Development of a Numerical Model for Prediction of the Cooling Load of Nutrient Solution in Hydroponic Greenhouse (수경온실의 양액 냉각부하 예측모델 개발)

  • Cooling of nutrient solution is essential to improve the growth environment of crops in hydroponic culture during summer season in Korea. This study was carried out to provide fundamental data for development of the cooling system satisfying the required cooling load of nutrient solution in hydroponic greenhouse. A numerical model for prediction of the cooling load of nutrient solution in hydroponic greenhouse was developed, and the results by the model showed good agreements with those by experiments. Main factors effecting on cooling load were solar radiation and air temperature in weather data, and conductivity of planting board and area ratio of bed to floor in greenhouse parameters. Using the model developed, the design cooling load of nutrient solution in hydroponic greenhouse of 1,000$m^2$(300pyong) was predicted to be 95,000 kJ/hr in Suwon and the vicinity.

Molasses clarifying method used by lime-phosphate for yeast culture (효모배지용 당밀의 청징법에 관하여)

  • The constituents of molasses and effect of pH precipitate formation in molasses solution, vary according to its producing districts. The formation of precipitation is not so changeable in the range of buffering zone of molasses solution(pH4.3-6.3) in philippine molasses according to the change of pH value. On lower or higher than the range of buffering zone, the precipitation is increased from pH 4.3 to 2.8 and from 6.3 to 8.1, it is decreased when pH value is lower or higher than the pH value range. For molasses clarifying, it had better adjust the pH of molasses solution to neutral or weak alkali range out of the alkai side of the buffering zone, with lime solution. And then, add the calcium super phosphate solution to pH value of alkali side in buffering zone, as much as the pH of clarified molasses solution can reach to middle value in buffering zone. For the equilibrium of pH value on clarifying molasses, it takes plenty of time more than 6 hours.

Continuous Production of Pullulan by Aureobasidium pullulans HP-2001 with Feeding of High Concentration of Sucrose

  • In this study, glucose, sucrose, and dextrin were found to be better carbon sources for the production of pullulan by Aureobasidium pullulans HP-2001. Maximal production of pullulan with 200 g/l sucrose as a carbon source was 54.2 g/l. The highest yield of pullulan from sucrose was 0.40, when the sugar concentration was 100 g/1. Optimal conditions for the continuous production of pullulan by A. pullulans HP-2001 in a 7-1 bioreactor were determined by studying the effects of composition of feed solution, dilution rate, and concentration of sucrose in the feed solution. Pullulan concentration and productivity with 100 g/l glucose and 2.5 g/l yeast extract were 38.1 g/l and 0.53 g/l h for 72 h, respectively, in a batch culture of A. pullulans HP-2001. When the substituted medium contained 100 g/l sucrose, 2.5 g/l yeast extract, and mineral salts, which is the same composition as the medium for the production of pullulan, the pullulan concentration and productivity were 74.9 g/l and 0.55 g/l h for 120 h, respectively. The production of pullulan at the steady state increased with a dilution rate up to 0.015/h, and its concentration was 78.4 g/l with a weight average molecular weight ($M_w$) of $4.0{\times}10^5$. Unlike a batch culture, however, the decline of the $M_w$ and the number average molecular weight ($M_n$) of pullulan was not found in the continuous culture of A. pullulans HP-2001. When the concentration of sucrose in the feed solution was 200 g/l, 113.5 g/l of pullulan was obtained at the steady state. The steady state was maintained longer in the continuous culture fed with the feed solution containing 200 g/l sucrose than when fed with the feed solutions containing either 100 or 150 g/l sucrose.

Analysis of Domestic Patent Information on Hydroponics Field (수경재배 분야의 국내 특허 정보 분석)

  • This study was conducted to collect the basic data fo the development ot hydroponic technique and the efficient promotion of water culture. We have also analysed the water culture associated domestic patent application informations searching for on-line internet site with an admission fee (http://www2.wips.co.kr) from the year 1983 to January, 2006. Fifty nine patent applications related to the hydroponics were searched for in this study. Main patent contents applied were as followed in the order of that nineteen applications about the hydroponic equipments by 32.2%, thirteen applications about the materials used in water culture by 22.0%, eleven applications about the composition of nutrient solution by 18.6%, nine applications about the cultural methods by 15.3%, and seven applications about the cultural media by 11.9%. The compositions of nutrient solution and the cultural methods intended for specific crops were increased in number following five applications about tomato plants and four applications about potatoes. Thirty three patents (55.9%) were mainly applied before the year 2000. Main patent contents were the compositions of nutrient solution before the year 2000 in contrast to the cultural methods and substrates after the year 2000.

Management of Nutrient Solution Based on $\textrm{NH}_4\textrm{H}_2\textrm{PO}_4$Concentration in Deep Flow Culture of Cherry Tomato (방울토마토 담액재배시 $\textrm{NH}_4\textrm{H}_2\textrm{PO}_4$ 농도에 기초한 배양액 조절)

  • This study was carried out to investigate the effect of NH$_4$H$_2$PO$_4$ on pH stabilization in deep flow culture system using tap water, and to determine the optimum range of NH$_4$H$_2$PO$_4$ in culture solution. The pH of tap water is 7.5. The higher the concentration of NH$_4$H$_2$PO$_4$ was, the more the pH of nutrient solution was decreased. In NH$_4$H$_2$PO$_4$ 4/3-5/3 me/$\ell$, the pH of nutrient solution was 6-7.5 during the experiment. The highest brix(%) was obtained in NH$_4$H$_2$PO$_4$ 5/3-6/3 me/$\ell$. Leaf length, leaf width and stem-base diameter were highest in NH$_4$H$_2$PO$_4$ 2/3 me/$\ell$. The L and b* values were highest and the a* value was lowest in NH$_4$H$_2$PO$_4$ 8/3 me/$\ell$. Toxicity symptom of ammonium appeared in NH$_4$H$_2$PO$_4$ 8/3 me/$\ell$. It suggests that there was the relationship between leaf color and growth condition. It was concluded that NH$_4$H$_2$PO$_4$ 2/3 me/$\ell$ was good before harvest stage and NH$_4$H$_2$PO$_4$ 5/3-6/3 me/$\ell$ at harvest stage.

Development of Optimal Nutrient Solution of Cherry Tomato (Lycopersicon esculentum Mill. 'KoKo') in a Closed Soilless Culture System (순환식 수경재배에 적합한 방울토마토 '꼬꼬' 배양액 개발)

  • The experiment was conducted to investigate the nutrition absorption pattern in the growth stage and develope the optimal nutrient solution hydroponically grown the cherry tomato 'Koko' in closed substrate culture system with the nutrient solution of National Horticultural Research Station in Japan into 1/2S, 1S, and 2S. When plant was grown in 1/2S, the growth and yield were high and the pH and EC in the root zone were stable. Suitable composition of nutrient solution for cherry tomato was $NO_3-N\;6.8,\;PO_4-P\;2.7,\;K 3.2,\;Ca\;3.6\;and\;Mg\;1.1\;me\;L^{-1}$ in the early growth stage, $NO_3-N\;7.3,\;PO_4-P\;2.2,\;K\;3.7,\;Ca\;3.6;and\;Mg\;1.1\;me\;L^{-1}$ in the late growth stage by calculating a rate of nutrient and water uptake. To estimate the suitability for the nutrient solution in a development of cherry tomato developed by Wongkwang university in Korea (WU), plant was grown in perlite substrate supplied with different solution and strengths(S) by research station for greenhouse vegetable and floriculture in the Netherlands (Proefstation voor tuinbouw onder glas th Mssldwijk; PTG) of 1/2S, 1S, and 2S, respectively. The growth was good at the PTG and WU 2S in the early stage and the PTG of 1S and WU of 1S and 2S in the late stage. The highest yield of cherry tomato obtained in the WU of 2S. pH and EC in root zone of WU of 2S were stable during the early and late growth stage. Therefore when cherry tomato plant was grown in WU of 2S of EC $1.6{\sim}2.0\;dS\;m^{-1}$in the nutrient solution, not only stable growth and yield but also fertilizer reduction can be obtained than that of PTG.

Hydroponic Culture of Leaf Lettuce Using Mixtures of Fish Meal, Bone Meal, Crab Shell and the Pig Slurry Leachate of Woodchip Trickling Filter (목편살수여상 침출액비와 어분, 골분, 게껍질 혼합액을 이용한 상추의 수경재배)

  • The pig slurry leachate was dark brown-colored solution that leaches out of woodchip trickling filter. The purpose of this research was to investigate the effect of pig slurry leachate and byproduct on growth characteristics of leaf lettuce in hydroponics culture. The effects of addition of fish meal, bone meal and crab shell for the growth of leaf lettuce were investigated. Leaf lettuce were grown in each of six combination treatment solutions; slurry leachate, slurry leachate + fish meal, slurry leachate+bone meal, slurry leachate + crab shell and chemical hydroponic solution for lettuce based on EC content. The chemical nutrient solution was the solution of National Horticulture Research Station for the growth of lettuce. The all of nutrient solution was adjusted 1.5 mS/cm in EC in hydroponics culture. 1. The pH level of leachate of trickling filter was increased and EC decreased gradually during treatment. Pig slurry leachate was low in suspended solids (SS), phosphorus (P), but rich in potassium (K). 2. The plot of slurry leachate (SL) was lowest in the growth characteristics of lettuce. The leaf length and width of lettuce treated with mixture plot of slurry leachate and fish meal (SL + FM) was higher compared with plot in slurry leachate. The chlorophyll reading was reduced in plot treated with slurry leachate, but that in plot of SL+FM was similar compared with control plot. 3. The fresh weight of lettuce showed lowest in the plot treated with slurry leachate. The addition of fish meal increased the yield of comparing plot of slurry leachate, but plots of bone meal and crab shell addition were not significantly difference. The fresh weight of leaf lettuce in plot of SL+FM was 87% as 400.0g compared with control. In conclusion, the mixture solution of pig slurry leachate and fish meal could be used as a nutrition solution of organic lettuce hydroponics.

A Study on Implementation of Culturenomics based on Placeness (장소성을 바탕으로 한 컬처노믹스 구현에 관한 연구)

  • The study is motivated by methodological approach on how to apply the existing traditional placeness to the present age. Moreover, its purpose lies in establishment of character of place that not only embraces the existing meaning of place but also reflects on the new paradigm of space through culturenomics which embraces the core indexes of modern values such as economics, culture, and everything. Culture has a potential to create various values through its convergence with other fields, one of which is place. Place is essentially an original center of human existence. Culture is no more separated from daily life and people's way of life becomes a culture. In the a culture-oriented social system, design of place will be developed in a multi-dimensional way that accepts diversity and centers on relation. At our places experiencing placelessness through uniformity and modernization, this study has a significant meaning since it shows culturenomics may offer a solution to the direction of growth in the future.

Isolation and Culture of Protoplasts from Hypocotyl-derived Callus of Soybean (Glycine max) (대두 (Glycine max) 부배유 유래 칼루스의 원형질체 분리 및 배양)

  • The isolation and culture of protoplasts from hypocotyl-derived calluses of Glycine max (L.) Merr. cv. Jangyeop were obtained by digestion for 6 hrs in an enzyme solution containing 3.5% cellulase, 1.5% macerozyme, 10% sorbitol and 0.1% CaCl2.2H2O at pH 5.8. Newly formed cell wall of protoplasts cultured in MS agar medium containing 10 $\mu$M $\alpha$-naphthaleneacetic acid (NAA) and 32 $\mu$M N6-benzylaminopurine (BAP) could be observed after 24 hrs culture. The first cell division of the protoplasts was observed after 3 days of culture; cell clusters after 2 weeks of culture. When transferred to solid media, the protoplasts formed cell clusters gave rise to proliferating calluses.

Reduction of Aluminum Toxicity by Calcium and Magnesium in Pines (소나무속 식물에서 칼슘과 마그네슘에 의한 알루미늄 독성의 경감)

  • Alleviation of Al toxicity by Ca and Mg was studied with seedings of Pinus densiflora, P. rigida and P. thunbergii under the solution culture. The seedlings were cultivated in the enriched Can and Mg nutrient solution with 2, 000 ${\mu}M$ Al for three weeks. The hightest total root lengths of P. densiflora, P. rigida and P. thunbergii increased by 21, 33 and 133% in Ca enriched solution, and 23, 44 and 107% in Mg enriched solution, respectively. Ratios of lateral root to main root length increased significantly in Mg enriched solution, and redution of Al toxicity was more affected by enriched Mg than by enriched Ca. Al content in tissue was reduced by enriched Ca and Mg.
