을 관찰하였다. 또한 골쇄보열수추출물 약침액이 항산화활성에 미치는 영향을 관찰하기 위하여 1,1-diphenyl-2-pciry hydrazyl(DPPH) radical 소거능을 관찰하였다. 결과 : 골쇄보열수추출물 약침액은 $50{\sim}400{\mu}g/ml$ 농도에서 세포독성이 나타나지 않았다. 골쇄보열수추출물 약침액은 200 및 $400{\mu}g/ml$ 농도에서 LPS로 유도된 NO 및 $PGE_2$ 생성을 통계학적으로 유의하게 감소시켰다. 골쇄보추출물은 $50{\sim}400{\mu}g/ml$ 농도에서 DPPH 소거능을 관찰한 결과 농도의존적으로 활성화되는 것을 관찰하였다. 결론 : 골쇄보열수추출물 약침액은 항염증 및 항산화 효과가 있을 것으로 사료된다.

Quality Evaluation of Cheongpomuk Added with Chlorella Powder

  • This study examined the quality characteristics to evaluate the optimal mixing ratio of a chlorella powder in Cheongpomuk. The total polyphenol and total flavonoids contents of Cheongpomuk and chlorella powder was 279.90 ㎍/mL, 489.50 ㎍/mL and 48.79 ㎍/mL, 687.85 ㎍/mL respectively. The DPPH free radical scavenging activities and ABTS radical scavenging activities of chlorella powder was 65.39%, 91.28% respectively. The L values (58.85~26.23), a values (-0.44~-5.84) of the Hunter's color values decreased and b value (-10.98 ~4.12) increased significantly in proportion to the amount of chlorella powder. The total polyphenol contents (55.00~734.20 ㎍/mL), total flavonoids contents (41.00~88.29 ㎍/mL), DPPH free radical scavenging activities (35.10~65.50%) and ABTS radical scavenging activities (50.29~92.86%) of Cheongpomuk increased significantly in groups of chlorella powder. In the physical properties, the hardness (135.93~103.93 g/㎠), chewiness (6.76~3.76 g), springiness (4.92~4.36 %), and gumminess (147.70~94.23) of Cheongpomuk decreased significantly in proportion to the amount of chlorella powder. The Cheongpomuk containing 15% chlorella powder showed a high score with regard to color, flavor, moistness, chewiness and overall acceptance. The appropriate addition amount of chlorella powder for making Cheongpomuk is 15%. This study indicate that Cheongpomuk treated with chlorella powder had the highest functional component and antioxidant activity.

매실나무(Prunus mume) 겨우살이 추출물의 항산화활성 (Antioxidant Activity of Extracts from Mistletoe Cultivated on Prunus mume)

  • 최근 매실나무를 기주로 한 겨우살이의 인공재배에 성공함에 따라, 식 의약품과 화장품의 소재 및 산업화 가능성을 확인하기 위해 80% 에탄올 및 증류수 초음파추출물의 총 폴리페놀 플라보노이드 함량 및 DPPH free radical 소거능을 측정하였다. 경기도 남양주시에서 인공재배한 매실나무겨우살이(동결건조) 및 강원도 홍천에서 채취한 자연산 굴참나무겨우살이(열풍건조, $30^{\circ}C$, 7day)의 80% 에탄올, 증류수 초음파추출물에 대한 총 폴리페놀 및 플라보노이드함량 그리고 DPPH free radical 소거능을 측정한 결과, 시료의 농도에 비례하여 총 폴리페놀과 플라보노이드 함량 그리고 DPPH free radical 소거능이 증가하였다. 총 폴리페놀 함량은 매실나무겨우살이의 80% 에탄올 추출물이 5,000ppm에서 $192.63{\mu}g/ml$로 가장 많았으며, 굴참나무겨우살이의 물 추출물이 100ppm에서 $4.86{\mu}g/ml$로 가장 낮고 매실나무겨우살이가 모든 용매에서 더 많은 폴리페놀을 함유한 것으로 확인되었다. 총 플라보노이드 함량은 굴참나무겨우살이의 80% 에탄올 추출물이 5,000ppm에서 $83.59{\mu}g/ml$로 가장 높았으며, 매실나무겨우살이 물 추출물이 $4.6{\mu}g/ml$로 가장 낮았고 굴참나무겨우살이가 모든 용매에서 더 많은 플라보노이드를 함유한 것으로 확인되었다. DPPH free radical 소거능은 80% 에탄올 추출물 5,000ppm에서 매실나무겨우살이 81.75%로 시료 중 가장 높았고, 굴참나무겨우살이 물 추출물 100ppm이 3.55%로 가장 낮음을 확인할 수 있었다. 본 연구 결과에 따르면, 겨우살이를 초음파추출법으로 추출할 때, 80% 에탄올 초음파추출물이 더 많은 폴리페놀을 함유하여 80% 에탄올 추출물이 물 추출물보다 비교적 적합하였다. 80% 에탄올로 초음파추출 하였을 때 동결건조 한 매실나무겨우살이가 열풍건조한 자연산 굴참나무겨우살이보다 더 많은 폴리페놀을 함유하고 있고, 더 높은 DPPH free radical 소거능을 보여줌에 따라 식 의약품 그리고 화장품의 소재화 및 산업화의 가능성이 높음을 확인할 수 있었다.

In-Vitro Anticancer and Free Radical Scavenging Potential of Compound Formulation Used in Unani System of Medicine

  • Cancer is one of the leading cause of mortality in India as well as worldwide. The management of cancer by conventional therapy has shown life threatening adverse effects. The researchers are now exploring the natural way of treatment. Unani system of medicine have rich literature for cancer and many compound formulations have been described in this system. Unani system of medicine is based on holistic approach and treat human being as a unit with natural herbs, mineral and animal origin drugs. An important compound Unani formulation (CUF) from the literature has been chosen to explore the Unani claim of its anticancer activity. The phytochemical constituents were assessed using standard phytochemical screening method. Antioxidant property of this formulation was assessed by DPPH assay. The DPPH free radical scavenging assay was carried out by colorimetric method and ascorbic acid was taken as a positive control. Three different extracts of CUF on different concentrations were used to screening on human breast cancer (BCC) MCF-7 cell line. For the estimation of in-vitro cytotoxic potency of the investigated extracts was assessed on MTT assay by using trypan blue method and paclitaxel was used as the standard. Hydro-ethanolic (HE) extract showed highest free radical scavenging activity among all extracts. DPPH Assay showed substantial antioxidant activity of these extracts in hydro-ethanol extract at 1㎍ concentration of CUF. The CUF showed antioxidant and anticancer activity. The claim made by Unani physician has been proved.

녹용과 한약재료로 조제된 녹중탕의 유리기 소거 활성 (Free Radical Scavenging Activity of NokJoongTang Prepared from Antler and Various Oriental Medicinal Materials)

  • Two different NokJoongTangs were prepared by boiling antler and various oriental medicinal materials. The phenolic contents and free radical scavenging effects against DPPH(1,1-diphenyl-2-picrylhydrazyl) radical and hydroxyl radical of the NokJoongTangs which are used as the health supplement foods, were investigated. NokJoongTang-II prepared dried antler with various oriental medicinal materials, had higher phenolic content than that of NokJoongTang-I prepared 1mm raw antler with the same materials. In addition, NokJoongTang-II exhibited higher scavenging activities than these of NokJoongTang-I against DPPH radical and hydroxyl radical. These result indicate that free radical scavenging activity of NokJoongTang showed a difference by using raw and dry antler for NokJoongTang preparation, and that the findings also show a positive correlation between radical scavenging activity and total phenolic contents.

식물유래 천연색소의 항산화 활성 (Antioxidant Activity According to Each Kind of Natural Plant Pigments)

  • 선발된 13종의 천연색소 추출물에 대하여 free radical 소거활성 및 아질산염 소거활성, 그리고 항산화효소 활성에 대하여 조사하였다. 천연색소 종류별 free radical 소거 활성을 보면 적양배추(red cabbage), 양파껍질(onion peel), 레드비트(redbeet), 흑미(black rice), 오디(mulberry) 색소 등에서 상대적으로 높게 나타났고, 청치자(blue gardenia) 및 황치자(yellow gardenia), 완숙여주(mature bitter melon) 색소 등에서는 비교적 낮은 활성을 보였다 또한 아질산염에 대한 소거능을 조사한 결과, 반응용액 pH 1.2의 경우 양파껍질, 레드비트, 치자황, 지황(chinese foxglove), 치자청 색소에서 높은 소거 활성을 나타냈으며, 적양배추와 오디 색소에서는 상대적으로 낮은 소거활성을 보였다. 그리고 반응용액 pH 6.0에서는 대부분의 색소 추출물에서 아질산염 소거활성을 나타내지 않았다. 그리고 항산화효소활성 결과를 보면, SOD 활성의 경우 여주, 흑미, 포도과피 등에서 높은 활성을 나타냈고, 자색고구마, 지황에서도 비교적 높은 활성을 보였다. 또한 APX 활성은 뽕잎, 오디, 지황의 순으로 높은 활성을 보였으며, CAT 활성도 APX 결과와 비슷한 양상으로 나타났다. 이는 천연색소에 있어 항산화 효소의 종류에 따라 그 활성에 차이가 있음을 알 수 있다. 본 연구결과를 통해 다양한 생리활성을 지니고 있는 천연식물색소에 대해 높은 항산화 기능성을 확인함으로서 향후 고부가가치의 향장소재 및 식품소재로서의 개발가능성이 매우 높음을 시사해 주고 있다.

In vitro Free Radical Scavenging and Hepatoprotective Compound from Sanguisorbae Radix

  • In the course of searching for hepatoprotective agents from natural products, four compounds were isolated from the MeOH extract of Sanguisorbae Radix, as guided by their DPPH free radical scavenging activity. The structures were determined as 4,5-dimethoxy-3-hydroxybenzoic acid methyl ester (1), (+)-gallocatechin (2), methyl $6-O-galloyl-{\beta}-D-glucopyranoside$ (3), and pomolic acid $3-O-[{\alpha}-L-arabinopyranoside]-28-O-[{\beta}-D-glucopyranosyl]$ ester (ziyu-glycoside I) (4). Compounds 2 and 3 showed significant DPPH free radical scavenging effects, exhibiting $IC_{50}$ values of 11.4 and $13.0\;{\mu}M$, respectively. L-Ascorbic acid was used as a positive control and exhibited the $IC_{50}$ value of $50.3\;{\mu}M$. In evaluation of the hepatoprotective activity of the isolated compounds on drug-induced cytotoxicity, compound 2 showed the significant hepatoprotective effect with the $EC_{50}$ value of $91.84\;{\pm}\;11.0\;{\mu}M$ on tacrine-induced cytotoxicity in Hep G2 cells, while silybin, a positive control, exhibited $EC_{50}$ value of $122.4\;{\pm}\;12.5\;{\mu}M$.

Effect of Kaempferol on the Cytotoxicity Induced Oxygen Free Radicals in Skin Fibroblast Derived from Human In Vitro

  • In order to evaluate on the effect of kaempferol on the cytotoxicity of oxygen tree radicals, XTT assay was performed to determine the cell viability after skin fibroblasts derived from human (Detroit 51) that were treated with various concentrations of hydrogen peroxide $(H_2O_2)$. And also, the effect of kaempferol on the cytotoxicity induced by H202 that was examined by cell viability, lactate dehydrogenase (LDH) activity and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity in these cultures. $H_2O_2$ decreased cell viability in dose-dependent manner in these cultures and the $XTT_{90}\;and\;XTT_{50}$ values were determined at concentration of $35{\mu}M\;and\;90{\mu}M$ of $H_2O_2$ after skin fibroblasts derived from human were treated with $15{\sim}90{\mu}M$ of $H_2O_2$ for 6 hours, respectively. $H_2O_2$ was highly toxic on cultured skin fibroblasts derived from human by toxic criteria of Brenfreund and Puerner (1984). In the protective effect of kaempferol on $H_2O_2$-induced cytotoxicity, kaempferol increased DPPH radical scavenging activity and significantly decreased LDH activity. From these results, it is suggested that oxygen tree radical, $H_2O_2$, was highly toxic on cultured skin fibroblasts derived from human, and also kaempferol of flavonoid showed the protection on $H_2O_2$-induced cytotoxicity.
