In this study, we investigated the antioxidative effects of AY by measuring 1.1-Diphenyl-2-picrylhydrazyl (DPPH), hydroxyl radical ($^{\cdot}OH$), and superoxide radical ($O_2{^-}$) scavenging activities in vitro. AY was extracted with ethanol and then partitioned with n-hexane, methylene chloride ($CH_2Cl_2$), ethylacetate (EtOAc) and n-butanol (n-BuOH). In the DPPH radical scavenging assay, AY at concentrations of 10 to $100{\mu}g/mL$ dose-dependently increased inhibition of DPPH oxidation, with the EtOAc fraction of AY showing the highest DPPH radical scavenging activity fractions. The $^{\cdot}OH$ radical scavenging activities of the extract and four fractions of AY increased by over 80% at a concentration of $50{\mu}g/mL$. In particular, the IC50 value of the EtOAc fraction was $0.03{\mu}g/mL$, which was the lowest value among all fractions. We also found that the EtOAc fraction of AY was better at $O_2{^-}$ radical scavenging than other fractions. Taken together, these results suggest that AY, especially the EtOAc fraction, can be used as a natural antioxidant against free radicals.

Present studies were conducted to investigate the effects of water and 80% ethanol solvent on antioxidant activities of flower extracts of Chrysanthemum frutescens, Coreopsis lanceolata, Matricaria recutica, and Hieracium pilosella. Extraction yields of water extracts ranged 27.15~40.25% and 80% ethanol extracts 24.92~42.84%, respectively. In all species polyphenol and flavonoid contents were higher with 80% ethanol extraction, especially in C. lanceolata. Same results were obtained with scavenging effects on DPPH and ABTS radicals and ferrous ion chelating effects. Scavenging activity of flower extracts on DPPH radicals was highest in H pilosella - higher than that of synthetic antioxidant DHT. Higher scavenging activity on ABTS radicals was observed with M. recutica. Generally Compositae flower extracts exhibited higher scavenging activity on ABTS radicals than DPPH. Scavenging activity on ABTS radicals of M. recutica and P pilosella were superior to those of ascorbic acid and BHT. Ferrous ion chelating effects were much higher with H. pilosella flower extracts by 80% ethanol. Chelating effects of 4 species were much lower than those of EDTA. In conclusion, to develop natural antioxidant from above 4 Compositae species, 80% ethanol is recommended for efficient solvent to obtain maximum antioxidant isolation and activity.
As part of our continuing search for bioactive natural products, the antioxidant and ${\alpha}$-glucosidase inhibitory activities of an 80% methanolic extract and organic solvent soluble-portions of Aruncus dioicus var. kamtschaticus roots were investigated by using a bioassay system. The antioxidant activity of A. dioicus var. kamtschaticus roots extract and organic solvent soluble-portions were assessed by examining with 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) ($ABTS^+$) radical scavenging. In addition, anti-diabetic effects of the A. dioicus var. kamtschaticus root extract and organic solvent soluble-portions were tested via ${\alpha}$-glucosidase inhibition assay. The total phenolic contents of the products were determined by applying UV-VIS spectrophotometry. All tested samples showed dose-dependent radical scavenging and ${\alpha}$-glucosidase inhibitory properties. In particular, the ${\alpha}$-glucosidase inhibitory and radical scavenging effects of the ethyl-acetate (EtOAc)-soluble portion from the roots of A. dioicus var. kamtschaticus were greater than those from other solvent-soluble portions. These results indicate that A. dioicus var. kamtschaticus could be considered a new effective source of natural antioxidants and anti-diabetic materials. More systematic research of the constituents of the roots of this A. dioicus variety will be conducted to further develop its antioxidative and anti-diabetic properties.
The major objective of this study was to investigate the antioxidant activities of methanolic extracts from different parts (flower, leaf stem, and root) of Chrysanthemum zawadskii by employing various in-vitro assay systems. The extraction yields from the flower, leaf stem, and root were 18.347, 12.93, and 11.33-----, respectively. The total polyphenol content was highest in the flower (17.16 mg/100 g) and lowest in the root (11.33 mg/100 g). The antioxidant activities were raised within creasing amounts of extracts, and the extracts from the flower showed the highest effect on the superoxide anion radical scavenging, metal chelating on ferrous ions and reducing power. In addition, the leaf stem also showed good antioxidant activity in various systems. These results suggest that the methanolic extracts from the flower and leaf stem possess excellent antioxidant activities and may thus serve as potential sources of natural antioxidants.

The principal objective of this study was to investigate the antioxidant and synergistic effects of fruit and vegetable concentrates. Ten foods from two categories, including fruits (raspberry, blackberry, blueberry, acai berry, aronia, cranberry, wild berry, and red grape) and vegetables (spinach and cabbage) were combined in pairs. The antioxidant activity of the individual and combined samples was measured using DPPH (2,2-diphenyl-1-picryl-hydrazyl-hydrate) and FRAP (ferric reducing antioxidant power) assays. Synergistic antioxidant activities of the combinations of cabbage and acai berry, and blueberry and cranberry showed the most significant (p<0.05) increase in the DPPH and FRAP assays, respectively. In addition, the combination of cabbage and red grape demonstrated significantly high synergistic interaction in both DPPH and FRAP assays (p<0.05). These results indicate the importance of strategic selection of foods and their composition ratio for maximum synergistic antioxidant activity.

We investigated the antioxidant activities and effects of soy-sprout extracts (SE) against $H_2O_2$-induced oxidative stress in HepG2 cells. The major free amino acids were asparagine, valine, pheylalanine, histidine, isoleucine, and leucine in SE. Both soy-spout extract by enzyme (SEE) and soy-spout extract by ultra high pressure (SEP) showed higher contents compared with soy-sprout water extract (SEW). The total polyphenol and isoflavone contents were highest in SEE. SEE had the highest DPPH and ABTS radical scavenging activities as well. To determine the effects of SE on $H_2O_2$-induced oxidative damage, cell viability was measured using XTT assay. Pre-treatment with SEE and SEP significantly increased cell viability compared with $H_2O_2$-treated control cells by 29% and 32%, respectively. These results indicate that SEE and SEP possess antioxidant activity.

Desmodium heterocarpon is one of vines belongs to Fabaceae family, mainly distributed in Asian countries such as Korea and Japan. This study was conducted to explore new nutraceutical resources from the plant kingdom possessing biological activities. To fulfill this purpose, the anti-oxidative and anti-inflammatory activities of D. heterocarpon ethanol extract (DHEE) were evaluated by 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging activity assay, reactive oxygen species (ROS) scavenging activity assay, nitric oxide (NO) inhibitory activity assay, and the analysis of related protein expressions by Western blot hybridization. DHEE exhibited potent anti-oxidative activity as confirmed by DPPH radical scavenging capacity against DPPH similar with ascorbic acid, a well-known anti-oxidative agent, used as a positive control. DHEE also effectively suppressed hydrogen peroxide ($H_2O_2$)-induced ROS on RAW 264.7 murine macrophage cells. Furthermore, DHEE induced the expression of the anti-oxidative enzyme heme oxygenase 1 (HO-1), and its upstream transcription factor, nuclear factor-E2-related factor 2 (Nrf2) as a dose dependent manner. DHEE inhibited lipopolysaccharide (LPS) induced nitric oxide (NO) formation as a consequence of inducible NO synthase (iNOS) down regulation. Taken together, these results suggest that DHEE has anti-oxidative and anti-inflammatory activities and thus appears to be useful sources as potential anti-oxidant and anti-inflammatory agents. The identification of active compounds that confer biological activities of DHEE might be needed.
In this study, the antioxidant and tyrosinase inhibitory activities of aqueous ethanolic extract from the immature fruits of Malus pumila cv. Fuji were evaluated. The antioxidant capacities of the extract was investigated employing radical scavenging assays using DPPH and $ABTS^+$ radicals. The skin-whitening effect of M. pumila cv. Fuji extract was tested using mushroom tyrosinase assay. In addition, the total phenolic content was measured by a spectrophotometric analysis. All tested samples showed a dose-dependent radical scavenging and tyrosinase inhibitory activities. Among the tested samples, the ethyl acetate (EtOAc)-soluble portion from the immature fruits of M. pumila cv. Fuji was showed the significant DPPH and $ABTS^+$ radicals scavenging activities. Also, the tyrosinase inhibitory activity of the ethyl acetate (EtOAc)-soluble portion from immature apples was higher than other solvent-soluble portion. These results suggest that unripe fruits of M. pumila cv. Fuji could be considered as a new valuable source of natural antioxidant and skin-whitening agents. Systematic investigation of immature fruits of Malus pumila cv. Fuji will be performed for the further development of its biological properties.
In this study, we analyze the antioxidant and anti-inflammatory activities of various extracts of Cryptoporus volvatus. Three extracts were prepared using hot-water, 70% ethanol, and 70% methanol. Compared to the hot-water and 70% methanol extracts, the 70% C. volvatus ethanol extracts showed significantly higher DPPH radical scavenging activities (76.1%), nitrite scavenging activities (29.5%), and total polyphenol content (9.17 mg GAE/g). The DPPH radical and nitrite scavenging activities of C. volvatus both correlated significantly with the total polyphenol content. The Pearson's correlation coefficient of DPPH radical and nitrite scavenging activities with total polyphenol content were r = 0.744 (p<0.05) and r = 0.921 (p<0.01), respectively. To test their anti-inflammatory effects, nitric oxide production and cell viability were measured by performing nitric oxide (NO) and MTT assays on lipopolysaccharide-treated RAW 264.7 cells. The NO production levels of the 70% ethanol extract-treated cells were slightly lower than those of the others. In our MTT assay, the hot-water extracts showed higher cell viabilities (81.4%) than those of 70% ethanol and 70% methanol extracts. The ${\beta}$-glucan content of C. volvatus was 37.8%. From the results of this study, it is suggested that C. volvatus, an easily accessible wild mushroom, has potential to be used in the development of medicines, cosmetics, and functional health foods.

In this study, the antioxidant and pancreatic lipase inhibitory activities of aqueous methanolic (70% methanol) extract from the roots of Anemarrhena asphodeloides were investigated. The extracts of four solvent fractions (the n-hexane layer, EtOAc layer, n-BuOH layer, and $H_2O$ layer) of the 70% methanol extract were also investigated. Furthermore, the total phenolic content was quantified using a spectrophotometric method. All the tested samples showed dose-dependent radical scavenging and pancreatic lipase inhibitory activities. In particular, the pancreatic lipase inhibitory activity of the ethyl acetate soluble portion (the EtOAc layer) from the rhizomes of the A. asphodeloides was higher than that of the other solvent-soluble portions. The antioxidant property of the extracts was evaluated using radical scavenging assays with DPPH and $ABTS^+$ radicals. 1000 mg/ml of the n-BuOH layer extract showed 91.2% DPPH radical scavenging activity. The EtOAc layer extract and the n-BuOH layer extract showed $IC_{50}=20.5{\pm}1.7mg/ml$ and $IC_{50}=50.5{\pm}0.7mg/ml$$ABTS^+$ radical scavenging activities, respectively. The anti-obesity efficacy of the A. asphodeloides extract was tested via porcine pancreatic lipase assay. A pancreatic lipase inhibitory activity ($IC_{50}$) of $31.3{\pm}0.1mg/ml$ was obtained from the EtOAc layer extract. These results suggest that A. asphodeloides can be considered a new potential source of natural antioxidant and anti-obesity agents.
