Antifungal Activity of Methanolic of Centella asiatica and Andrographis panicuiata

  • The antifungal activity of methanolic extracts of Centella asiatica and Andrographis paniculata leaves was observed against fourteen fungi, viz., Alternaria alternata, A. brassicae, A. brassicicola, A. solani, A. tenuissima, Cercospora blumae, Curvularia lunata, C. penniseti, and Drechslera monoceras, D. oryzae, D. turitica, Fusarium albizziae and F. udum. Different concentrations of the methanolic extract (1000, 2000, 3000, 5000, 7000, 10000ppm) were used. The effect of mixed leaf extract (1500 ppm of C. asiatica + 1500 ppm of A. paniculata) and its 1:2 ad 1:4 dilutions were also studied. The individual extracts of both the plants showed significant inhibitory effect on spore germination of all the fungi tested. F. udum, F. albizzae, D. oryzae, D. turtica, and D. monoceras were particularly sensitive to these extracts. In general, the extract of C. asiatica showed a higher inhibitory effect in all concentrations against all the fungi as compared to A. paniculata, except for A. brassicae A. solani, D. oryzae, D. penniseti and Curvularia sp. The inhibitory effect of extracts increased when they were used in combination with or without dilutions against A. brassicicola, A. solani A. brassicae, A. alternata, A. tenussima, C. blumae, C. lunata, C. penniseti and Curvularia species. Higher efficacy of active ingredient of these extracts under field condition is envisaged against plant pathogens.

벼 이삭마름병해에 관여하는 균류에 관한 연구 (Studies on the Fungi Associated with Ear Blight of Rice)

  • 벼 이삭마름병해에 관여하는 균류와 이들 균류에 대한 약제별 살균효과를 알고저 시험한 결과를 요약하면 다음과 같다. 1. 일반 장려품종 및 통일품종의 변색된 벼알에서 균류를 조사한 결과 일반 장려품종에서는 Pyricularia oryzae, Helminthosporium oryzae, Epicoccum purpurascens, Curvularia lunata, Cladosporium cladosporoides, Alternaria tenuis균 등이 주로 많이 검출되었고 통일 품종에서는 Trichoconis padwickii, Fusarium dimerum, Nigrospora oryzae균 등이 많이 검출되었다. 2. 벼 이삭도열병에 이병된 이 삭의 각 부위로부터 균류를 조사한 결과 이삭목에서는 Pyricularia oryzae, Nigrospora oryzae, Curvularia lunata 균등이 이삭가지에서는 Helminthosporium eryzae, Trichoconis padwickii, Stemphylium sp., Cladosporium cladosporoides, Alternaria tenuis균 등이 많이 검출되었으며 벼알에서는 Phema spp. 균이 주로 많이 검출되었다. 3. 각종 균류에 이병된 이삭가지 및 벼 알에서의 Difolatan 수화제, Dithane M-45 수화제의 살균효과를 실내에서 시험한 결과 Difolatan 800배 액은 Helminthosporium oryzae, Cladosporium cladosporoides, Alternaria tenuis 균 등에 살균효과가 좋았고 Dithane M-45 500배 액은 Pyricularia oryzae, Trichoconis padwickii, Curvularia lunate 균등에 비교적 효과가 좋았다. 4. 약제 방제시기는 유숙기살포구, 출수개화기살포구의 순으로 효과가 좋았으며 약제별 방제효과는 본 시험포장에 이삭도열병 발생이 많았던 관계로 도열병 방제 약제인 히노산유제, 가스가민유제가 Difolatan 수화제, Dithane M-45 수화제에 비하여 다소 효과가 좋은 것으로 나타났다.

The Effect of Seed-borne Mycoflora from Sorghum and Foxtail Millet Seeds on Germination and Disease Transmission

  • The seed-borne mycoflora of sorghum and foxtail millet collected from different growing areas in South Korea were isolated and taxonomically identified using dry inspection, standard blotter and the agar plate method. We investigated the in vitro and in vitro germination rates of disinfected and non-disinfected seeds of sorghum and foxtail millet using sterilized and unsterilized soil. The percent recovery of seed-borne mycoflora from the seed components of sorghum and foxtail millet seeds was determined and an infection experiment using the dominant species was evaluated for seedling emergence and mortality. A higher number of seed-borne fungi was observed in sorghum compared to that of foxtail millet. Eighteen fungal genera with 34 fungal species were identified from the seeds of sorghum and 13 genera with 22 species were identified from the seeds of foxtail millet. Five dominant species such as Alternaria alternata, Aspergillus flavus, Curvularia lunata, Fusarium moniliforme and Phoma sp. were recorded as seed-borne mycoflora in sorghum and 4 dominant species (Alternaria alternata, Aspergillus flavus, Curvularia lunata, Fusarium moniliforme) were observed in foxtail millet. The in vitro and in vitro germination rates were higher using disinfected seeds and sterilized soil. More seed-borne fungi were recovered from the pericarp compared to the endosperm and seed embryo. The percent recovery of seed-borne fungi ranged from 2.22% to 60.0%, and Alternaria alternata, Curvularia lunata and 4 species of Fusarium were isolated from the endosperm and embryo of sorghum and foxtail millet. Inoculation of the dominant seed-borne fungi showed considerable mortality of seedlings. All the transmitted seed-borne fungi might well be a primary source of infection of sorghum and foxtail millet crops.

Antifungal Activity of Withametelin, a Withanolide Isolated from Datura metel

  • Withametelin, a steroidal compound isolated from leaves of Datura metel L.(Solanaceae), showed antifungal activity against some plant pathogenic as well as saprophytic fungi tested in vitro. Except Curvularia maculans and Colletotrichum sp., spore germination of all the other 23 fungi was inhibited significantly at 125 to 1000 ppm. Out of the 25 fungi tested, Curvularia sp., Cercospora abelmoschi, Heterosporium sp., Erysiphe cichoracearum and Ustilago cynodontis were most sensitive as there was complete inhibition of germination at 1000 ppm. Similarly, spores of Alternaria brassicae, Curvularia lunata and Helminthosporium pennisetti showed less than 3% germination at the same concentration. Rest of the fungi showed more than 15% spore germination at the same concentration.

Effect of 1-corydalmine, an Alkaloid Isolated from Corydalis chaerophylla Roots on Spore Germination of Some Fungi

  • 1-Corydalmine, an alkaloid isolated from roots of Corydalis ehaerophylla inhibited spore germination of some plant pathogenic as well as saprophytic fungi e.g. Alternaria brassicae, A. brassicieola, A. solani, Curvularia lunata, C. maculans, C. sp., C. pallseens, Erysiphe pisi, Fusarium udum, Helminthosporium species, H. penniseti and a Heterosporium species. 1-Corydalmine significantly inhibited spore germination of all the fungi at 100 to 1500 ppm. It was effective against all the fungi at 1500 ppm. C. lunata was highly sensitive to this chemical even at 250 ppm.

  • 1. 벼 종자(種子)의 변색정도(變色程度)에 따른 균류(菌類)의 검출(檢出)을 조사(調査)한 결과(結果) 변색(變色)이 심한 종자(種子)에서 Alternaria tenuis, Cladosporium cladosporioides, Curvularia lunata, Fusarium semitectum, Gerlachia oryzae, Phoma sp. Trichoconiella padwickii 등이 높은 비율(比率)로 검출(檢出)되었고 특히 반점종자(斑點種子)에서는 Helminthosporium 균(菌)이 높은 비율(比率)로 검출(檢出)되었다. 2. 벼 종자상(種子上)에서 진균(眞菌)의 검출(檢出)에 미치는 광선(光線)의 영향(影響)을 조사(調査)한 결과(結果) Alternaria tenuis, Helminthosporium oryaze, Trichoconiella padwickii, Fusarium moniliforme, Fusarium semitectum 등(等)은 암흑구(暗黑區)에서 보다 광선조사구(光線照射區)에서 검출율(檢出率)이 높았고 Phoma sp. Curvularia lunata, Epicoccum purpurascens, Verticillium sp.는 광선조사구(光線照射區)나 암흑구(暗黑區)나 거의 차이가 없었으며 Gerlachia oryzae는 암흑구(暗黑區)에서 오히려 검출율(檢出率)이 높았다. 3. 벼 종자상(種子上)의 배양온도(培養溫度)가 균류(菌類)의 검출(檢出)에 미치는 영향(影響)을 조사(調査)하였던 바 Alternaria tenuis, Cladosporium cladosporioides는 저온구$(15^{\circ}C)$에서 검출율(檢出率)이 높았고 Phoma sp.는 고온구$(30^{\circ}C)$에서 검출율(檢出率)이 높았다. 4. 검출(檢出)된 각(各) 균류(菌類)가 종자(種子) 변색(變色)및 수량(收量)에 미치는 영향(影響)을 조사(調査)하였던 바 공시(供試)한 7게(個)의 균류(菌類) 모두가 종자(種子) 변색(變色)에 관여(關與)하여 인공접종(人工接種)한 이삭에서의 천립중(千粒重)은 대조구의 천립중(千粒重)에 비해 모두 낮았고 특히 공시(供試)한 7개(個)의 균(菌)을 혼합접종(混合接種)한 이삭의 천립중(千粒重)과 등숙비율(登熟比率)은 대조구보다 저하(低下)됨을 알 수 있었다.

  • 변색미(變色米) 발생(發生)에 관여(關與)하는 병원균(病原菌)과 해충을 분류동정(分類同定)한 결과(結果) 해충으로는 허리노린재과(科)에 속하는 Leptocorisa oratorius가 우점종(優占種)이었고 노린재과(科)인 Menida varipennis. Stollia ventralis 및 Nezara viridula 등이 관여(關與)하였으며 병원균(病原菌)으로는 Drechslera oryzae. Curvularia lunata, Trichoniella padwickii, Sarocladium oryzae, Alternaria tenuis 및 Fusarium solani 등이 관여(關與)하였다. 병원균(病原菌)과 해충의 복합발생시(複合發生時)에 변색미발생(變色米發生)이 더 심하였고 병원균(病原菌)만의 발생시(發生時)는 변색미(變色米) 발생(發生)에 주로 영향(影響)을 미쳤으며 노린재류만의 발생시(發生時)는 수량감수(收量減收)에 더 큰 영향(影響)을 주었다. 그리고 노린재류에 의한 벼 유숙기간(乳熟期間)의 흡즙(吸汁)은 병원균침입(病原菌侵入)을 조장(助長)하여 벼의 질적(質的) 변화(變化)와 양적(量的) 감소(滅少)에 크게 영향(影響)하였다.

Screening Plant Growth-Promoting Bacteria with Antimicrobial Properties for Upland Rice

  • This study explores beneficial bacteria isolated from the roots and rhizosphere soil of Khao Rai Leum Pua Phetchabun rice plants. A total of 315 bacterial isolates (KK001 to KK315) were obtained. Plant growth-promoting traits (phosphate solubilization and indole-3-acetic acid (IAA) production), and antimicrobial activity against three rice pathogens (Curvularia lunata NUF001, Bipolaris oryzae 2464, and Xanthomonas oryzae pv. oryzae) were assessed. KK074 was the most prolific in IAA production, generating 362.6 ± 28.0 ㎍/ml, and KK007 excelled in tricalcium phosphate solubilization, achieving 714.2 ± 12.1 ㎍/ml. In antimicrobial assays using the dual culture method, KK024 and KK281 exhibited strong inhibitory activity against C. lunata, and KK269 was particularly effective against B. oryzae. In the evaluation of antimicrobial metabolite production, KK281 and KK288 exhibited strong antifungal activities in cell-free supernatants. Given the superior performance of KK281, taxonomically identified as Bacillus sp. KK281, it was investigated further. Lipopeptide extracts from KK281 had significant antimicrobial activity against C. lunata and a minimum inhibitory concentration (MIC) of 3.1 mg/ml against X. oryzae pv. oryzae. LC-ESI-MS/MS analysis revealed the presence of surfactin in the lipopeptide extract. The crude extract was non-cytotoxic to the L-929 cell line at tested concentrations. In conclusion, the in vitro plant growth-promoting and disease-controlling attributes of Bacillus sp. KK281 make it a strong candidate for field evaluation to boost plant growth and manage disease in upland rice.

Antifungal Activities of Ethanolic Extract from Jatropha curcas Seed Cake

  • Phorbol ester extraction was carried out from Jatropha curcas seed cake, a by-product from the biodiesel fuel industry. Four repeated extractions from 5 g of J. curcas seed cake using 15 ml of 90% (v/v) ethanol and a shaking speed of 150 rpm gave the highest yield of phorbol esters. The ethanolic extract of J. curcas seed cake showed antifungal activities against important fungal phytopathogens: Fusarium oxysporum, Pythium aphanidermatum, Lasiodiplodia theobromae, Curvularia lunata, Fusarium semitectum, Colletotrichum capsid, and Colletotrichum gloeosporioides. The extract contained phorbol esters mainly responsible for antifungal activities. The extract could therefore be used as an antifungal agent for agricultural applications.

벼 깨씨무늬병 및 잎집썩음병에 항진균 활성을 갖는 길항 미생물의 탐색 (Screening of Antagonistic Bacteria Having Antifungal Activity against Brown Spot and Sheath Rot of Rice)

  • 벼의 깨씨무늬병과 잎집썩음병의 원인균인 Cochliobolus miyabeanus와 Sarocladium oryzae에 의해 발병되며, 깨씨무늬병이 이삭에 발생할 경우Curvularia lunata에 의해 이삭마름병이 추가로 발병하게 되고 한국인의 주식인 쌀의 수확량 감소 및 쌀의 품질 저하와 같은 손실을 초래하여 세계적으로 벼 재배 국가에서는 큰 문제로 장기적으로는 심각한 문제를 초래한다. 따라서, 이러한 식물 병원성 곰팡이의 생물학적 방제를 위해 순창군 논 토양에서 세포외 효소 활성이 우수하고 siderophore를 생산하는 유용미생물 5종을 선별하였다. 5종의 선별 미생물은 벼의 식물 병원성 곰팡이 3종에 대하여 우수한 항진균 활성을 갖고 있었으며, 특히 JSRB 177균주는 가장 우수한 활성을 지녀 최종 균주로 선별되었다. 최종 선별된 JSRB 177균주는 16S rRNA 염기서열 분석을 통하여 Bacillus subtilis로 동정되었으며, 최종적으로 JSRB 177의 당 이용성 및 효소 생산에 대한 분석을 통하여 생리학적 특성을 확인하였다. 향후 포트 시험 및 생산 공정 확립 등 산업화에 연관된 추가 연구가 필요하지만 앞선 결과를 토대로 JSRB 177 균주는 벼 병원성 곰팡이에 대한 생물학적 방제를 위한 소재로 높은 활용이 기대된다.
