Production of Water-Solubled Pigment from Mycelial Culture of Cordyceps scarabaeicola KEFC-C252 and Its Antimutagenic Effect (Cordyceps scarabaeicola KEFC-C252의 균사체 배양에 의한 수용성 색소의 생산과 색소의 항돌연변이 효과)

  • Cultural conditions for the production of water-soluble pigment from mycelial culture of Cordyceps scarabaeicola KEFC-C252 and antimutagenic activity of the pigment were investigated. To obtain the maximum productivity of the pigment from mycelial culture of C. scarabaeicola KEFC-C252, the optimized medium was made with 1.5% sucrose, 2.5% yeast extract and initial pH 5.5. C. scarabaeicola KEFC-C252 was cultivated to reach the maximum concentration of the pigment at $26^{\circ}C$ for 108 hrs. C. scarabaeicola KEFC-C252 produced about 1.2 g/liter pigment under the optimized condition. The pigment was isolated from the culture filtrate by ethylacetate extraction, acidic precipitation and crystallization. The isolated pigment was scarlet hexagonal column crystal, and the color of the pigment was changed according to pH of the solution. The pigment showed violet in the alkaline water but showed red color in the acidic water. The pigment showed inhibitory activity against mutagenic activity induced by 4-nitroquinoline N-oxide. Furthermore, the pigment showed inhibitory activity against spontaneous mutation on Salmonella typhimurium TA98 and TAlOO.

  • Universal Design is comprised of three main concepts known as barrier-free, adaptive and life-span design. It does not focus on accessibility for the disabled, but rather expands its breadth to the comfortable use and easy access in ail physical environments and facilities. In this study, the researcher views the present and future of universal design through the cases of kitchen design. Based on the market changes in design guidelines and the changes of time uncovered from past studies, this researcher suggests that universal design would develop through mass customization for personalized use, application of new technology and consideration of psychological aspects. Additionally, for the diffusion of universal design appropriate solution in economic cost problem, social recognition and conversion of problem cognizance in designing is needed. In Korea design models of universal design concepts are not yet in production over various fields. In order to develop and study universal design which suits our country, cultural and regional characteristics should be considered as well as physical, perceptual, psychological aspects.

Design of Measuring Trays in the Irrigation System Using Drainage Electrodes for Tomato Perlite Bed Culture (토마토 펄라이트 베드재배시 배액전극 제어법에 적합한 측정틀 설계)

  • Measuring tray as a component in irrigation control system using drainage electrodes was designed and applied for tomato perlite bed culture, and the effectiveness of the irrigation control system was investigated in terms of cultural development and cultivation costs. Five different types of measuring trays equipped with drainage electrodes were tested and the traditional tray was used as the control equipped with time clock. After the first experiment, "Tube-2" was removed because of instability of water content in the substrate. After second experiment, "Tube-1" was removed because of instability of water content in the substrate and low plant yields. In third experiment, "Up-Board" exhibited the best stability in water contents and yields as well as efficiencies in water and fertilizer utilization. The "Up-Board" was the most economical and the easiest system among the tested trays. Therefore, the "Up-Board" system was concluded as the excellent design to apply for the control method using drainage electrodes for tomato perlite bed culture.

  • The cultural grounds of laver has been surveyed using SPOT-5 satellite images to calculate the facilities of laver cultivation area in the coastal waters of Korea 10m resolution multispectral images of SPOT-5 are adopted for the south area of Daebu Island, Hwaseong city to develop an automatic detection approach of laver nets that consists of the following: band difference technique, canny edge detector and morphological analysis. The satellite-based facilities number was relatively high as compared with the licensed number in 2005, 676,749 chaek and 572,745 chaek(柵, unit of measure for laver farm), respectively. The data could be applied to achieve a good harvest for laver seaweed growers and to control its national production keeping a stable market price for the government body.

  • This study aims to analyze how the media represents marriage immigrant women in the reality of Korean multicultural society. In particular, by analyzing an EBS documentary titled, 'Multicultural Fierce Conflicts between Mother-in-law and Daughter-in-law', this study clarifies that this content adopted travel narrative to show conflicts between mother-in-law and daughter-in-law and solution of these conflicts. The documentary apparently shows mutual understanding and cultural diversity but still keeps representing these conflicts as individual's 'trivial' problems. Under this way of representation, social and structural problem or institutional problem in reality can be covered up by the medium of travel. It makes such a conflict look like an individual's 'trivial' problem can be solved by individuals.

  • After observing that most tourists plan to complete their visits to multiple cultural heritage sites within one day, we surmised that for many museum visitors, the foremost thought is with regard to the amount of time is to be spent at each location and how they can maximize their enjoyment at a site while still balancing their travel itinerary? Recommendation systems in e-commerce are built on knowledge about the users' previous purchasing history; recommendation systems for museums, on the other hand, do not have an equivalent data source available. Recent solutions have incorporated advanced technologies such as algorithms that rely on social filtering, which builds recommendations from the nearest identified similar user. Our paper proposes a different approach, and involves providing dynamic recommendations that deploy social filtering as well as content-based filtering using term frequency-inverse document frequency. The main challenge is to overcome a cold start, whereby no information is available on new users entering the system, and thus there is no strong background information for generating the recommendation. In these cases, our solution deploys statistical methods to create a recommendation, which can then be used to gather data for future iterations. We are currently running a pilot test at Chao Samphraya national museum and have received positive feedback to date on the implementation.

  • The area of domestic urban design and architecture has established its original characteristics as various programs and functions were created in the drastically changing society. Such features have been presented in a certain type of patterns or unique formations, which demand a gradual development of designs in harmony with the functions and roles of the city and thus designers are forced to continue thinking deeply on this matter in their own perspectives. In particular, such demands are outstandingly high in reality in historical cities such as Andong, which this study works on. As the design contest winner is based on the geographical characteristics and locality, Confucianism and seonbi(Korean classical scholar) spirit, which are regarded as a philosophy of value, how the traditional concepts could be reflected in the plan was the focus of the discussion. The conclusions deduced from such a background include the following. First, it is necessary to make an approach on the basis of the concept in traditional architecture at the complex planning level based on the ideology in the implementation plan that requires traditionality for a complex design with contemporary functions. Second, it is important to approach this as an exploration of a solution to globalize Confucian culture in the future based on the cultural characteristics of the region and the environmental characteristics of the surroundings. Third, beginning from a concept that explains the complex to a concept that explains individual buildings, it is necessary to reflect characteristics in which the realization of a traditional space is planned in three dimensions.

Stability of Soybean Isoflavone Isomers According to Extraction Conditions

  • Stability of soybean isoflavone isomers according to extraction conditions such as temperature, pH, and extracting solvents was investigated. Heating induced three chemical reactions to occur for malony1 derivatives of isoflavones, namely decarboxylation of malony1 groups into acety1 derivatives, deesterification of malony1 residues, and hydrolysis of $\beta$-glycosidic bonds. Among the twelve isoflavone isomers, change in concentrations of acety1glycosides were most pronounced: Acety1 derivatives were present only in trace amounts in unheated hypocotyls, but the content increased dramatically during heating. As for the glycosides, concentrations of daidzin and glycitin increased due to heat treatment, though that of genistin remained almost unchanged. Heat decomposition rates and the patterns differed among the three malony1 derivatives. After 120 min at $80^{circ}C$, the relative concentrations of daidzin, glycitin and genistin were increased from $9.2\%$, $12.4\%$ and $3.3\%$ to $19.3\%$, $21.9\%$ and $6.2\%$, respectively. When crude isoflavones were solubilized in glycine buffer (pH 10.0) and incubated at $80^{circ}C$, deesterification occurred faster than at pH 7.0. When the pH of isoflavone solution was increased, the malony1glycosides were hydrolyzed to their respective glycosides at increased rate. Both acetyl and aglycone forms were unchanged and only de-esterification reactions occurred. At the acidic pH, malonylglycosides were much stable both at 60 and $80^{circ}C$. However at pH 10, $80^{circ}C$ and 1 hr, $75-80\%$ of malonylglycosides were transformed to their deesterified glycosides. When isoflavones were extracted with $60\%$ aqueous ethanol at $60^{circ}C$, isoflavone isomers were stable and the deesterification reactions did not occur in these conditions. However, at $80^{circ}C$ deesterification of malonyiglycosides occurred significantly with $15-20\%$ of malonylglycosides being hydrolyzed into their respective glycosides. This experiment showed that malonylglycosides undergo decomposition when heated or exposed to alkaline conditions. Also, aqueous ethanol was preferred to aqueous methanol as solubilizing media for obtaining extract with minimum degradation of malonylglycosides.

  • Today, people are bombarded with information, high-technology and multimedia. With that in mind, museum can no longer attract visitor with traditional concept, so it must find some solution or stimulation to attract their attention back. New museum is becoming a experimental space for new culture, new education, and most of all a place to fulfill one's satisfaction of life. Over the past centuries, Guggenheim Foundation has done a great job in providing place for art and place to get emotional stimulation. Out of all of their museums, Frank L. Wright's Solomon R. Guggenheim in New York and frank O. Gehry's Guggenheim Museum in Bilbao had most unique character yet share similar feature to be recognized as the architectural landmark of the 20th & 21st century Although there are close to 50 years of gap between two museums, their unique, attractive, site-specific, emotional value comes from the two most innovative architect of the past and present century. For Wright the Guggenheim was his last project but it had one of the greatest impact on his career and for Gehry the museum gave him confidence that anything is possible. This study will focus on the spatial organization as well as the architectural formation of both museum to compare and analyze. The goal of the research is to give fundamental data which will reveal emotional elements as well as concepts from each work. The content of this research will mainly focused on their architectural philosophy and it will reveal their concept on human emotion as well as the characteristics of the spatial organization. The result of the study will be a valuable reference for people designing art & cultural facilities in the future.

A Study on the Failure Factors of Popular Use of International Domain Names (IDNs): Focusing on the International Standardization Process (다국어도메인의 대중화 실패 요인 탐색: 국제표준화 과정 분석을 통하여)

  • This research analyzes the international standardization process by observing the international communication sources in order to understand the reason of its current poor use. Using the 'organizing theory' frame, we observe the initial discourse on the need of internationalized domain names in ICANN and the process of negotiation for technical choice of layers. Lastly, we take Korean domain names as a case study in order to understand the conflict and the cooperation of different stakeholders. We summarize the factors of failure of IDN as follows. First, the need of IDN in the beginning was raised around non-English speaking countries, in Asia and Middle East, with the discourse on 'digital divide solution and cultural value' ICANN rather pursues the 'technical stability of Internet Infrastructure', which made its standardization take as long as 10 years. As a result, a variety of standards and services are proposed in the marketplace, which engendered inefficient competition and domain name-related disputes such as cybersquatting, technical instability and confusion of users. In addition, the government agencies fail to present the appropriate policies adjusting multiple interests of different stakeholders.
