악성종양환자(惡性腫瘍患者)에서 혈청(血淸) Tissue Polypeptide Antigen 치(値)의 임상적(臨床的) 의의(意義)에 관(關)한 연구(硏究) (A Study on the Clinical Significance of Serum Tissue Polypeptide Antigen Levels in Patients with Malignancy)

  • To evaluate the clinical significance of serum tissue polypeptide antigen (TPA) levels in patients with malignancy, serum TP A levels were measured by radioimmunoassay in 49 normal controls, 9 patients of postoperative colon cancer without recurrence and 68 patients with various untreated malignancy, who visited Seoul National University Hospital from February, 1983 to September, 1983. The results were as follows; 1) Serum TPA levels in 49 normal controls were in the range of 22-135 U/L $(74{\pm}28U/L,\;mean{\pm}S.D.)$. There was no sex or age difference. Normal upper limit of serum TPA was defined as 130 U/L (mean+2S.D.). 2) Serum TPA levels in 68 patients with various untreated malignancy (stomach cancer 33 cases, colon cancer 11 cases, lung cancer 10 cases, primary liver cancer 9 cases and metastatic cancer of unknown primary site 5 cases) were in the range of 10-800 U/L $(189{\pm}170U/L,\;mean{\pm}S.D.)$ and significantly elevated, compared with those of normal controls (p<0.005). 3) The sensitivities of serum TPA in various untreated malignancy were 39% in stomach cancer, 55% in colon cancer, 50% in lung cancer, 67% in primary liver cancer and 80% in metastatic cancer of unknown primary site respectively. 4) The sensitivities of serum TPA related to resectability in stomach and colon cancer were 32% in resectable stomach cancer, 50% in unresectable stomach cancer, 29% in resectable colon cancer and 100% in unresectable colon cancer respectively. 5) The mean value of serum TPA levels in 9 patients of postoperative colon cancer without recurrence was $70{\pm}39U/L$ and significantly decreased, compared with that of untreated colon cancer, $180{\pm}150U/L$ U/L (p<0.05). 6) In patients with stomach or colon cancer, there was no significant correlation between serum TP A and serum CEA levels, but simultaneous measurement of serum TPA and serum CEA levels increased sensitivities. From above results, we concluded that serum TPA level is a useful indicator reflecting tumor activity and responses to anticancer treatment in patients with malignancy.

Aspergillus nidulans의 유성분화에 관여하는 nsdC 유전자의 분리 및 분석 (Isolation and Characterization of the nsdC Gene in Sexual Development of Aspergillus nidulans)

  • 사상성 진균인 Aspergillus nidulans에서 유성분화초기단계, 또는 유성분화유도를 위한 세포내 조건 형성과정에 관여할 것으로 예상되는 유전자를 탐색하였다. 선행연구결과를 통해 유성분화를 전혀 하지 못하는 NSD (never in sexual development) 돌연변이주가 분리되어 nsdA, nsdB, nsdC, 그리고 nsdD의 4상 보군으로 동정된 바 있다. 본 연구에서는 이들 유전자 중 nsdC 유전자를 분리하고자 A. nidulans AMAl-Not I Genomic DNA library로 nsdC6 돌연변이균주를 형질전환하여 야생형처럼 유성분화를 할 수 있는 형질전환체를 분리하고 이들로부터 약 10 kb genomic DNA가 삽입된 library DNA를 분리하였다. Genomic priming system (GPS)을 이용하여 nsdC6 돌연변이를 상보하는 유전자의 부분 서열을 확보한 후 전체 DNA 염기서열을 결정하였다. 유전자분석 결과 nsdC는 intron 없이 1,929염기(643개의 아미노산)로 구성된 Open reading frame (ORF)를 가지며, 약 1kb 정도의 비교적 긴 5'-UTR 부위에 2개의 intron을 가지고 있음이 확인되었다. 또한 NsdC polypeptide의 중앙에 $C_2H_2C_2H_2C_2HC$ 형의 zinc finger DNA binding domain과 C 말단 부위에 coiled-coil domain이 존재하였다. nsdC6 돌연변이는ORF의 407 bp와 408 bp사이에 엽기 T가 삽입되어 frameshift가 일어난 것으로 밝혀졌다. 따라서 nsdC6 돌연변이균주는 단지 139개 아미노산만 갖고 있는 결실 단백질이 생산됨을 알 수 있었다.

  • 고슴도치의(Erinaceus koneanus)췌강 각 부위별에 출현하는 glucagon(A)세포,insulin(B)세포, somatostatin(D)세포 및 pancreatic polypeptide(PP)세포 등 4종의 내분비세포의 출현빈도, 분포상태, 모양 및 크기 등을 관찰하고자 췌장을 비장부와 십이지장부로 구분하고 이를 각각 근위부, 중간부 및 원위부로 세분한 후 면역세포화학적 방법을 시도하여 광학현미경하에서 검경하였던 바 다음과 같은 결론을 얻었다. Glucagon(A)세포의 크기는13 $\mu$ m x 9.5 $\mu$ m였고 그 모양은 원형, 난원형 및 추체형을 띠고 있었으며 췌도의 주변부와 외분비 실질사이에 분포하고 있었다. A세의 출현빈도는 비장부의 모든 부의에서 24.5%-30.5%, 십이지장부의 원위부에서 30.1%로 관찰되었으나 십이지장부의 근위부와 중간부에서는 2.6%-5.2%로 관찰되었다. Insulin(B)세포의 크기는 11.6$\mu$m x 9.4$\mu$m로서 원형 또는 난원형을 띠었고 췌도내에 균등하게 분포하고 있었으며, 비장부의 모든 부위와 십이지장의 원위부에서는 63.5%-68.9%, 십이지장부의 근위부와 중간부에서는 35.9%-55.6%로 출현하였다. Somatostatin(D)세포는 크기가 12.6$\mu$m x 9.1 $\mu$m로서 주변부와 외분비 실질사에 나타났으며 세포의 출현빈도는 비장부와 십이장부의 모든 부위에서 2.6%-5.9%로서 소수 관찰되었다. Pancreatic polypeptide(PP)세포는 크기가 12.8$\mu$ m x 8.5 $\mu$m였고 세포의 모양은 원형, 난원형 및 추체형 등 다양하였으며 췌도의 주변부와 외분비실질간에 주로 분포하고 있었다. PP세포의 출편빈도는 십이지장부의 근위부와 중간부에서는 각각 42.5%, 55.4%로서 관찰된 반면 비장부의 모든 부위와 십이지장부의 원위부에서는 0.5%-2%로서 소수 출현되었다.

한국산 고슴도치(Erinaceus koreanus)의 장관 내분비세포에 관한 면역조직화학적 및 전자현미경적 연구 (An Immunohistochemical ana Ultrastructural Studies on the Gut Endocrine Cells in the Hedgehog, Erinaceus koreanus)

  • In order to know the distribution, relative frequencies, types and morphology, endocrine cells in the intestinal tract of the hedgehog(Erinaceus koreanus) were studied by light microscopy, immunohistochemistry and electron microscopy. The results obtained are summarized as follows: 1. Two kinds of endocrine cells were demonstrated with two specific staining methods. Argyrophil cells(reactive cells for Grimelius method) were found most frequently in the intestinal region, and were infrequent in the rectum, whereas argentaffin cells (reactive cells for Masson-Hamperl method) were found most frequently in the rectum and in the other legions were infrequent. These reacting cells were mainly found in the intestinal glands, whereas a small number in the mucosa. 2. Twelve kinds of endocrine cells, gastrin(Gas)-, somatostatin (Som)-, serotonin(5-HT)-, glucagon(Glu)-, bovine pancreatic polypeptide(Bpp)-, cholecystokinin(Cck)-, secretin(Sec)-, motilin(Mot)-, glicentin(Gli)-, gastric inhibitor polypeptide(GIp)-, substance P(Sp)-, and neurotensin(Neu)-immunoreactive cells, were identified by immunohistochemical method. Gas-, Som-, 5-HT-, Glu-, Cck-, Sec-, Mot-, Gli-, Sp-, and Neu-reactive cells were observed in the duodenum, and among these Gas- and 5-HT-reactive cells were moderately found while the others were infrequent. In the jejunoileum Gas-, Som-, 5-HT-, Glu-, Cck-, Mot-, Gli-, GIp-, SP-, and Neu-reactive cells were found, and among these 5-HT- and GIp-reactive cells were moderately found while the others were infrequent. In the colon Sec-reactive cell was not detected. 5-HT-reactive cells were found most frequently and the others were infrequent in this region. 5-HT-, Bpp-, GIi- and Neu-reactive cells were found in the rectum. Among these 5-HT-reactive cells were found most frequently. 3. Electron microscopically, five types of endocrine cells, EC, ECL, D, G, A-like cell, were identified in the intestinal region. EC and ECL cells in the duodenum, EC, D and G cells in the jejuno-ileum, EC and A-like cells in the colon and EC cell in the rectum were observed respectively.

Nucleotide Sequence of Leghemoglobin cDNA from Canavalia lineata

  • Poly(A)+ RNA was selected from Canavalia lineata root nodule RNA through oligo(dT) cellulose column and used for construction of a cDNA library using λgt10-EcoRI arms. The size of the library was 7.2$\times$105 pfu/mL. A full length leghemoglobin (Lb) cDNA clone, pCILb1(687 bp) isolated with soybean Lb probe, contained one open reading frame (ORF) of 447 bp with 54 bp plus 186 bp at 5' and 3' untranslated region, respectively. A consensus sequence of plant translation start region (AAAATGGG) was found at 5' untranslated region, and two polyadenylation-related sequence (AATAAA, AATAAG) and a conserved motif between them (gACTTGTT) were found upstream of poly(A)+ tail consisted of 13 (A)s at 3' untranslated region. The ORF encoded a polypeptide consisted of 149 amino acids with a molecular weight of 16.2 kD. Deduced amino acid sequences showed high degree of homology values with those of other Lbs ranging from 66% (Casuarina glauca) to 85% (Glycine max).

  • Several parameters of u -glycerol-3-pholphate dehydrogenase (GPDH) such as activity, content and translatable mRNA levels were measured to elucidate mechanism underlving developmental and tissue specific regulation of 6PDH activity in Drosophila melonogastrr. In adult segments, most of total GPDH activity (62%1 Iwas detected in thorax where GPDH-1 resided, while 32% of total GPDH aUiviD was only detected in abdomen where GPDH-3 resided. The relative synthesis of GPDH was, however, similar in both tissues, although 58% of total GPDH was synthesized in abdomen. These results strongly suggest that the turnover rate of the abdominal enzyme (GPDH-3) was much more rapid than that of thoracic enzymes (GPDH-1). In nitro translation and immunoblotting experiments also indicate that GPDH-3 was arised by posttranslational modification from a single polypeptide (GPDH-1).

  • 돌연변이유기를 통한 유용견전형질의 해조류품종을 개발하기 위해 화학적돌연변이유발원인 Ethyl-methanesulphonate(EMS)에 대한 구멍갈파래(Ulva pertusa Kjellman) 엽체의 감수성을 검토하여 효과적인 돌연변이유기조건을 찾고자 하였다. EMS를 처리한 배지에 엽체를 생장시키며 변이를 유발시키고자 하였을 때, $0.05{\%}$$0.025{\%}$의 EMS 처리구에서 각각 $100{\%}$와 약 $20{\%}$의 생장억제가 일어났다. 한편 고농도의 EMS액에 엽체를 일정시간 침적한 후 정상배양함으로서 변이를 유발코자 했을 때, $1.0{\%}$ EMS액에 40분과 $0.5{\%}$ EMS액에 80분 처리한 경우 각각 $100{\%}$와 약 $10{\%}$의 생장억제가 일어났다. EMS 처리된 엽체는 생장형과 체색이 모체와 전혀 다른 변이체로 나타나기도 하였는데 이들의 polypeptide 일부에 변화가 있음이 SDS-PAGE로 조사되었다.

  • Cephalosporin-C deacetylase (CAH) catalyzes the deacetylation of cephalosporin derivatives. A novel gene encoding the CAH from Bacillus sp. KCCM10143 was cloned and sepuenced. The uncleotide sequence contained an open reading frame encoding a polypeptide consisting of 217 amino acids and a molecular weight of 24 kDa which was in good agreement with the value obtained by sodium dodecylsulfate-polyacrylamide gel electrophoresis. An expression plasmid was constructed by inserting the CAH gene into the region of the pTrc99A expression vector. An active from of the CAH protein was expressed in the soluble fraction obtained after cell disruption. in fermentation using a 5-1 jar fementer, the transformant E. coli JM109 (pDST654) produced 4.12 U of CAH per ml of culture during 16 h of incubation.

  • 유구낭미충(이하 낭미충)(Cysticercus cellulosae)에서 추출한 조항원으로 낭미충증(cysticercosis), 스파르가눔중(sparganosis) 및 포충중(hydatidosis) 환자 및 정상인 혈청내 IgG 항체와의 혈청학적 반응을 ELISA 및 EITB 방법으로 추적하였다. ELISA에서 낭미충의 조항원 단백은 스파르가눔중 환자 혈청과의 교차 반응도 보였고, 포충증 환자 혈청에 대해서는 낭미충 환자 혈청보다 오히려 높은 흡광도(O.D.)를 보였다. 낭미충 조항원을 SDS-PAGE로 전기영동한 결과 260 KDa∼22 KDa 범위에서 31개의 polypeptide band를 얻었으며 이 중 11개의 분획이 강한 염색성을 나타내었다. 낭미충 조항원과 낭미충중 환자 혈청과의 EITB 반응에서 22개의 항원대가 관찰되었으며 이 중 104 KDa, 82 KDa, 78 KDa 및 59 KDa는 모든 검사 혈청에서 인지되었다. 또 22개의 항원대 중 12개는 정상인 혈청에서도 반응이 관찰되어 교차반응하는 비특이성 항원 성분으로 해석되었다. 낭미충 조항원과 스파르가눔증 환자 혈청과의 반응에서 11개의 항원대가 인지되었으며 이 중 낭미충증 환자 혈청과 교차반응하는 9개의 항원대가 인지되었다. 나머지 163 KDa 및 131 KDa 항원대는 스파르가눔 환자에서만 동정되었다. 낭미충 조항원과 포충증 환자 혈청과의 반응에서는 21개의 항원대가 인지되었는데, 이 중 63 KDa는 포충증 환자에서만 인지되었고 나머지는 낭미충증 환자 혈청과 교차반응하는 항원대이었다. 한편 낭미충 조항원 중 104 KDa, 82 KDa, 72 KDa, 59 KDa및 34 KDa는 낭미충증, 스파르가눔증 및 포충중 환자와 정상인의 혈청에서도 모두 교차반응을 일으키는 항원이었다.

Functional Implications in Apoptosis by Interferon Inducible Gene Product 1-8D, the Binding Protein to Adenovirus Preterminal Protein

  • Adenovirus (Ad) precursor to the terminal protein (pTP) plays an essential roles in the viral DNA replication. Ad pTP serves as a primer for the synthesis of a new DNA strand during the initiation step of replication. In addition, Ad pTP forms organized spherical replication foci on the nuclear matrix (NM) and anchors the viral genome to the NM. Here we identified the interferon inducible gene product 1-8D (Inid) as a pTP binding protein by using a two-hybrid screen of a HeLa cDNA library. Of the clones obtained in this assay, nine were identical to the Inid, a 13-kDa polypeptide that shares homology with genes 1-8U and Leu-13/9-27, most of which have little known functions. The entire open reading frame (ORF) of Inid was cloned into the tetracycline inducible expression vector in order to determine the biological functions related with adenoviral infection. When Inid was introduced to the cells along with adenoviruses, fifty to sixty percent of Ad-infected cells expressing Inid had rounded morphology, which was suggestive of apoptosis. Results from the terminal deoxynucleotidyl transferase (TdT) and DNA fragmentation assays confirmed that Inid induces apoptosis in Ad-infected or in uninfected cells. The Inid binding to pTP may target the cell for apoptotic destruction as a host defense mechanism against the viral infection.
