Symptom Determinant as RNA3 of Lily Isolates of Cucumber mosaic virus on Zucchini Squash

  • Three isolates of Cucumber mosaic virus (CMV) from lily plants showing mosaic and distortion symptoms were detected by reverse-transcriptase polymerase chain reaction (RT-PCR) using primers specific to Cucumovirus genus namely, LK-CMV, LK4-CMV, and LKS-CMV. Restriction enzymes patterns of the RT-PCR products revealed that the lily isolates belonged to subgroup IA of CMV. In terms of biological properties, the lily isolates have highly similar but distinct pathogenicity as reported in other lily strains and ordinary strains of CMV. To characterize the molecular properties, cDNAs containing coat protein (CP) gene and 3' non-coding region (NCR) of RNA3 for the isolates were cloned and their nucleotide sequences were determined. The CP similarity (218 amino acids) was highly homologous (>97%) with that of subgroup I CMV strains. However, an additional 20-nulcleotide long segment was only present in 3' NCR of lily isolates, which form an additional stem-loop RNA structure. By using chimeric construct exchange cDNA containing 3'NCR of LK-CMV into the full-length cDNA clone of RNA3 of Fny-CMV, this additional segment may prove to be significant in the identification and fitness of the virus in lily plants. The pathology of zucchini squash infected by F1F2L3-CMV, a pseudorecombinant virus was showed to change drastically the severe mosaic and stunting symptom into a mild chlorotic spot on systemic leave, compared with Fny-CMV. To delimit the sequence of RNA3 affected the pathology, various RNA3 chimeras were constructed between two strains of CMV. The symptom determinants of F1F2L3-CMV were mapped to the positions amino acid 234, 239, and 250 in 3a movement protein (MP). RNA3 chimeras changed the sequences encoding three amino acids were resulted in alteration of systemic symptom.

Characterization and Sequence Analysis of a Lily Isolate of Cucumber mosaic virus from Lithium tsingtauense

  • A new isolate of Cucumber mosaic virus (CMV), identified as Li-CMV was isolated from a diseased Korean native lily (Lithium tsingtauense Gilg). Biological and serological properties of Li-CMV were characterized, and reverse transcription-polymerase chain reaction (RT-PCR) analysis, restriction enzyme profiling of RT-PCR products, and nucleotide sequence analysis of RNA3 of the virus were performed in this study. Remarkable differences in symptoms between Li-CMV and ordinary CMV strains were found in tobacco plants and Datura stramonium. Li-CMV-infected tobacco plants (cv. Xanthi-nc and cv. Samsun) induced chlorotic ringspots on uninoculated upper leaves, and the symptom expression was delayed or faint whereas, ordinary CMV strains induced green mosaic symptoms on the plant. Systemic infections were observed on Nicotiana benthamiana with severe mosaic symptom. Restriction mapping analysis of RT-PCR products using MspI showed that Li-CMV belonged to CMV subgroup I. A full-length CDNA copy of RNA3 for the virus was amplified by RT-PCR, cloned, and its complete nucleotide sequence was determined. The RNA3 of Li-CMV was 2, 232 nucleotides long, and consisted of two open reading frames of 843 and 657 bases encoding 3a protein (movement protein) and coat protein, respectively. Results of this study indicate that Li-CMV is a novel strain and belongs to subgroup I of CMV in the genus Cucumovirus.

Cucumber Mosaic Virus 1a Protein Interacts with the Tobacco SHE1 Transcription Factor and Partitions between the Nucleus and the Tonoplast Membrane

  • The transcription factor SHE1 was identified as an interacting partner with the cucumber mosaic virus (CMV) 1a protein in the yeast two-hybrid system, by a pull-down assay, and via bimolecular fluorescent complementation. Using fluorescent-tagged proteins and confocal microscopy, the CMV 1a protein itself was found distributed predominantly between the nucleus and the tonoplast membrane, although it was also found in speckles in the cytoplasm. The SHE1 protein was localized in the nucleus, but in the presence of the CMV 1a protein was partitioned between the nucleus and the tonoplast membrane. SHE1 expression was induced by infection of tobacco with four tested viruses: CMV, tobacco mosaic virus, potato virus X and potato virus Y. Transgenic tobacco expressing the CMV 1a protein showed constitutive expression of SHE1, indicating that the CMV 1a protein may be responsible for its induction. However, previously, such plants also were shown to have less resistance to local and systemic movement of tobacco mosaic virus (TMV) expressing the green fluorescent protein, suggesting that the CMV 1a protein may act to prevent the function of the SHE1 protein. SHE1 is a member of the AP2/ERF class of transcription factors and is conserved in sequence in several Nicotiana species, although two clades of SHE1 could be discerned, including both different Nicotiana species and cultivars of tobacco, varying by the presence of particular insertions or deletions.

아마리리스에 모자익병을 일으키는 CMV에 관한 연구 (Isolation and Properties of Cucumber Mosaic Virus Inducing Mosaic Symptoms in Hippeastrum hybridum Hort)

  • 아마리리스에 모자익병을 일으키는 병원바이러스의 기주범위, 물리적성질, 혈청반응을 조사하고 바이러스 입자를 전자현미경으로 관찰을 하였다. 바이러스의 지표식물 검정 결과 C. amaranticolor, V. unguiculata, C. sativus G. globosa둥 21종의 CMV감수성식물에 병징이 나타났다. 바이러스의 내열성은 55C(10분간)이며 내희석성은 $10^{-3}$이고 내보존성은 실온에서 2일이었다. 한천내 확산법을 이용한 항혈청반응결과 뚜렷한 반응대가 형성되었으며, 바이러스흡광도는 260nm에서 최고였으며, 245nm에서 최저였다. 전자현미경에 의한 바이러스입자의 검경결과 직경이 90nm내외인 소구형 입자가 관찰되었다. 이상의 결과를 종합하면 아마리리스에 모자익병을 일으키는 것은 CMV에 의한 것으로 보인다.

수박에 오이녹반모자이크바이러스의 토양전염과 예방대책 (Soil Transmission of Cucumber green mottle mosaic virus and Its Control Mensures in Watermelon)

  • 오이녹반모자이크바이러스(CGMMV)가 오염된 포장에서 이 바이러스병의 토양전염율은 0.2∼3.5%였다. 경작지 토양과 같이 습도가 적당히 유지된 토양에서 CGMMV의 생물활성 지속기간은 17개월 후에는 바이러스의 활성이 손실되었으나, 담수 토양에서는 33개월 이상 이 바이러스의 생물활성이 유지되었다. CGMMV의 토양전염을 억제하기 위하여 정식직전 수박 묘의 뿌리를 10% 탈지분유액에 침지하여 정식한 처리구에서는 이 바이러스병이 발병되지 않았으나, 무처리구에서는 5.0∼7.6%가 발병되었다. 탈지분유액을 처리한 뿌리를 주사전자현미경으로 관찰한 결과 뿌리 주위로 얇은 막이 형성되어져 있었다.

  • Virus disease occurred up to 62% in average in the greenhouse production of table tomato Seokwang in Suwon, Korea. From symptomatic transition of the labeled tomatoes, two different symptoms, mosaic and bud necrosis, were developed independently. Cucumber mosaic virus necrosis strain (CMV-N) was isolated from table tomato showing bud necrosis symptoms. The isolate caused the bud necrosis on four tomato cultivars and locally infected Chenopodium spp. and Vicia faba by mechanical inculation. The 5th RNA segment, satellite RNA, was identified from CMV-N-infected plants by dsRNA analysis. Crystals of virus particles were observed in cytosols and vacuoles. The virus particles of CMV-N presented abundantly in xylem vessel.

Involvement of RNA2 for systemic infection of Cucumber mosaic virus isolated from lily on zucchini squash

  • A lily strain of Cucumber mosaic virus (LK-CMV) was not able to systemically infect zucchini squash (Cucurbita pepo), while Fny strain of CMV (Fny-CMV) caused systemic mosaic and stunting symptom at 4 days post-inoculation on the same host species. The pathogenicity of LK-CMV in zucchini squash was investigated by reassortments of genomic RNAs of LK-CMV and Fny-CMV for infection, as well as by pseudorecombinants generated from biologically active transcripts of CDNA clones of LK-CMV and Fny-CMV, respectively. The assessments of pathogenicity for LK-CMV indicated that RNA2 of LK-CMV was responsible for systemic infection in zucchini squash. In the protoplast of zucchini squash, the RNA accumulations of all constructed pseudorecombinants were indistinguishable and LK-CMV replication was slightly lower than that of Fny-CMV, suggesting that the inability of LK-CMV to infect squash plants was responsible for the poor efficiency of virus movement, rather than the reduction of replication function.

동부(Vigna sinensis)의 국부병반을 이용한 Cucumber mosaic virus와 Broad bean wilt virus 2의 구별 (Discrimination of Cucumber mosaic virus and Broad bean wilt virus 2 Using Local Lesions on Vigna sinensis)

  • 용담에서 전형적인 모자이크 증상을 나타낸 바이러스 이병주 및 황화증상을 보이는 식물체를 채집하였다. 이들 중, 모자이크 증상의 잎을 접종하여 전신감염된 Nicotiana benthamiana로부터의 접종원을 동부의 초생엽에 접종하였을 때, 2가지 형태의 괴사 국부병반이 형성되었다. 즉 하나는 작은 크기 pinpoint 형태의 국부 괴사병반(SNS)이며, 다른 하나는 SNS보다 약간 큰 halo 형의 국부 괴사 병반(LNS)으로 뚜렷하게 구분되었다. 한편 황화 병징의 용담으로부터 증식된 접종원은 동부의 초생엽에 LNS만 형성하였다. 동부에 형성된 SNS 및 LNS형 병반을 각각 단일병반 분리하여 N. bentamiana에 접종하였다. 그 결과 SNS를 접종한 N. bentamiana에서는 접종엽에 무병징, 상위엽에는 전형적인 모자이크 증상이 발현되었다. 반면 LNS를 접종한 식물체에서는 접종엽에 퇴록 윤문이 형성되었으며, 상위엽에는 괴저를 동반한 모자이크 증상이 발현되었다. 이들 SNS 및 LNS 병반으로부터 분리한 바이러스를 dsRNA, RT-PCR 및 혈청학적 유연관계 분석을 통하여 SNS 병반은 CMV에 의해서, LNS는 BBWV2에 의해서 유도된다는 것을 확인하였다. 이 연구의 결과로부터 자연계에서 CMV와 BBWV2가 복합감염되어 병징의 구분이 어려운 경우, 동부에 접종하여 유도된 병반을 이용하여 두 바이러스의 감염을 쉽게 구분할 수 있는 방법으로 활용할 수 있을 것으로 생각되었다.

Construction of Antibodies for Detection and Diagnosis of Cucumber green mottle mosaic virus from Watermelon Plants

  • We immunized BALB/c mice with purified Cucumber green mottle mosaic virus isolate HY1 (CGMMV-HY1). Through the selection of positive clones that were grown on the HAT medium, four sensitive monoclonal clones (CG99-01, CG99-02, CG99-03, and CG99-04) were selected from 500 Hypoxanthine-guanine phosphoribosyltransferase positive hybridoma cells. Four sensitive clones of CGMMV-HYI were determined as IgM type of the subclass of mouse immunoglobulins Ig group. The titer of monoclonal antiserum against CGMMVHY1 was estimated 1:12,800 by the indirect ELISA. Although monoclonal antibodies (MAbs) from CG99-01 and from CG99-04 cross-reacted with Zucchini green mottle mosaic virus and Kyuri green mottle mosaic virus, MAb from the cell line CG99-03 was highly specific to CGMMV. No MAbs cross-reacted with Cucumber mosaic virus-Fny. Only CG99-04 reacted with Pepper mild mottle virus weakly and CG99-02 reacted with both CGMMV and KGMMV. CGMMV was detected from the rind of watermelon fruit by DAS-ELISA of CGMMV-HY1, but not from the flesh of watermelon. Average seed transmission rate of CGMMV in watermelon was $24\%$ from symptomatic watermelon collected from 5 regions of Gyeongnam province. CGMMV was detected by DAS-ELISA with specific MAb of CGMMVHY1 periodically from root stock, during the sequential process for nursery seedling in Haman. Necrotic spots on cotyledons of root stock seedling progressed to reveal the typical symptomatology on the primary leaves of scion upon grafting. Here, we have established MAb based ELISA system, which could accurately detect CGMMV from watermelon seeds, nursery seedlings, transplants and field samples from greenhouse or open out door field as well.

  • Tomato aspermy virus(TAV-B)는 두 개의 다른 CMV균주 D 및 K에서 분리한 satellite RNA의 증식 및 encapsidation에 대하여 helper virus의 역할을 하였다. 이 두 satellite RNA는 역시 ??? system에서 보이는 바와 같이 TAV가 감염된 담배에서 TAV 병징을 크게 감소시켰으며, 이는 감염된 식물내의 Virus 함량의 감소와 관계가 있었다. 또한 CMV가 감염된 토마토에서 lethal necrosis를 유발시킴을 볼 수 있었다.
