Effects of low temperature and salicylic acid on chilling tolerance in cucumber seedlings

  • The present study was undertaken to Investigate the effects of low temperature and salicylic acid (SA) on chilling tolerance In acclimated and nonacclimated cucumber seedlings. Acclimation significantly affected survival and shoot dry weights. Injuries of acclimated seedlings at the third leaf stage were on the average smaller by half than those of the nonacclimated ones. Chilling caused a large increase in free proline levels, regardless of acclimation status. Exogenous treatment with SA resulted in improvement in growth and survival of acclimated, chilled seedlings, indicating SA and acclimation have common effects. Cycloheximide treatment In the presence of SA restored acclimation-induced chilling tolerance. An elevated proline level was observed in cold-treated and SA- treated plants and the level was more pronounced in the light than in the dark at chilled temperature, indicating that endogenous proline may play a role in chilling tolerance.

Association of Anti-apoptotic Mechanism Due to House Dust Mite in Neutrophils with Protein Synthesis and Bad

  • House dust mite is an essential allergen in the pathogenesis of allergic diseases. Abnormal regulation of neutrophil apoptosis is an important pathogenic process in allergic diseases. In the present study, we investigated the effects of house dust mites on spontaneous apoptosis of neutrophils and its associated mechanisms. Extract of Dermatophagoides pteronissinus (DP) inhibited neutrophil apoptosis in a time-dependent manner. Cycloheximide (CHX), an inhibitor of translation, increased apoptosis of DP-treated neutrophils as well as control cells. The pro-apoptotic effect of CHX was blocked by DP in neutrophils. In addition, DP increased the phosphorylation of Bad in a time-dependent manner, indicating that it exerted an inhibitory effect on the function of Bad. These results suggest that DP has anti-apoptotic effects of neutrophils and may regulate protein synthesis and activation of Bad. Moreover, these findings may shed light on elucidation of allergy pathogenesis due to house dust mites.

Heat-Shocked Drosophila Kc Cells Have Differential Sensitivity to Translation Inhibitors

  • The heat shock response is a universal stress response observed in all organisms and cultured cells. The response is regulated at both the transcriptional and translational level. Heat shocked Drosophila melanogaster Kc cells are used as the system for the study of translational regulation. In this system non-heat shock messages are associated with polysome but are not translated in a heat shocked condition. To figure out the change in the translation machinery. the effects of translation elongation inhibitors were tested on Kc cells. The result showed that the sensitivity of translation to these drugs changed in heat shocked cells. The significant changes were the decreased inhibition of heat shock protein synthesis by cycloheximide, emetine. and puromycin. and the increased inhibition of heat shock protein synthesis by verrucarin A. implying that the translation elongation mechanism in heat shocked cells changed.

  • This study was conducted to investigate the effect of recipient activation time on the nuclear remodeling, chromatin structure, pronuclear formation and in vitro development of bovine nuclear transfer embryos derived from adult ear skin cells. Somatic cells were transferred to enucleated oocytes after quiescent treatments by serum starvation or culture to confluency. Nuclear transfer embryos were activated with a combination of Ca/sup 2+/-ionophore and cycloheximide at 1, 1.5, 2, 2.5, 3, and 5 h after electrofusion. (omitted)

  • Chimerism has become an important tool for investigating fundamental aspects of early embryonic development and differentiation in mammals for producing transgenic animals. The objective of this study was to evaluate the developmental capacity of chimeric embryos reconstructed with parthenotes and IVF bovine embryos into empty zona pellucida.   • The effects of several compounds related to signal transduction cascade were determined to induce the production of alizarin and purpurin in the hairy root culture system of Rubia cordifolia var. pratensis. It was found that out of five tested compounds jasmonic acid(1 mg/l) and methyl jasmonate(1 mg/l) stimulated strongly the biosynthesis of the pigments while linolenic acid(1 mg/l) induced no significant increase of the product. Yeast extract(600 mg/l) and arachidonic acid(1 mg/l) showed relatively mild inducing effects on production of alizarin. The effects of jasmonic acid and methyl jasmonate were reduced by treatment with cycloheximide(2.8 mg/l).

  • 최근 수년 전부터 학습(learning), 기억(memory)의 분자약리학적 기전과 치매(dementia)의 신경생리학적 원인 규명에 대한 연구가 사회적 요구에 부응하여 증가하고 있다. 이에 본 연구에서는 횐쥐를 이용하여 실험실적 건망모델을 설정하여 항치매 물질의 효력 screening을 시도하였다. 실험실적 건망모델은 다음 두 가지 방법으로 설정하였다. 첫째, Acetylcholine 길항제인 Scopolamine을 사용하여 중추신경계중 기억, 학습기전과 관련된 것으로 알려진 cholinergic신경계률 차단하여 유발한 실험적 건망모델과, 둘째, 단백질합성 저해제인 cycloheximide를 사용하여 기억, 학습경로에 관여하는 수종의 중추 단백질듈을 비선택적으로 저해하여 유발한 실험적 건망모델을 이용하여 인삼, 오미자등의 항치매 효과를 검색하고자 수동적 회피학습능, 능동적 회피학습능, 자발운동량을 측정하여 기억, 학습, 행동의 상관 관계를 고찰하였다.

  • To produce reconstituted rabbit embryos with fetal fibroblasts, the present study was evaluated the efficiencies of the activation conditions as assessments of subsequent development and chromosome in the embryos. New Zealand White rabbits were used throughout the study. Fetal fibroblasts collected from 22-d of fetuses were cultured in DMEM+10% FBS in 5% CO₂ in air. The culture was maintained for 10 passages. In every passage half of cell suspension were kept in frozen. (omitted)

개구리 난자의 성숙촉진요인에 관한 연구

  • 북방산개구리, 참개구리 및 옴개구리를 사용하여 성숙된 난자의 세포질에서 활성을 띠는 성숙촉진요인( maturation promoting factor, MPF)을 미세주입 법으로 확인하고 이들의 성질을 조사하였다. 핵붕괴(germinal vesicle breakdown, GVBD)된 난자의 세포질을 미성숙 난자(GV난자)에 주입하고(75-100 nl) 이들을 15-24시간 배양했을 때 대부분의 GV 난자들이 핵붕괴를 일으켰으나(약 80%) 미성숙 난자(GV 난자)의 세포질을 주입했을 때에는 약 200nl의 난자들만이 핵붕괴를 일으켰다. 핵붕괴된 난자들의 crude extract를 주입했을 때에도 역시 성숙유도 효과가 있었으며 이종간에도 효과가 있었다. 난자의 성숙을 잘 일으키지 않는 옴개구리의 난자를 사용하여 MPF를 가진 세포질을 계대주입(serial transfer)하였을 때에도 MPF가 계속 활성을 띠는 것을 확인할 수 있었다. 아울러 개구리 난자의 MPF생성과 증폭과정에 CAMP의 증가나 단백질 합성의 저해가 미치는 영향을 조사한 결과. MPF의 작용이 유의하게 이들에 의해 억제되는 것을 알 수 있었다.

Canavanine Effects on the Amylase Activity and Protein Content in Barley Half Seeds (Canavanine에 의한 보리 무배부 종자의 Amylase 활성과 단백질 함량의 변화)

  • L-canavanine was added to GAs treated barley seeds, and induced amylase activity, soluble protein content, and arginine content were mesured. Canavanine, added at the beginning of the incubation period, inhibited amylase activity and protein accumulation. Amylase activity decreased markedly by addition of canavanine at 6 hr after incubation, where soluble protein content was not affected. The addition of canavanine after 12 hr incubation did not show serioud inhibited effect on the amylase activity and protein accumulation. GAs incubation caused decrement in arginine content per mg protein, but it was somewhat recovered by canavanine treatment. The longer the time between GAs and canavanine addition was, the less the recovery ration was. Arginine content in the $\alpha$-amylase fraction (ammonium sulfate 20~50% saturation) was lower than in 0~20% fraction, but higher than in 50~80% fraction. These results and control expreiments, using cordycepin and cycloheximide, support the idea that canavanine might incorporate into protein.
