Optimization of Culture Medium for the Production of an Exopolysaccharide (p-CY02) with Cryoprotective Activity by Pseudoalteromonas sp. RosPo-2 from the Antarctic Sea

  • When cells are exposed to freezing temperatures, high concentrations of cryoprotective agents (CPA) prevent ice crystal formation, thus enhancing cell survival. However, high concentrations of CPAs can also cause cell toxicity. Exopolysaccharides (EPSs) from polar marine environments exhibit lower toxicity and display effects similar to traditional CPA. In this study, we sought to address these issues by i) selecting strains that produce EPS with novel cryoprotective activity, and ii) optimizing culture conditions for EPS production. Sixty-six bacteria producing mucous substances were isolated from the Ross Sea (Antarctic Ocean) using solid marine agar plates. Among them, Pseudoalteromonas sp. RosPo-2 was ultimately selected based on the rheological properties of the produced EPS (p-CY02). Cryoprotective activity experiments demonstrated that p-CY02 exhibited significantly cryoprotective activity at a concentration of 0.8% (w/v) on mammalian cells (HaCaT). This activity was further improved when combined with various concentrations of dimethyl sulfoxide (DMSO) compared to using DMSO alone. Moreover, the survival rate of HaCaT cells treated with 5% (v/v) DMSO and 0.8% (w/v) p-CY02 was measured at 87.9 ± 2.8% after freezing treatment. This suggests that p-CY02 may be developed as a more effective, less toxic, and novel non-permeating CPA. To enhance the production of EPS with cryoprotective activity, Response Surface Methodology (RSM) was implemented, resulting in a 1.64-fold increase in production of EPS with cryoprotective activity.

  • 명태 actomyosin 용액에 대한 옥수수전분 가수분해물의 동결 보호 효과는 전기영동상 패턴, 용해도 및 $Ca^{2+}$-ATPase 활성 변화로 알아보았다. 먼저 전기영동상 패턴에서 0일차와 저장 30일차를 비교하였을 때, control의 경우 저장온도에 관계없이 각 band들의 변화가 수반되었고, 특히 MHC의 변화가 두드러졌다. Sucrose 첨가구의 경우, 저장온도에 상관없이 band상의 변화가 거의 없는 것으로 나타났다. 전분가수분해물이 첨가된 시료들에서는 온도 의존성이 현저하여 $-5^{\circ}C$에서 저장한 시료들은 MHC band가 나타나지 않았고 반면에 $-20^{\circ}C$에서는 저장 30일 째에도 0일차와 거의 같은 양상을 보였다. 시료들을 $-5^{\circ}C$에서 저장할 때 용해도 변화는 sucrose의 경우 저장 30일 후에도 $90\%$ 이상의 높은 값을 유지한 반면에 나머지 시료들은 저장과 함께 급속히 감소하였다 $-20^{\circ}C$에서는 sucrose, D.E. 10, 15 및 20을 함유한 시료들은 약 $90\%$ 정도를 나타내었다. 잔존 활성 변화에서도 용해도 변화와 거의 유사한 경향을 나타내었다. 이상에서 살펴보았듯이 명태 actomyosin 용액의 동결변성에 대한 옥수수전분가수분해물들의 보호효과는 저장온도에 따라 많은 차이를 나타내었고, 특히 D.E. 10의 경우는 기존의 동결변성 방지제인 sucrose와 비교하여 $-12^{\circ}C$ 이하에서는 거의 같은 효과를 나타내는 것을 알았고, 저감미, 저열량의 동결변성 방지제로서 가능한 것으로 사료된다.

Cryoprotective Properties of Exopolysaccharide (P-21653) Produced by the Antarctic Bacterium, Pseudoalteromonas arctica KOPRI 21653

  • Twenty-five bacterial strains that secrete mucous materials were isolated from sediment obtained from King George Island, Antarctica. Seven of these strains proved capable of producing cryoprotective exopolysaccharides. The strain KOPRI 21653 was selected for the further study of an anti-ice-nucleating polysaccharide (ANP), which originated from a polar region. KOPRI 21653 was identified as Pseudoalteromonas arctica as the result of 16S rRNA analysis. The exopolysaccharide, P-21653, was purified completely from the KOPRI 21653 cell culture via column chromatography and protease treatment. The principal sugar components of P-21653 were determined to be galactose and glucose, at a ratio of 1:1.5, via GC-MS analysis. The cryoprotective activity of P-21653 was characterized via an E. coli viability test. In the presence of 0.1% (w/v) P-21653, the survival ratio of E. coli cells was as high as 82.6% over three repeated freeze-thaw cycles. The survival ratio decreased drastically to 71.5 and 48.1 %, respectively, in five and seven repeated cycle conditions; however, the survival ratios were greater over three (96.6-92.1%) to seven (100.5-91.6%) freeze-thaw cycles in the presence of 0.5 and 1.0% (w/v) P-21653. In addition, at much lower concentrations (0.1-1.0%), P-21653 resulted in survival ratios (83.1-98.4%) similar to those of two commercially available cryoprotectants ($V_{EG}$ plus X-1000, 92.9% and $V_{M3}$, 95.3%), which were utilized at the recommended concentrations (90%). The biochemical characteristics of exopolysaccharide P-21653 reflect that this compound may be developed as a useful cryoprotectant for use in medical applications and in the food industry.

Effect of lactoferrin on ram sperm motility after cryopreservation

  • Objective: The objective of this study was to analyse the differentially abundant proteins caused by freeze-thawing of ram sperm and explore candidate proteins of interest for their ability to improve ram sperm cryopreservation outcomes in vitro. Methods: Sperm were from three mature Dorper. Fresh and frozen sperm proteins were extracted, and the differentially abundant proteins were analysed by mass spectrometry. Among these proteins, lactoferrin (LTF) was selected to be added before cryopreservation. Next, sperm samples were diluted in Tris extender, with the addition of 0, 10, 100, 500, and 1,000 ㎍/mL of LTF. After thawing, sperm quality was evaluated by motility, plasma membrane integrity, mitochondrial activity and reactive oxygen species (ROS). Results: Cryopreservation significantly altered the abundance of 40 proteins; the abundance of 16 proteins was increased, while that of 24 proteins was decreased. Next, LTF was added to Tris extender applied to ram sperm. The results showed that sperm motility and plasma membrane integrity were significantly improved (p<0.05) by supplementation with 10 ㎍/mL LTF compared to those in the control group. There was no significant difference in mitochondrial activity between the 0 ㎍/mL group and other groups (p>0.05). Supplementation of the cryoprotective extender with 10 ㎍/mL LTF led to decreased ROS levels compared with those in the control and other groups (p<0.05). Conclusion: The LTF is an important protein during cryopreservation, and the addition of 10 ㎍/mL LTF to a cryoprotective extender can significantly improve the function of frozen ram sperm.

  • 결빙억제제(CPA)의 종류, 농도 및 침지시간이 참전복 Haliotis discus hannai 발생배의 생존활성에 미치는 영향을 조사하였다. 4세포, 담륜자 및 피면자를 dimethyl sulfoxide, ethylene glycol, propylene glycol 농도별로 10분, 20분 및 30분간 침지하였다. CPA 침지 후 각 발생배의 발생진행률은 각 CPA 농도가 낮을수록, CPA 침지시간이 짧을수록 높았다. 또한, 피면자에서 비틀림전까지의 발생진행률은 4세포에서 담륜자까지의 것보다 높았다. CPA 침지 후 발생진행률 및 유생활성지수는 ethylene glycol에서 가장 높았다. 참전복 발생배의 냉동보존 효과를 높일 수 있는 CPA 및 농도는 ethylene glycol 2.0 M, 평형시간은 30분인 것으로 추정된다.

  • This study aimed to find an optimal diluent and cryoprotective agent (CPA) during cryopreservation of common Korean bitterling Acheilognathus signifier sperm. The bitterling is an endangered species in Korea, and this study will enable conservation and further technical development of artificial seed production. We tested the effects of cryopreservation and toxicity by the type of diluent and CPA. The optimal combination of diluent and CPA for cryopreservation was 300 mM glucose+10% methanol, resulting in a survival rate and sperm activity index (SAI) of $96.3{\pm}1.5%$, and $3.0{\pm}0.0$ respectively, and no significant difference compared to fresh sperm. The survival rate and SAI of post-thawed sperm was $9.7{\pm}1.5%$, $0.8{\pm}0.3$ respectively, which was significantly higher than had been achieved with other diluents and CPAs.

Effect of diluent variation on cryopreservation of large yellow croaker Larimichthys crocea

  • The aim of this research was to investigate different factors, including cryoprotective agents (CPAs), diluents, dilution ratios, equilibrium times, freezing rates, and thawing methods to optimize cryopreservation protocols for large yellow croaker (Larimichthys crocea). The parameters evaluated were sperm motility, sperm activity index (SAI), survival rate, and DNA damage. Different types of CPAs, such as dimethyl sulfoxide (DMSO), propylene glycol (PG), ethylene glycol (EG), methanol, and glycerol, were tested for sperm preservation. The highest motility, SAI, and survival rate were observed when EG was used. Different diluents such as Stein's solution, Hank's balanced salt solution, marine fish Ringer's solution, artificial seminal plasma (ASP) of small yellow croaker, and Cortland solution were investigated. The highest post-thaw motility was observed upon using ASP as the diluent. Different concentrations of EG were then mixed with ASP to identify the optimal EG concentration. Experimental results showed that the motility (70.33 ± 1.20%), SAI (5), and survival rate (78.30 ± 0.42%) of post-thaw sperm were optimum when 10% EG and ASP were used as the CPA and diluent of cryopreservation, respectively. Post-thaw sperm motility was high at equilibration times below 150 s and at an optimum dilution ratio of 1:1 (sperm: CPA + diluent) and was not significantly different compared with fresh sperm motility. The freezing rate was found to be slow below -10℃/min. The thawing temperature of 45℃ was identified as ideal. The percentage of tail DNA in post-thaw sperm at 10% EG and ASP was also investigated and was found to have more significant DNA damage than that in fresh sperm but significantly lower damage than that in post-thaw sperm at EG concentrations of 5%, 15%, and 20% (p < 0.05). The cryopreservation protocols obtained in this study will be useful in large yellow croaker hatcheries.

담수순화 감성돔(Acanthopagrus schlegeli) 정자의 냉동보존 조건별 활성평가 (Assessment of Sperm Activity of Black Porgy(Acanthopagrus schlegeli) Acclimated in Freshwater on Cryopreservation Condition)

  • 담수에서 사육한 감성돔 Acanthopagrus schlegeli(담수감성돔) 냉동/해동 정자의 생존율이 가장 높았던 결빙억제제(cryprotective agent, CPA)는 dimethyl sulfoxide(DMSO)였으며, 다음으로 glycerol, ethylene glycol(EG) 그리고 methanol 순이었다. 해동 후 정자운동성은 glycerol, DMSO, EG, methanol 순으로 높았으며, 이상의 결과들은 해수에서 사육한 감성돔(해수감성돔)에서도 나타났다. 담수 및 해수감성돔 정자의 냉동보존시 적정 CPA는 DMSO와 glycerol이었으며, 생존율과 정자운동성이 가장 높았던 적정 농도는 모두 10%였다. 특히 10% DMSO로 냉동보존한 담수 및 해수감성돔 해동정자의 생존율과 정자운동성은 10% glycerol로 냉동보존한 해동정자보다 높게 나타났다. 냉동보존기간에 따른 담수 및 해수감성돔 해동정자의 생존율과 정자운동성을 측정한 결과, 대부분 보존기간이 길어질수록 감소하였으며, CPA의 농도가 높을수록 생존율과 정자운동성은 급격히 감소하였다. 그러나 5% DMSO로 냉동보존한 해동정자의 생존율 및 정자운동성은 차이는 보이지 않았다.

아미노산 혼합용액이 Lactobacillus casei YIT 9018의 동결건조 및 저장성에 미치는 영향 (Effect of the amino acid mixture on freeze-drying and preservation of Lactobacillns casei YIT 9018)

  • Lactobacillus casei YIT9018은 국내의 발효유제조에 널리 사용되고 있으며, 이에 대한 효과적인 동결건조 방법을 확립하기 위하여 glycine과 glutamic acid를 함유한 amino산 혼합액을 기본 현탁 배지로 사용하여 동결건조 및 rehydration을 실시하고 그 효과를 기존의 보호제들과 비교하였다. 아미노산 혼합액은 대조구에 비해서 현저한 동결보호 효과를 가지고 있었다. 또 glycine과 DL-glutamic acid는 단독 사용시보다 혼합할 경우 더욱 효과적이었으며 이때의 최적농도는 0.1M이었다. L. casei YIT9018주는 MRS 배지상에서 접종후 18시간만에 정상기에 들어갔으며 24시간 후에는 최대생육을 나타냈다. 또 이 24시간 균체가 동결건조에 대하여 가장 저항성이 큰 것으로 판명되었다. 아미노산 혼합액에 sucrose, lactose등의 이당류 및 NF-MS를 첨가하면 85% 이상의 높은 생존율을 기대할 수 있으며, 최적농도는 당류의 경우 7.5-10%(w/v), NFMS의 경우는 10%정도였다. 이상의 결과로 보아 동결후의 생존율을 높이고 활력을 유지하기 위해서는 아미노산 혼합액에 NFMS의 첨가가 바람직하다고 생각되었다. 한편, rehydration media에 따라서도 생존율이 변하였는데 아미노산 혼합액은 대조구에 비해 약 3배의 동결 보호효과를 나타냈다.

동자개 Pseudobagrus fulvidraco 정자 동결보존 (Sperm Cryopreservation of Korean Bullhead Pseudobagrus fulvidraco)

  • 본 연구는 동자개 정자의 동결보존을 위해 동결보존제의 최적 농도와 적정 희석액을 구명하여 정자를 최상의 상태로 보존하여 인공종자를 생산하는데 목적이 있다. 실험은 3종의 희석액(I: 300 mM glycose, II: Kurokura extender, III: Li extender), 4종의 동결보존제(dimethyl sulfoxide, ethylene glycol, methanol and glycerol)와 4개의 동결보존제 농도(5, 10, 15, 20%)를 조합하여 대하여 조사하였다. 동결보존한 정자를 해동한 후 정자의 생존율과 정자활성지수로 동결보존제의 효과를 평가하였다. 희석액 III(Li extender)과 10과 15%의 methanol을 조합했을 때 동자개 정자의 생존율과 정자활성지수는 각각 66.9 ± 8.7, 67.3 ± 13.1%과 2.6 ± 0.4, 2.6 ± 0.5로 다른 희석액과 동결보존제보다 높았다.
