An Isolation of Crude Saponin from Red-Ginseng Efflux by Diaion HP-20 Resin Adsorption Method (홍삼유출액으로부터 Diaion HP-20 수지 흡착법에 의한 조사포닌의 분리)

  • This study was carried out to isolate saponin compounds from red-ginseng efflux, which was produced during the industrial processing of red-ginseng from fresh ginseng. We isolated crude saponin from the efflux extract (moisture content 35.0%) by using Diaion HP-20 adsorption method. Non-saponin fraction, which was adsorbed on Diaion HP-20 resin, was removed by eluating with $H_{2}O$ and 25% spirit. Then crude saponin was eluated with 95% spirit, continuously. Saponin in the eluated fractions was confirmed by TLC analysis. Crude saponin isolated from red ginseng efflux extract contained 12.10% of saponin. whereas those of white ginseng and red-ginseng were 3.30 and 3.39%, respectively. Ginsenoside contents showed the highest contents kin crude saponin from red ginseng efflux extract. Expacilly, the ginsenoside-$Rb_{1}$ and Re showed the highest contents in red-ginseng efflux extract when compared with those of white ginseng and red ginseng crude saponins. And the other ginsenosides except ginsenoside-$Rb_{1}$ and -Re also showed the highest contents in red ginseng efflux extract. However, the ratio of PD saponin (Panaxadiol saponin: $Rb_{1}+Rb_{2}$+Rc+Rd) to PT saponin (panaxatriol: $Re+Rg_{1}$) showed almost the same level when compared with those of ginseng saponin fractions. Ratio of PD/PT from red ginseng efflux extract was 1.99. Ratios of PD/PT from white ginseng and red ginseng were 1.85 and 1.84, respectively. Saponin purity, which was calculated by ratio percent of total ginsenoside to curde saponin content, was 45.90%. In case of white ginseng and red ginseng, the purities were 35.50 and 36.00%, respectively. However, by PHLC analysis, we confirmed that crude saponin isolated from red ginsengs. It suggested that crude saponin isolated from red ginseng ellux also would be useful component as ginseng saponins.

The Effect of Crude Saponins of Korean Red Ginseng against Airway Inflammation and Airway Hyperresponsiveness Induced by Diesel Exhaust Particles in Mice (생쥐에서 디젤배기가스 입자에 의한 기도염증과 기도 과민성에 미치는 홍삼 조사포닌의 영향)

  • The objective of this study was to investigate the effect of crude ginseng total saponins (CGS) against airway inflammation (AI) and airway hyperresponsiveness (AH) induced by diesel exhaust particles (DEP) in mice. AI and AH were induced by the intratracheal instillation with 0.1 $mg/m{\ell}$ of DEP suspension once a week for 10 weeks combined with ovalbumin (OVA) sensitization. Mice were also treated orally with 75 $mg/m{\ell}$ of CGS, 5 days a week for 10 weeks. Oral CGS treatment decreased in the level of serum immunoglobulin (IgE) and histamine increased by DEP and OVA, and declined respiratory resistance. It also dropped an enhanced infiltration of eosinophils in the bronchoalveolar lavage fluid (BALF) of mice, and an increased T helper type 2 cell derived cytokine levels such as of interleukin (IL)-4, IL-13 and IL-5 in the BALF. However, it did not influence T helper type 1 cytokine such as interferon-gamma in the BALF. These results indicate that CGS may alleviate allergen-related AI and AH in mice and may play an important role in the modulation of asthmatic inflammation.

Comparison of Non-saponin Composition and Contents in Fresh Ginseng Roots Cultivated in Different Areas and at Various Ages (수삼의 지역별 연근별 인삼 비사포닌 성분 함량 비교)

  • This study was carried out to obtain the basic information for non-saponin contents that can be used to index fresh ginseng roots (Panax ginseng C. A. Meyer) cultivated in the Republic of Korea and China. Non-saponin components in fresh gingeng roots which were cultivated in various areas and ages in Korea were determined. Acidic polysaccharide, total polysaccharide, crude polyacetylene were quantitatively analyzed by using the method of spectrophotometric determination, while the total protein was analyzed by using Lowry method. The results show that there were no statistically significant differences for the average contents of four non-saponins among 4-years-old, 5-years-old, and 6-years-old fresh ginseng roots. Additionally, this study assessed the average contents of non-saponin components in 4-years-old fresh ginseng roots (Panax ginseng C. A. Meyer) which were cultivated in Korea and China. The result showed that the average contents of crude polyacetylene and acidic polysaccharide were statistically significant. Four-years-old fresh ginseng roots cultivated in Korea had the higher average contents of crude polyacetylene and acidic polysaccharide than those cultivated in China. However the average contents of total polysaccharide and total protein had no statistically significant difference.

Effect of Saponin Fraction from Platycodon grandiflorum on Clinical Chemical Changes in TCDD (2,3,7,8-Tetrachlorodibenzo-ρ-dioxin)-induced Rat Toxicity

  • This study was carried out to investigate the protective effect of crude saponin from Platycodon grandiflorum on Clinical chemical parameters in male rats acutely exposed to 2,3,7,8-tetrachlorodibenzo-ρ-dioxin (TCDD). Crude saponin was prepared from Korean Platycodon grandiflorum with Diaion HP-20 adsorption chromatography after extraction of 80% ethanol at 75℃. The crude saponin was confirmed by thin layer chrmatography. When compared with ginseng saponins, the crude saponin had both a few number of saponins and a broad distribution. Forty male rats (200±20 g) were divided into 4 groups. Normal control (NC) group received vehicle and saline; TCDD-treated (TT) group received TCDD (40 ㎍/kg, single dose) intraperitoneally; Platycodon grandiflorum saponin (PG5 and PG10) groups received crude saponin 5 mg/kg and 10 mg/kg (p.o), respectively, for 2 weeks before 1 week of TCDD-exposure. Increase of body weight was retarded greatly by TCDD-exposure. Body weight of animals in TT group was significantly decrease after 2 days of TCDD-exposure. However, body weights of animals in PG groups increased through the experimental perimental period, although the increasing rate was slower than that of NC group. Increases in contents of blood glucose, total cholesterol and triglyceride (TG) and activities of amylase, lipase, AST, ALT and LDH by toxic action of TCDD were significantly attenuated by crude saponin from Platycodon grandiflorum (P<0.05). In conclusion, these results suggest that crude saponin prepared from Korean Platycodon grandiflorum might be a member of useful protective agents against TCDD, which is one of the environmental hormones.

Stem-leaves of Panax as a rich and sustainable source of less-polar ginsenosides: comparison of ginsenosides from Panax ginseng, American ginseng and Panax notoginseng prepared by heating and acid treatment

  • Background: Ginsenosides, which have strong biological activities, can be divided into polar or less-polar ginsenosides. Methods: This study evaluated the phytochemical diversity of the saponins in Panax ginseng (PG) root, American ginseng (AG) root, and Panax notoginseng (NG) root; the stem-leaves from Panax ginseng (SPG) root, American ginseng (SAG) root, and Panax notoginseng (SNG) root as well as the saponins obtained following heating and acidification [transformed Panax ginseng (TPG), transformed American ginseng (TAG), transformed Panax notoginseng (TNG), transformed stem-leaves from Panax ginseng (TSPG), transformed stem-leaves from American ginseng (TSAG), and transformed stem-leaves from Panax notoginseng (TSNG)]. The diversity was determined through the simultaneous quantification of the 16 major ginsenosides. Results: The content of ginsenosides in NG was found to be higher than those in AG and PG, and the content in SPG was greater than those in SNG and SAG. After transformation, the contents of polar ginsenosides in the raw saponins decreased, and contents of less-polar compounds increased. TNG had the highest levels of ginsenosides, which is consistent with the transformation of ginseng root. The contents of saponins in the stem-leaves were higher than those in the roots. The transformation rate of SNG was higher than those of the other samples, and the loss ratios of total ginsenosides from NG (6%) and SNG (4%) were the lowest among the tested materials. In addition to the conversion temperature, time, and pH, the crude protein content also affects the conversion to rare saponins. The proteins in Panax notoginseng allowed the highest conversion rate. Conclusion: Thus, the industrial preparation of less-polar ginsenosides from SNG is more efficient and cheaper.

Changes in Effective Components of Ginseng by Puffing (팽화에 의한 인삼 유효성분의 변화)

  • This study was conducted to investigate the effect of moisture content and pressure on extraction yield, crude saponins and ginsenoside contents of puffed Korean ginseng. Puffed ginsengs showed relatively higher extraction yield ($50.0{\sim}62.1%$) and amounts of crude saponins ($19.6{\sim}48.8$ mg/g ginseng) than no-puffed ginseng ($37.6{\pm}0.8%$ and $11.0{\pm}1.0$ mg/g ginseng), respectively. The highest extraction yield and amounts of crude saponins were obtained in 8.0% moisture content sample puffed at 10 $kg_f/cm^2$. In HPLC analysis, amounts of measured major ginsenosides (Rb1, Rb2, Rc, Rd, Re, and Rg1) decreased with increasing puffing pressure, yet contents of almost all major gin senosides were higher than control (no-puffed). On the other hand, ginsenoside Rg3 were produced after puffing suggesting that chemical structure of some ginsenosides might be altered during the puffing process. These results indicate that puffing can increase the extraction yield and crude saponin contents and it could influence the ginsenoside composition.

  • This study was carried out to investigate effects of fat-soluble solvents on the purification against nan-saponin substances such as chlorophylls and other pigments and on the yields of saponins in separating saponins from ginseng root, leaf and stem. Ginseng root saponins were effectively purified by various fat-soluble solvents while ginseng leaf stem saponins were by chloroform. And alternative extractions of ethyl acetate, ethyl ether, chloroform and benzene there more effective for ginseng leaf stem saponins than that by any single solvent. Contents of crude saponin fractions and total ginsenosides in ginseng leaf were $18.5{\sim}19.5%\;and\;10.8{\sim}11.4%$, which were very high compared with $4.6{\sim}5.1%\;and\;2.0{\sim}2.6%$ in ginseng root or $2.2{\sim}2.5%\;and\;0.63{\sim}0.67%$ in ginseng stem. Therefore, ginseng leaf is good resources for total saponin or $ginsenosides-Rg_1,\;.Re,\;-Rc,\;-Rd,\;-Rb_2\;and\;-Rf$.

Effect of Extraction on Chemical Composition of Rcd Ginseng Extract (추출 회수에 따른 홍삼 extract의 성분 조성에 관한 연구)

  • Red ginseng was extracted with water and analyzed for yield, saponin, pectin and other chemical composition. It was found that: (1) The total solid content in extract after 6 times of extraction was 46.8%,: including 13.6% of centrifugal residue; (2) 83.7% of total extractable solids and 86% total saponin was extracted after the initial three runs of extraction. (3) No significant changes were observed in HPLC pattern of extracted saponins over a range of extractions; (4) The ratio of centrifugal residue to total solids increased as the number of extractions increased; (5) The ratios of fat, protein, reducing sugar and pectin contents decreased with repeating extraction while those of crude fiber, total sugar and 35% alcohol insoluble residue increased when they were compared with total solids.

Effects of Ginseng Saponins on the Induction of Differentiation in Mammary Epithelial Cells and Mammary tumor Cells (홍삼 사포닌에 의한 유선상피 및 유선암세포의 분화 유도 효과 연구)

  • Using Ginseng saponins (crude saponin and total saponin) and ginsenoside Rbl Rb2, Rc, Rd, Re, Rhl, and Rh2 in this study, we have examined the effects of the compounds on the induction of differentiation in normal rat mammary epithelial cells and 7,12-dimethylbenz[a]anthracene (DMBA)-induced mammary tumor cells in culture. When normal rat mammary organoids were cultured in 100-mm culture plates in the presence or absence of ginseng saponins, there were four different cell colonies after two weeks in culture: cobble stone, spindle, honey comb, and senescence type colonies. Ginseng saponins showed different effects on the development of each colonies. Scrape-loading dye transfer tech-nique was performed to measure the effects of total saponin, Rhl, and Rh2 on intercellular junctional communication. Intercellular communication was not observed at short cultilral time, e.g., four or seven days, but when it cultured it up to two weeks, cell to cell communication was observed in saponin-treated cells. Reconstituted basement membrane, Matrigel, supported the growth and development several different multicellular structures from normal mammary organoids (e.g., ductal, webbed, stellate, and squamous colonies) or DMBA-induced mammary tumor (e.g., alveolar unit, foamy alveolar unit, squamous metaplasia, lobule-ductal, stellate, and webbed colony). In ginseng saponin-treated groups, webbed colonies were more and squamous colonies were less than control group. Moreover, the ductal colonies, marker tructure of well-differentiate mammary epithelial cells, were developed more in saponin-treated group than in control group. In conclusion, ginseng saponins affected on the differentiation of normal rat mammary epithelial cells and DMBA-induced mammary tumor cells in culture.
