Phenolic Content, DPPH Radical Scavenging, and Tyrosinase Inhibitory Activities of Ecklonia cava Extracted with the Ultrasonic Wave Method (초음파 추출법에 의한 감태 추출물의 페놀성 화합물 함량, DPPH 라디칼 소거 활성 및 tyrosinase 저해 활성 분석)

  • The applicability of the ultrasonic wave method to the extraction of useful components from seaweeds was investigated. Extracts from freeze-dried Ecklonia cava powder were prepared with hot water ($65^{\circ}C$), water ($24^{\circ}C$), 50% ethanol, and 100% ethanol, and ultrasonic extraction was also performed. The content of phenolic compounds and the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity and tyrosinase inhibitory activity of the extracts were analyzed, and differences in the data obtained by the ultrasonic extraction and the traditional extraction methods were compared. The phenolic content in the E. cava extract by ultrasonic extraction (142.80 mg/g) was approximately 14 times higher than the phenolic content in the hot water extract (10.03 mg/g). The DPPH radical scavenging and the tyrosinase inhibitory activities of the ultrasonic extract were approximately 4 times and 14 times higher than the hot water extracts, respectively. The correlation between the phenolic content and the DPPH radical scavenging activity ($R^2$=99.47) and between the phenolic content and the tyrosinase inhibitory activity ($R^2$=99.99) was very high. These results indicate that ultrasonic extraction is more suitable than traditional extraction for the extraction of useful components from E. cava.

Inhibitory Effect of DPPH Radical Scavenging Activity and Hydroxyl Radicals (OH) Activity of Hydrocotyle sibthorpioides Lamarck (피막이풀의 DPPH 라디칼과 hydroxyl radicals (OH) 항산화 활성 및 리폭시게나아제 저해 효과)

  • In this study the hot water extract was prepared from Hydrocotyle sibthorpioides (Araliaceae) leaves and stems to study antioxidant activities and lipoxygenase inhibition. The extract showed the protective hydroxyl radical (-OH) which can damage virtually all types of macromolecules: carbohydrates, nucleic acids (mutations), lipids (lipid peroxidation), and amino acids. Hydroxyl radical scavenging activity of H. sibthorpioides was 78.6%. The extract showed strong activity against 1, 1- diphenyl 2-picrylhyorazyl (DPPH) which is a well-known radical and a trap (scavenger) for other radicals. DPPH scavenging activity of leaves of H. sibthorpioides was evaluated at 8.0 mg/ml was 86.0%. Lipoxygenases (LOXs) constitute a heterogeneous family of lipid peroxidizing enzymes capable of oxygenating polyunsaturated fatty acids to their corresponding hydroperoxy derivatives. The inhibitory effect of 15-LOX by H. sibthorpioides was assayed using a Morgan microplate assay. The extract of H. sibthorpioides was 55.5% inhibitory effects on the inhibition of LOX at 8.0 mg/ml. The IC50 values for OH activity, DPPH activity, and LOX inhibition from leaves 5.23 mg/ml, 6.44 mg/ml, and 3.71 mg/ml, respectively. Antioxidative activity assay showed that the water extracts from leaf and stem had a strong reducing power. These results show that H. sibthorpioides has some phytochemical constituents which may be active against the free radicals (OH and DPPH) and lipoxygenase enzyme.

Compositions of Astragali Radix and Angelicae Radix by DPPH Radical Scavenging Activity (당귀(當歸)와 황기(黃芪)의 배합 변화가 DPPH 자유기 소거에 미치는 영향 연구)

  • Objectives : This study was conducted to compare antioxidant activity of ratio-related Angelicae Radix and Astragali Radix compositions by DPPH radical scavenging activity. Methods : The species of Angelicae Radix was compared as Angelica gigas, A. sinensis and A. acutiloba. Then various compositions of Angelicae Radix with Astragali Radix were prepared with different ratio. Diphenyl-picryl-hydrazyl (DPPH) radical scavenging activity was measured after 10, 20 and 30 minutes. Each extracts were tested by 1, 5, 10, 50, 100, 500 and 1,000 ${\mu}g/ml$ concentrations. Results : The results showed that Angelica gigas, A. sinensis and A. acutiloba scavenged DPPH radical with 66.8, 61.7 and 56.7% in 1,000 ${\mu}g/ml$ concentration, respectively. According to this result, Angelicae Radix and Astragali Hadix compositions with 100:0, 10:1, 5:1, 3:1, 1:1, 1:3, 1:5, 1:10 and 0:100 ratio were scavenged DPPH radical with 56.3, 49.9, 61.2, 55.7, 54.7, 55.0, 52.8 ,md 50.2% in 1,000 ${\mu}g/ml$ concentration, respectively. The extract of Angelicae Radix and Astragali Radix compositions with 100:0, 10:1, 5:1, 3:1, 1:1, 1:3, 1:5, 1:10 and 0:100 ratio scavenged DPPH radical with the $IC_{50}$ being 85,21, 1016.26, 656.51, 785.67 and 956.74 ${\mu}g/ml$ respectively. Conclusion : In conclusion, the extract of Angelicae Radix and Astragali Radix compositions with 1:1 ratio showed dose-dependent DPPH radical scavenging activity.

Determination of Antioxidant Activity of Edible Calendula Flowers by Hot-air Drying Time (열풍건조 처리시간에 따른 식용꽃 금잔화의 항산화능 검정)

  • This experiment was conducted to determine optimal hot-air drying time for producing antioxidative calendula tea. The edible calendula was dried in hot air at $60^{\circ}C$ for 5, 10, 15 hours and Hunter's color value was measured and extracted with 95% ethanol as a solvent. The contents of total polyphenol, total flavonoid, total carotenoid and DPPH (2,2 Dipheny-1-picrylhydrazyl) radical scavenging activity were investigated. The correlation between the amount of antioxidant and antioxidant activity was analyzed. As the treatment time increased, the Hunter values of L, a, and b tended to decrease, and the L value and b value decreased significantly at 15 hours. The color change was occurred in all treatments compared with the control, and the total color difference (${\Delta}E$) was the largest at 15 hours treatment. The contents of total flavonoids, total carotenoids and DPPH scavenging activity increased in the 5 hours, but decreased in the 15 hours compared to the 10 hours. The content of total polyphenol was not changed with different treatment time. A positive correlation ($p{\leq}0.01$, r = 0.610) between carotenoids and DPPH radical scavenging activity was demonstrated. The DPPH radical scavenging activity was increased at 5 and 10 hours compared to the control and decreased at 15 hours. Therefore, 5 hours or 10 hours of treatment is appropriate and further studies are needed to determine the specific treatment time.

Comparative Study of Antioxidant Activity of Apple and Pear Peel

  • Apple and pear are popular fruits consumed in Korea and are common fruit in daily diet. In order to compare the antioxidant activity of the apple and pear peels, total polyphenol contents, total flavonoid contents, $ABTS^+$ free radical scavenging activity, and DPPH free radical scavenging activity were measured from hot water, ethanol, and methanol extracts of the two fruit peels. The total polyphenol and flavonoid contents were highest in 95% methanol extracts of the apple peelsand 70% ethanol extract of the pear peels, respectively. Total polyphenol contents of the pear peels were higher than that of apple peels, and total flavonoid contents of the apple peels were higher than that of pear peels. The apple and pear peels had the highest $ABTS^{+{\cdot}}$ and DPPH free radical scavenging activity in 95% methanol extracts and 70% ethanol extracts, respectively. $ABTS^{+{\cdot}}$ and DPPH free radical scavenging activity of pear peels was higher than that of apple peels, and the DPPH free radical scavenging activity of apple and pear peels were detected in hot water, 95% methanol, and 70% ethanol extracts, respectively. Ascorbic acid, a synthetic antioxidant used as positive control, had significantly higher scavenging activity than the apple and pear peels. In conclusion, the apple and pear peelshave great potential as natural antioxidants. Therefore, above results should be considered to provide the possibility for the development of high functional antioxidants.

Effects of $Angelica$ $Gigantis$ Pharmacopuncture Extracts on the Elastase Activity and DPPH and NO Scavenging Activities (당귀약침액의 Elastase 효소 활성과 DPPH, NO 소거능에 미치는 영향)

  • 목적 : Elastin 섬유는 피부, 폐, 동맥, 정맥 등 조직에서 발견된다. Elastin 섬유의 손실은 대동맥, 동맥, 세동맥의 탄력 등과 연관되어 폐기종, 혈액순환 부전 및 폐동맥 고혈압을 악화시킬 수 있다. 본 연구는 당귀약침액의 elastase 효소활성에 대한 효과와 항산화 효과에 대하여 연구하고자 계획되었다. 방법 : 당귀약침액의 elastase 억제효과와 DPPH, 및 NO 소거능을 측정하여 항산화 효과를 측정하였다. 결과 : 당귀약침액에서 통계적으로 유의한 elastase 활성 억제효과를 관찰할 수 있었다. 또한 통계적으로 강한 DPPH free radical 소거 효과가 관찰되었다. 그리고 NO 소거효과를 관찰할 수 있었으며, 그 효능은 pH 6.0에서 가장 강한 효능을 보였다. 결론 : 본 연구 결과에 의하면, 당귀약침액에는 유효한 elastase 활성 억제효과와 강한 항산화 효과가 있어, 폐기종 및 폐동맥 고혈압 치료에 유효한 작용이 있을 것으로 사료된다.

Convergence study of oxidative stress from fraction of Xanthium strumarium L. (도꼬마리 추출물의 산화적 스트레스에 대한 융합연구)

  • Xanthium strumarium L. is an annual plant belongs to the family Asteraceae which is is called a 'Cocklebur' that is used for medicinal purposes. Convergent phyto-activity of various extracts of Xanthium strumarium L. (Asteraceae) was examined. We estimated antioxidant activity from ground part and fruit extract of X. strumarium using 1, 1-diphenyl-2-picrylhydrazyl radical (DPPH) and ABTS assay. The extract of X. strumarium was separated each fraction that of ethanol, petroleum ether, and ethyl acetate. It showed potent radical scavenging effect against the DPPH radical and ABTS. The study revealed that X. strumarium could be used as a potential source of natural antioxidant.

  • Objective : This study was undertaken to determine the inhibitory effect of Whallak-tang against Nitric Oxide, the scavenging effect of Whallak-tang on DPPH radical. Methods : Both nitric concentration 1, 3, 6 and 24 hours after treatment with Vit. C and Whallak-tang at 0.1, 1, 10 mg/ml and scavenging effect 10, 20, 30 and 60 minute after treatment with Vit. C and Whallak-tang at 0.1, 1, 10 mg/ml were estimated. Results : The inhibitory effect of Whallak-tang against Nitric Oxide was dose-dependent. The scavenging effect of Whallak-tang on DPPH radical was dose-dependent. Conclusion : These results would provide important basic data on the possibility of the clinical treatment of Whallak-tang in musculoskeletal disease.

Measurement of Antioxidation Substances in Basil (바질의 항산화 물질 측정에 관한 연구)

  • Basil is known to contain six types of polyphenols that engage in physiological activation; protocateuic acid, caffeic acid, chlorogenic acid, courmaric acid, rosmarinic acid and quercetin. In this study, the antioxidants in eight types of basil were evaluated. Specifically, the antioxidative activation of basil was evaluated based on the relationship between active oxygen scavenging (DPPH radical-scavenging), which was used as an index showing the content and functionality of the polyphenol compounds in basil, and $Fe^{2+}$/ascorbate (FTC) and 2-thiobarbituric acid (TBA). The total polyphenol content of the different types basil occurred in the following order: Dark-opal> Lettuce> Bush> Greek> Lemon> Sweet> Geno> Holy. The highest content Dark-opal was 173.3 mg, which was about three times greater than the lowest content Holy, which was 49.85 mg. In addition, DPPH radical-scavenging by Dark-opal 51%, which was the highest scavenging activity occurred in the folling order: Dark-opal> Lettuce> Bush> Greek> Lemon> Sweet> Geno> Holy. The antioxidative activation values measured using the FTC and TBA were the same as the value obtained using the DPPH method. Finally, the level of antioxidative activation measured using FTC, TBA and DPPH methods showed that the higher the content of polyphenol substance was, the stronger the antioxidative activation became.

Antioxidant Activity of Rosa rugosa (해당화의 항산화 효과)

  • An antioxidant activity of Rosa rugosa extract and its solvent-partitioned fractions was determined not only by measuring lipid peroxide produced when a mouse liver homogenate was exposed to the air at 37$^{\circ}C$, using thiobarbituric acid (TBA) but also by evaluating the free radical scavenging effect against DPPH radical, authentic peroxynitrite, and 3-morpholinsydnonimine (SIN-1). All its partitioned fractions including crude extract showed potent scavenging effect against DPPH radical, peroxynitrite, and lipid peroxidation. n-BuOH fraction, in particular, was found to be the most effective in DPPH radical scavenging ability as well as inhibition against lipid peroxidation. The 15% aqueous MeOH fraction also showed a strong potency which was slightly lower than n-BuOH fraction. Based on these results, we suggest that Rosa rugosa could be useful for preventing an oxidative damage.
