Isolation of Intestinal Glucose Uptake Inhibitor from Punica granatum L.

  • Inhibition of intestinal glucose uptake is beneficial in reducing the blood glucose level for diabetes. To search for an effective intestinal glucose uptake inhibitor from natural sources, 70 native edible plants, fruits and vegetables were screened using Caco-2 cells and fluorescent D-glucose analog 2-[N-(7-nitrobenz-2-oxa-1,3-diazol-4-yl) amino]-2-deoxy-D-glucose (2-NBDG). A compound that was able to inhibit glucose uptake was isolated from methanol extract of Punica granatum L. and called PG-1a. PG-1a appears to be a phthalic acid-diisononyl ester- like compound (PDE) with molecular weight of 418. The inhibitory effect of PG-1a on intestinal glucose uptake was dose-dependent with 89% inhibition at $100\;{\mu}g$/mL. Furthermore, the intestinal glucose uptake inhibitory effect of PG-1a was 1.2-fold higher than phlorizin, a well known glucose uptake inhibitor. This study suggests that PG-1a could play a role in controlling the dietary glucose absorption, and that PG-1a can effectively improve the diabetic condition, and may be used as an optional therapeutic and preventive agent.

Phelligridin D maintains the function of periodontal ligament cells through autophagy in glucose-induced oxidative stress

  • Purpose: The objective of this study was to investigate whether phelligridin D could reduce glucose-induced oxidative stress, attenuate the resulting inflammatory response, and restore the function of human periodontal ligament cells (HPDLCs). Methods: Primary HPDLCs were isolated from healthy human teeth and cultured. To investigate the effect of phelligridin D on glucose-induced oxidative stress, HPDLCs were treated with phelligridin D, various concentrations of glucose, and glucose oxidase. Glucose-induced oxidative stress, inflammatory molecules, osteoblast differentiation, and mineralization of the HPDLCs were measured by hydrogen peroxide (H2O2) generation, cellular viability, alkaline phosphatase (ALP) activity, alizarin red staining, and western blot analyses. Results: Glucose-induced oxidative stress led to increased production of H2O2, with negative impacts on cellular viability, ALP activity, and calcium deposition in HPDLCs. Furthermore, HPDLCs under glucose-induced oxidative stress showed induction of inflammatory molecules (intercellular adhesion molecule-1, vascular cell adhesion protein-1, tumor necrosis factor-alpha, interleukin-1-beta) and disturbances of osteogenic differentiation (bone morphogenetic protein-2, and -7, runt-related transcription factor-2), cementogenesis (cementum protein-1), and autophagy-related molecules (autophagy related 5, light chain 3 I/II, beclin-1). Phelligridin D restored all these molecules and maintained the function of HPDLCs even under glucose-induced oxidative stress. Conclusions: This study suggests that phelligridin D reduces the inflammation that results from glucose-induced oxidative stress and restores the function of HPDLCs (e.g., osteoblast differentiation) by upregulating autophagy.

  • 경골어류 4종(조피볼락, 용치놀래기, 송곳니베도라치 및 졸복) 장관의 복합당질 성상의 차이를 밝히기 위해 장을 네 부위로 나누워 아홉 종류의 biotinylated lectin들(DBA, SBA, PNA, BSL- I , RCA- I , sWGA, UEA- I 및 Con A)로 반응시켰다. 조피볼락을 제외하면 부위에 따른 차이는 없었다. 용치놀래기 장 상피는 모든 부위에서 D-glucose/mannose 잔기를 함유하고 있었다. 조피볼락, 송곳니베도라치 및 졸복 장 모든 부위에서는 정도의 차이는 있지만 ${\beta}$-N-acetyl-D-gal-actosamine가 함유되어 있었다. 또한 조피볼락의 근위장, 중간장 및 원위장에는 galactose-${\beta}$-1,3-N-acetyl-D-galactosamine 및 ${\alpha}$-D-galactose도 함유되어 있었으나 직장에는 이들 당 잔기들이 함유되어 있지 않았다. 송곳니베도라치에는 ${\beta}$-N- acetyl-D-galactosamine 외에 ${\beta}$-N-acetyl-D-glucosamine 및 D-glucose/mannose도 발견되었으며, 졸복에서는 Galactose-${\beta}$-1,3-N-acetyl-D-galactosamine, D-galactose 및 D-glucose/mannose도 발견되었다.

금식 여부에 따른 혈당치 분포와 당뇨병 선별을 위한 혈당조절지표의 평가 (The Assessment of Blood Glucose Distribution according to the Fasting State and Glycemic Control Indicators for Diabetes Screening)

  • 본 연구는 금식 여부에 따른 혈당치 분포 차이를 분석하였고 공복 혈당치와 혈당조절지표인 당화혈색소, 프록토사민, 1,5-AG의 상관성을 분석하여 전당뇨병 선별에 필요한 추가 정보를 제공하고자 하였다. 단국대학교병원에 내원한 707명의 외래 환자를 금식 군과 비금식 군으로 나누어 혈당 검사를 시행한 후 각 군의 혈당치 평균을 산출하고 성별, 연령별 및 임상과 별 결과치 분포를 분석을 하였다. 또한 건강검진을 목적으로 내원한 금식 상태의 153명에 대해 혈당치와 당화혈색소, 프록토사민, 1,5-AG을 측정하여 혈당치와 혈당조절지표의 상관성을 평가하였다. 비금식 군의 혈당 평균치는 111.9 mg/dL, 금식 군은 103.6 mg/dL로서 유의한 차이가 있었고 (p<0.05) 평균치 차이는 여자가 4.8 mg/dL로서 남자의 12.2 mg/dL보다 적었다. 연령대에 따른 중앙값의 차이는 비금식 군에서만 유의한 차이가 있었고(Kruskal-Wallis test, p<0.01) 금식 군에서는 유의하지 않았다. 공복 시 포도당 장애 선별 구간에서 혈당 농도와 상관성이 높은 검사 종목은 1,5-AG로 평가되었다. 저자들은 금식 여부에 따른 혈당치 분포의 차이를 외래 환자를 대상으로 분석하였으며, 건강검진 대상자의 결과치 분석을 통해 1,5-AG가 프록토사민, 당화혈색소보다 공복 혈당치와 유의한 상관성이 있는 것으로 평가하였다. 당뇨병 선별 시 혈당 측정과 함께 단기 혈당 조절능력을 반영하는 지표가 활용될 수 있을 것으로 사료된다.

Guar gum을 첨가한 쑥설기가 Type-ll 당뇨환자의 혈액성분에 미치는 영향 (Effects of Guar Suksolgi on the Blood Glucose and Lipids in Type-ll Diabetic Subjects)

  • To determine the effects of guar suksolgi on blood glucose and lipids in type-ll diabetic subjects, a piece of guar suksolgi(36.3g, 54Kcal) was administered to eight patients for 3 weeks every meal. No significant differences occured indietary intakes and body weight before and after the treatment. Fasting blood glucose levels were decreased from 132.38mg/dI to 114.75mg/dI after the treatment, but not statistically significant. Blood TG levels were increased from 159.13mg/dI to 175.00mg/dI after the treatment, but not statistically significant. Excluding one patient who had extremely high TG level, blood TG levels tended to be decreased from 148.00mg/dI to 121.00mg/dI. TC LDL-c, HDL-c levels were decreased after the treatment, but not statistically significant. HbAIC concentrations were decreased from 8.54mg/kI to 7.80mg/dI after the treatment, but not statistically significant. In the case of three patients who had had normal fasting blood glucose levels, blood glucose levels tended to be decreased at postprandial 30, 60 minutes, and blood insulin levels tended to be decreased at postprandial 30, 60, 90, 120 minutes, although none of the levels were statistically significant. Therefore, if guar suksolgi is adinistered to type-ll diabetic subjects being more hyperglycemic than our patients, their blood glucose and lipids will be decreased significantly.

Effect of Glucose on Listeria monocytogenes Survival under Sequential Sublethal Stresses of Gamma Irradiation and NaCl

  • This study evaluated glucose effect on Listeria monocytogenes survival under gamma irradiation and NaCl stress. L. monocytogenes in phosphate buffered saline (PBS) plus glucose (0-4%) was treated with gamma irradiation (0-0.5 kGy), and the samples were then exposed to NaCl (0-9%) in tryptic soy agar plus 0.6% yeast extract. $D_{10}$ and $t_{3D}$ values were determined, and a model for prediction of $D_{10}$ values was developed. Cell counts of L. monocytogenes reduced as irradiation dose increased, and L. monocytogenes in PBS (no glucose) was more sensitive to irradiation and NaCl compared to those in PBS (2 or 4% glucose). $D_{10}$ values were 0.07-0.1, 0.12-0.16, and 0.13-0.15 kGy for 0, 2, and 4% glucose, respectively. The $t_{3D}$ values were 0.22-0.3 (0% glucose), 0.35-0.48 (2% glucose), and 0.40-0.44 (4% glucose). A model performance was acceptable. These results indicate that glucose in foods would increase the resistance of L. monocytogenes to gamma irradiation and NaCl stress.

한국산 마의 당질 분석 (Carbohydrates Analyses of Korean Yam(Dioscorea) Tubers)

  • 한국산 마의 주요 품종인 D. batatas, D. aimadoimo 및 D. japonica 등 3가지 품종의 일반성분을 분석하여 비교하였고 각 시료들을 동결건조하여 분말로 만든 다음 분획하여 HPLC에 의한 탄수화물의 조성과 함량, DNS법에 의한 환원당을 정량하였다. 탄수화물 중 유리당을 구성하는 주된 조성은 fructose, glucose, sucrose 및 maltose 등이었고 이 중 D. batatas에서는 glucose와 sucrose, D. aimadoimo와 D. japonica에서는 fructose와 sucrose가 가장 많은 부분을 차지하고 있었다. 총당은 주로 mannose와 glucose로 구성 되었고 3가지 품종 모두 mannose가 80% 이상으로, 이는 마가 감자나 고구마보다 끈끈한 점질물로 알려진 mannan이 많기 때문인 것으로 생각된다. Soluble fraction과 전체 마에서의 환원당 정량결과 soluble fraction에서 당함량은 D. aimadoimo가 가장 많았고, insoluble fraction에서 당함량은 D. japonica가 가장 많았다.

심혈관 수술 신생아의 혈당수준에 따른 환아 결과 (Patient Outcomes according to Blood Glucose Level in Neonates with Cardiovascular Surgery)

  • Purpose: The purpose of this study was to provide basic data for suitable neonate blood glucose maintenance by investigating and analyzing the blood glucose level of post cardiovascular surgery neonates in the ICU for the effect of blood glucose levels on the following outcomes; ICU stay, hospital stay, mechanical ventilation time, morbidity, and mortality. Methods: The participants were 143 neonates in the ICU after having had cardiovascular surgery. The design for this study was an investigation of the blood glucose levels of the neonates and retrospective analysis of patient outcomes according to blood glucose level. Results: The results for the neonate groups showed that the factors of hospital stay, ICU stay, mechanical ventilation time and mortality, for the group with a blood glucose level over 140 mg/dL were longer and higher than for the group with blood glucose of less than 100 mg/dL or the group between 100-139 mg/dL. Conclusion: The results of this study indicate that when caring for neonates after cardiovascular surgery, it is important to recognize the influence of blood glucose levels on patient outcomes like hospital days, ICU stay, length of time on mechanical ventilation and mortality. Further, care guidelines for neonates' glucose level management need to be developed.

Evaluation of three glucometers for whole blood glucose measurements at the point of care in preterm or low-birth-weight infants

  • Purpose: We evaluated three blood glucose self-monitoring for measuring whole blood glucose levels in preterm and low-birth-weight infants. Methods: Between December 1, 2012 and March 31, 2013, 230 blood samples were collected from 50 newborns, who weighed, ${\leq}2,300g$ or were ${\leq}36$ weeks old, in the the neonatal intensive care unit of Eulji University Hospital. Three blood glucose self-monitoring (A: Precision Pcx, Abbott; B: One-Touch Verio, Johnson & Johnson; C: LifeScan SureStep Flexx, Johnson & Johnson) were used for the blood glucose measurements. The results were compared to those obtained using laboratory equipment (D: Advia chemical analyzer, Siemens Healthcare Diagnostics Inc.). Results: The correlation coefficients between laboratory equipment and the three blood glucose self-monitoring (A, B, and C) were found to be 0.888, 0.884, and 0.900, respectively. For glucose levels ${\leq}60mg/dL$, the correlation coefficients were 0.674, 0.687, and 0.679, respectively. For glucose levels>60 mg/dL, the correlation coefficients were 0.822, 0.819, and 0.839, respectively. All correlation coefficients were statistically significant. And the values from the blood glucose self-monitoring were not significantly different from the value of the laboratory equipment, after correcting for each device's average value (P>0.05). When using laboratory equipment (blood glucose ${\leq}60mg/dL$), each device had a sensitivity of 0.458, 0.604, and 0.688 and a specificity of 0.995, 0.989, and 0.989, respectively. Conclusion: Significant difference is not found between three blood glucose self-monitoring and laboratory equipment. But correlation between the measured values from blood glucose self-monitoring and laboratory equipment is lower in preterm or low-birth-weight infants than adults.

Efficacy and safety of Panax ginseng berry extract on glycemic control: A 12-wk randomized, double-blind, and placebo-controlled clinical trial

  • Background: Antihyperglycemic effects of Panax ginseng berry have never been explored in humans. The aims of this study were to assess the efficacy and safety of a 12-wk treatment with ginseng berry extract in participants with a fasting glucose level between 100 mg/dL and 140 mg/dL. Methods: This study was a 12-wk, randomized, double-blind, placebo-controlled clinical trial. A total of 72 participants were randomly allocated to two groups of either ginseng berry extract or placebo, and 63 participants completed the study. The parameters related to glucose metabolism were assessed. Results: Although the present study failed to show significant antihyperglycemic effects of ginseng berry extract on the parameters related to blood glucose and lipid metabolism in the total study population, it demonstrated that ginseng berry extract could significantly decrease serum concentration of fasting glucose by 3.7% (p = 0.035), postprandial glucose at 60 min during 75 g oral glucose tolerance test by 10.7% (p = 0.006), and the area under the curve for glucose by 7.7% (p = 0.024) in those with fasting glucose level of 110 mg/dL or higher, while the placebo group did not exhibit a statistically significant decrease. Safety profiles were not different between the two groups. Conclusion: The present study suggests that ginseng berry extract has the potential to improve glucose metabolism in human, especially in those with fasting glucose level of 110 mg/dL or higher. For a more meaningful benefit, further research in people with higher blood glucose levels is required.
