New Phytoformula (CAS) Containing the Roots of Cyathula officinalis, Achyranthes japonica and Sophora subprostrata Inhibits Collagen-induced Arthritis in Mice

  • The combined new phytoformula (CAS), a mixture (5 : 3 : 2, w/w/w) of the ethanol extracts of the roots of C. officinalis and A. japonica, and the n-butanol fraction of the S. subprostrata strongly inhibited arthritic severity score as well as IL-6 production in serum of collagen-induced arthritic mice. Histological observation also indicated that the CAS-treated group showed less breakdown of joint cartilage of the collagen-treated mice. In contrast, C. officinalis alone or a combination of A. japonica and S. subprostrata did not show significant inhibitory action on the same animal model. Thus, it is thought that CAS possesses a synergistic inhibitory action on arthritic condition. All these results strongly suggest that CAS may be a potential anti-arthritic agent.

우슬(牛膝)의 형태(形態) 감별에 관한 연구 (A Study on a Morphological Identification of Achyranthes and Cyathula Root)

  • Objectives : A morphological classification among Achyranthes japonica Nakai (produced in Korea), A. bidentata Blume (imported from China), and Cyathula officinalis Kuan (used in China exclusively) was made through microscopic observation. Method : The slice of the tested material made by paraffin section technique was colored with Safranine Malachite Green contrast methods, and then observed and photographed by olymphus-BHT. Result : 1. Korean A. japonica Nakai has slim roots, whose diameter is $1{\sim}5\;mm$. 1) Most of its intersection is stele, which includes a lot of vascular bundles. The inside of vascular bundles shows the arrangement of $2{\sim}4$ cycles, the innermost wheel of which is divided into two parts. 2) Parts of parenchymatous cell include Crystal sand of calcium oxalate. 2. The diameter of the roots of Chinese A. bidentata Blume is $0.4{\sim}1\;cm$. 1) The xylem in the vascular bundle in the middle of its intersection is rather big, around which the inner parts of the vascular bundles are arranged in the shape of $2{\sim}4$ cycles around. The most outside part of it is small, and the middle part is gathered into $2{\sim}3$ groups. 2) Parts of parenchymatous cells include Crystal sand of calcium oxalate. 3. The roots of Chinese C. officinalis Kuan is thick, whose diameter is $0.5{\sim}3\;cm$. 1) Vascular bundle of its intersection is spotted, arranged in the shape of $4{\sim}11$ concentric circles, whose center is divided into $2{\sim}9$ groups. 2) Parenchymatous cells include crystal sand and square crystal of calcium oxalate. Conclusion : It is concluded that Achyranthes japonica Nakai, A. bidentata Blume, and C. officinalis Kuan have different shapes in both external forms and internal intersections, so that they can be easily distinguished from one another through microscopic observation.

HPLC/DAD를 이용한 6종(種) 우슬(牛膝)의 분류기준 연구;우슬(牛膝)(쇠무릎, Achyranthes japonica $N_{AKAI}$)로부터 20-hydroxyecdysone 분리.동정 및 산지별 우슬의 HPLC 패턴 비교 (A Study on Discriminative Criteria of 6 Kinds of Achyranthis Radix Using HPLC/DAD;Isolation and Identification of 20-hydroxyecdysone from Aclryranthes japonica $N-{AKAI}$ and Comparison of Patterns of Achyranthis Radix from Different Locations by HPLC)

  • Objectives : This study was performed to investigate the discriminative criteria of 6 kinds of Achyranthis Radix by HPLC/DAD. Methods : 20-hydroxyecdysone is isolated by silica gel column chromatography ($CHCl_3$:MeOH, 7:1-1:1 v/v) and identified by nuclear magnetic resonance, A high-performance liquid chromatographic method with diode array detection was used to identify 20-hydroxyecdysone in A. japonica. The analysis was performed using $C_{18}$ column with isocratic elution consisted of 18% acetonitrile and 82% water and the detection was carried out by DAD at 254 nm. 6 kinds of Achyranthis Radix from different locations were extracted in MeOH. Each extracts was analyzed by HPLC in same condition as used in analysis of 20-hydroxyecdysone. The identities of each extracts were determined by comparing the retention time and UV spectrum with that of reference compound. Results : 1. A. japonica and A. bidentata showed the similar patterns of HPLC chromatogram and 20-hydroxycedysone was present in both of them because the peaks having the same retention time and UV spectrum as 20-hydroxyecdysone were shown in the HPLC chromatograms of A. japonica and A. bidentata 2. Cyathula officinalis and C. capitata showed the similar patterns of HPLC chromatogram. The peak having the same retention time and UV spectrum as 20-hydroxyecdysone was shown in the HPLC chromatogram of C. capitata but not shown in the HPLC chromatogram of C. officinalis. 3. Two species of medicinal drugs from Sacheon province showed similar patterns of HPLC chromatogram. Achyranthis Radix from Sacheon(wild) did not have 20-hydroxycedysone but Achyranthis Radix from Sacheon(cultivated) showed the peak having the same retention time as 20-hydroxyecdysone but UV spectrum of the peak was different from that of 20-hydroxyecdysone. Conclusions : These results suggested that 20-hydroxyecdysone could be the discriminative criteria for Achyranthis Radix contain 20-hydroxyecdysone though they belong to different genus and species. And the patterns of HPLC chromatogram also could be the discriminative criteria as the different species of Achyranthis Radix belonging to the same genus showed similar patterns of HPLC chromatogram.

  • 1983년(年) 7월(月)부터 84년(年) 10월(月)까지 지리산(地異山), 속리산(俗離山), 덕유산(德裕山), 칠갑산(七甲山), 계룡산(鷄龍山) 지역(地域) 및 충남일원(忠南一圓)에서 젖버섯 속(屬)에 속하는 균류(菌類)들을 채집(探集)하여 분류(分類)한 결과(結果) Lactarius 19종, 4변종 및 1 form이 동정(同定)되었다. 이들 Lactarius를 Hesler 및 Smith(1979)의 체계(體係)에 따라 6 section으로 나누었으며 이중 한국(韓國)에서 처음 발견된 버섯은 다음과 같다. L. lignyotus var. nigroviolascens (Atk.) Hesler & Smith 보라잿빛 헛대 젖버섯, L. sumstinei Pk. 우산주름 젖버섯, L. subvellereus var. subdistans Hesler & Smith 노란주름융단 젖버섯, L. vellereus var. virescens Hesler & Smith 푸른주름융단 젖버섯, L. piperatus var. glaucescens (Crossl.) Hesler & Smith 녹색젖 굴털이, L. luteolus Pk. 갈색끈적 젖버섯, L. pyrogalus (Fr.) Fr. 제브라 젖버섯, L. argillaceifolius var. argillaceifolius Hesler & Smith 흑갈색주름 젖버섯, L. repraesentaneus Britz. 보라빛주름 젖버섯, L. mutabilis Pk. 진갈색무늬 젖버섯, L. thejogalus (Fr.) S.F. Gray 갈황색 젖버섯, L. gracilis Hongo 애기 털 젖버섯, L. cyathula f. japonicus Hongo 애기낙엽 젖버섯.

우슬(牛膝)의 관절염(關節炎) 치료(治療) 효능(效能)에 관(關)한 서지학적(書誌學的) 고찰(考察) (A Bibliographic Study on the Therapeutic Effects of Achyranthis Radix in Arthritis)

  • According to herbalogical bibligraphy and literature, therapeutic effects of Achyranthis Radix on arthritis was as follows, 1. Achyranthis Radix of Amaranthaceae is divided into five species-Achyranthes japonica, A. bidentata, A. longifolia, A. fauriei, Cyathula capitata, C. officialis and the characteristic, taste, channel entry, effects and main treatments were alike. 2. Winefrying stood for repairing treatement method for Achyranthis Radix Before repairing treatment method, Achyranthis Radix had three tastes(bitter, sour, and sweet) and calm and not poisoning characteristic. After repairing treatment method, the bitter taste was disappeared, and calm and not poisoning characteristic was changed into warm characteristic. 3. Effects of Achyranthis Radix were quicking the blood and dispelling stasis, liver-kidney supplement and strengthening musculo-skeletal system. Main treatments were relief of lumbar and knee joint pain, static menstrural block and wind-cold- damp impediment. 4. Contraindication of Achyranthis Radix was sympthom caused by spleen-kidney yang vacuity, upper burner disease and lower burner hemorrhage etc. Being used in pregnant woman, it could incur abortion. 5. Contraindication of Achyranthis Radix was beef, milk and mutton. It's fear was Radix Cynanchi Stauntonii and Semen Plantaginis. It's aversion was the firefly. Herba Taraxaci, Carapax Amydae, Carapax Testudinis and Radix Cynanchi Stauntonii. From above results, I suppose Achyranthis Radix has enough herbalogical foundation and could be used to treat arthritis and it is necessary to make a profound study of it.
