Synthesis, Spectroscopic Studies of Binuclear Ruthenium(II) Carbonyl Thiosemicarba-zone Complexes Containing PPh3/AsPh3 as Co-ligands: DNA Binding/Cleavage

  • The ruthenium(II) ferrocenyl heterocyclic thiosemicarbazone complexes of the type $[RuCl(CO)(EPh_3)]_2L$ (where E = P/As; L = binucleating monobasic tridendate thiosemicarbazone ligand) have been investigated. Strutural features were determined by analytical and spectral techniques. Binding of these complexes with CT-DNA by absorption spectral study indicates that the ruthenium(II) complexes form adducts with DNA and has intrinsic binding constant in the range of $3.3{\times}10^4-1.2{\times}10^5M^{-1}$. The complexes exhibit a remarkable DNA cleavage activity with CT-DNA in the presence of hydrogen oxide and the cleavage activity depends on dosage.

Structural, Electrochemical, DNA Binding and Cleavage Properties of Nickel(II) Complex [Ni(H2biim)2(H2O)2]2+ of 2,2'-Biimidazole

  • A nickel(II) complex $[Ni(H_2biim)_2(H_2O)_2](ClO_4)_2{\cdot}H_2O$ (1) of biimidazole ligand has been synthesized and characterized (Where $H_2biim$ = 2,2'-biimidazole). The single crystal X-ray diffraction of the complex shows a dimeric structure with six coordinated psudo-octahedral geometry. The cyclic voltammograms of complex exhibited one quasireversible reduction wave ($E_{pc}=-0.61V$) and an irreversible oxidation wave ($E_{pa}=1.28V$) in DMF solution. The interaction of the complex with Calf-Thymus DNA (CT-DNA) has been investigated by absorption and fluorescence spectroscopy. The complex is an avid DNA binder with a binding constant value of $1.03{\times}10^5M^{-1}$. The results suggest that the nickel(II) complex bind to CT-DNA via intercalative mode and can quench the fluorescence intensity of EB bind to CT-DNA with $K_{app}$ value of $3.2{\times}10^5M^{-1}$. The complex also shown efficient oxidative cleavage of supercoiled pBR322 DNA in the presence of hydrogen peroxide as oxidizing agent. The DNA cleavage by complex in presence of quenchers; viz. DMSO, KI, $NaN_3$ and EDTA reveals that hydroxyl radical or singlet oxygen mechanism is involved. The complex showed invitro antimicrobial activity against four bacteria and two fungi. The antimicrobial activity was nearer to that of standard drugs and greater than that of the free ligand.

New Unsymmetric Dinuclear Copper(II) Complexes of Trans-disubstituted Cyclam Derivatives: Spectral, Electrochemical, Magnetic, Catalytic, Antimicrobial, DNA Binding and Cleavage Studies

  • Six new binuclear copper(II) complexes have been prepared by template condensation of the dialdehydes 1,8-[bis(3-formyl-2-hydroxy-5-methyl)benzyl]-l,4,8,11-tetraazacyclotetradecane (PC-a) and 1,8-[bis(3-formyl-2-hydroxy-5-bromo)benzyl]-l,4,8,11-tetraazacyclotetradecane (PC-b) with appropriate aliphatic diamines, and copper(II) perchlorate. The structural features of the complexes have been confirmed by elemental analysis, IR, UV-vis and mass spectra etc. The electrochemical behavior of all the copper(II) complexes show two irreversible one electron reduction process. The room temperature magnetic moment studies depict the presence of an antiferromagnetic interaction in the binuclear complexes. The catechol oxidation and hydrolysis of 4-nitrophenylphosphate were carried out by using the complexes as catalyst. The antimicrobial screening data show good results. The binding of the complexes to calf thymus DNA (CT DNA) has been investigated with absorption and emission spectroscopy. The complex [$Cu_2L^{1a}$] displays significant cleavage property of circular plasmid pBR322 DNA in to linear form. Spectral, electrochemical, magnetic and catalytic studies support the distortion of the copper ion geometry that arises as the macrocyclic ring size increases.

Human Topoisomerase I-DNA 절개가능 복합체에 대한 Indenoisoquinoline 유도체들의 결합양상 연구 (Binding Mode Studies of Indenoisoquinoline Analogues into Human Topoisomerase I-DNA Complex Using Flexible Docking)

  • Topoisomerase I (Topo I) participates in the DNA replication, transcription, and repair. Binding of Topo I inhibitor to the Topo I-DNA cleavage complex forms stabilized ternary complex which blocks DNA religation and ultimately causes cell death. Camptothecin (CPT) and its derivatives have been among the most effective anticancer drugs by inhibition of topo I. However, efforts to synthesize non-CPT drugs have been actively going on because the CPT derivatives have several limitations such as poor solubility, short half-life, and side effects. As an indenoisoquinoline, NSC314622 is not as potent as CPT, but its chemical stability and slower reversibility of the cleavage complex made it a good lead compound. Recently, a series of indenoisoquinoline analogues were synthesized with substituted dimethoxy or methylenedioxy on the aromatic ring and alkylamino on the lactam nitrogen. Some of them showed quite good Topo I inhibitory activity. Using the computer docking program, Surflex-Dock, indenoisoquinoline analogues were docked into the human Topo I-DNA cleavable complex. The docking results showed that the compounds with activity better than NSC314622 intercalated between the -1 and +1 base pairs at the cleavage site, but those with little or no activities did not appear to intercalate. These results could be useful to design new Topo I inhibitors improved than CPT.

Camptothecin 유도체의 Human Topoisomerase I-DNA 복합체에 대한 Docking 연구 (Docking Studies of Camptothecin Analogues into Human Topoisomerase I-DNA Complex)

  • Human topoisomerase I (Topo I) plays a pivotal role in cell replication, transcription and repair and, therefore, is an important anti-cancer target. 20S-camptothecin (CPT) is a representative Topo I inhibitor. Compounds belonging to CPT family inhibit the religation step of Topo I-DNA by binding to the DNA cleavage site. Computational docking studies with Surflex-Dock were carried out to investigate the binding modes between Topo I-DNA binary complex structure and the ligand such as 20S-CPT and 9,10-substituted 20S-CPT analogues. The docking results demonstrated that most of the compounds with $IC_{50}$ value under $0.5{\mu}M$ intercalated exactly between the -1 and +1 DNA bases, deeply toward the cleavage site. The complex was stabilized by hydrogen-bonding and hydrophobic interactions with both the enzyme and the DNA. The compounds with $IC_{50}$ value above $0.5{\mu}M$ were poorly docked and did not intercalate. In addition, the docking results confirmed the overall correlation between the $IC_{50}$ values and docking scores, indicating the possible use of the modeling for the prediction of biological activity and design of potential inhibitors.

Sequencing of the RSDA Gene Encoding Raw Starch-Digesting $\alpha$-Amylase of Bacillus circulans F-2: Identification of Possible Two Domains for Raw Substrate-Adsorption and Substrate-Hydrolysis

  • The complete nucleotide sequence of the Bacillus circulans F-2 RSDA gene, coding for raw starch digesting a-amylase (RSDA), has been determined. The RSDA structure gene consists of an open reading frame of 2508 bp. Six bp upstream of the translational start codon of the RSDA is a typical gram-positive Shine-Dalgarno sequence and the RSDA encodes a preprotein of 836 amino acids with an Mr of 96, 727. The gene was expressed from its own regulatory region in E. coli and two putative consensus promoter sequences were identified upstream of a ribosome binding site and an ATG start codon. Confirmation of the nucleotide sequence was obtained and the signal peptide cleavage site was identified by comparing the predicted amino acid sequence with that derived by N-terminal analysis of the purified RSDA. The deduced N-terminal region of the RSDA conforms to the general pattern for the signal peptides of secreted prokaryotic proteins. The complete amino acid sequence was deduced and homology with other enzymes was compared. The results suggested that the Thr-Ser-rich hinge region and the non-catalytic domain are necessary for efficient adsorption onto raw substrates, and the catalytic domain (60 kDa) is necessary for the hydrolysis of substrates, as suggested in previous studies (8, 9).

Effects of Takrisodokyeum Water Extracts on LNCaP Prostate Cancer Cells

  • Androgen receptors (AR) play a crucial role in the development and progression of prostate cancer. Many studies have suggested that prostate cancer cell proliferation is inhibited by AR downregulation, and it has been reported that Takrisodokyeum (TRSDY) induced apoptotic cell death and suppressed tumorigenesis in human leukemia cells. Therefore, this study was conducted to elucidate the mechanism by which TRSDY affects cell growth and AR expression in androgen-dependent prostate cancer cells (LNCaP cells). We investigated the proliferation and apoptosis of LNCaP cells using MTT and DNA fragmentation assays. In addition, we used western blot analysis to assess the effects of TRSDY on the expression of the AR target gene, prostate-specific antigen (PSA). Furthermore, the mechanism of AR downregulation by TRSDY was investigated using EMSA to analyze the binding activity of AR to androgen response elements (ARE). TRSDY significantly suppressed proliferation and induced apoptosis in LNCaP cells. In addition, TRSDY-induced apoptotic cell death was accompanied by activation of caspase-3 and cleavage of its substrate, poly(ADP-ribose) polymerase. TRSDY also inhibited the constitutively expressed- or 5a-dihydrotestosterone (DHT)-induced AR/PSA protein levels. However, these effects were mediated by inhibition of the binding of AR to ARE. TRSDY-mediated AR/PSA downregulation contributes to the inhibition of cell proliferation and the induction of apoptosis in LNCaP human prostate cancer cells. Our findings suggest that TRSDY may be used as a chemopreventive or chemotherapeutic agent for the treatment of prostate cancer.

새로운 Anthraquinone 유도체, DHAQ-97의 항암작용: 아폽토시스에 의한 인체 유방암세포 사멸 유도 (Anticarcinogenic Activity of a Novel Anthraquinone Derivative DHAQ-97: Induction of Apoptosis in Human Breast Cancer Cell Line MCF-7)

  • DHAQ-97, (2-(3-[p-bis(2-chloroethyl)aminophenyl]-2 formylaminopropanoyloxy) methy1-1,4-dihy-droxy-9,10-anthraquinone), is a novel anthraquinone derivative synthesized for use as an anti-neoplastic agent. In the present study, we have evaluated the selective cytotoxicity of DHAQ-97 by comparing its effects on viability and proliferation of human breast cancer cell line (NCF-7) versus normal immortalized breast epithelial cell line (MCF-10A). Thus, DHAQ-97 reduced both viability and proliferation of MCF-7 cells to a much greater extent than did for MCF-10A cells. The growth inhibitory and anti-proliferative properties of DHAQ-97 appear to be attributable to its ability to induce apoptosis as revealed by positive staining after in 냐셔 nick-end labeling (TUNEL), cleavage of poly(ADP-ribose)polymerase, release of mitochondrial cytochrome c into cytoplasm, and increased expression of pro-apoptotic Bax protein. Recent studies have indicated possible involvement of the ubiquitous eukaryotic transcription factor, NF-kappa B (NF-kB) in the regulation of apoptotic cell death. In line induced cytotoxicity in cultured MCF-7 cells. Furthermore, mild activation of NF-kB, as determined by its increased DNA binding capability, was observed 30 min after treatment with 10$\mu\textrm{m}$ DHAQ-97. Taken together, the above findings suggest that DHAQ-97 exerts selective cytotoxicity towards cancer cells through induction of apoptosis, which appears to be regulated by NF-kB.

전사활성 인자인 Sox4의 단백질 분해효소에 의한 표적 부위에 관한 연구 (A Novel Glycine-Rich Region in Sox4 is a Target for the Proteolytic Cleavage in E. coli)

  • Sox4는 DNA 결합 도메인인 HMG-box와 전사 활성 도메인인 serine rich region (SRR)과 아직 그 기능이 알려져 있지 않은 glycine rich region (GRR)등의 세 개의 기능 도메인을 가지는 전사인자이다. 전사인자인 Sox4는 생체 내 초기 분화 시 중요한 역할을 하는 유전자로 알려져 있으나 여전히 그 정확한 생리적 기능 및 세포 내에서 이 유전자 산물이 전사 활성 에 어떻게 관여하는지 그 정확한 기전은 잘 밝혀져 있지 않다. 이에 본 연구에서는 Sox4의 생리적 기능을 이해하고 세포 내에서 Sox4의 기능 연구에 유용하게 사용할 수 있는 항체를 제조하기 위해 Sox4를 대장균에서 발현, 정제하였다. 모든 기능 도메인을 다 포함하는 Sox4는 대장균에서 발현 시 대부분 절단되는 양상을 나타내었다. Sox4의 각 도메인을 대장균에서 GST-융합 단백질로 발현, 정제하여 그 발현 양상을 비교해 본 바 N-말단을 제거한 Sox4 ($\Delta$HMG)의 경우 67 kDa 크기의 단백질이 생성되므로 이 단백질을 항원으로 이용하여 Sox4의 GRR에 특이적으로 반응하는 항체를 제조하였다. 또한, 67 kDa 크기의 단백질 외에 34 kDa 크기에서 GST-융합 단백질이 관찰되었다. 이 밴드는 Sox4 (GRR)를 발현, 정제시에도 관찰되는 동일한 크기의 밴드이며 thrombin과의 반응을 통해 7 kDa 크기로 절단되는 Sox4 밴드이다. 그러므로 이 들 결과로부터 GRR 내에 단백질 분해효소의 표적 부위가 존재함을 확인할 수 있었다. 본 연구결과는 아직 그 기능이 밝혀져 있는 않은 Sox4의 새로운 도메인인 GRR이 단백질 분해 효소의 표적 부위로 작용하여 Sox4의 안정성을 조절함으로써 Sox4의 활성에 중요한 역할을 수행할 수 있다는 가능성을 제시하고 있다.은 억제 회복 효과는 $Mg^{2+}$에 의한 리보자임의 td intron 구조적 안정성에 기인하는 것으로 추정된다.력이 뛰어난(adaptable) 인간적 요구사항을 충족시켜야 한다. 셋째, 다이내믹 시미트리는 역(逆, reciprocity)의 원리와 보상(補賞, complement)의 원리를 제 1의 구성원리로 하며 공간에서 서로에 대한 역과 공통성(common property)을 갖고 자기유사를 지닐 때 연속체(continuum)를 손상하지 않고 전체공간을 유기체적으로 분절한다.같은 pattern 이었다. 그러나 JR89주에서는 280kb 가 나타나지 않아 다른 분리균주와 구분되었다.과 밀가루국(麴) 사이에 차(差)가 별(別)로 없었다.果)에서 총지질(總脂質)을 구성(構咸)하는 지방산(脂肪酸) 조성(組成)은 $C_{18:2}$산(酸), $C_{16:0}$산(酸)의 순(順)으로 그 함량(含最)이 맞은데 비(比)하여 각획분(各劃分)의 지질(脂質)을 구성(構成)하는 지방산(脂肪酸) 조성(組成)은 $C_{16:0}$산(酸), $C_{18:2}$산(酸)의 순(順)으로 그 함량(含量)이 많은 것으로 나타났으며 동결건조후(凍結乾燥後) 저장(貯藏)하는 동안에$C_{18:2}$산(酸), $C_{18:3}$산(酸)의 함량(含量)이 계속(繼續) 감소(減少)하고 있었다. 5. 4-monomethylsterol fraction에는 cycloartenol(20.6%)이 비교적(比較的) 높은 함량(含量)으로 함유(含有)되어 있었고, 그 외(外) cyclolaudenol, cycloeucalenol 및 citrostadienol 등이 함유(含有)되어 있었다. 6. 4-desmethylsterol fraction에
