Diversity and Pathogenicity of Fusarium Species Associated with Grain Mold of Sorghum (수수 이삭곰팡이 증상에서 분리한 Fusarium속 균의 다양성 및 병원성)

  • Sorghum (Sorghum bicolor Moench) was traditionally grown on a small scale, however, at present its cultivation is getting momentum in terms of food and animal feed crop throughtout the Korea. Grain mold symptoms of the plant were frequently observed during disease surveys in Korea from 2007 to 2009. The symptoms were highly variable. Severely infected grain was fully covered with mold and partially infected grain may look normal or discolored. Ninety isolates of Fusarium species were obtained from the diseased plants collected from several locations in the country. Among the collected Fusarium isolates, 41 were identified as Fusarium thapsinum, 23 as F. proliferatum, 12 as F. graminearum, 5 as F. incarnatum, and 3 as F. equiseti based on their morphological and cultural characteristics. Elongation factor 1 alpha gene sequences of the isolates were used for phylogenetic analysis. Analyses of the sequences revealed that the isolates were confirmed to be identical with related species of NCBI GenBank. Pathogenicity tests showed that three dominant species, F. thapsinum, F. proliferatum and F. graminearum were strongly virulent to grains of sorghum. This study is the first report of sorghum grain mold caused by Fusarium species in Korea.

Tuber Enlargement and Chemical Components of Yams (Dioscorea opposita Thunb.) (둥근마(Dioscorea opposita Thunb.)의 괴경비대 및 성분특성)

  • Tuber yield and content of general component and diosgenin which is a main bioactive property were investigated in order to determine the growth characteristics of round typed yam(Dioscorea opposita L.) and the potential of artificial culture at Suwon, Korea. Tubers of round yam were initiated to form at 60 days after planting and then enlargement of tubers lasted by 160 days after planting. Compared to short typed yam(108g), tuber weight of round yam was higher(127g) on the basis of dry weight at 200 days after planting. In comparison of general component between round yam and short yam, protein of round yam$(3.62\%)$ was higher than short yam$(2.10\%)$. Water content in round yam$(64.5\%)$ was lower in short yam$(79.4\%)$, indicating a higher dry weight ratio of round yam. Hardness of round yam was 2787.6 while short yam showed about two times higher hardness(4946.9). Lightness was higher in round yam(77.4). In tuber extracts analysis, diosgenin content was respectively $3.32\%$ in round yam and $2.61\%$ in short yam.

Chemical Analysis on Biologically Active Substances among Habitats of Allium victorialis for a High Income Crop (산마늘의 고소득 작물화를 위한 기능성 물질 분석)

  • When the contents of the constituents such as total amino acids, free amino acids, volatile organosulfuric compounds and steroidal saponins among three origins in the aerial-and underground parts of Allium victorialis, it was suggested that the characteristic components regarding to quality evaluation could be differed according to the purpose of utilization. For the utilization of amino acids, underground parts of this plant was shown to be better than aerial part. In addition, Ulung island origin was found to contain the highest amino acids content among the three origins though the difference was small. The amino acids showing remarkably high contents were appeared to be arginine, glutamine and asparagine. In the volatile organosulfuric compounds, the origina of Mt. Odae and Mt. Chiri positioned in inland showed higher contents than Ulung island origin geographically positioned in the ocean. Inland origins were shown to contain higher organosulfurie component contents in aerial parts than in underground parts while those of Ulung island origin were higher in underground parts than aerial parts. Underground parts, regarding to saponin constituents, showed higher contents than aerial parts. Underground parts of Ulung island origin were shown to contain more saponins than those of other two origins and the sequence of the contents was in the order of Ulung island>Mt. Chiri>Mt Odac.

Characteristics of Agronomy Traits to Transgenic Rice Selected by Molecular Breeding Method (분자육종기법에 의해 선발된 형질전환 벼 계통의 작물학적 특성)

  • This study was carried out to develop new cultivars using the $T_5$ generation of transformed rice by PCR analysis with DNA marker in each generation $(from\;T_3\;to\;T_5)$. In the previous study, we successfully developed the transgenic rice plants over-expressing the Arabidopsis $H^+/Ca^{2+}$ antiporter CAX 1 (accession no. U57411) gene. The calcium concentration in brown rice of transgenic plants was higher than that of donor plants, Iipum, and was selected 3 lines out of 25 lines at cultured GMO field. The major agronomic traits such as culm length, panicle length and panicle number of 3 lines at transgenic plants $(T_5)$ were similar to wild type. Also these lines appeared to have disease resistance to rice blast, cold resistance as compared with donor types. The grain shape was similar to donor plant, however, the 1000 grain weight of brown rice was different from transgenic plants. These finding would be used for basic data of new variety registration.

Effects of Transgenic Soybean Cultivation on Soil Microbial Community in the Rhizosphere (형질전환 콩 재배가 근권 토양 미생물상에 미치는 영향)

  • BACKGROUND: Soybean [Glycine max (L.) Merrill] is a legume and an important oil crop worldwide. This study was conducted to evaluate the possible impact of transgenic soybean cultivation on the soil microbial community. METHODS AND RESULTS: Microorganisms were isolated from the rhizosphere soils. Microbial community was identified based on the culture-dependent and molecular biology methods. The total numbers of bacteria, fungi, and actinomycete in the rhizosphere soils cultivated with transgenic and non-transgenic soybeans were similar to each other, and there was no significant difference between transgenic and non-transgenic soybeans. Dominant bacterial phyla in the rhizosphere soils cultivated with transgenic or non-transgenic soybeans were Actinobacteria, Firmicutes, and Proteobacteria. The microbial communities in transgenic and non-transgenic soybean soils were characterized using the denaturing gradient gel electrophoresis (DGGE). The DGGE profiles showed the different patterns, but didn't show significant difference to each other at 0.05 significance level. DNAs were isolated from soils cultivating transgenic or non-transgenic soybeans and analyzed for persistence of transgenes in the soil by using PCR. PCR analysis revealed that there were no amplified ${\gamma}$-tmt and bar gene in soil DNA. CONCLUSION(S): The results of this study suggested that microbial community of soybean field were not significantly affected by cultivation of the transgenic soybeans.

Spatio-temporal Distribution of Phytoplankton Community in the Jangsu Bay and Adjoining Sea of South Sea, Korea (장수만 식물플랑크톤 군집의 시.공간적 분포 특성)

  • This study describes about the spatio-temporal distributions in phytoplankton community such as species composition, standing crop and dominant species from May 2006 to February 2007 in the Jangsu bay and the northwestern parts of Gamak bay. Based on the principal component analysis (PCA) of the environmental factors as well as biological parameters, the bio-oceanographical characteristics were analysed. A total of 83 species of phytoplankton belonging to 47 genera were identified. Whereas diatoms and dinoflagellates occupied more than 65% and 30% of total species, respectively. The annual dominant species were Chaetoceros affinis, Paralia sulcata and Bacillaria paxillifera in spring, Chaetoceros didymus, Ch. affinis and Octactis octonaria in summer, Skeletonema costatum-like species and B. paxillifera in autumn. Moreover phytoplankton cell density was ranged between 3.1 $cells{\cdot}mL^{-1}$ in spring and 521.0 $cells{\cdot}mL^{-1}$ in winter. It fluctuated with an annual mean of 76.0 $cells{\cdot}mL^{-1}$ between the lowest value of 7.6 $cells{\cdot}mL^{-1}$ in spring and the highest value of 220.2 $cells{\cdot}mL^{-1}$ by Skeletonema costatum-like species in winter. Briefly, the phytoplankton cell density in the mixing seasons was higher in comparison with the other seasons. According to the PCA, the biological oceanographic characteristics of the Jangsu bay was affected by the introduction of outside seawater particularly in temperature increasing seasons, and the other seasons, it may be described the light intensity, and mix between inner and outer bay sea waters.

Factors Affecting Genetic Transformation of Italian Ryegrass (이탈리안 라이그래스의 형질전환에 미치는 몇 가지 요인의 영향)

  • A system for the production of transgenic plants has been developed for Italian ryegrass(Lolium mult리orum Lam.) via Agrobacterium-mediated transformation of embryogenic callus. Mature seed-derived calli were infected and co-cultured with Agrobacterium EHA101 carrying standard binary vector pIG121Hm encoding the hygromycin phosphotransferase(HPT), neomycin phosphotransferase II (NPTII) and intron-oontaining $\beta$g1ucuronidase( intron-GUS) genes in the T-DNA region. The effects of several factors on transformation and the expression of the GUS gene were investigated. Inclusion of 200${\mu}M$ acetosyringone(AS) in inoculation and co-cultivation media lead to a significant increase in stable transformation efficiency. Increasing Agrobacterium cell density up to 1.0 in $OD_{600}$ during infection increased transfonnation efficiency of embryogenic calli. The highest transfonnation efficiency was obtained when embryogenic calli were incoulated with Agrobacterium in the presence of 0.1% Tween20 and 200${\mu}M$ AS. Hygromycin resistant calli were developed into complete plants via somatic embryogenesis. GUS histochemical assay and PCR analysis of transgenic plants demonstrated that transgenes were integrated into the genome of Italian ryegrass.

Notes on Rare Species of Mycoparasitic Forming Fungus in Korea (국내에서 발견된 균류에 기생하는 희귀종 버섯)

  • One interesting parasitic forming fungus, being grown on the eggs of Phallus impudicus was collected from Mulchajoreum and on woods of Saryeoni in Jeju island. This taxon identified as a Podostroma solmsii (E. Fisch.) S. Imai f. octospora Yoshim. Doi (Hyporeaceae) based on morphological characteristics. According to the interpretation of Doi(1978), this taxon hsving the stipitate, genus Podostroma, family Hypocreaceae, Hypocreales, Ascomycota was identified as Podostroma solmsi f. octospora and its korean common name designated by authors. This species is characterized in having the fusiforme to ellipsoidal, unicellular and warted ascospores without septate and with blunt apiculus at each end and mycoparasitic habitat. Although there is some mycologists suggest that this taxon having above characters needs to be derived from Hypomyces. But authors are primarily going to record this species as a named "Podostroma solmsii (E. Fisch.) S. Imai f. octospora Yoshim. Doi" for the fungus flora of Korea. In a future, it is need in detail to compare their DNA sequencing data on the type species and some related species. All collections cited here are deposited in the Herbarium Conservation Center of National Academy of Agricultural Sciences.

A Study on the Development of Model on Environmental Education Program for Livestock Farmer -Focused on Hongsung-gun, Chungcheungnamdo- (축산업자 환경교육 프로그램 개발 연구 - 충청남도 홍성군을 중심으로 -)

  • Livestock agriculture plays an essential role Hongsung's economy. Unfortunately, if manure is manage properly it can contaminate water and pollute the air. The purposes of this study were to provide awareness of environmental impact and to design educational programs with sustainable agriculture. I used the specific steps of processes. First, collecting informations through survey on environmental awareness of livestock farmers in Hongsung-gun and recognizing the tendencies. Second, analyzing on educational status of livestock farmers. Third, finding objectives of environmental educational programs. At the end of processes, establishing programs which concerning on environmental education. I used two research methods to gather and analyze the data for developing an environmental education program. The first one is a general survey among 50 livestock farmers from Hongseong-gun, Chungcheongnam-do. The second one is the in-depth interviews with specialists in environmental education and government officials from the central government - the Ministry of Agriculture and Forestry - and the local government - Hongsung-gun who are in charge of livestock farming. I found the following results from this research. First, most livestock farmers replied with 'highly likely' to the five items on the recognition of the necessity of environment preservation, whereas only 38.7% answered with 'likely' to the item on the environmental pollution caused by livestock farming. This shows the tendency that they have less recognition of the reality than necessary. Second, most of the contents among 5 areas(knowledge, recognition, function, attitude, and participation) are the knowledge areas. That results are analyzed training materials for livestock farmers made by Hongsung-gun in 2004 and in 2005 by using goal categories suggested in the Tbilisi Resolution. Third, after analyzing the survey and in-depth interviews, I found that the need for education to recognize the environmental pollution, environmental protection and recycling of the livestock waste. We can summarize conclusions from this research. First, we need on educational program improve the livestock farmers' recognition of importance in environmental protection. Second, it is necessary to develop educational program with the viewpoint that livestock waste is not one of the culprits of the environmental pollution, but a recyclable resource. Third, contents of the environmental education program, should include all the educational elements, namely, knowledge, recognition, attitude, function and participation. The education for livestock farmers requires a research on the environmental education program that can be used with crop farmers who can use the resources recycled from the livestock waste from the viewpoint of recycling resources. This research is focused on the collection and analysis of the basic data for developing an environmental education program and the composition of the program. A further research on the implementation and assessment of this program is required.

Quantitative analysis of rutin with mulberry leaves (II) (뽕나무 유전자원의 rutin 함량 변이(II))

  • We analyzed rutin content using mulberry genetic resources. They were grown under the same environment and conditions. Mulberry leaves were collected and then freeze-dried and powdered for rutin test. As a result, mean content of mulberry strains was $0.38{\pm}0.17%$, and the coefficient of variation (CV) was 45.7%. The variation was greatly severe. Among the tested strains, 'Jangloe' was showed the highest content of 0.97%, whereas 'Sungsu 3' was showed the lowest content of 0.01% respectively. Finally we selected rutin high-containing 6 strains. They are as follows. 'Jangloe', 'Pumbo 6', 'Youjin', 'Kaeryangjeonjeon', 'Baekhak', and 'Pumbo 32' which are more than twice of the overall average content.
