First Report of Pseudomonas viridiflava Causing Leaf Spot of Cucumber in Korea (Pseudomonas viridiflava에 의한 오이 점무늬병의 발생 보고)

  • A severe disease with leaf spots and necrotic symptoms was observed in cucumber (Cucumis sativus L.) seedlings in April 2018 at a nursery in Kimjae, Korea (35o 47'09.8"N 127o 2'24.3"E). The infected plants initially showed spots on water-soaked cotyledons which, at later stages, enlarged and spread to the leaves, which the lesions becoming dry and chlorotic. The symptomatic samples were collected from cucumber and the isolates were cultured on LB agar. The representative bacterial strain selected for identification showed fluorescent on King's medium B, was potato rot-positive, levan and arginine dihydrolase-negative, oxidase-negative and tobacco hypersensitivity-positive in LOPAT group 2 as determined by LOPAT tests. A pathogenicity test was carried out on a 3-week-old cucumber. After 3 days of inoculation, leaf spots and necrotic symptoms appeared on the cucumber, similar to the originally infected plants. The infecting bacterial strain was identified as Pseudomonas viridiflava, by 16S rDNA sequence analysis. This is the first report of leaf spot diseases on cucumber caused by P. viridiflava.

Zn Acquisition by Extraradical Hyphae in Arbuscular Mycorrhizal Plant Depending on Zn Nutritional Status of Cucumber(Cucumis sativus cv. Baekdadagi)

  • The contribution of plant nutrition status in arbuscular mycorrhizal (AM) plant to the nutrient acquisition by extraradical hyphae of AM fungi was investigated using cucumber colonized with Glomus intraradicies (BEG 110) focusing on the Zn. Compartmentalized pots with separated Bones for hyphal growth were used to determine the contribution of extraradical AM hyphae to Zn uptake from hyphal zones. $0.5\;{\mu}M$ Zn was supplied into the hyphal zones as nutrient solution (10 mL/day) with a form of $ZnSO_4$. Zn foliar application was made two times for one week before harvest (8 mL/plant). The colonization rate by AM were high in all of Zn treatments. The dry weight of cucumber increased by AM colonization compared to those of non-mycorrhizal counterpart. However: Zn foliar application resulted in no significant difference in dry weight between mycorrhizal- and non-mycorrhizal plant. In addition, the enhancement of Zn content in cucumber shoot by AM colonization were also reduced by Zn foliar application. These results indicate that the interaction between host plant and AM fungus for nutrient uptake might be related to plant nutritional status and nutrient contents. In consequence, higher Zn contents in host plant by foliar application of Zn could restrict the role of extraradical hyphae of AM fungus on the Zn acquisition and transfer from fungus to host plant.

Response to Plant Hormones of Senescence-related Genes for Cucumis sativus L. in Cotyledon Development (오이 떡잎에서 노쇠화 관련 유전자들의 식물 호르몬에 대한 반응 연구)

  • This study was carried out to discover the response of cucumber (Cucumis sativus L.) senescence- associated genes (SAGs) to several plant hormones in detached and developing cotyledon. Accordingly, a collection of cucumber SAGs were examined to characterize their gene expression response through semi-quantitative RT-PCR. Cotyledons were excised at day 14 after seed sowing from plantlets, then incubated in 100 μM each of IAA or zeatin solution for up to 4 days in light and darkness. They were collected at 2-day intervals and used for total RNA extraction and subjected to RT-PCR. Gene expression levels of several cucumber SAGs were significantly changed during the incubation period. More than five cucumber SAGs involving SAG 60 responded to the IAA and zeatin treatment. In the ethylene response study, cotyledons were exposed up to 10 days by ethylene gas. Most of the cucumber SAGs did not show immediate response to ethylene in green cotyledon. The exceptions were PCK, SAG 158, and SAG 288 genes, which responded after 1 day of exposure to green cotyledon, while ICL and SAG 281 revealed strong responses after 10 days of being exposed to yellowing cotyledon. These results suggest that several cucumber SAGs react actively in response to starvation or senescence against exogenously applied stimulus. This induced senescence response is able to understand the SAGs role in lipids and amino acids metabolism partly and function in organ senescence during development.

Comparison of Growth and Yield Characteristics for the Desert Climate Adaptability of European Long- and Medium-sized Cucumber Varieties (유럽계 장과형과 중과형 오이 품종의 사막기후 적응성 검증을 위한 생육 및 수량 특성 비교)

  • This study aimed to examine cucumber (Cucumis sativus) varieties adaptive to the desert climate by comparing and analyzing the growth, yield, and water consumption. Two long-sized cucumber varieties, 'Gulfstream' and 'Imea' and two medium-sized cucumbers, 'Nagene' and 'Sausan' were cultivated in coir substrate hydroponics under hot and humid greenhouse conditions from March 2 to June 20, 2020. On the 113 DAT, 'Nagene' had the longest plant height and the highest internode number. The marketable fruit number per plant was higher in the medium-sized varieties, which had more internode number. The marketable fruit number was 31.3 for 'Gulfstream', 30.7 for 'Imea', 57.8 for 'Nagene', or 56.0 for 'Sausan' with no significant difference in total fruit weights per plant. The water consumption required to produce 200 g of fruit was lower in the 'Nagene' (2.39 L) with the highest water use efficiency (WUE). Therefore, 'Nagene' variety may have higher adaptability to desert high temperature compared to the long-sized varieties, and it is going to be necessary to verify more medium-sized cucumber varieties.

Determination of Daily Amount of N and K Required in Various Growth Stages and Establishment of Diagnostic Criteria Using Petiole Sap Analysis in the Semi-Forcing Culture of Cucumber (반촉성 관비재배 오이의 생육단계별 시비관리를 위한 일일시비량 및 엽병즙액의 농도 기준 설정)

  • This study was conducted to determine the daily application rate and amount of N and K with fertigation during different growth stages in semi-forcing culture of cucumber plants (Cucumis sativus L. cv. Eunseongbaekdadagi). The diagnostic criteria for N and K also investigated based on petiole sap analysis. The dry weight increased slowly until 30 days after transplanting. The highest dry weights were observed at 60 days after planting, then it decreased. As the plant grew, the contents of N and K in the petiole sap and fruit of cucumber decreased. The daily uptake of N and K were highly correlated with the growing days. The $NO_3$ concentrations in petiole sap were in the range from 3,500 to 4,500 mgㆍ$L^{-1}$ in the early growth stage, but those were in the range from 2,000 to 3,000 mgㆍ$L^{-1}$ after then. However, K concentration in petiole sap were in the rang from 5,000 to 7,000 mgㆍ$L^{-1}$ The fluctuation in petiole sap concentration of K was severe in the monthly fertigation and moderate in the daily fertigation. The fertigation by petiole sap diagnosis forced EC of soil to be low and yield to increase compared to the control.

  • DNA markers can determine the genotype of many species. Single nucleotide polymorphism (SNP) detection is difficult without sequencing but it becomes easier with sdCAPS method. Here an experiment was performed for developing molecular markers using two SNPs, CSatpB-SNP and CSycf1-SNP, of chloroplast in cucumber plants. Properly designed primers with nucleotide sequences for restriction enzymes proved success of PCR and efficacy of digestion by the restriction enzymes. Then these markers were used to study the genotyping of cucumber breeding lines and cultivars obtained from various sources in respect of their chilling stress response. We confirmed that a U.S. cucumber line, 'NC76' known to possess a nuclear factor for the chilling tolerance showed the chloroplast genotypes related to chilling tolerance. However all Korean cucumber cultivars tested in this study showed the chloroplast genotypes related to chilling susceptibility. In conclusion, to develop chilling tolerant cucumber, both maternal and a nuclear factors related to chilling tolerance should be transferred from 'NC76' when 'NC76' is used as a female source and other elite lines as recurrent parents.

Oviposition and Feeding Preference of the Cotton caterpillar, Palpita indica(Lepidoptera: Pyralidae), in Cucurbitaceae (박과류에서 목화바둑명나방(Palpita indica)의 산란 및 식이선호성)

  • The adult of the Cotton caterpillar, Palpita indica (Saunder) laid eggs more on mature leaves than on aged and developing leaves. They laid more on leaves than on petiole and stem, and more on the adaxial than on the abaxial surface side of the leaves. Larvae of the Cotton caterpillar showed their preference in the order of the cucumber (Cucumis sativus), gourd (Lagernaria siceraria), watermelon (Citrullus lanatus) > oriental melon (Cucumis melon L. var makuwa), wax gourd (Benincasa hispida), melon (Cucumis melo), star cucumber (Sicyos angulatus) > sponge cucumber (Lufa cylindrica), cotton (Gossypium indicum). There were no distinct differences among varieties and lines that were collected and hybridized at the Gochang Watermelon Experiment Station in Korea. There was a positive relationship between the leaf area and the degree of damage by the Cotton caterpillar. The feeding amounts of the Cotton caterpillar excrement were gradually increased to 16-18 days after hatching, after that the amounts of excrement were rapidly decreased.

Characterization of an Isolate of Cucumber mosaic virus from Raphanus sativus L. (열무에서 분리한 오이모자이크바이러스 분리주의 특성)

  • Cucumber mosaic virus (CMV)-like isolate was collected from Raphanus sativus (cv. Choon-hyang), which showed mosaic symptoms. The isolate was confirmed to a strain of CMV by host responses in Vigna unguiculata, Chenopodium amaranticolor and Gomphrena globosa, by viral genome composition with RT-PCR and PCR-RFLP, and by serological analysis. Symptom developed by the strain of CMV was severe in Nicotiana benthamiana, N. glutinosa, N. tabacum (cv. Samsun, cv. Xanthi), Cucumis melo (cv. Early hanover), Cucumis sativus (cv. White wonder), Capsicum annuum (cv. Chung-yang and cv. Geum-top), but mild symptom was developed in Raphanus sativus (cv. Choon-hyang), Brassica rapa ssp. pekinensis (cv. Bul-Am No. 3), and B. juncea (cv. Daenong Jukgot). Newly isolated strain of CMV could infect diverse crops including Solanaceae, Cucurbitaceae and Brassicaceae. We designated the new strain of CMV as Gn-CMV based on the novel infectivity of Brassicaceae. In double-stranded (ds) RNA analysis, Gn-CMV consisted of 3.3, 3.0, and 2.2 kb genomes likewise other strains of CMV. SDS-polyacrylamide gel electrophoresis (SDS-PAGE) showed 28 kDa of the CMV coat protein. By restriction enzyme mapping using Cac8I, ClaI and MspI of RT-PCR products indicated that Gn-CMV belongs to CMV subgroup I.

  • Effect of arsenite, arsenate and dimethylarsinic acid (DMA) on the growth of seedling plants were investigated in order to compare the toxicity of arsenic species in soil environments. Test plants were mung bean (Phaseolus radiatus), wheat (Triticum aestivum), barely (Hordeum vulgare), cucumber (Cucumis sativus L.). Seedling growth in As-contaminated soil were significantly reduced in all test species. Arsenite was more toxic than arsenate and DMA. Among the test plants, mung bean was most sensitive to arsenic, followed by cucumber, wheat, and barely.

Effects of Soil Moisture Content according to Irrigation Methods in Culture on Storability of Cucumber(Cucumis sativus L.) Fruit (관수방법에 따른 토양내 수분함량의 차이가 수확후 오이의 저장에 미치는 영향)

  • This study was made to investigate the effects of soil moisture content according to irrigation methods on the storability and quality of cucumber. The fresh weight loss of cucumber fruit harvested in drip irrigation plot was more than that in conventional irrigation plot at both 13$^{\circ}C$ and 24$^{\circ}C$ storage temperature. Dry weight ratio decreased during storage, and was higher in conventional irrigation plot than drip irrigation plot both 13$^{\circ}C$ and 241 storage. The decrease of dry weight ratio was higher at 24$^{\circ}C$ than 13$^{\circ}C$. Vitamin C was not influenced by soil moisture content, but decreased during storage at 13$^{\circ}C$ and 24$^{\circ}C$ The decrease of vitamin C at 24$^{\circ}C$ in 8 days after storage was twice as much at 13$^{\circ}C$. Firmness was measured differently in two parts of cucumber ; fruit stalk and blossom part. The firmness of fruit stalk part was higher than that of blossom part. This phenomena was observed continuously at until final day at 13$^{\circ}C$ and 24$^{\circ}C$ storage. But the difference of firmness was not showed in soil moisture content. Vitamin C, firmness and other quality characteristics were not influenced by soil moisture content during cultivation. The different soil moisture content according to irrigation methods did not affect the storability and quality of cucumber.
