인삼 (Panax sp.) 재배를 위한 미세기상조건 (The Micrometeorological Requirements for the Culture of Ginseng (Panax sp.))

  • 인삼은 한국, 일본, 소련, 북미에서 재배되고 있다. 이들 생산지역의 미세기상을 각각 조사해 보면, 인삼의 생육에 필요한 필수조건을 갖고 잇으며 각 지역에 따라 미세기상은 다른 방법으로 조절되고 있다, 한국, 중국에서의 최근연구자료를 비교해 보면 인삼생산에 맞는 여러가지 미세기상 조건들을 구명 할 수 있을 것이며, 여러가지 해가림 재료에 따른 일광차단에 관한 연구도 미세기상 조건구명의 한가지 방법이 되고 있다. 북미에서는 나무와 polypropylene 해가림 재료가 사용되지만, 중국에서는 마른 풀을 엮은 이엉사이에 polyvinylchloride film을 넣어 해가림을 한다. 이러한 해가림 재료들이 인삼생육과 수량에 미치는 효과는 뿌리생육에 있는 것 같으며, 생육기간 중 대부분 뿌리의 생육 적온에 미치지 못하고 있다. 년근별 생육자료를 분석해 보면, 근권의 토양온도가 지상부와 뿌리의 건물 분배에 미치는 효과를 구명하는데 유용할 것이다.

송이의 세포외 분비 $\beta$-Glucosidase 효소의 특성 (Characteristics of Extracellular $\beta$-Glucosidase in Tricholoma matsutake)

  • Cellulose 분해에 관련된 $\beta$-Glucosidase 효소 활성의 특성 파악을 위하여 송이균사(Tricholoma matsutake DGUM 26001)의 액체 배양시 세포외로 분비되는 $\beta$-Glucosidase 효소를 부분정제하여 그 특성을 조사하였다. 효소 활성에 미치는 적정 온도는 55-$70^{\circ}C$이었고 최적 온도는 $65^{\circ}C$이었다. 적정 효소활성에 영향을 주는 적정 pH는 3.0-5.0 범위였으며 최적 pH는 4.0이었다. Salicin을 기질로 최적 조건하에서 $\beta$-Glucosidase 효소의 비활성도는 18.7 unit/mg protein이었다. 열안정성은 $60^{\circ}C$이하의 온도에 60분간 열처리시 약 90%이상의 효소활성을 유지하였다. $Fe^{++}$이온은 효소활성을 촉진하였으나, $Hg^{++}$$Cu^{++}$이온은 효소활성을 매우 억제하였다. Salicin에 대한 효소활성을 100으로 하였을 때, cellobiose는 48.6%의 상대적 효소활성을 나타내었으며, cellobiose에 대한 Km값 및 Vmax값은 각각 0.12mM과 0.02umol/min었다.

  • Poly-${\beta}$-hydroxybutyrates(PEB)는 토양에서 완전히 이산화탄소와 물로 분해되는 세포 내에 생성되는 고분자 물질로 잘 알려져 있다. Alcaligenes eutrophus의 세포 성장과 PHB의 생산성을 증가시키기 위해서 가압배양법이 사용되었다. 실험 데이타를 다양한 가압과 온도하에서 탄소 공급원과 기체성분의 영향에 관해서 분석한 결과 0.0075vvm의 기체수소를 공급한 배양이 수소를 공급하지 않은 배양에 비해 더 나은 PEB 생산성을 나타내었고, 6atm, $30^{\circ}C$에서 산소의 성분비를 2%에서 8%로 변화시켰을 때 더 높은 수율과 PEB의 생산성을 얻을 수 있었다.

Streptomyces griseus Trypsin (SGT) Has Gelatinase Activity and Its Proteolytic Activity Is Enhanced by Manganese

  • Gelatinase is a proteolytic enzyme that hydrolyzes gelatin. Gelatinolytic activity was detected from culture broths of Streptomyces griseus IFO13350 and HH1 by paper disc assays on 0.5% agar plates containing 1% gelatin. The concentrated extracellular protein from the S. griseus was analyzed by SDS polyacrylamide gel, and two proteins, with molecular weights of 30 and 28 kDa, respectively, were identified to have gelatinase activity by gelatin zymography. The protein with a molecular weight of 28 kDa was confirmed to be S. griseus trypsin (SGT). The effects of metal ions and metal chelators on the protease activity of the SGT were studied. Of the metal ions tested, only manganese was found to enhance the protease activity, 2.6 times, however, $Co^{2+},\;Cu^{2+},\;and\;Zn^{2+}$, and metal chelators, such as EDTA and EGTA, inhibited the SGT activity. When the protease activity of the SGT was measured at various pHs, in the presence of 5 mM $MnCl_2$, its highest activity was at pH 11.0, whereas only 60% of the maximum activity was observed between pHs 4.0 and pH 6.0, and almost 80% activity between pHs 7.0 to pH 10.0. The protease activity was measured at various temperatures in the presence of 5 mM $MnCl_2$. The SGT was found to be stable up to $60^{\circ}C$ for 30 min, while only 16% of the enzyme activity remained at $60^{\circ}C$, and at $80^{\circ}C$ almost all the activity was lost. The optimal temperature for the protease activity was $50^{\circ}C$.

Antioxidant Activity of Yogurt Fermented at Low Temperature and Its Anti-inflammatory Effect on DSS-induced Colitis in Mice

  • This study was performed to evaluate the antioxidant activity of yogurt fermented at low temperature and the anti-inflammatory effect it has on induced colitis with 2.5% dextran sodium sulfate (DSS) in Balb/c mice. Yogurt premix were fermented with a commercial starter culture containing Lactobacillus acidophilus, Bifidobacterium lactis, Streptococcus thermophilus, and Lactobacillus delbrueckii subsp. bulgaricus at different temperatures: $22^{\circ}C$ (low fermentation temperature) for 27 h and $37^{\circ}C$ (general fermentation temperature) for 12 h. To measure antioxidant activity of yogurt samples, DPPH, $ABTS^+$ and ferric reducing antioxidant potential (FRAP) assays were conducted. For animal experiments, inflammation was induced with 2.5% DSS in Balb/c mice. Yogurt fermented at low temperature showed higher antioxidant activity than that of the yogurt fermented at general temperature. In the inflammatory study, IL-6 (interleukin 6) was decreased and IL-4 and IL-10 increased significantly in DSS group with yogurt fermented at general temperature (DYG) and that with yogurt fermented at low temperature (DYL) compared to that in DSS-induced colitic mice (DC), especially DYL had higher concentration of cytokines IL-4, and IL-10 than DYG. MPO (myeloperoxidase) tended to decrease more in treatments with yogurt than DC. Additionally, yogurt fermented at low temperature had anti-inflammatory activity, although there was no significant difference with general temperature-fermented yogurt (p>0.05).

Field experiment on effect of butaphosphan and cyanocobalamin complex on the immunity and stress of olive flounder at low temperature

  • In this study, a fish metabolic accelerator (a combination of butaphosphan and cyanocobalamin [BPC]) was injected into the muscle of the olive flounder, Paralichthys olivaceus, to investigate its effect on immunity and stress in fish maintained at low temperatures. A single dose of BPC was injected (100 mg/kg body weight) into the olive flounder, and its immunity and stress were observed after one and two weeks. Immunity tests revealed the presence of lysozyme (LZM), nitroblue tetrazolium (NBT), myeloperoxidase (MPO), anti-protease (AP), glutathione peroxidase (GPx), and total immunoglobulin (TIg). BPC injection was found to increase immunity activity compared to the control group. In particular, there was significantly high GPx activity. There was similarly high activity for MPO and GPx in the first week following the injection, followed by significant differences between the BPC-injected and control groups in the second week. There was a reduced low water-temperature stress response in the BPC-injected fish, as evidenced by the cortisol and glucose levels of the control and BPC groups. Lower levels were also observed in the BPC group than the control group during the second week. Cortisol levels were significantly lower in the BPC group than the control group. Histological examinations were conducted in the first and second weeks after the intramuscular injection of the recommended BPC dose to confirm the safety of BPC in aquaculture. There were no abnormalities observed in any tissue samples. This study confirms that the injection of BPC is safe even when used in a culture situation. BPC helps relieve stress and improves non-specific immune responses (innate immunity) in the olive flounder.

항곰팡이능 보유 유산균의 숙성치즈 적용 연구 (Application of Lactic Acid Bacteria to Inhibit Fungal Contamination of Cured Cheeses)

  • 항균활성을 가진 유산균은 생균제 등의 형태로 식품 중 부패나 식중독을 유발하는 유해균을 저해하는데 효과적으로 활용할 수 있다. 이에 본 연구에서는 된장에서 분리한 유산균인 P. pentosaceus M132-2 균주를 주요 오염 곰팡이 3종을 대상으로 분석한 결과, 이들의 생장을 모두 저해하여 항곰팡이능을 보유한 것을 확인하였다. 또한 M132-2 균주는 우수한 내염성과 저온내성을 가지고 있어 치즈의 염도와 저온의 숙성온도에서도 생존이 가능할 것으로 판단된다. 마지막으로 M132-2 상등액을 실제 고다 치즈에 적용한 결과, 오염 곰팡이 생장이 현저하게 저해되는 것을 확인하였다. 결과적으로 P. pentosaceus M132-2 균주는 치즈를 비롯한 다양한 식품의 부패 방지에 활용할 수 있는 유용한 수단이 될 수 있다.

Subcritical water extraction of Gracilaria chorda abbreviates lipid accumulation and obesity-induced inflammation

  • Obesity-induced inflammation is crucial in the pathogenesis of insulin resistance and type 2 diabetes. In this study, we investigated the effects of the Gracilaria chorda (GC) on lipid accumulation and obesity-induced inflammatory changes or glucose homeostasis in cell models (3T3-L1 adipocytes and RAW 264.7 macrophages). Samples of GC were extracted using solvents (water, methanol, and ethanol) and subcritical water (SW) at different temperatures (90, 150, and 210℃). The total phenolic content of GCSW extract at 210℃ (GCSW210) showed the highest content compared to others, and GCSW210 highly inhibited lipid accumulation and significantly reduced gene expressions of peroxisome proliferator-activated receptor-γ, CCAAT/enhancer-binding protein-α, sterol regulatory element-binding protein-1c, and fatty acid synthase in 3T3-L1 adipocytes. In addition, GCSW210 effectively downregulated the pro-inflammatory cytokine regulator pathways in RAW 264.7 macrophages, including mitogen-activated protein kinase, signal transducers and activators of transcription and nuclear factor-κB. In co-culture of 3T3-L1 adipocytes and RAW 264.7 macrophages, GCSW210 significantly reduced nitric oxide production and interleukin-6 levels, and improved glucose uptake with dose-dependent manner. These findings suggest that GCSW210 improves glucose metabolism by attenuating obesity-induced inflammation in adipocytes, which may be used as a possible treatment option for managing obesity and associated metabolic disorders.

QTL Analysis of Plant Height in Rice Using CNDH Population

  • Rice is a staple crop used by more than 50% of the world's population. However, in rapidly changing climates such as abnormal high temperatures and typhoons, the food security of rice is greatly threatened. Plant factories have the advantage of being able to grow crops regardless of climate change, so they can be a response to climate change. However, in plant factories, crops are grown by placing the culture bed vertically, so shorter crops are more efficient. Therefore, in order to search for genes related to the height of rice, QTL analysis was performed by investigating the plant height of Cheongcheong/Nagdong doubled haploids from 2017 to 2021. Plant height of rice investigated for five years showed a normal distribution, meaning that genes related to rice height are quantitative traits. As a result of QTL analysis, a total of 12 QTLs were detected, and QTLs overlapped for 5 years in RM12285-RM212 on chromosome 1. Also, The QTLs of plant height detected in 2019 has a LOD score of 17.64 in RM12285-RM212 region of chromosome 1. As a result of QTL analysis, 44 height-related genes were searched from the detected chromosomes, and among them, Os01g0757200 in RM 12285-RM212 on chromosome 1 region, named OsGA2ox3q1, were selected as genes related to the height of rice. The relative gene expression level of OsGA2ox3q1 was highly expressed in cultivar with short culm lines, and was low expressed in cultivar with long culm lines. OsGA2ox3q1 can be used to breed semi-dwarf cultivar in rice more efficiently.

온도를 달리한 누룩 발효 기간별 배양 유산균 변화 및 분리 유산균들의 효소 활성 (Change in the composition and enzyme activity of culturable lactic acid bacteria in Nuruk during fermentation at different temperatures)

  • 발효 온도를 달리한 전통방식으로 제조된 두 가지 형태(TN-A와 TN-B)의 누룩들로부터 발효 기간별 배양된 유산균 총균수, 유산균 종들의 변화 및 분리된 유산균들의 효소활성을 비교하였다. TN-A의 유산균 총균수는 $10^4{\sim}10^5$ log CFU/mL로 발효 3일차부터 발효 기간에 관계없이 유지되었고, TN-B에서는 발효 6일차까지 $10^2$ log CFU/mL로 유지되다가 6일차 이후 유산균이 검출되지 않는 특징을 보였다. 유산균 종들의 변화에 있어서, TN-A는 발효 3일차에 우점종 유산균으로 나타난P. pentosaceus 종이 발효 종료 시점이 30일차 까지 우점하고 있음을 확인할 수 있었고, TN-B는 발효 3일차와 6일차에 각각 P. pentosaceus와 E. hirea로 나타나 유산균이 검출된 발효 기간까지 TN-A 보다는 많은 변화를 보였다. TN-A와 TN-B으로부터 분리된 유산균들의 효소 활성은 단백질분해효소와 당화효소 활성을 분석하였다. 단백질분해효소평가는 티로신 생성량으로, 당화효소 활성은 아밀로스 분해에 따른 아밀로스 잔여 함량으로 평가한 결과, 분리된 유산균(9종)들은 0.49~0.89 mg/mL의 티로신 생성 범위의 단백질분해 활성과 0.64~0.79 mg/mL의 아밀로스 분해 활성 범위를 보였고, 유산균들 중 가장 높은 단백질분해효소와 당화효소 활성을 보이는 균주는 각각 P. acidilactici와 L. sakei로 확인되었다. 이상의 결과들은 온도를 달리하여 전통적인 방법으로 누룩을 제조하였을 시 누룩 내에 유산균들을 조절할 수 있음을 시사한다. 이를 토대로 향후 많은 연구들이 진행된다면, 누룩관련 제품에 유산균들에 의해 만들어지는 산미와 풍미를 향상 시킬 수 있는 전통적인 누룩제조가 가능할 것으로 판단된다.
