Study on Soil Extraction Methods for the Human Health Risk Assessment of Crop Intake Pathway around Abandoned Metal Mine Areas (폐금속광산 지역 농작물섭취경로의 인체위해도 산정을 위한 생물농축계수와 토양분석방법에 관한 연구)

  • Generally, the contribution of crop-intake pathway (CIP) is remarkable in human health assessment (HHA) of heavy metal contamination. Although the crop exposure concentrations (Cp) should directly be used for calculating the average daily dose (ADD) of CIP, the soil exposure concentration (Cs) multiplied by soil-crop bio-concentration factor (BCF) has frequently been used instead of using Cp values. Thus, the BCF values are significant in the HHA, and care should be taken to ensure the reasonable acquisition of BCF values. Meanwhile, the BCF values are known to be significantly affected by analytical methods. Nevertheless, they have been calculated from the concentrations of soil and crop analyzed by only one method: total digestion (aqua regia extraction). For this reason, this study was initiated to seek appropriate soil analysis methods for effective computation of the ADD of CIP. The concentrations of 5 metal contaminants (As, Cd, Cu, Pb, and Zn) in 127 soil samples obtained from 4 abandoned metal mine areas were analyzed by several methods including total digestion and partial digestions using 0.1/1 N HCl, 1M $NH_4NO_3$, 0.1 M $NaNO_3$, and 0.01M $CaCl_2$. The heavy metal concentrations in 127 crop samples (rice grains) were analyzed by total digestion as well. Using the concentrations of soils and crops, the BCF values of each contaminant were calculated according to the kind of soil extraction methods applied. Finally, the errors between Cp and $C_s{\times}BCF$ were computed to evaluate the relevance of each method. The results indicate that the partial extraction using 0.1 N and 1 N HCl was superior or equivalent to total digestion. In addition, the 0.1M $NaNO_3$ method combined with total digestion is recommended for improving the reliability of BCF values.

  • Objectives : AMP-activated protein kinase (AMPK) is a key metabolic regulator that reduces lipogenesis. AMPK is mainly activated via phosphorylation of liver kinase B (LKB) 1 under energy stress. Here, we highlighted the anti-obesity effect and underlying mechanism of Coix lacryma-jobi var. mayuen Stapf sprout water extract (CSW) sprout extract in connection with the LKB1/AMPK signaling pathway. Methods : C57BL/6 mice (20~25 g) fed HFD to induce obesity and at the same time administered CSW 100 mg/kg (CSWL; (CSWL; CSW low concentration) or CSW 200 mg/kg (CSWH; CSW high concentration) or Garcinia extract (Garcinia) 200 mg/kg orally for 6 weeks. Body weight and food intake were measured at the same time each day. After 6 weeks of CSW administration, liver tissue and serum were obtained through an autopsy. After the end of the experiment, biochemical analysis (triglycerides (TG), total cholesterol (TC), HDL-cholesterol, and LDL-cholesterol) was performed on the serum. And then, protein levels related to TG and TC synthesis were measured through western blot analysis in liver tissue. Results : As a result, serum TG, TC, and LDL-cholesterol levels were significantly increased in the control group and significantly decreased in the CSW administration group. On the other hand, the HDL-cholesterol level was increased in the CSW-administered group. And as a result of Western blot analysis, CSW significantly increased the phosphorylation of LKB1 & AMPK, and remarkably decreased the expression of factors related to TG and TC synthesis. Conclusions : Our findings suggest that CSW influences the TG and TC synthesis to positively affect HFD-induced obesity in C57BL/6 mice.

Comparison of Human Health Risk Assessment of Heavy Metal Contamination from Two Abandoned Metal Mines Using Metal Mine-specific Exposure Parameters (국내 폐금속 광산에 특화된 노출인자를 이용한 두 폐금속 광산 중금속 오염에 대한 인체위해성평가 비교)

  • There are numerous closed and abandoned mines in Korea, from which diverse heavy metals (e.g., As, Cd, Cu, Pb, Zn) are released into the surrounding soil, groundwater, surface water, and crops, potentially resulting in detrimental effects on the health of nearby residents. Therefore, we performed human risk assessments of two abandoned metal mines, Yanggok (YG) and Samsanjeil (SJ). The exposure parameters used in this assessment were specific to residents near mines and the included exposure pathways were relevant to areas around metal mines. The computed total excess carcinogenic risks for both areas exceeded the acceptable carcinogenic risk ($1{\times}10^{-6}$), indicating that these areas are likely unsafe due to a carcinogenic hazard. In contrast, the non-carcinogenic risks of the two areas differed among the studied receptors. The hazard indices were higher than the unit risk (=1.0) for male and female adults in YG and male adults in SJ, suggesting that there are non-carcinogenic risks for these groups in the study areas. However, the hazard indices for children in YG and female adults and children in SJ were lower than the unit risk. Consumption of groundwater and crops grown in the area were identified as major exposure pathways for carcinogenic and non-carcinogenic hazards in both areas. Finally, the dominant metals contributing to carcinogenic and non-carcinogenic risks were As and As, Cu, and Pb, respectively. In addition, the carcinogenic and non-carcinogenic risks of YG were evaluated to be 10 and 4 times higher than those of SJ, respectively, resulted from the relatively higher exposure concentration of As in groundwater within SJ area. Because of lacking of several exposure parameters, some of average daily dose (ADD) could not be computed in this study. Furthermore, it is likely that the ADDs of crop-intake pathway included some errors because they were calculated using soil exposure concentrations and bioconcentration factor (BCF) rather than using crop exposure concentrations.
