Association between Diabetes Mellitus and Fatty Liver Based on Ultrasonography Screening in the World's Highest Cholangiocarcinoma Incidence Region, Northeast Thailand

  • Fatty liver disease (FLD) can be a precondition for other liver pathology including cholangiocarcinoma (CCA). Diabetes mellitus (DM) has been suggested in some studies to be a risk factor for FLD as well as cancers, including cholangiocellular carcinoma; however, there are currently very few studies on FLD in DM subjects, although the rate of FLD continues to increase annually. To determine the association between DM and FLD ultrasonographic data were analyzed from the Cholangiocarcinoma Screening and Care Program (CASCAP), in northeast Thailand. DM was reported by the subjects based on the CASCAP health questionnaire. Factors that were associated with FLD were determined by prevalence, odds ratio (ORs) and its 95% confidence intervals (CIs) using multiple logistic regression. There were 45,263 subjects with a mean age of 53.46 (${\pm}9.25$) years. FLD was found in 36.3% of DM subjects but only in 20.7% of non-DM subjects. The association between DM and FLD was adjusted for all other factors including gender, age, education level, relatives diagnosed with CCA, smoking, alcohol consumption, and hepatitis B and C. The risk of DM in subjects having FLD was highly significant compared with the non-DM subjects (OR 2.13; 95%CI: 1.92 to 2.35; p-value < 0.001). Thus DM is significantly associated with FLD which in turn may facilitate the development of several diseases including CCA. DM should be taken into consideration in future ultrasonic investigations of FLD and CCA.

Potent Anticancer Effects of Lentivirus Encoding a Drosophila Melanogaster Deoxyribonucleoside Kinase Mutant Combined with Brivudine

  • Objective: Deoxyribonucleoside kinase of Drosophila melanogaster (Dm-dNK) mutants have been reported to exert suicide gene effects in combined gene/chemotherapy of cancer. Here, we aimed to further evaluate the capacity of the mutanted enzyme and its potential for inhibiting cancer cell growth. Methods: We altered the sequence of the last 10 amino acids of Dm-dNK to perform site-directed mutagenesis and constructed active site mutanted Dm-dNK (Dm-dNKmut), RT-PCR and western bloting studies were used to reveal the expression of lentivirus mediated Dm-dNKmut in a breast cancer cell line (Bcap37), a gastric cancer cell line (SGC7901) and a colorectal cancer cell line (CCL187). [3H]-labeled substrates were used for enzyme activity assays, cell cytotoxicity was assessed by MTT assays, cell proliferation using a hemocytometer and apoptosis induction by thenannexin-V-FITC labeled FACS method. In vivo, an animal study was set out in which BALB/C nude mice bearing tumors were treated with lentivirus mediated expression of Dm-dNKmut with the pyrimidine nucleoside analog brivudine (BVDU, (E)-5-(2-bromovinyl)-(2-deoxyuridine). Results: The Dm-dNKmut could be stably expressed in the cancer cell lines and retained its enzymatic activity. Moreover, the cells expressing Dm-dNKmut exhibited increased sensitivity in combination with BVDU, with induction of apoptosis in vitro and in vivo. Conclusion: These findings underlined the importance of BVDU phosphorylated by Dm-dNKmut in transduced cancer cells and the potential role of Dm-dNKmut as a suicide gene, thus providing the basis for future intensive research for cancer therapy.

  • Duckweed, an aquatic plant of the family Lemnaceae, is a rich source of protein and also contains cell wall materials. Spirodela, Lemna and Wolffia, the most available species of duckweeds were evaluated in terms of their chemical composition, the rate and extent of digestion of their dry matter(DM) and crude protein(CP) in the rumen and also their acceptability to the cattle. The three species contained CP of 284, 399 and $299g{\cdot}kg^{-1}$ DM, respectively; NDF of 471, 574 and $476g{\cdot}kg^{-1}$ DM, respectively; ADF of 215, 203 and $227g{\cdot}kg^{-1}$ DM, respectively. The rumen digestibilities of DM of the three species for 24 h were 410, 570 and $731g{\cdot}kg^{-1}$ DM, respectively and of CP were 528, 740 and $778g{\cdot}kg^{-1}$ DM, respectively. The rates of digestion of DM of the three duckweeds were 2.22, 3.63 and $5.73%h^{-1}$, respectively and of CP were 5.14, 4.22 and $6.05%h^{-1}$, respectively. Similarly, the extent of digestion of DM were 853, 723 and $926g{\cdot}kg^{-1}$ DM, respectively and of CP were 801, 874 and $943g{\cdot}kg^{-1}$ DM, respectively. Mixed duckweeds as a component of a concentrate mixture were eaten by the cattle at the rate of 10% of their live weights. It may be concluded that the dry matter and crude protein of the available duckweeds wee highly degradable in the rumen and may be fed to cattle mixing with concentrates. For the effective utilization of duck weeds as cattle feed their effect on the rumen digestion kinetics of a roughage diet need to be studied carefully.

  • ${\beta}-PbO_2$ electrode, which was electrodeposited on Ti madras, was prepared and for the decomposition of cyanide in electroplating wastewater. After the investigation of temperature and pH conditions for no hydrogen cyanide evolution during the decomposition of cyanide, the optimum current densities of ${\beta}-PbO_2$ electrodeposition and cyanide decomposition were determined in 500ppm NaCN solution, and durability of ${\beta}-PbO_2$ electrode was also investigated. Hydrogen cyanide was actively generated above $40^{\circ}C$ and was not evolved above pH 13. ${\beta}-PbO_2$ electrode electodeposited at $5A/dm^2$ showed the best cyanide decomposition efficiency The decompostion efficiency was about 70% at low decomposition current density ($0.08A/dm^2$), and it decreased gradually to about 10% as the decomposition current density increased up to $4A/dm^2$. The film of ${\beta}-PbO_2$ electrodeposited was corrosive at $20A/dm^2$ and was broken at $50A/dm^2$.

Effects of Dietary Oligosaccharide on the Blood Glucose and Serum Lipid Composition in Streptozotocin-Induced Diabetic Rats (Streptozotocin 유발 당뇨쥐의 혈당 및 혈중 지질조성에 미치는 Oligosaccharide의 영향)

  • This study was conducted to examine the effects of dietary oligosaccharide on the blood glucose and serum lipid composition in streptozotocin (STZ)-induced diabetic rats. Sprague-Dawley male rats weighing 150$\pm$10g were randomly assigned to one normal and four STZ-induced diabetic groups. Diabetic groups were classified to basal diet(DM group) 10% xylooligosaccharide diet(DM-XO group) 10% isomaltooligosaccharide(DM-IMO group) and 10% fructooligosaccharide (DM-FO). Diabetes was experimentally induced by intravenous injection of 50 mg/kg of body weight of STZ in citrate buffer (pH 4.3) after feeding of experimental diets for 4 weeks. The rats were fed with experimental diet for further 4 weeks in diabetic state. The oligosaccharide diets were not effected on the body weight food intakes and food efficiency ratio. The oligosaccharide diets were also not effected on the body weight food intakes and food efficiency ratio. The oligosaccharide diets were also not effected on the weights of liver kidney and small intestine but the weight of cecum was significantly increased on the groups of xylooligosaccharide and isomaltooligosaccharide diet. The levels of oral glucose tolerance test was more effectively improved by DM-XO group. The levels of blood glucose were markedly lower in oligosaccharide supplemented groups than that of DM group. The levels of blood glucose were markedly lower in oligosaccharide supplemented groups than that of DM group. Activities of two intestinal enzymes such as lactase and sucrase in DM-XO and DM-FO groups were lower than that of DM group while activity of maltase was lower only in DM-XO in DM-FO groups than that of DM-group respectively. The levels of serum triglyceride in DM-XO group were lower than that of DM-group respectively. The levels of serum triglyceride in DM-XO groups were lower than that of DM group however was no significant differences among the oligosaccharide groups. These results suggest that dietary oligosaccharide may act as functional food to be capable of improving carbohydrate and lipid metabolism in diabetic rats.

A Study on DM-RS Structure for LTE V2V Communications (LTE 차량 간 통신을 위한 DM-RS 구조 연구)

  • The study on 'support for V2V(Vehicle to Vehicle) services based on LTE sidelink' has been carried out in 3GPP RAN working group. The conventional LTE sidelink technology is not adequate for high speed V2V communications because the conventional DM-RS(Demodulation Reference signal) structure is developed for pedestrian environment. While the typical speed of pedestrian environment is 3km/h, that of V2V communications is 120 km/h and the information on channel estimation is not sufficient with legacy DM-RS structure. In this paper, improved DM-RS structures for V2V communications that have rapid channel variation are introduced and the performance of channel estimation accuracy is analyzed. Simulation results show that the performance improvement of channel estimation can be achieved based on extended sub-carrier spacing structure resulted from reduced inter-carrier interference. However, the extended sub-carrier spacing requires the longer cyclic prefix and higher overhead. As a results of considerations on DM-RS modification, the sub-carrier spacing is maintained and the high density of DM-RS is applied for V2V communications.

Effects of Mulberry Juice and Cake Powders on Blood Glucose and Lipid Lowering and Erythrocytic Antioxidative Enzyme Activities in Streptozotocin-Induced Diabetic Rats (오디즙 및 오디박 분말이 Streptozotocin 유발 당뇨쥐의 혈당 및 혈청지질 강하와 적혈구 항산화 효소계에 미치는 영향)

  • The aim of this study was to investigate the effects of mulberry juice and cake powder on blood glucose and lipid status along with intestinal disaccharidase and erythrocyte antioxidative enzyme system in streptozotocin (STZ)-induced diabetic rats. Sprague-Dawley male rats weighing $100{\pm}10g$ were randomly assigned to one normal group, and eight STZ-induced diabetic groups: control diet group without mulberry juice and cake powders (DM-C), three mulberry juice powder groups (0.5%: DM-0.5J, 1%: DM-1J, 2%: DM-2J) and low mulberry cake powder groups (0.25%: DM-0.25C, 0.5%: DM-0.5C, 1%: DM-1 C, 2%: DM-2C). After three-week feeding of each experimental diet, diabetes was induced by intravenous injection of 50 mg/kg body weight of STZ in sodium citrate buffer (pH 4.3) via tail vein of eight DM groups. Rats were sacrificed at the 9th day of diabetic states. Level of blood glucose was 505 mg/dl in DM-C group but it was 28% and 39% lower in mulberry juice and cake powder fed groups, respectively, than the DM-C group. Activities of maltase, sucrase and lactase in proximal part of small intestine were significantly lower in the mulberry juice and cake powder groups by $42{\sim}47%$ than those of DM-C group. Erythrocytic superoxide dismutase, glutathione peroxidase and catalase activities were significantly reduced by STZ but increased close to normal levels along with less accumulation of thiobarbituric acid reactive substances (TBARS). Serum levels of triglyceride and total cholesterol and HDL-cholesterol by STZ-DM were reduced and increased respectively, to the norma] levels by the mulberry juice and cake powder. Except the levels of TBARS, the effects on the other measurements by the various dietary levels of mulberry juice and cake powder were almost same and the effect of the cake powder was most significant at the lowest level. These results indicate that mulberry juice and cake powders have consityerable hypoglycemic effect and strengthening antioxidant defense systems at the low levels in diabetic state and may be able to reduce diabetic complications.

Comparative Studies of Seven Solvents for the Extraction of Neutral Lipid in Corn Embryo (옥수수 배의 중성 지질의 추출을 위한 7종 용매의 비교 연구)

  • The comopositon of lipids extracted from corn embryo with various solvents were analyzed. The solvents for the extraction were benzene(BZ), n-hexane(HX), pet. ether(PE), trichlorethylene (TE), chloroform-methoanol(2:1, v/v) (CM), dichlormethane - methanol(2:1, v/v)(DM) and hexanediethyl ether(5:1, v/v)(HD). The lipids were than fractinated by silicis acid column chromatography(SACC) into three lipid fractions. The Neutral lipid fractons were further separated by thin layer chromatography(TLC) and the individual lipid spots were quantitatived by TLC scanner. And then the fatty acid compositions of total lipids and neutral lipids were determined by gas chromatography(GC). Crude oil contents of corn embryo were most efficient by using DM, CM and neutral lipid was extracted much HX, BZ, HD systems than did PE, DM, DM an CM were most efficient solvent systems for extracting glycolipid and phospholipid. The major component of the neutral lipid fractions was found to be triglyceride, and it was superior DM to PE. Linoleic acid was the predominant fatty acid in the total lipids, and it was most efficient with BZ and TE. The major fatty acid in neutral lipids was also linoleic acid and it was superior BZ to PE, CM, HD and oleic acid was similar to seven solvents and palmitic acid was found much superior in using CM.

Effects of Green Tea Catechin on the Superoxide Dismutase, Glutathione Peroxidase and Xanthine Oxidase Activites of Kidney in Diabetic Rats (녹차 Catechin이 당뇨쥐 신장조직의 Superoxide Dismutase, Glutathione Peroxidase 및 Xanthine Oxidase)

  • The purpose of this study was to investigate the effect of green tea catechins on the antioxidative defense enzyme activity of kidney in diabetic rats. Sprague-Dawley male rats weighting 100$\pm$10g were randomly assigned to one normal and three STZ-induced diabetic groups; catechin free diet(DM-0C group), 0.25% catechin diet(DM-0.25C group) and 0.5% catechin diet(DM-0.5C group). Diabetes was induced by intravenous of 55mg/Kg body weight of STZ in sodium citrate buffer(pH 4.3) after 4 weeks feeding of experimental diets. Rats were sacrified at the 6th day of diabetic states. Superoxide dismutase(SOD) activity in kidney was decreased by 25% and 20% in DM-0C and DM-0.25C groups compared with normal group, DM-0.5C group was not significantly different when compared with normal group. Glutathione peroxidase(GSHpx) activity in kidney was were no significant differences the diabetic groups compared to normal group. Xanthin oxidase(XOD) activity was increased by 110% and 63% in DM-0C and DM-0.25C groups compared with normal group, DM-0.5C group was not significantly different when compared with normal group. The contents of superoxide radical(O$_2$)in kindney were 116% and 33%, respectively, higher in DM-0C and DM-0.25C groups than normal group. DM-0.5C group and normal groups were similar levels in their superoxide radical contents of kidneys. Levels of TBARS(thiobarbituric acid reactive substance) in kidney were increased by 62% in DM-0C group, when compared with normal group, but those of DM-0.5C group were similar to that of normal groups. These results indicate that free radical generation system was weakened and free radical scavenger system was enhance in kidney of STZ-induced diabetics rats by dietary catechin. Thereby it may reduce renal disorders such as oxidative damage and aging of tissue.

Chemical Changes during Ensilage and In sacco Degradation of Two Tropical Grasses: Rhodesgrass and Guineagrass Treated with Cell Wall-degrading Enzymes

  • Effects of the cell wall-degrading enzymes derived from Acremonium cellulolyticus and Trichoderma viride on the silage fermentation and in sacco degradation of tropical grasses i.e. rhodesgrass (Chloris gayana Kunth. cv. Callide) and guineagrass (Panicum maximum Jacq. cv. Natsukaze) were investigated in laboratory-scale experiments. These two grasses were either treated with or without the enzymes before ensiling. Untreated rhodesgrass produced acetate fermentation silage (lactate, $13.0\;g\;kg^{-1}$ DM; acetate, $38.7\;g\;kg^{-1}$ DM) with high final pH value and $NH_3$-N content (5.84 and $215\;g\;kg^{-1}$ DM). Addition of enzymes significantly increased (p<0.01) the lactate production (lactate, 45.6; acetate, $34.0\;g\;kg-^{1}$ DM) and decreased (p<0.01) the pH and $NH_3$-N (4.80 and $154\;g\;kg^{-1}$ DM) in the ensiled forages when compared with the control silages. Untreated guineagrass was successfully preserved with a high lactate proportion (lactate, 45.5; acetate, $24.1\;g\;kg^{-1}$ DM), and the addition of enzymes further enhanced the desirable fermentation (lactate, $57.5\;g\;kg^{-1}$ DM; acetate, $19.4\;g\;kg^{-1}$ DM). The content of NDF was lowered (p<0.05) by enzymes in both silages, but the extent appeared greater in the enzyme-treated rhodesgrass (rhodesgrass, $48\;g\;kg^{-1}$ DM; guineagrass, $21\;g\;kg^{-1}$ DM). Changes in the kinetics of in sacco degradation showed that enzyme treatment increased (p<0.01) the rapidly degradable DM (rhodesgrass, 299 vs. $362\;g\;kg^{-1}$ DM; guineagrass, 324 vs. $343\;g\;kg^{-1}$ DM) but did not influence the potential degradation, lag time and degradation rate of DM and NDF in the two silages.
