Anti-inflammatory, anti-ulcer and hypoglycaemic activities of ethanolic and crude alkaloid extracts of Madhuca indica (Koenig) Gmelin seed cake

  • Madhuca indica has been used ethnomedically in Indian folks. In the present study we have investigated anti-inflammatory, anti-ulcer and hypoglycaemic effect of ethanolic extract (EE) and crude alkaloid extract of Madhuca indica seed cake on albino rats. The study showed that the EE had a significant, dose dependent anti-edematogenic, anti-ulcerogenic and hypoglycaemic activity, whereas the crude alkaloid extract exhibited a significant only. Both the extracts possess dose dependent inhibitory activity on carrageenan-induced edema, inhibiting prostaglandins or mediators involved in prostaglandin synthesis, the second phase of inflammation. The EE was significantly effective in protecting pylorus-ligation-induced gastric ulcers at a higher dose level. The active principle of EE seems to be a selective inhibitor of the COX II (prostaglandin synthesis) without important effect on COX I since, EE exhibited both anti-edematogenic and anti-ulcerogenic effect. The EE was effective in reducing the plasma glucose level in normal albino rats in a dose dependent manner, producing hypoglycaemic effect by stimulating the release of insulin from the ${\beta}-cells$ and/or increasing the uptake of glucose from the plasma.

  • A new aromatic amide, methyl-N-(3-carbamoylpropionyl) anthranilate was isolated for the first time as a natural compound and one known $C_{19}-diterpene\;alkaloid$, avadharidine was also obtained from the root of Aconitum pseudolaeve var. erectum. The\;LD_{50}$ values of water extract and MeOH extract of the root of Aconitum pseudolaeve var. erectum in mice were 1. 23 g (13. 6 g crude drug) and 0. 77 g(5. 13 g crude drug)/kg, p.o., respectively.

Proximate, Phytochemical, and In Vitro Antimicrobial Properties of Dried Leaves from Ocimum gratissimum

  • Ocimum gratissimum is a common plant in the tropics and has been used in food and medicine. Its usage in food and medicine could be attributed to its phtyochemical and antimicrobial properties. In this study we investigated the proximate, phytochemical, and antimicrobial attributes of air dried leaves of O. gratissimum. The aqueous extract was found to contain phtyochemicals with alkaloid and saponin present in appreciable amounts. The proximate analysis (crude protein and crude fibre content were 15.075% and 17.365%, respectively) showed that the leaf could be a good source of protein and fibre. The aqueous ethanolic extract of the leaf exhibited activity against a wider range of organisms when compared to the aqueous extract at the investigated concentrations. Aqueous ethanolic extracts of O. gratissimum leaf was active against Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, and Bacillus cereus and the aqueous extract of the leaf was active against P. aeruginosa.

An Acetophenone Derivative, Clavatol, and a Benzodiazepine Alkaloid, Circumdatin A, from the Marine-Derived Fungus Cladosporium

  • The crude extract of the mycelium of Cladosporium was found to exhibit antimicrobial activity against the Staphylococcus aureus, methicillin-resistant S. aureus, and multidrug-resistant S. aureus. Bioassayguided fractionation of an organic extract led to the isolation of an acetophenone derivative, clavatol (2',4'-dihydroxy-3',5'-dimethylacetophenone) (1), and a benzodiazepine alkaloid, circumdatin A (2). Compound 1 showed moderate antibacterial activity against S. aureus, methicillin-resistant S. aureus, and multidrug-resistant S. aureus with minimum inhibitory concentration (MIC) values of 62.5, 62.5, 31.0 $\mu$g/mL, respectively, but compound 2 was inactive. Compounds 1 and 2 exhibited UV-A protection activity with ED$_{50}$ values of 227.0 and 82.0 $\mu$M, respectively, indicating that they were more potent than the positive control, oxybenzone (ED$_{50}$ 350 $\mu$M), a common sunscreen agent.

A Study on Antimicrobial Activity of Veratri Herba (여로의 항균작용에 관한 연구)

  • This study was carried out to investigate the efficacy of antifungal and antibacterial activity of Veratri Herba, which was used as insecticide in the clinical part of oriental dermatology for a long time. Sample herb was selected with Veratrum maackii in the middle of 5 veratrum species. The experimental results of this herb were as follows. Crude extract showed various relative inhibitory rates from 68.9% to 95.6% in 2mg/disc against Fusarium oxysporum, Botrytis cineria, Saccharomyces cerevisiae, Escherichiae coli, and Bacillus subtilis on the basis of inhibitory range against B. cineria, and the inhibitory effect was decreased depending on density. In the extract of various organic solvent, chloroform and ethyl acetate fraction show the various size of dear zone against all microbes. And in the subfraction assay from ethyl acetate fraction, crude alkaloids show the antibacterial and antifungal effect significantly.

Studies on the Cattle Tick killing and Repellent Effects of the Ingredients of Radix Jingyu (진구성분의 소진드기 구제(驅除)(살충(殺蟲) 및 기피(忌避))효과(效力)에 관한 연구(硏究))

  • In the previous report it was demonstrated that aqueous extracts of two Korean indigenous crude drugs, Radix Jingyu and Fructus ponciri, have an excellent tick repellent effect, and also the need for further analytical study of these crude drugs was suggested. In the experiments to be reported herein, attempts were made to find out the active ingredients from Radix Jingyu, since the aqueons extract of Radix Jingyu proved to be mone effective than that of Fructus ponciri. It will be shown that the active ingredient is indicative of Lycaconitine, a known alkaloid. An aqueons solution containing 1.5 Gms of Lycaconitine in tartrate when sprayed on a cow exerted tick repellent effect against Boophilus microplus which lasted approximately 25 days without any harmful influences on cattle. It will also be shown that the oil-fat components of Radix Jingyu act synergistically with Lycaconitine in accelerating and lengthening the effect, although the components alone have no effect under the conditions tested. It was of interest to note that the oil-fat components of Radix Jingyu showed definite repellent effect against the larvae of Boophilus microplus in laboratory experiments by the method newly devised. On the basis of these observations, it was concluded that Lycaconitine can be used as cattle tick repellent, and the evidence seems to warrant a large scale production of Lycaconitine for its widespread use. Further studies on the possible effect of the alkaloid on other genera or species of ticks and related vermins were also indicated.

  • The present communication attempts to evaluate the physicochemical and preliminary phytochemical studies on the fruit of Terminalia chebula Retz. Combretaceae family. Haritaki is one of the most celebrated herbs in the Indian traditional medicine system, Ayurveda. Terminalia chebula is known to exhibit different properties like anticancer, anti-inflammatory, anti-protozoal, antimicrobial, antioxidant, hepato and renal protective activities, and in the management of metabolic syndrome. As there is no detailed standardisation work reported on fruit, the physicochemical parameters, preliminary phytochemical constants, heavy metals, analysis are carried out. The phytochemical screening indicated the presence Tannin, Alkaloid, Phenol, Carbohydrate, Steroids, Protein and Resin compounds in CO2 extract of Haritaki. The present investigation will helpful in assessing the quality and purity of a crude drug. Thus, the study provides facts that CO2 extract of Haritaki contains medicinally important bioactive phytochemical compounds which justifies the use of plant species as conventional medicine for treatment of many diseases.

Correlation between Tobacco Leaf and Smoke Compositions (잎담배 조성과 연기성분과의 관계)

  • Chemical compositions of Flue - cured and Burley tobacco leaf and smoke were analyzed to find out simple correlation between compositions. Chemical Constituents of Bark of Beilschmiedia kunstleri Gamble with Anticancer, Anti-Streptococcus pyogenes, Anti-Bacillus cereus and Anti Plesiomonas shigelloides Activities

  • Lauraceae is a family medicinal plant whose tubers possesses antimicrobial, and cytotoxic, such as antiparasitic and anti-inflammatory special effects and has been used for the medicine in the cure of hepatitis and rheumatism. The antimicrobial activities of bioactive compounds including one neolignan; kunstlerone (1) and two alkaloids include isocaryachine (2) and noratherosperminine (3) as well as crude hexane, methanol and dichloromethane extracts were evaluated. Additionally, the effect of compounds 1, 2 and 3 were evaluated on A549, PC-3, A375, HT-29 and WRL-68 cell lines. In conclusion, kunstlerone 1 showed moderate cytotoxicity against various cancer cell lines such as A549, PC-3, A375, HT-29 and WRL-68, respectively with $EC_{50}$ values of 28.02, 26.78, 33.78, 33.65 and $16.46{\mu}g/mL$. The crude methanol extract showed antigrowth activity against S. pyogenes II and B. cereus, with MICs of $256{\mu}g/mL$. The compounds kunstlerone (1), isocaryachine (2) and noratherosperminine (3) showed complete inhibition against P. shigelloides, with MIC ${\leq}60{\mu}g/mL$ compare to ampicillin, as a positive control, which showed antigrowth activity against P. shigelloides at MIC $10{\mu}g/mL$.

An 5-Lipoxygenase Inhibitor Isolated from the Roots of Cynanchum wilfordi Hemsley (은조롱뿌리의 5-Lipoxygenase활성 억제성분)

  • The effects of the extract of the root of Cynanchum wilfordi (Asclepiadaceae), the alkaloid fraction, and the isolated main constituent (gagaminine) on 5-lipoxygenase(5-LOEC 1.13.11.34) in bovine PMNL have been studied. The effect of the crude extracts of wildand cultivated plant was also compared each other. Furthermore, the inhibitory effect of gagaminine on 5-Lo was compared with those of the standard drugs. Gagaminine inhibited 5-Lo activity with an $IC_{50}$ value of $26\;{\mu}M$, this result indicates that gagaminine may be useful for in vivo experiments as 5-LO inhibitor.
