Cytotoxic Quassinoids from Simaba cedron

  • During a survey of new antitumor substances from higher plants, we have found that the crude extract of Simaba cedron Planchon (Simaroubaceae) showed cytotoxic activity (IC$\sub$50/ 0.7 $\mu\textrm{g}$/mL) against P388 leukemia cells. Activity-guided chromatographic purification using P388 cells led to the isolation of five novel quassinoids, cedronolactones A-E (1-5) and nine known quassinoids, simalikalactone D (6), chaparrinon (7), chaparrin (8), glaucarubolone (9), glaucarubol (10), samaderine Z (11), guanepolide (12), ailanquassin A (13), and polyandrol (14). In this seminar, the structural elucidation of 1-5 and the cytotoxic activity of the isolated compounds are discussed.

  • Ginseng leaf/stem extract produced by subcritical water extraction at high temperature ($190^{\circ}C$) posses higher cytotoxic activity against human cancer cell lines than ethanol extract. Subcritical water extraction can be a great candidate for extraction of functional substance from ginseng leaves/stems.

  • Comparative cytotoxic activities of petroleum ether soluble fraction from various ginsengs of Panax species were evaluated using A549 (human lung adenocarcinoma) and SK-OV-3(human ovary carcinoma) cancer cell lines. Korean red ginseng, Korean white ginseng, American ginseng and Canadian ginseng were found to show more potent cytotoxicitles on A549 and SK-OV-3 cell lines than Chinese red ginseng, Japanese red ginseng and Sanchi ginseng. It is noteworthy that especially, red ginseng prepared from the root of Panax ginseng cultivated in Korea shows relatively stronger cytotoxic activities than those cultivated in China and Japan.

  • Some known coumarins, decursin, nodakenetin, umbelliferone, 7-demethylsuberosin, columbianetin, decursinol angelate and decursinol, showing significant cytotoxic activities against P388 cell lines,were isolated from the roots of Angelica gigas (Umbelliferae) . 7-Demethylsuberosin and columbianetinwere obtained from Angelica gigas for the first time. Chernotaxonornic difference about coumarins com-ponents between the roots of Angelica gigas and those of A. acutiloba is also discussed.

  • A minor cytotoxic compound was isolated by bioassay-guided fractionation from Asiasari Radix and identified as aristolactam III(1) on the basis of spectral data and chemical evidence. This is the first report on the isolation of compound 1 from Asiasarum genus. Compound 1 exhibited a significant cytotoxic activity against the three kinds of human cancer cell lines (A 549, SK-MEL-2 and SK-OV-3).

  • A cytotoxic coumarin against L1210 cell was isolated from the unripe fruit of Poncirus trifoliata ($ED_{5}$) = 10.2 $\mu$ g/ml. Its structure was identified as aurapten, 7-geranyloxycoumarin. Hydrolysis of the substance gave umbelliferone and geraniol. Only geraniol showed the cytotoxic activity ($ED_{53}$ = 6.5 $\mu$g/ml) while umbelliferone and its methyl or allyl derivatives were not active.

  • Natural killer (NK) cells are a heteroguneous subpopulation of lymphocytes that spontaneously exhibit cytotoxic activity against various virus-Infected and neoplastic target cells without prior exposure to a specific antigen. It was thought that NK calls play an important role in immunosurvrillanre against viral agents and tumors, and in prevention of metastasis. Recently, several reports have indicated evidence that ginseng extracts show a significant stimulatory effect on the humoral and cellular immune responses. This evidence gives support to the suggestion that the anticarcinogenic effect of ginseng may be due to the effect of ginseng on the immunological system. Treatment with total, diol, and triol saponin resulted in an increase in NK cytotoxic activity, but no enhancement of the lytic activity due to the natural killer cytotoxic factor (NKCF). Therefore, these results suggest that the augmentation of NK activity by ginseng saponin fractions may not be due to the activation of NKCF lytic activity.
