본 연구에서는 온라인 마켓에서 판매되는 50개 복어제품의 종판별 및 표시사항 일치여부 모니터링을 수행했다. 복어의 종판별을 위해 cytochrome c oxidase subunit I 및 cytochrome b 유전자의 염기서열을 분석하여 NCBI GenBank 데이터베이스에 등록되어있는 생물종의 염기서열과 비교 후 계통 분석을 수행했다. 참복, 흰점참복, 까치복, 복섬, 검복, 국매리복, 흑밀복 총 7종이 동정되었으며, 35개 제품(70%)에서 표시사항과의 불일치를 나타냈다. 12개의 제품(24%)에서는 식품공전에서 제시한 식용 가능한 복어 21종의 국명 대신 일반명(복어)을 사용하였다. 가공 정도별 불일치율은 다중가공 제품(n=9, 81.8%)이 단순가공 제품(n=26, 66.7%)보다 높은 비율을 보였으며, 원산지별 불일치율의 경우 중국산 제품(n=8, 80%)이 국산 제품(n=26, 66.7%)보다 높은 비율을 보였다. 시장명, 방언 등의 이름이 혼용되어 다수의 복어종을 밀복, 졸복으로 표시하였다. 이러한 분류체계의 어려움으로 인해 흰점참복, 국매리복과 같은 식용불가 복어가 식용 가능한 복어인 졸복으로 혼용되어 판매되는 것이 확인되었다. 따라서 식용 가능한 복어를 정확히 분류할 수 있는 방법의 개발이 필요하며, 수입 및 국내 유통 복어 제품의 주기적인 모니터링이 필요하다.
Background: In this study, we investigated how to convert the Panax ginseng DNA sequence code and chemical fingerprints into a two-dimensional code. In order to improve the compression efficiency, GATC2Bytes and digital merger compression algorithms are proposed. Methods: HPLC chemical fingerprint data of 10 groups of P. ginseng from Northeast China and the internal transcribed spacer 2 (ITS2) sequence code as the DNA sequence code were ready for conversion. In order to convert such data into a two-dimensional code, the following six steps were performed: First, the chemical fingerprint characteristic data sets were obtained through the inflection filtering algorithm. Second, precompression processing of such data sets is undertaken. Third, precompression processing was undertaken with the P. ginseng DNA (ITS2) sequence codes. Fourth, the precompressed chemical fingerprint data and the DNA (ITS2) sequence code were combined in accordance with the set data format. Such combined data can be compressed by Zlib, an open source data compression algorithm. Finally, the compressed data generated a two-dimensional code called a quick response code (QR code). Results: Through the abovementioned converting process, it can be found that the number of bytes needed for storing P. ginseng chemical fingerprints and its DNA (ITS2) sequence code can be greatly reduced. After GTCA2Bytes algorithm processing, the ITS2 compression rate reaches 75% and the chemical fingerprint compression rate exceeds 99.65% via filtration and digital merger compression algorithm processing. Therefore, the overall compression ratio even exceeds 99.36%. The capacity of the formed QR code is around 0.5k, which can easily and successfully be read and identified by any smartphone. Conclusion: P. ginseng chemical fingerprints and its DNA (ITS2) sequence code can form a QR code after data processing, and therefore the QR code can be a perfect carrier of the authenticity and quality of P. ginseng information. This study provides a theoretical basis for the development of a quality traceability system of traditional Chinese medicine based on a two-dimensional code.
자리돔과(Teleostei: Pomacentridae) 어류는 열대 및 아열대 해역에 널리 분포하고 있으며 특히 산호해역의 해양생물 군락을 이루는 주요구성 어류이다. 현재 우리나라에는 본과에 5종이 분포하는 것으로 알려져 있는데 본 논문에서는 genus Chromis와 Dascyllus 2속을 대상으로 분자계통학적 위치를 규명하는데 그 목적이 있다. 분자계통분석에서 일본산 D. aruanus는 계통도상에서 폴리네시아산 D. aruanus와 함께 유집되었고, 우리나라의 Chromis fumea는 호주의 C. nitida와 함께 유집되었으며, 두 종간의 유전적 거리를 표시하는 P 값은 0.047로 상대적으로 매우 낮았다. 우리나라의 C. notatus는 뉴칼레도니아산 C. flavomaculata와 함께 유집되었는데, 특히 제주의 D. melanurus와 인도-태평양의 D. melanurus 사이에는 유전자 염기서열의 차이가 전혀 관찰되지 않았다. 그러나 이 두 지역의 D. melanurs와 인도네시아의 D. melanurus간에는 한개의 염기서열 차이가 있었다. 일본산 Dascyllus aruanus의 염기서열은 폴리네시아산 Dascyllus aruanus와 2개의 염기서열 차이가 있었고, 한국산 Chromis fumea와 C. notatus 사이에는 다소 많은 수의 염기서열 차이가 있었다. 본 논문에서 도입한 미토콘드리아 cytochrome DNA의 염기서열 정보 및 분석은 이 유전자가 종 수준에서 뿐만 아니라 지역집단의 구분 및 확인을 위한 DNA 바코드로서 이용될 수 있음을 보여준다.
동절기(1 ~ 2월) 시설재배지 대파에 파총채벌레(Thrips tabaci)가 발생하였다. 파총채벌레의 동정은 트랩에 포획된 개체의 DNA 바코드를 중심으로 확인하였다. 주별 파총채벌레 발생은 끈끈이판 하나당 약 240 ~ 700 마리의 포획 밀도를 나타냈다. 포획 효율은 트랩 색상에 따라 차이를 보여 황색이 청색 트랩에 보다 우수하였다. 또한 대부분(90% 이상) 대파는 이들 파총채벌레의 식흔을 보였다. 이러한 파총채벌레 발생은 특정 비닐하우스에 국한되었다. 이러한 국부적 파총채벌레의 발생 양상을 분석하고자 이들 행동을 실내외에서 관찰하였다. 실내 분석은 약 1.5 mm 정도 몸길이의 성충이 약 5 cm 까지 도약하였다. 야외에서는 이들 성충이 시설재배지 최대 높이인 2 m 까지 비행 행동을 보였다. 이러한 비행 행동은 인근(2 m 이내) 시설재배지까지 이동이 가능할 것으로 추정되었으나 실제로 전파되지 않은 것은 야외 저온 조건이 물리적 장벽을 제공하여 준 것으로 해석되었다. 따라서 겨울기간 파총채벌레의 대발생은 특정 소지역에 국한되었다.

Red algal mitochondrial genomes (mtDNAs) can provide useful information on species identification. mtDNAs of Pyropia / Porphyra (Bangiales, Rhodophyta) have shown diverse variation in their size and gene structure. In particular, the introns and intronic open reading frames found in the ribosomal RNA large subunit gene (rnl) and cytochrome c oxidase subunit 1 gene (cox1) significantly vary the mitochondrial genome size in Pyropia / Porphyra species. In this study, we examined the exon / intron structure of rnl and cox1 genes of Pyropia yezoensis at the intraspecific level. The combined data of rnl and cox1 genes exhibited 12 genotypes for 40 P. yezoensis strains, based on the existence of introns. These genotypes were more effective to identify P. yezoensis strains in comparison to the traditional DNA barcode cox1 marker (5 haplotypes). Therefore, the variation in gene structure of rnl and cox1 can be a novel molecular marker to discriminate the strains of Pyropia species.

Twelve plant species were collected from the east coast of Korea to identify culturable endophytes present in their roots. The fungal internal transcribe spacer (ITS) region (ITS1-5.8SrRNA-ITS2) was used as a DNA barcode for identification of fungi. A total of 194 fungal strains were identified and categorized into 31 genera. The genus Penicillium accounted for the largest number of strains, followed by the genus Aspergillus. Furthermore, using 5 statistical methods, the diversity indices of the fungi were calculated at the genus level. After comprehensive evaluation, the endophytic fungal group from Phragmites australis ranked highest in diversity analyses. Several strains responsible for plant growth and survival (Penicillium citrinum, P. funiculosum, P. janthinellum, P. restrictum, and P. simplicissimum), were also identified. This study provides basic data on the sheds light on the symbiotic relationship between coastal plants and fungi.
Fishes in genus Sardinella are small pelagic species, which plays an important role in marine ecosystem as the first consumer. Those species are also commercially important, whose total catch reaches 278,600 tons in 2011 in Indonesia, but their identification has been difficult for their morphological similarity. In this study, we reported Sardinella jussieu for the first time in Indonesian coastal area (Banten Bay, Indonesia, $6^{\circ}\;0^{\prime}\;50.00^{{\prime}{\prime}}\;S-106^{\circ}\;10^{\prime}\;21.00^{{\prime}{\prime}}\;E$). We were able to confirm the species by both its morphological characteristics including the black spot at dorsal fin origin, the dusky pigmentation at caudal fin, 31 total scute numbers, and DNA sequence identity in the GenBank database by the molecular analysis. Its total mitochondrial genome was determined by the combination of next-generation sequencing and typical PCR strategy. The total mitochondrial genome of Sardinella jussieu (16,695 bp) encoded 13 proteins, 2 ribosomal RNAs, 22 transfer RNAs, and the putative control region. All protein-coding genes started with ATG and typical stop codon and ended with TAA or TAG except for ND4 in which AGA is used. Phylogenetic analyses of both COI region and full mitochondrial genome showed that S. jussieu is most closely related to Sardinella albella and Sardinella gibbosa

We conducted five times surveys, in June, September and October in 2012; June and September 2013, to catalog the mushroom flora in Ulleung-gun, Republic of Korea. More than 400 specimens were collected, and 317 of the specimens were successfully sequenced using the ribosomal DNA internal transcribed spacer barcode marker. We also surveyed the morphological characteristics of the sequenced specimens. The specimens were classified into 2 phyla, 7 classes, 21 orders, 59 families, 122 genera, and 221 species, and were deposited in the herbarium of Korea National Arboretum. Among the collected species, 72% were saprophytic, 25% were symbiotic, and 3% were parasitic. The most common order was Agaricales (189 specimens, 132 species), followed by Polyporales (47 specimens, 27 species), Russulales (31 specimens, 22 species), Boletales (10 specimens, 7 species), and so on. Herein, we also reported the first Bovista species in Korea, which was collected from Dokdo, the far-eastern island of Korea.
Boxwood is a representative ornamental shrub that is widely used in landscaping horticulture. After pruning, damaged leaves or stems of boxwoods are unavoidably vulnerable to infection by various plant pathogens. Several boxwood diseases caused by fungi, such as Volutella blight and Macrophoma leaf spot, have been reported worldwide including Republic of Korea. In this study, we isolated and identified fungal pathogens of boxwood diseases that occurred in Korea and characterized their morphological and taxonomic characteristics. Boxwood samples showing blight symptoms were collected in Seoul, Republic of Korea, and the putative fungal pathogens Pseudonectria buxi, P. foliicola, and Neofusicoccum buxi were successfully identified. Investigation of the morphological features of the field isolates, including mycelial growth and conidial morphology, and phylogenetic analysis of multiple DNA barcode loci revealed that there were some morphological and genetic variations among isolates, but all of the analyzed isolates were closely related to the corresponding reference strains. We also found that P. foliicola strains were more virulent than P. buxi, and the N. buxi strains isolated in this study were weak pathogens or saprophytes. The results of our study will contribute to the development of control strategies for boxwood diseases caused by fungi and accelerate research on the complex ecology of boxwood diseases.
Zoysia 속 잔디는 학교운동장 및 공원, 골프장, 스포츠경기장과 같이 다양한 장소에 식재되고 있는 중요한 잔디이다. 해안가에서 자생하는 Zoysia 속 들잔디와 갯잔디는 외부 형태적 특성이 유사하여 외부 형태적 분류 뿐 만 아니라 분자생물학적 분류도 필요하다. 본 연구에서는 nrDNA-ITS(Internal Transcribed Spacer)의 DNA 바코드 분석을 통해서 자생하는 들잔디와 갯잔디의 분자생물학적 신속한 분류체계를 확립하고자 하였다. 이를 위해 난지형 잔디인 Zoysia 속 들잔디(Z. japonica) 및 갯잔디(Z. sinica)와 한지형 대표 잔디인 크리핑 벤트그라스(A. stolonifera) 및 켄터키 블루그라스(P. pratensis)의 nrDNA-ITS 염기서열을 확보하였다. 확보된 들잔디및 갯잔디, 크리핑 벤트그라스, 켄터키 블루그라스의 ITS 염기서열 전체 구간은 각 686bp와 687bp, 683bp, 681bp으로 확인되었으며, nrDNA-ITS 내부 염기서열구간 분석 결과, ITS1의 크기는 248-249bp, ITS2는 270̵-274bp, 5.8S rDNA는 163-164bp의 차이로, 각 4종의 잔디가 ITS 염기서열을 이용하여 식별되었다. 특히, 들잔디와 갯잔디 nrDNA-ITS 염기서열은 19 염기(2.8%) 차이를 나타냈으며, ITS1과 ITS2의 G + C 함량은 55.4-63.3% 임을 확인하였다. 이러한 들잔디와 갯잔디의 ITS 염기서열 차이를 바탕으로 CAPS 마커로 전환하여 대조구 및 수집된 자생 Zoysia 속 잔디 영양체 62개체를 분석한 결과, 외부형태학적 분류법으로 들잔디 개체, 갯잔디 개체로 동정되었지만, ITSCAPS 마커를 이용한 분자생물학적 분류법으로 들잔디 36개체와 갯잔디 22개체 뿐만 아니라 들잔디와 갯잔디간의 자연교배종 4개체도 식별하였다. 이상의 결과에서 들잔디와 갯잔디는 ITS 염기서열 및 ITS 기반 CAPS를 통하여 식별할 수 있을 것으로 판단된다.
