The purpose of present study was to investigate the effects of physiologically active compound (SD62-122) from Phlomidis Radix on the cell cycle progression and its molecular mechanism in human gingival fibroblasts(HGFs). For this purpose, fibroblasts were isolated and cultured from excisioned gingiva during crown lengthening procedure in healthy adult. The following parameter were evaluated that there are cell number counting, MIT assay, cell cycle progression, western blot analysis. The cell number and MIT assay of primary cultured fibroblast was not increased at 2 days but significant increased compare to negative control at 3days(p<0.05). S phase was increased and G1 phase decreased in both $10^{-8}M$ and $10^{-9}M$ of SD62-122 in cell cycle analysis. The cell cycle regulation protein levels of Cyclin $D_1$, Cyclin E, cdk 2, cdk 4 and cdk 6 were increased compare to control in both $10^{-8}M$ and $10^{-9}M$ of SD62-122. The protein levels of p21 and p53 were decreased compare to control, but the level of pRb was not changed compare to control in $10^{-9}M$ of SD2-122. These results suggested that physiologically active compound (SD62-122) isolated from Phlomidis Radix increases the cell proliferation and cell cycle progression in HGFs, which is linked to increased cell cycle regulation protein levels of Cyclin $D_1$, Cyclin E, cdk 2, cdk 4 and cdk 6, and decreased the levels of p21, p53.
Recently, in proportion to increased demand on environmentally-friendly heat source, efficient management of district heating(DH) system becomes one of important issue. The objectives of this paper are to systematize data processing of transition temperature, investigate the effect of temperature variations on thermal fatigue and find out a way to improve design fractures of Korean DH pipes. For this purpose, reliable fatigue lift evaluation procedures are examined and applied to quantify thermal fatigue lives. Also, as a prototypal optimization analysis results, mean value of original cross sectional area of selected pipes was reduced 18.6% sustaining their sufficient margins against fatigue failure. So, it is anticipated that the output of this research can be used as useful information of optimal design and operation in the future.
The objectives of this study were to analyze the load acting on the transmission when tractor performed rotary operations in the poorly drained paddy fields and to compare its severeness with those obtained under different operational conditions. The loads were measured at the input shaft of the transmission and the load spectrum was constructed using the rain-flow cycle counting method. The severeness of the loads was represented by the partial damage sum which was determined by suing the modified Miner's rule. In the field with a shallow hardpan PTO speed affected severeness of the rotary operations greater than the forward speed did. In the field with deep hardpan, on the other hand, the effect of the forward speed was greater. the severeness in the well drained fields did not differ significantly from that in the field with a shallow hardpan. The severest load was recorded in the field with a deep hardpan.
In the present study the flow fields around pitching foils have been visualized by using a Schlieren method with a high speed camera in a wind tunnel at low Reynolds number regions. It has been observed that small vortices are shed discretely from the leading and trailing edge and that they stand in line on the integrated streakline of separation shear layer. By counting vortices in the VTR frames it was clarified that the number of vortex shedding from the leading and trailing edge during one pitching cycle strongly depends on the non-dimensional pitching rate.
This study aims to estimate the fatigue resistance of small wind composite blade using the fatigue life estimation formula in the GL guideline. For this, firstly, we estimated a turbine blade's bending moment spectrum by using wind profile wind profile and BEMT. And fatigue tests were performed to obtain the S-N curve of composite materials used in blade. In addition, a finite element analysis was used to identify fatigue critical locations and fatigue stress spectrum. And the fatigue resistance of composite blade were evaluated using the rainflow cycle counting, and Goodman diagram and the fatigue life estimation formula in the GL guideline.

Quantification of sensitive material is of vital importance when it comes to the movement of nuclear fuel throughout its life cycle. Within the electrorefiner vessel of electrochemical separation facilities, the task of quantifying plutonium by neutron analysis is especially challenging due to it being in a constant mixture with curium. It is for this reason that current neutron multiplicity methods would prove ineffective as a safeguards measure. An alternative means of plutonium verification is investigated that utilizes the (𝛼,n) signature that comes as a result of the eutectic salt within the electrorefiner. This is done by utilizing the multiplicity variable a and breaking it down into its constituent components: spontaneous fission neutrons and (𝛼,n) yield. From there, the (𝛼,n) signature is related to the plutonium content of the fuel.
Human UHRF1 (ubiquitin-like PHD and RING finger domain-containing 1) has been reported to be over-expressed in many cancers, but its role in ovarian cancer remains elusive. Here, we determined whether knockdown of UHRF1 by lentivirus-mediated shRNA could inhibit ovarian cancer cell growth. Lentivirus-mediated short hairpin RNAs (lv-shRNAs-UHRF1) were designed to trigger the gene silencing RNA interference (RNAi) pathway. The efficiency of lentivirus-mediated shRNA infection into HO-8910 and HO-8910 PM cells was determined using fluorescence microscopy to observe lentivirus-mediated GFP expression and was confirmed to be over 80 percent. UHRF1 expression in infected HO-8910 and HO-8910 PM was evaluated by real-time PCR and Western blot analysis. The Cell Counting Kit-8 (CCK-8) assay was used to measure cell viability; flow cytometry and Hoechst 33342 assay was applied to measure cell cycle arrest and apoptosis. Cell invasion was assessed using transwell chambers. Our results demonstrated that the loss of UHRF1 promoted HO-8910 and HO-8910 PM cell apoptosis, while inhibiting cell proliferation. In addition, UHRF1 knockdown significantly inhibited the invasion of human ovarian cancer cells. In the present study, we also showed that depleting HO-8910 cells of UHRF1 caused activation of the DNA damage response pathway, with the cell cycle arrested in G2/M-phase. The DNA damage response in cells depleted of UHRF1 was illustrated by phosphorylation of CHK (checkpoint kinase) 2 on Thr68, phosphorylation of CDC25 (cell division control 25) on Ser 216 and phosphorylation of CDK1 (cyclin-dependent kinase 1) on Tyr 15.
Concerns are emerging in marine industry on the additional fatigue damages induced by hydroelasticity, and large container carriers, among others, are considered to be susceptible to this hydroelastic response due to its large size, deck openings and high speed. This study focuses on the fatigue damage estimation of 9,400TEU container carrier based on the full scale measurement data via long-base strain gage installed on the ship. Some correlation analyses have been also done to check whether there was significant torsional response during the voyage. Direct cycle counting method was used to derive stress histogram and the long-term fatigue damage was estimated based upon that analyzed data. It turned out that the fatigue damage of this particular ship during the measurement period increased by more than 60% due to the hydroelastic response of the hull, and main contribution is considered to come from vertical bending mode.
강철도교에 대한 확률기반 피로 수명 평가를 위한 많은 연구들이 그간 있어 왔지만, 대부분 상대적으로 단순한 피로 균열 진전 모델을 기반으로 한 연구들이었다. 이 모델은 최소 응력이 0이고 일정한 응력변동 진폭을 가정하기 때문에, 철도교의 피로수명 평가에는 적합하지 않다. 따라서 본 연구에서는 보다 고도화된 균열 진전 모델을 이용해 강철도교의 피로 수명을 평가하는 새로운 확률기반 기법을 제안하였다. 또한 이 기법은 철도교에서 흔히 발생하는 다양한 하중 변동 진폭을 rainflow cycle counting algorithm을 사용해 고려할 수 있어, 보다 현실적인 피로 수명을 평가할 수 있다. 제안된 기법을 강철도교 예제 모델에 적용하여 피로 수명을 주요 부재 및 시스템에 대해 평가하였다. 또한 다양한 활하중-사하중 비가 피로 수명에 끼치는 영향을 분석하였으며, 그 결과 활하중-사하중 응력 비가 0에서 5/6까지 증가함에 따라 부재와 시스템 수준 모두에서 피로 수명이 30년 내외까지 줄어드는 것을 확인하였다.
Purpose : This study was aimed to evaluate the cell cycle arrest and apoptosis induction after irradiation and epidermal growth factor (EGF) treatment in three human epithelial tumor cell lines (A431, Siha, KB). Materials and Methods: Single irradiation of 2, 5 and 10 Gy was done on three cell lines with 5.38 Gy/min dose rate using Cs-137 irradiator at room temperature. Also, EGF of 10 ng/ml was added immediately after 10 Gy irradiation. Cell growth was evaluated by counting the living cell number using a hemocytometer at 1 day, 2 days, 3 days, 4 days and 5 days after irradiation. Cell cycle arrest and apoptosis induction were assayed with the flow cytometry at 8 hours, 12 hours, 1 day, 2 days, 3 days, 4 days and 5 days after irradiation. Results : Growth of irradiated three cell lines were inhibited in proportion to radiation dose. EGF treatment after irradiation showed various results according to cell lines. On all cell lines, G2 arrest was detected after 8 hours and maximized after 12 hours or 1 day. Amount of G2 arrest was positively dose dependent. However, EGF showed no significant change on G2 arrest. G2 arrest was recovered with time at 2 Gy and 5 Gy irradiation. However, at 10 Gy irradiation, G2 arrest was continued. Apoptosis was detected at 10 Gy irradiation. On EGF treated group after irradiation, A431 and Siha cell lines showed slightly increased apoptosis but there was no statistically significant difference. KB cell line showed no marked change of apoptosis induction. Conclusion : Irradiation effects on cell cycle arrest and apoptosis induction in three human epithelial tumor cell lines, however epidermal growth factor doesn't effect on.
