방사선 방어작용이 있는 인삼 단백분획의 CHO-KI 세포에 대한 세포 독성 (Cytotoxic Effect of Radioprotective Ginseng Protein Fraction on CHO-KI Cells)

  • Radioprotective ginseng protein fraction was isolated from Korean white ginseng and its cytotoxic effect on CHO-K1 cells was studied by the method of measuring the relative cell survival and total cellular protein content (FRAME method). When ginseng protein at the dose of 300, 600, 900, $1200{\mu}g/ml$ was treated to cells for 24 hrs, the relative survival was significantly decreased at the concentration of above $600{\mu}g/ml$, indicating the presence of cytotoxic effect of the protein at certain concentration. When cellular protein content was measured after ginseng protein at the dose of 300, 600, 900, $1200\;{\mu}g/ml$ was treated, the amount of cellular protein was significantly reduced at the concentration above $600{\mu}g/ml$ in the case of 24 hr treatment and at all concentrations including $300{\mu}g/ml$ in the case of 72 hr treatment. The data suggest that the protein may inhibit cell growth, resulting in the reduction of live cells in culture. $ID_{50}$ value which is the concentration of ginseng protein that reduces the total cellular protein content to 50% of the control was calculated as 2276.86 and $1323.32\;{\mu}g/ml$ in groups treated for 24 and 72 hr, respectively. Since $ID_{50}$ value of above $1000{\mu}g/ml$ indicates very weak cytotoxicity, the ginseng protein seems to exert very weak cytotoxic effect on CHO-K1 cells.

Studies on Antimutagenic and Cytotoxic Effects of Seaweeds Protein-Polysaccharides

  • Polysaccharide content in protein-polysaccharides (PPS) extracted from sea mustard, sea tangle and fusiform were 40.61, 38.42 and 52.80% , respectively. 5% of sea tangle PPS showed highest inhibitory activity on 4-nitorquinoline -1-oxide(4-NQO) against Salmonella typhimurium TA100 compared to the other seaweed PPS. 5% of sea mustard PPS showed highest inhibition ration of 62% on 2-nitrofluorene(2-NF)against Salmonella typhimurium TA98. These PPS extracts showed cytotoxic activity against human colon cancer cell (SW-480), and showed mild cytotoxic activity on human stomach cancer cell(SNU-1) and human hepatic cancer cell(HepG 2).

  • The calyx extract of Campsis grandiflora displayed inhibitory activity against protein kinase C from the bovine brain. Separation guided by protein kinase C enzyme assay and bleb forming assay led to isolation of a potent protein kinase C inhibitor that was identified as a known phenylpropanoid glycoside, verbascoside. It suppressed completely bleb-formation of K562 cell surface induced by phorbol 12,13-dibutylate at the concentration of 60 $\mu\textrm{g}$/ml and IC$_{50}$ of the protein kinase C occured at 20 $\mu{M}$. This compound was tested for cytotoxic activity against ten human tumor cell lines in vitro. it exhibited moderate cytotoxic activity against skin tumor cell line M14 (IC$_{50}$ 2.2 $\mu\textrm{g}$/ml) and very weak cytotoxicity against other cell lines (IC$_{50}$>10 $\mu\textrm{g}$/ml)

  • Some studies of dandelion have been carried out on bioactivities, however, no comparative analysis on antioxidant and cytotoxic activities in the Korean dandelion (T. coreanum NAKAI.; KD) and dandelion (T. officinale WEB.) have been reported. In this study, the extracts of KD and dandelion analyzed relative phenolic contents and free radical scavenging, antioxidant enzyme and cytotoxic activities. The extract of Korean dandelion leaves (KDL) exhibited a higher phenolics content ($368{\pm}11.5\;mg/l00\;g$) and a strong free radical scavenging activity ($RC_{50}$value;87.89 ug/ml) than other parts and BHT (120.12 ug/ml), synthetic antioxidant. At ascorbate peroxidase (APX) activity, the dandelion root (DR) had a greater (38.8 U/mg protein) antioxidant enzyme compare to the dandelion leaves (12.2 U/mg protein). The catalase (CAT) and superoxide dismutase (SOD) followed higher enzyme activity in Korean dandelion root (KDR) than other parts. In a cytotoxic activity against human cancer cell, the extracts of KDR was found to be active against Calu-6, HCT-116, and SNU-601 cell lines, with $IC_{50}$ values of 522.34, 532.74 and 614.85 ug/ml, respectively. These results suggest that KD and dandelion would be an alternative antioxidant source, based on natural plant resources.

Induction of apoptosis by protein fraction isolated from the leaves of Mirabilis jalapa L on HeLa and Raji cell-line

  • The leaves of Mirabilis jalapa L contains protein fraction presumed ribosome-inactivating protein (RIP). RIP is a group of protein that has RNA N-glycosidase activity that is capable to inhibit protein synthesis. Protein fraction of the plant was shown to be cytotoxic on HeLa cell-line, however, the mechanism by which the protein kill the cells is not identified yet, whether trough apoptosis, necrosis, or other mechanism. This research aim to study the mechanism of cell death caused by the protein fraction isolated from the leaves of this plant on HeLa and Raji cell-line, as representative of different kind of cancer cells. Results showed that protein fraction isolated from the leaves of Mirabilis jalapa L was more cytotoxic to HeLa cell-line (LC50: 0.65 mg/ml) than to Raji cell-line (1.815 mg/ml) on 48 hours incubation time. Moreover, it was demonstrated that the death of HeLa cells caused by the protein fraction was due to induction of apoptosis, while on Raji cell-line was due to non-apoptosis way, presumably via necrosis.

TAGLN2-mediated actin stabilization at the immunological synapse: implication for cytotoxic T cell control of target cells

  • Actin dynamics is critical for the formation and sustainment of the immunological synapse (IS) during T cell interaction with antigen-presenting cells (APC). Thus, many actin regulating proteins are involved in spatial and temporal actin remodeling at the IS. However, little is known whether or how actin stabilizing protein controls IS and the consequent T cell functions. TAGLN2 − an actin-binding protein predominantly expressed in T cells − displays a novel function to stabilize cortical F-actin, thereby augmenting F-actin contents at the IS, and acquiring leukocyte function-associated antigen-1 activation following T cell activation. TAGLN2 also competes with cofilin to protect F-actin in vitro and in vivo. During cytotoxic T cell interaction with cancer cells, the expression level of TAGLN2 at the IS correlates with the T cell adhesion to target cancer cells and production of lytic granules such as granzyme B and perforin, thus expressing cytotoxic T cell function. These findings identify a novel function for TAGLN2 as an actin stabilizing protein that is essential for stable immunological synapse formation, thereby regulating T cell immunity. Association of Killer Cell Ig-like Receptor (KIR) with an Adaptor Protein Shc

  • Background: Cytotoxic function of killer cells is inhibited by specific recognition of class I MHC molecules on target cells by inhibitory killer Ig-like receptors (KIR) expressed on NK cells and some cytotoxic T cells. The inhibitory effect of KIR is accomplished by recruitment of SH2-containing protein tyrosine phosphatase (SHP) to the phosphotyrosine residues in the cytoplasmic tail. Methods: By in vitro coprecipitation experiments and transfection analysis, we investigated the association of KIR with an adaptor protein Shc in Jurkat T cells. Results: The cytoplasmic tail of KIR appeared to associate with an adaptor protein Shc in Jurkat T celilysates. Similar in vitro experiments showed that phosphorylated KIR cytoplasmic tail bound SHP-1 and Shc in Jurkat T cell lysates. The association of KIR with Shc was further confirmed by transfection analysis in 293T cells. Interestingly, however, Shc appeared to be replaced by SHP-2 upon engagement of KIR in 293T cells. Conclusion: Our data indicate that KIR associate with an adaptor protein Shc in Jurkat T cells, and suggest that KIR might have an additional role which is mediated by this adaptor protein.

  • The aim of this study was to investigate the cytotoxic effect and its mechanism on Radix Aconiti(RA) extract in lung cancer cell lines. RA extract treatment decreased the cell viability in a dose-dependent fashions in lung cancer cells including A549, H460, H23 and H157 cells. Many investigators reported that A549 and H460 cells expressed wild-type p53, but H23 and H157 cells preserved mutated p53. After treatment with RA extract in A549 and H460 cells, we measured the expression of p53 protein levels using Western blot. analysis. In both cells treated with RA extracts, p53 protein expressions were increased in a dose-dependent manner. In our experiments, RA extracts also have cytotoxic effects in H23 and H157, which have mutated p53. Treatment with RA extract decreased bcl-2 protein expressions in both cells. These results suggest that RA extracts have cytotoxic effects via p53 expression increase and bcl-2 inhibitable pathways in A549, H460 cells and H23, H157 cells, respectively.

  • The cytotoxic activities of Pinellia ternate, Aconitum carmichaeli, Arisaema amurense, Aconitum kusnezoffii and Scutellaria baicalensis on monkey kidney cell (Vero) and human liver cell (WRL68) were evaluated by Sulforhodamine B Protein (SRB) and Tetrazolium-based (MTT) colorimetric assay methods The results were as fellows : 1 The Pinellia ternate and Arisaema amurense did not show the cytotoxic activities at any concentration without processing or natural drugs. 2 The extracts of Aconitum carmichaeli, Aconitum kusnezoffii and Scutellaria baicalensis showed cytotoxic activities. However, the cytotoxic activities of processing drugs were less effective than the natural drugs. 3. The cytotoxic activities on monkey kidney cell (Vero) and human liver cell (WRL68) of Aconitum carmichaeli was determined by MTT assay. The Kyungpo(京?) of Aconitum carmichaeli showed less concentrate than that of the Dangpo(唐?). The $IC_{50}$ value on monkey kidney cell (Vero) and human liver cell (WRL68) of Kyungpo was $937{\pm}29\;and\;731{\pm}31{\mu}g/ml$, respectively 4. The cytotoxicity on monkey kidney cell (Vero) and human liver cell (WRL68) of Scutellaria baicalensis showed strong activities without processing or natural drugs.
