The comparative analysis of smoke components delivered from cigarette papers manufactured by flax and wood pulp (궐련지의 섬유종류가 담배 연기성분에 미치는 영향)

  • This study was conducted to evaluate the effect of cigarette papers, flax and wood, on the delivery of mainstream smoke. The main components of cigarette papers were cellulose, hemicellulose, lignin, and pectin. Lignin contents, known as precursor of smoke's phenolic compounds, of the flax and wood cigarette papers were 5.8% and 10.6%, respectively. The pyrolysis products of cigarette papers were similar by the profile of total ion chromatogram. But, the area % of some components, such as 1,3-cyclopentanedione, 3,5- dimethyl cyclopentane-1,2-dione, 2-hydroxy-3-methyl-2-cyclopentenone, dihydro-2(3H)-furanone, 3-methyl-2(5H)-fruanone, and 5-methyl-2-furaldehyde delivered through pyrolysis of the flax cigarette paper were higher than that of wood cigarette paper. Otherwise, the area % of some components, such as 2-methyl-cyclopentene-l-one), 2,3-butanedione, 2-cyclopentene-l-one, and 5-hydroxy-2-methyl-furaldehyde, 2-furaldehyde delivered through pyrolvsis of the wood cigarette paper were higher than that of flax cigarette paper. To identify the difference between two cigarette papers, we used the cigarette column filled with the cut cigarette paper instead of the cut tobacco leaf. The amounts of semi-volatile fraction delivered from flax cigarette paper was more than that of wood cigarette paper. But, by using the cut tobacco, there was no big difference of delivery amount between flax and wood cigarette papers. Also, aroma of TPM by collecting from brening cut tobacco wrapped in flax and wood papers showed a different pattern by the electonic nose system. Although the difference between two cigarette papers by using the cut tobacco was smaller than that of cut cigarette paper, this result indicated that the fax and wood had the different effects on the delivery of smoke components as shown in the sensory test results.

Synthesis of 2(5H)-Furanones and 2-Cycloalkenones and Their Fungicidal Activities (2(5H)-Furanone 및 2-Cycloalkenone 유도체의 합성과 살균활성)

  • We have developed an efficient synthesis of 2(5H)-furanones using the microwave-promoted Suzuki-Miyaura coupling reaction and evaluated their fungicidal activities against six fungal pathogens in vivo. In addition, 2-cyclopentenones or 2-cyclohexenones possessing one or two additional methylene groups instead of the oxygen in the 2(5H)-furanone skeleton were also prepared by using the similar method. As the results of in vivo fungicidal screening against 6 plant diseases, a few derivatives displayed specific fungicidal activities against rice blast and tomato late blast. Further studies toward the optimization of the chemical structures are necessary for the development of novel fungicides with high potency.

Role of STAT3-Interacting Protein (STIP1) in ${\Delta}^{12}-Prostaglandin$ $J_2-Induced$ Cell Death

  • ${\Delta}^{12}-Prostaglandin$ $J_2\;({\Delta}^{12}-PGJ_2)$ is one of cyclopentenone prostaglandins. The ${\Delta}^{12}-PGJ_2$ is known to induce apoptosis of tumor cells, however, it's action mechanism is not clear. It has recently been reported that STAT3 is involved in tumorigenesis. In the present study, we investigated the role of STAT3-interacting protein (STIP1) in the cytotoxicity of ${\Delta}^{12}-PGJ_2$, since STIP1 was recently reported as a modulator of STAT3 activation by specifically binding to inactive (unphosphorylated) STAT3. The effect of ${\Delta}^{12}-PGJ_2$ was observed in stably overexpressing Neuro-2A cells transfected with full cDNA of STIP1, and cytotoxicity of ${\Delta}^{12}-PGJ_2$ in the transfected cells was increased, compared with the vector control cells. The cytotoxicity of ${\Delta}^{12}-PGJ_2$ treatment was significantly accentuated by pretreatment of the STIP1-transfected cells with protein kinase inhibitor, genistein, and less activation of STAT3 in STIP1-transfected cells was shown, compared with the vector control cells. Expression of bax was also increased in the STIP1-transfected cells. These data suggest that STIP1 inhibits cell growth via inhibition of STAT3 activation in ${\Delta}^{12}-PGJ_2$ treatment.

Upregulation of Lipopolysaccharide-Induced Interleukin-10 by Prostaglandin $A_1$ in Mouse Peritoneal Macrophages

  • The cyclopentenone prostaglandins (cyPGs) prostaglandin $A_1$ ($PGA_1$) and 15-deoxy-${\Delta}^{12,14}$-prostaglandin $J_2$ (15d-$PGJ_2$) have been reported to exhibit antiinflammatory activity in activated monocytes/macrophages. However, the effects of these two cyPGs on the expression of cytokine genes may differ. In this study, we investigated the mechanism of action of $PGA_1$ in lipopolysaccharide (LPS)-induced expression of inter leu kin (IL)-10 mRNA in mouse peritoneal macrophages. 15d-$PGJ_2$ inhibited expression of LPS-induced IL-10, whereas $PGA_1$ increased LPS-induced IL-10 expression. This synergistic effect of $PGA_1$ on LPS-induced IL-10 expression reached a maximum as early as 2 h after simultaneous $PGA_1$ and LPS treatment ($PGA_1$/LPS), and did not require new protein synthesis. The synergistic effect of $PGA_1$ was inhibited by GW9662, a specific peroxisome proliferator-activated receptor ${\gamma}(PPAR{\gamma})$ antagonist, and Bay-11-7082, a NF-${\kappa}B$ inhibitor. The extracellular signal-regulated kinases (ERK) inhibitor PD98059 increased the expression of $PGA_1$/LPS-induced IL-10 mRNA, rather than inhibiting the IL-10 expression. Moreover, $PGA_1$ inhibited LPS-induced ERK phosphorylation. The synergistic effect of $PGA_1$ on LPS-induced IL-10 mRNA and protein production was inhibited by p38 inhibitor PD169316, and $PGA_1$ increased LPS-induced p38 phosphorylation. In the case of stress-activated protein kinase/c-Jun $NH_2$-terminal kinase (SAPK/JNK), the SAPK/JNK inhibitor SP600125 did not inhibit IL-10 mRNA synthesis but inhibited the production of IL-10 protein remarkably. These results suggest that the synergistic effect of $PGA_1$ on LPS-induced IL-10 expression is NF-${\kappa}B$-dependent and mediated by mitogen-activated protein (MAP) kinases, p38, and SAPK/JNK signaling pathways, and also associated with the $PPAR{\gamma}$ pathway. Our data may provide more insight into the diverse mechanisms of $PGA_1$ effects on the expression of cytokine genes.

Compostion of Constituents of Commercial Wood Vinegar Liquor in Korea (시판 목초액의 성분조성)

  • Two commercial wood vinegar liquors prepared from Cryptomeria japonica and Quercus sp., which are used as a mineral fertilizer in Korea, were extracted using dichloromethane as a solvent, respectively. The extracts were separated into acidic, phenolic, neutral and basic fraction by acid or alkali treatment, and the compositions of each fraction were analyzed by means of GC and GC-MS. A total of 103 compounds including 26 acids, 32 phenols and 45 neutral compounds were identified. The major components were acetic, propionic and n-butyric acid, representing of $41{\sim}58%$ of the acidic fraction, guaiacol, 4-methylguaiacol and phenol, repersenting of $53.2{\sim}63.9%$ of the phenolic fraction, and furfural, 3-methyl-2-cyclopenten-2-one, 2,3-dimethyl-2-cyclopenten-1-one and 5-methyl-2-furfural in the neutral fraction. In addition to these compounds, phenolic fraction in dichloromathane extract from wood vinegar liquor of C. japonica included large amounts of vanillin, acetovanillone and tentatively identified ethylvanillyl ether while that of Quercus sp. included some amounts of syringol and 4-methylsyringol.
