We have previously demonstrated that the redox reactant pyruvate prevents apoptosis in the oxidant model of bovine pulmonary artery endothelial cells (BPAEC), and that the anti-apoptotic mechanism of pyruvate is mediated in part via the mitochondrial matrix compartment. However, cytosolic mechanisms for the cytoprotective feature of pyruvate remain to be elucidated. This study investigated the pyruvate protection against endothelial cytotoxicity when the glycolysis inhibitor 2-deoxy-D-glucose (2DG) was applied to BPAEC. Millimolar 2DG blocked the cellular glucose uptake in a concentration- and time-dependent manner with >85% inhibition at $\geq$5 mM within 24 h. The addition of 2DG evoked BPAEC cytotoxicity with a substantial increase in lipid peroxidation and a marked decrease in intracellular total glutathione. Exogenous pyruvate partially prevented the 2DG-induced cell damage with increasing viability of BPAEC by 25-30%, and the total glutathione was also modestly increased. In contrast, 10 mM L-lactate, as a cytosolic reductant, had no effect on the cytotoxicity and lipid peroxidation that are evoked by 2DG. These results suggest that 2DG toxicity may be a consequence of the diminished potential of glutathione antioxidant, which was partially restored by exogenous pyruvate but not L-lactate. Therefore, pyruvate qualifies as a cytoprotective agent for strategies that attenuate the metabolic dysfunction of the endothelium, and cellular glucose oxidation is required for the functioning of the cytosolic glutathione/NADPH redox system.
Cup-plant (Silphium perfoliatum L.) has potential to produce high biomass and highly digestible forage in the wetlands where other productive forages do not grow or produce well. However, high moisture content at harvest is a considerable disadvantage of cup-plant for the production of high quality silage. This study was conducted to determine the effect of moisture content on the characteristics of cup-plant silage. Harvested cup-plant was ensiled in farm scale plastic bag silos and laboratory silos. In the plastic bag silos, first growth (FG) and regrowth (RG) cup-plant was harvested, wilted and ensiled. Dry matter content of FG and RG was 280 g/kg and 320 g/kg after 48 hr of wilting. The silage made with FG had pH 5.3 and 5.63 g/kg DM of acetate as a major volatile fatty acid. The composition of lactate, butyrate and acetate production was 1.0: 0.9: 2.3. The pH of silage made with RG was 4.5 and lactate was a major fermentation end product (16.8 g/kg DM). In the laboratory silos, wilted and unwilted first growth cup-plant material was ensiled to compare the early fermentation end products at days 2, 4, 11, and 40. Wilting increased dry matter content by 42% in the harvested material. Wilted silage showed about one unit lower pH until day 11. The contents of ammonia nitrogen and acetate were higher in un wilted silage, while that of lactate was higher in wilted silage (p<0.05). Butyrate and propionate were not detected in the wilted silage until day 40. We conclude from the results that moisture control is essential for the production of high quality cup-plant silage and high pH of cup-plant silage is due to low concentrations of fermentation end products.

The present study was aimed to estimate the effect of ensiling period and bacterial inoculants on chemical compositions and fermentation characteristics on rye silage harvested at delayed stage. Rye (Secale cereale L.) was harvested after 20 days of heading stage (29.4% dry matter, DM). The harvested rye forage was applied with different inoculants following: applications of distilled water (CON), Lactobacillus brevis (LBB), Leuconostoc holzapfelii (LCH), or mixture of LBB and LCH at 1:1 ratio (MIX). Each forage was ensiled into 20 L mini bucket silo (5 kg) for 50 (E50D) and 100 (E100D) days in triplicates. The E50D silages had higher in vitro digestibilities of DM (IVDMD, p<0.001) and neutral detergent fiber (IVNDFD, p=0.013), and lactate (p=0.009), and acetate (p=0.011) than those of E100D, but lower pH, lactic acid bacteria (LAB), and yeast. By inoculant application, LCH had highest IVDMD and IVNDFD (p<0.05), while MIX had highest lactate and lowest pH (p<0.05). The CON and LCH in E50D had highest LAB and yeast (p<0.05), whereas LBB in E100D had lowest (p<0.05). Therefore, this study concluded that LCH application improved the nutrient digesbility (IVDMD and IVNDFD) of lignified rye silage, and longer ensiling period for 100 days enhanced the fermentation characteristics of silage compared to ensiling for 50 days.
The ubiquitous plasma membrane $Na^+/H^+$ exchanger 1 (NHE1) is rapidly activated in response to various extracellular stimuli and maintains normal cytoplasmic pH. Yeast two-hybrid screening was used in order to identify proteins interacting with NHE1 using its cytoplasmic domain as a bait from HeLa cDNA library. One of the interacting cDNA clones was human Lactate dehydrogenase B (LDHB). In vitro translated LDHB was pulled down together with GST-NHE1.cd protein in the GST pull down assay, confirming the interaction in vitro. LDHB antibody immunoprecipitated endogenous LDHB together with NHE1 from H9c2 cells, validating cellular interaction between NHE1 and LDHB. Subsequent analysis revealed that the overexpression of LDHB increased intracellular PH, implying opening of the NHE1 transporter. Moreover, overexpression of LDHB activated caspase 3 and induced cell death, consistent with the expected phenotype of hyper-activation of NHE1. Collectively, our data indicate that LDHB modulates NHE1 activity via physical interaction.
This study was designed to investigate the effects of sports drink including the extract from Prunus mume on the changes of oxygen uptake, minute ventilaton, heart rate, and blood lactate concentration during 1 hour treadmill running exercise corresponding to 75%VO$_2$max. Subjects were 16 male athletes(long distance runners and tennis players). The changing patterns of oxygen uptake and minute ventilation showed no significnat difference among all types of sports drink, but the minute vetilation of the placebo group showed a higher value than type I group during submaximal exercise. Although the changing patterns of heart rate and blood lactate concentration showed no significnat difference among all types of sports drink, but type E group showed a lower heart rate compared to Placebo group. And blood lactate concentration of placebo group showed a higher value compared to the value corresponding to lactate threshold during submaimal exercise. but the other types of sports drink showed a lower value of blood lactate concentration. Especially blood lactate threshold of type D and E groups showed a lower value (range from 1.44 to 2.00mM) during submaximal exercise. In these results, the sports drink including the extract from Prunus mume showed no significant effects to the changes of oxygen uptake, minute ventilaton, heart rate, and blood lactate concentration during 1 hour treadmill runnuing exercise corresponding to 75%VO$_2$max. But it could be suggested the positive effects of the intake of sports drink including the extract from Prunus mume on the inhibition of exercise-induced fatigue during submaximal exercise, because of the lower changing Patterns of heart rate, blood lactate concentration, and ventilation efficiency in this results.
Batch fermentations of lactic acid were performed with Lactobacillus casei to investigate the effect of pH on cell growth and production of lactic acid and by-products. Maximum productivity of lactic acid increased with increasing pH from 5.0 to 6.5, and the extent of D-lactate production was different at each pH. Acetate and D-lactate concentrations increased even after the complete consumption of glucose in the medium. While a pH range of 6.0-6.5 was optimal for cell growth and lactic acid production, superior results were achieved at pH 6.0 when both maximum lactic acid productivity and minimum by-product formation were considered.

Microbiologically influenced corrosion (MIC) is an electrochemical process where the participation of microorganisms initiates, facilitates, or accelerates the corrosion reaction. Sulfate-reducing bacteria (SRB) reduce sulfate to sulfide and are known to be the most destructive microorganisms in anaerobic MIC. Accordingly, the current study attempted to elucidate the mechanisms involved and the relative importance of the corrosive products in SRB-induced corrosion. The measured rate of anaerobic corrosion of iron coupons by Desulfovibrio desulfuricans was $89.9{\;}\mu\textrm{g}{\;}\textrm{m}^{-2}{\;}d^{-1}$. Direct contact between the cells and the iron coupon did not seem to be necessary for corrosion to occur, since the corrosion rate was similar ($100.8{\;}\mu\textrm{g}{\;}\textrm{m}^{-2}{\;}d^{-1}$) when the coupon was enclosed in a dialysis bag. The participation of sulfide in the corrosion process was only marginal, as the specific corrosion rate was 2.5 times higher in a sulfate-free pyruvate medium than in an $H_2S-producing$ lactate medium. Acetate (18.8-22.1 mM), the end-product of pyruvate and lactate metabolism, was identified in the culture medium and thus presumed to play a major role in the corrosion process involving Desulfovibrio desulfuricans.

In this study, we investigated the effect of silkworm, and its by-products on exercise endurance, muscle mass, and fatigue recovery using ICR mice model. Powders of silkworm, pupae, dongchunghacho, and silk powder were suspended in water and feed to mice for 2 weeks. The forced swimming time was increased by 4.3, 4.1 and 2.8 seconds after silkworm, pupae, and dongchughacho administration compared to DW fed group. Increases in exercise ability were achieved by difference mechanism according to feeding materials. Pupae increased muscle mass by 129% compared with the control group which may effect on elongation of swimming time. Dongchunghacho improved the plasma concentrations of fatigue markers such as creatinine, blood urea nitrogen (BUN), and D-lactate. Silkworm administration showed dual effect, the muscle mass induction (114% vs. control) and anti-fatigue (plasma creatinine, BUN, and D-lactate were 63, 75, and 78% vs. con) effect which contributed most elongated swimming time. In conclusion, silkworm and its by-products including pupae and dongchunghacho with the predominant protein and bioactive components improved muscle mass and showed anti-fatigue effect which could promote exercise performance ability.
Based on the potential beneficial effects of growth hormone releasing peptide (GHRP)-6 on muscle functions, a newly synthesized GHRP-6-biotin conjugate was tested on cultured myoblast cells. Increased expression of myogenic marker proteins was observed in GHRP-6-biotin conjugate-treated cells. Additionally, increased expression levels of insulin-like growth factor-1 and collagen type I were observed. Furthermore, GHRP-6-biotin conjugate-treated cells showed increased metabolic activity, as indicated by increased concentrations of energy metabolites, such as ATP and lactate, and increased enzymatic activity of lactate dehydrogenase and creatine kinase. Finally, binding protein analysis suggested few candidate proteins, including desmin, actin, and zinc finger protein 691 as potential targets for GHRP6-biotin conjugate action. These results suggest that the newly synthesized GHRP-6-biotin conjugate has myogenic stimulating activity through, at least in part, by stimulating collagen type I synthesis and several key proteins. The effect of a homolactic inoculant containing a blend of Lactobacillus plantarum, Pediococcus acidilactici and Enterococcus faecium or, the anionic surfactant, sodium dodecyl sulphate (SDS), alone or in combination on fermentation characteristics, aerobic stability and in situ DM, OM and NDF degradability of barley silage was investigated. Barley (Hordeum vulgare, L.) was harvested (45% DM), chopped and treated with water at 24 ml/kg forage (Control), inoculant at $1.09{\times}10^5$ cfu/g forage (I), SDS at 0.125% (wt/wt) of forage (S) or with the inoculant ($1.09{\times}10^5$ cfu/g) plus SDS (0.125% wt/wt; I+S). The treated forages were ensiled in triplicate mini silos and opened for chemical and microbiological analyses on d 1, 2, 3, 7, 14, 42 and 77. Silage samples from d 77 were opened and aerobically exposed for 7 d. The in situ rumen degradability characteristics of silage DM, OM and NDF were also determined. The terminal concentration of NDF in S and I+S was lower (p<0.001) than in other treatments. Lactate concentration was higher (p<0.001) and the rate and extent of pH decline were greater (p<0.001) in I and I+S than S and Control silages. A homolactic pathway of fermentation in I and I+S was evidenced by reduced (p<0.001) water-soluble carbohydrates concentration, higher lactate (p<0.01), lower acetate (p<0.01) and lower pH values (p<0.001) than in S and Control silages. All silages remained stable over 7 d of exposure to air as indicated by lower temperatures and moulds, and by non-detectable yeast populations. The treated silages had lower DM and OM degradability than in the Control but NDF degradation characteristics of I+S were improved compared to other treatments. It is concluded that the inoculant alone improved the fermentation characteristics whereas the combination of the inoculant with SDS improved both fermentation and NDF degradability of barley silage.
