As part of ongoing study focused on the discovery of natural antioxidants from Korean plants by measuring the DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging effect and superoxide quenching activity, ethanol extract of the aerial parts of Curcuma longa (Zingiberaceae) was found to show potent antioxidant activities. Subsequent activity-guided fractionation of the ethanol extract led to the isolation of two phenolic compounds, 1,2,3,4,6-penta-O-galloyl-${\beta}$-D-glucopyranoside (1) and gallic acid (2), as antioxidant compounds. Their structures were elucidated by spectroscopic studies. Compounds 1 and 2 were isolated for the first time from this plant. These compounds showed the significant antioxidative effects during the DPPH free radical scavenging test, and the riboflavin- and xanthine-originated superoxide quenching activity tests.
Effects of methanol extract from turmeric (Curcuma longa L.) (CME) on underlying mechanisms of lipolysis were investigated in 3T3-L1 adipocytes. Compared to the control, lipid accumulation with 72 hr treatment of CME at the concentration $20\;{\mu}g/mL$ was significantly decreased by 19.9% as quantified by Oil red O dye. Intracellular triglyceride (TG) content was also lowered by 19.3%. To determine the mechanism for TG content reduction, glycerol release level was measured. Incubation of 3T3-L1 adipocytes with 15 and $20\;{\mu}g/mL$ of CME significantly elevated the level of free glycerol released into the cultured medium by 20.4 and 28.6%, respectively. In subsequent measurements using quantitative real-time polymerase chain reaction (PCR), mRNA levels of hormone-sensitive lipase (HSL) and adipose triglyceride lipase (ATGL) were significantly increased by 21.2 and 24.9%, respectively, at the concentration $20\;{\mu}g/mL$. Results indicated that CME stimulated lipolysis through induction of HSL and ATGL mRNA expressions, resulting in increased glycerol release.

Periodontal disease is triggered by the host immune response to pathogens in the microbial biofilm. Worsening of periodontal disease destroys the tooth-supporting tissues and alveolar bone. As oral inflammation can induce systemic diseases in humans, it is important to prevent periodontal disease. In this study, we demonstrated that Curcuma xanthorrhiza supercritical extract (CXS) and its active compound, xanthorrhizol (XAN), exhibit anti-inflammatory effects on lipopolysaccharide (LPS)-treated human gingival fibroblast-1 cells and anti-osteoclastic effects on receptor activator of nuclear factor kappa B ligand (RANKL)-treated RAW264.7 cells. LPS-upregulated inflammatory factors, such as nuclear factor kappa B p65 and $interleukin-1{\beta}$, were prominently reduced by CXS and XAN. In addition, RANKL-induced osteoclastic factors, such as nuclear factor of activated T-cells c1, tartrate-resistant acid phosphatase, and cathepsin K, were decreased in the presence of CXS and XAN. CXS and XAN inhibited the mitogen-activated protein kinase (MAPK)/activator protein-1 (AP-1) signaling pathway. Collectively, these results provide evidence that CXS and XAN suppress LPS-induced inflammation and RANKL-induced osteoclastogenesis by suppressing the MAPK/AP-1 pathway.
Plant extracts have been studied due to their potential as photoprotective agents against UV and blue light exposure. Previous studies have revealed that several plant extracts have photoprotection capacities and synergistic effects with synthetic products. However, such results for pu'er tea and Curcuma longa L. have not been reported yet for a cosmetic formulation. Thus, the objective of this study was to evaluate photoprotection capacities of pu'er tea and C. longa L. extracts for a sunscreen compound. The pu'er tea extract improved sun protection factor value of 2-ethyl-hexyl methoxycinnamate (a synthetic sunscreen compound) by 46% and showed a high antioxidant capability that could help skin recover from photo-induced damage. C. longa L. extract also showed a potential to protect skin from blue light-induced damage because it not only had a maximum absorption peak at the blue light range, but also protected human fibroblasts from blue light-induced damage. The addition of both extracts shifted the critical wavelength of 2-ethyl-hexyl methoxycinnamate from 350 nm to 386 nm, giving it a broad-spectrum feature. Thus, pu'er tea and C. longa L. extracts may enhance the photoprotection ability of synthetic sunscreen products.

The purpose of this study was to examine the potential of turmeric (Curcuma longa L.) to inhibit 7,12-dimethylbenz[a]anthracene (DMBA)-induced mammary carcinogenesis in rats. Female Sprague Dawley rats were fed a control diet (NC and DC) or an ethanol extract of turmeric (DT) diet until the end of the experiment. The rats in the DC and DT groups were administered a single dose of DMBA (50 mg/Kg) by oral gavages at 50 days of age. The turmeric ethanol extracts decreased the incidence and multiplicity of DMBA-induced mammary tumor. The turmeric ethanol extract significantly decreased the tumor cell proliferation. The turmeric also significantly decreased the tumor grade based on the degree of the tubule formation. The results suggest that the ethanol extract of turmeric has an inhibitory effect against mammary carcinogenesis, and that such chemopreventive effect may be related to the inhibition of the initiation and the proliferation of tumor cells.
Irradiated and non-irradiated Korean medicinal herbs were extracted by water and 70% ethanol. Antimicrobial activity of these extracts were invesigated against selected food hygiene microoganisms. The ethanol extracts of the non-irradiated Agrimonia pilosa ledebour japonica Nakai, Curcuma longa Linne and Angelica gigas Nakai were completely inhibited on four species of bacteria, such as Vibrio parahaemolyticus, Clostridium perfringenes, Bacillus subtilis and Staphylococcus aureus. Also, above four strains did not have antibacterial activity in the water-ethanol mixtures. Futhermore, the ehtanol extracts of the non-irradiated Agimonia pilosa ledebour Japonica Nakai, Curcuma Iedoaria Roscoe, Curcuma longa Linne and Scutellaria baikalensis George were shown inhibitory effects against Aspergillus flavus and Pen icillium islandicum. And the water extract of Scutellaria baikalensis George was the same effect to these molds. Essentially the same results were observed when samples irradiated at a dose of 10 kGy.
The methanolic extract of the Rhizome of Curcuma zedoaria exhibited a significant prolongation of hexobarbital (HB)-induced hypnosis. Through liquid chromatography of an ether soluble fraction. monitoring by bioassay, three sequiterpenes, germacrone (A), curzerenone (B) and germacrone epoxide (C) were isolated as active consituents. A single treatment (100-200 mg/kg, i.p.) of each compound showed not only a significant prolongation of HB-induced sleeping time but also a significant inhibition of aminopyrine N-demethylase activity in mice, and further exhibited a typical type I binding spectra with oxidized rat hepatic cytochrome P-450 induced by phenobarbital. All of the compounds provoked a sleep episode at a subhypnotic dose of HB, implying that they possess CNS depressant properties.
This study was conducted to evaluate the effects of adding gradually increasing concentrations of turmeric extract (0, 0.1, 0.2, and 0.4%) to smoked duck meat on its chromaticity, antibiosis, and antiseptic degree against food poisoning bacteria, number of bacteria, lipid rancidity, sensory evaluation, and preference. The brightness, red color intensity and yellow color intensity changed significantly when 0.2% turmeric extract was added and the sodium nitrate concentration was reduced. Additionally, no antibiosis or antiseptic activities against food poisoning bacteria were observed in any turmeric treated samples, whereas the number of bacteria was increased in control samples compared to turmeric treated samples after 10 days of preservation. The TBARS value decreased during storage when turmeric extract concentration increased, resulting in positive sensory evaluation of its color, succulence, taste and hardness. In the preference test, the surface and meat color increased as the concentration of turmeric extracts increased. Hardness was highest at 0.2%, whereas taste was highest between 0.2 and 0.4%. The overall preference test was highest for the 0.2% extract samples. Overall, the results indicated that addition of 0.2% turmeric to smoked duck meat will lead to better nutrition, function, and overall preference.

Previous work demonstrated that an ethanol extract (HS0608) of a mixture of three medicinal plants of Curcuma longae radix, Phellinus linteus, and Scutellariae radix markedly inhibits $A{\beta}$ (25-35)-induced neurotoxicity. The present study was performed to further verify the neuroprotective effect of HS0608 on oxidative and ischemic cerebral injury using cultured rat cortical neurons and rats. Exposure of cultured cortical neurons to $100\;{\mu}M$ hydrogen peroxide ($H_2O_2$) induced neuronal apoptotic death. At $10-100{\mu}g/ml$, HS0608 inhibited neuronal death, elevation of intracellular calcium concentration ($[Ca^{2+}]_i$), and generation of reactive oxygen species (ROS) induced by $H_2O_2$ in primary cultures of rat cortical neurons. In vivo, HS0608 prevented cerebral ischemic injury induced by 2-h middle cerebral artery occlusion (MCAO) and 24-h reperfusion. The ischemic infarct and edema were significantly reduced in rats that received HS0608 (200 mg/kg). These results suggest that the anti-oxidative properties of HS0608 may be responsible for its neuroprotective effect against focal cerebral ischemic injury and that HS0608 may have a therapeutic role in neurodegenerative diseases such as stroke.
