This study was conducted to verify the relationships between the expression values of stress-related markers and their production performances in 25 strains of Korean domestic chicken breeds. For stress response markers, the amount of telomeric DNA; expression levels of heat shock protein (HSP)-70, $HSP-90{\alpha}$, and $HSP-90{\beta}$; and comet scores were analyzed. Production performances were measured by the survival rate, body weights, days at first egg laying, egg weight and hen housed egg production. The results showed that the production traits and values of stress-related markers showed significant differences between strains. In general, the stress response of pure bred chickens with heavy weights was relatively high, while that of hybrid chickens with light weights was relatively low. The correlation coefficients between telomere contents and body weights showed that there were weak negative relationships. However, the correlations of telomere content with the survival rate and egg production were weakly positive after 20 weeks old. The expression levels of HSP genes and DNA damage rate (comet scores) were positively correlated to body weight, but were negatively correlated to the survival rate and egg production. The results implied that increasing body weight was associated with increasing HSPs expression and the DNA damage rate was associated with decreasing telomere content. In addition, increasing HSPs expression and the DNA damage rate decreased the survival rate and egg production, but the relationships with the telomere content was the reverse. Correlations among the stress-related markers showed that there were significant correlation coefficients between all of the marker values. HSPs expression was negatively correlated to the telomere content, while it was positively correlated to the DNA damage rate. There was a highly negative correlation between the telomere content and DNA damage rate. In conclusion, increasing the HSP values and DNA damage rate can promote telomere reduction, which led to a decrease in disease resistance and robustness of the chicken. Thus, increasing the stress response was verified to adversely affect the laying performance and viability of chickens.

In order to know the biological effect of effluent from shipyard and the adjacent stream water on four organisms (flatfish, rockfish, sea squirt and arkshell) cultured around the shipyard, lethal rate and DNA damage were measured after 48 hr exposure and carried out by a single cell gel electrophoresis, namely comet assay. $LC_{50}$ (48 hr) could not be calculated in any organism 48 hours after exposure to effluent from shipyard and stream water, because all organism showed a lethal rate lower than 20%. Regardless of no acute toxicity, DNA damage of flatfish and rockfish was detected higher in Jang-Pyoung stream than in control, whereas sea squirt revealed higher DNA damage in laundry waste water. From these results, Jang-Pyoung stream seemed to have a relatively higher genotoxicity rather than effluent from shipyard.

To establish a new synthetic Korean meat chicken breed, we tested $5{\times}5$ diallel cross mating experiment with domestic chicken breeds. Comparing stress responses among diallel crossed chicken breeds, we analyzed telomere length, DNA damage and expressions of heat shock protein genes (HSPs) as the markers of the stress response. The telomere length was measured by quantitative fluorescence in situ hybridization on the nuclei of lymphocytes. The expression levels of HSP-70, $HSP-90{\alpha}$ and $HSP-90{\beta}$ genes were analyzed by quantitative real-time polymerase chain reaction in lymphocytes. The DNA damage rate of lymphocytes was quantified by the comet assay known as the single cell gel electrophoresis. In results, there were significant differences in the values of the stress markers such as telomere length, HSPs and DNA damage rate, and also were significant differences in viabilities and body weights among the $5{\times}5$ diallel crossed chicken breeds. The telomere shortening rate, expression values of HSPs and DNA damage rate were significant low in W and Y crossed chickens compare to the others, but GG pure breed showed the highest values in the 25 crossed chickens. Estimating correlation coefficient, the survival rate positively correlated to telomere length, but negatively correlated to the expression levels of HSP-70, $HSP-90{\alpha}$, $HSP-90{\beta}$ genes and to the value of % DNA in tail as DNA damage rate. The expression levels of HSP-70, $HSP-90{\alpha}$ and $HSP-90{\beta}$ genes of dead chickens had significantly higher than those of survival chickens. According to the results on the stress marker analysis, it would be considered that the crossed breeds had more stress resistant than the pure breeds, and the crossed chickens with a light strain such as W or Y were relatively resistant to stress, but the crossed chickens with a heavy strain such as G, H, F were susceptible to stress.

This study was performed to investigate the investigated effects of dietary supplementation of vitamin C and E on the growth performance and stress response in broiler chickens. Stress response was analyzed by the quantity of telomeric DNA, the rate of DNA damage and the expression levels of heat shock proteins (HSPs) and hydroxyl-3-methyl-glutaryl coenzyme A reductase (HMGCR) genes on tissues and blood. The telomere length and telomere shortening rates were analyzed by quantitative fluorescence in situ hybridization on the nuclei of lymphocytes and tissues. The DNA damage rate of lymphocytes was quantified by the comet assay. The expression levels of HSP70, HSP90s and HMGCR genes were measured by quantitative real-time polymerase chain reaction in lymphocytes. In results, there was no significant difference among treatments in body weight, weight gain, feed intake and mortality. The telomere shortening rate of the lymphocytes was significantly lower in the vitamin E supplemented group than the control group. The DNA damage was also decreased supplemented with vitamin C and E, as compared to the control group. The vitamin E supplemented group had a significant positive effect on the expressions of HMGCR, HSP90-${\alpha}$ and HSP90-${\beta}$ in lymphocytes, but had no significance on HSP70, as compared to the control group. We concluded that the dietary supplementation of vitamin E (100 mg/kg feed) had reduced the individual physiological stress response without stunt growth in broiler chickens.
This study was conducted to investigate the effects of housing systems on the productivity and physiological response as stress indicators in White Leghorn chickens. The chickens subjected to the conventional cages had a significantly lower viability, hen-housed egg production, egg weight and body weight compared with those to the floor pens. However, the hens housed in the conventional cages had a shorter day of the first egg and a greater egg quality compared with those housed in the floor pens. In addition, this study was also investigated to identify biological markers for assessing the physiological response of chickens under stress conditions. As biological markers, the amount of telomeric DNA was analyzed by quantitative fluorescent in situ hybridization on the nuclei of cells. The DNA damage rate of lymphocytes was also quantified by the comet assay. The amount of telomeric DNA of the lymphocytes, kidney and spleen was significantly higher in the chickens under floor pens than those under conventional cages. The DNA damage also increased in chickens raised under conventional cages, as compared to the chickens under floor pens. As results, we conclude that the chickens housed in conventional cages have a greater stressful status than those housed in floor pens.

The DNA damage by linoleic acid hydroperoxide (LHPO) was investigated in a DNA-LHPO system at $37^{\circ}C$ to elucidate the DNA damage mechanism by lipid peroxidation products. LHPO shelved a great DNA damage with the increase of its concentrations. DNA was completely damaged in a LHPO-DNA(weight ratio, 2:3) system after incubation for 2 days. The degree of DNA ,damage by LHPO was greated than that of linoleic acid. In the quantitative analysis of DNA damage, the decreasing ratio of DNA content was $60\%$ in $84{\mu}g$ LHPO system incubated for 1 day compared to the control solution marked $30\%$. There were no participation of active oxygens on the DNA damage by LHPO.

With Single Comb White Leghorn (WL) and Korean Native Chicken (KNC) breeds, we compared the stress response with chicken breeds that were subjected to a high stocking density. The expression levels of HSP70, HSP90-${\alpha}$, HSP90-${\beta}$ and HMGCR genes were measured by quantitative real-time polymerase chain reaction in lymphocytes. There was no significant difference between KNC and WL in body weight, weight gain, telomere shortening rate and DNA damage rate. However, the growth rate significantly decreased in chickens raised under high stocking density conditions, as compared to the control group. The telomere-shortening rate, DNA damage and HSPs expression of the lymphocytes were significantly higher in the high stocking density group than the control. The stress condition and breeds had a significant effect on the expressions of HSP70, HSP90-${\alpha}$ and HSP90-${\beta}$ in lymphocytes, except HMGCR. The stress response of WL was higher than that of KNC, as analyzed to the expression of HSP70 and HSP90-${\alpha}$. Therefore, we concluded that the chickens which were exposed to a high stocking density had increased the individual physiological stress response regardless of breeds, and White Leghorns are more susceptible to stress condition than Korean Native Chickens.
This study was performed to investigate the effect of mixed rearing of male and female chickens on the stress response in Korean native chickens. To identify the degree of the stress response, heterophil-lymphocyte ratio (H/L ratio), heat shock protein genes (HSPs) expression, and intracellular nuclear DNA damage rate were analyzed before and after the mixed rearing of male and female chickens. The results showed that the H/L ratio of chickens after mixing males and females was more than thrice as higher than before mixing (P<0.001), but the differences between males and females were not significant. HSP-70, HSP90-α, and HSP90-β expression levels were 2.5 to 3.4 times higher after mixing male and female chickens, compared to before mixing (P<0.01). In the mixed rearing of male and female chickens, the increase in HSPs expression in females was higher than in males. Comet indicators in intracellular DNA damage rate analysis showed a significant increase after mixing male and female chickens compared to before mixing (P<0.001). However, there was no significant difference between males and females with respect to DNA damage rate. Taken together, these results suggest that male and female mixed rearing acts as a strong external stressor in both male and female chickens.
Dose response of DNA repair synthesis induced by bleomycin was dose-dependent in lower doses, and maximum rate of it at 5 $\\mu$g/ml represents about 15% of total cells analyzed. At higher doses DNA-repair synthesis was reduced and the rate of it remained unchanged even prolonged treatment. Pretreatment with BUdR or IUdR was found to enhance DNA repair synthesis and also to interfere with semiconservative DNA synthesis at higher doses. Time dependence study showed that DNA repair synthesis occurred as long as for 24 hours after removal of bleomycin. These results seem to suggest that bleomycin is not to be an effective chemical in inducing excision repair and that damages induced in DNA by this drug might include not only strand breaks but other types of DNA damage.
This study aimed to investigate the effects of beak trimming and crossbreeding-combinations on the productive performance and stress response levels of Korean native chickens. The study divided 248 individuals from six crossbreeding-combinations into two groups: one underwent beak trimming, and the other did not. The survival rate, body weight, egg production rate, egg quality, feather damage score, HSP-70 gene expression level, H/L ratio, and intracellular DNA damage rate were measured and analyzed. The results showed that the beak-trimmed group had significantly higher survival rates and hen-housed egg production compared to the non-beak-trimmed group (P<0.05). Feather damage and DNA damage rates were significantly lower in the beak-trimmed group (P<0.05). On the other hand, there were no significant differences between the two groups in adult body weight, hen-day egg production, egg quality, HSP-70 gene expression level, and H/L ratio. Among the crossbreeding-combinations, there were significant differences in survival rate, body weight, feather damage score, egg quality, and DNA damage rate (P<0.05), while egg production rate, HSP-70 gene expression level, and H/L ratio showed no significant differences. There was an interaction between beak trimming and crossbreeding-combinations in some traits. In conclusion, beak trimming in Korean native chickens has a positive impact on productive performance, and in terms of stress response, beak trimming may not act as a stress factor or may even reduce stress after the growing period. Furthermore, there were differences in productive performance and stress response levels among crossbreeding-combinations, but the effects of beak trimming were similar across these combinations.
