The objective of this study is to provide basic information for developing a high-value ginseng product using ginseng saponin and prosapogenin. In order to achieve the proposed objective ginsenoside compositions of Black (BG) and Red (RG) ginseng extract with 95% ethyl alcohol were examined by means of HPLC. The crude saponin and ginsenoside composition of processed ginseng products were analyzed and compared, with BG topping the list with a crude saponin content of 7.53%, followed by RG (5.29%). Ginseng prosapogenin (ginsenosides $Rg_2$, $Rg_3$, $Rg_5$, $Rg_6$, $Rh_1$, $Rh_4$, $Rk_1$, $Rk_3$, $F_1$ and $F_4$) in BG was found to be contained almost 2.6 times as much as that in RG. Ginsenosides $Rg_3$, $Rg_5$, $Rk_1$, $Rh_4$ and $F_4$ in BG in particular were found to be almost 3 times as much as those in RG. $Rg_6$ and $Rk_3$ in BG were also found to be almost 4 times as much as those in RG.
The effects of total ginseng saponin. extracts of Panax ginseng C.A. Meyer, on the differentiation of F9 teratocarcinoma stem cells were studied. F9 stem cells cultured in the presence of ginseng saponin together with dibutyric cAMP became parietal endoderm - like cells. Moreover, the expressions of differentiation marker genes. laminin. type IV collagen. and retinoic acid $receptor-{\beta}(RAR{\beta})$ were increased after treatment of ginseng saponin. Among various ginsenosides purified from crude ginseng saponin, $Rh_1\;and\;Rh_2$ caused the differentiation of F9 cells most effectively. Since ginsenosides and steroid hormone show resemblance in chemical structure. we studied the possibility of the involvement of a steroid receptor in the differentiation process induced by ginsenosides. According to Southwestern blot analysis, a 94 kDa protein regarding as a steroid receptor was detected in F9 cells cultured in the medium containing ginseng saponin. Based on these data, we suggest that ginseng saponin, especially ginsenosides $Rh_1\;and\;Rh_2$ cause the differentiation of F9 cells and the effects of ginsenosides might be exerted via binding with a steroid receptor or its analogous nuclear receptor.
Marine sponges have been a remarkably rich source of pharmacologically active and structurally diverse natural products. As a part of our continuing search for novel secondary metabolites of biomedical importance from marine invertebrate, we encountered the sponge Lipastrotethya sp. from Chuuk, Micronesia. The crude organic extract of this animal exhibited considerable cytotoxicity against the K562 cell line. Guided by the $^1H$ NMR analysis, flash chromatography of the crude extract followed by HPLC yielded a new triterpene glycoside, along with ten known saponins of the sarasinoside class. The structure of this new compound was determined by combined spectroscopic methods such as COSY, HSQC and HMBC experiment. Among these metabolites, six compounds exhibited moderate cytotoxicity against ACHN, MDA-MB-231, NCI-H23 and NUGC-3 cell lines.
In order to identify the sugars of saponin originated from Korean ginseng, experimentations were made on the four kinds of ginseng. The results are summarized as follows. 1. The starch content of Ginseng-tail, White-ginseng 3 and 6 years old-whole ginseng were 10.4, 31.5, 8.2, 25.6% and total sugar of its were 37.0, 61.5, 64.5, 62.5% and free sugar were 7.6, 10.5, 11.3, and 10.7% respectively. 2. Ginseng saponins have been known as main active principles and are quantified as the index components of ginseng and its products for quality control. However ginseng saponins are easily hydrolyzed in acidic solutions of crude drug preparations. Due to the hydrolysis of saponins in acidic condition, it is generally difficult to determine ginseng saponins In crude drug preparations. Ginseng saponins, prosapogenins and sapogenins of crude drug extracts were quantified by HPLC. Ginseng saponins were quantified by HPLC on $Lichrosorb-NH_2$ column with acetonitrile/water/1-butanol(80:20:10, v/v). Ginseng $prosapogenin-Rg_2$ and $-Rg_2$ were extracted with ethyl acetate from $50\%$ acetic acid hydrolyzates of saponin fractions and quantified by HPLC on $Lichrosorb-NH_2$ column with acetonitrile/water(90:10, v/v). Ginseng sapogenins, panafadiol and panaxatriol, were extracted with diethyl ether from $7\%-sulfuric$ acid hydrolyzates of saponin fractions and quantified by HPLC on ${\mu}-Bondapak\;C_{18}$ column with acetonitrile/methano1/chloroform(83:10:7, v/v). These methods of analyses of sapogenins and prosapogenins were more useful for quality control than those of ginseng saponins in some of crude drug preparations.
The effects of variable moisture content, screw speed and barrel temperature on the physicochemical properties of red ginseng powder extrudates were investigated. The raw red ginseng powders were processed in a co-rotating intermeshing twin-screw extruder. Primary extrusion variables were feed moisture content (20 and 30%), screw speed (200 and 250 rpm) and barrel temperature (115 and $130^{\circ}C$). Extruded red ginseng showed higher crude saponin contents (6.72~7.18%) than raw red ginseng (5.50%). Tested extrusion conditions did not significantly affect the crude saponin content of extrudates. Increased feed moisture content resulted in increased bulk density, specific length, water absorption index (WAI), breaking strength, elastic modulus and crude protein content and decreased water solubility index (WSI) and expansion (p<0.05). Increased barrel temperature resulted in increased total sugar content, but decreased reducing sugar content in the extrudate (p<0.05). Furthermore, increased barrel temperature resulted in increased amino acid content and specific length and decreased expansion and bulk density of extrudates only at a higher feed moisture content. The physicochemical properties of extrudates were mainly dependent on the feed moisture content and barrel temperature, whereas the screw speed showed a lesser effect. These results will be used to help define optimized process conditions for controlling and predicting qualities and characteristics of extruded red ginseng.
Relative content(% weight) of proteins, carbohydrates, lipids, and crude ashs in seed of korean green tea plant(Camellia simecis L.) are not different from those in seed of sunflower and safflower. However, the Camellia seed contains much higher crude saponin content(12.2%) than that of sesame(0.29%) or peanut(0.63%). It also contains 82% unsaturated fatty acids including oleic acid and contains tocopherol $(22\;{\mu}g/g,\;{\alpha}-form\;only)$ that is significantly less than of other oil-seed.

To overcome a decrease of Korean ginseng production caused by successive cropping, we have tried to develop a nutrient culture system for Korean ginseng production. For determining the optimal substrate, mixture of sand and TKS-2 (S+T), peatmoss (P), reused rockwool (RR), and granular rockwool (GR) were investigated. The overall physico-chemical properties of RR fell into the reported optimal range for the ginseng cultivation. However, bulk density of S+T was a little higher than that of soil in Korean ginseng fields. The top fresh weight of the ginseng was high in RR and S+T substrates. The root fresh and dry weights in the RR were remarkably greater than that in the conventional soil (CS) of Korean ginseng fields. In terms of ginseng quality, the vitamin C content of ginseng root in nutrient culture was higher than that in CS. However, the contents of crude saponin and total ginsenosides in ginseng between in the nutrient culture and in the soil culture did not show any significant differences.
Biological compounds such as crude saponin and total polyphenol were determined in Allium hookeri roots and leaves. In addition, activities of DPPH radical scavenging activity, hydroxyl radical scavenging activity, ${\alpha}$-amylase inhibition activity and nitrite scavenging were detected in concentrates of 70% ethanol extracts of Allium hookeri roots and leaves. The crude saponin content of the roots and leaves were 4.28 mg/g, and 4.17 mg/g, respectively. Total polyphenol and total flavonoid content was higher in leaves than roots. Furthermore the DPPH radical scavenging activity and hydroxyl radical scavenging activities of leaves was higher than roots. For ${\alpha}$-glucosidase inhibitory activity, 70% ethanol extracts of Allium hookeri roots and leaves showed 60% activity inhibition at a 10 mg/mL concentration. Nitrite scavenging abilities under acidic conditions were most effective for Allium hookeri leaves extracts. These results suggested that Allium hookeri leaves might be used as the components of health functional food.
