  • The OJIP transients were investigated from leaves of Crinum asiaticum var. japonicum, Osmanthus insularis, Chloranthus glaber and Asplenium antiquum, in order to screen the indicators for quantification of environmental stresses and to apply them to the plant vitality test. The obvious diurnal changes or OJIP transients were found in Crinum asiaticum var. japonicum, Osmanthus insularis growing in sunny habitats, showing lower Fp and higher Fo in day time. The values of SF $I_{no}$ and P $I_{NO}$ increased dramatically in day time in leaves of Crinum asiaticum var. japonicum, Osmanthus insularis, and Asplenium antiquum. Although the value of RC/CS decreased slightly in day time in leaves of 4 tropica $l_tropical plants, the obvious diurnal changes or OJIP transients were not seen in winter, suggesting that tropica $l_tropical paints used in this research seemed to be irreversibly damaged in winter. However, the values of ETo/RC and Ψo/(1-Ψo) increased significantly in leaves of Crinum asiaticum var. japonicum, indicating that ETO/RC and Ψo/(1-Ψo) were useful as indicator parameters for scensing of low temperature.ature.

The Mechanism of Crinum asiaticum var. japonicum on the Activation of Anagen (문주란의 모발 성장기 유도 기전)

  • Crinum asiaticum var. japonicum and its active component, norgalanthamine have been reported to have hair growth-promoting effect via the proliferation of dermal papilla cells. In this study, we investigated the other mechanisms of C. asiaticum extract var. japonicum and norgalanthamine on the hair growth. The C. asiaticum var. japonicum extract inhibited $5{\alpha}$-reductase activity by 16%, which converts testosterone to dihydrotestosterone (DHT), a main cause of androgenetic alopecia, whereas the C. asiaticum var. japonicum extract didn't function as an opener of the $K_{ATP}$ channel. On the other hand, we examined whether norgalanthamine can inhibit transforming growth factor-${\beta}$ (TGF-${\beta}$) signal pathway, which is essential in the regression induction of hair growth. Norgalanthamine inhibited the phosphorylation of Smad2/3 on TGF-${\beta}1$-induced canonical pathway in human keratinocyte HaCaT cells. These results suggested that the C. asiaticum var. japonicum extract and norgalanthamine had the potential to influence hair growth through the inhibition of $5{\alpha}$-reductase activity and TGF-${\beta}1$-induced canonical pathway.

Pharmacological Actions of Crinum folium (나군대 잎의 약리 효과에 관한 연구)

  • Crinum asiaticum var. japonicum is a wild plant growing only in Jeju-island, Korea, and in Japan. The whole part of this plant has been known to have the pharmacological actions such as analgesic, anti-inflammatory, platelet-aggregation inhibitory, antitussive, and expectorant. With these assumed actions, the leaves (Crinum folium) of this plant has been used in the folk remedies for arthritis and arthralgia. There is, however, no scientific evidences for the pharmacological actions of Crinum asiaticum var. japonicum. In the present study, the analgesic, anti-inflammatory, and platelet-aggregation inhibitory actions of Crinium folium were evaluated using writhing test, tail-flick test, carrageenin antiedema test, in vitro thromboxane $B_2$ quantitation assay and in vitro platelet aggregation test. In order to obtain the partially purified fraction whose pharmacological action is excellent, the methanol extract of Crinium folium was fractionated consecutively into four biological fractions such as ether, ethyl acetate, butanol, and water fractions and their pharmacological actions of the fractions were investigated. Putting our results together, Crinium folium, especially ethyl acetate fraction was proven to have significant analgesic, anti-inflammatory and platelet-aggregation inhibitory actions by inhibition of prostanoids biosynthesis as one of its mechanism of action.

Anti-inflammatory Activity of Crinum asiaticum Linne var. Japonicum Extract and its Application as a Cosmeceutical Ingredient (문주란의 항염효과와 화장료적 특성)

  • Crinum asiaticum Linne var. japonicum has long been used as a rheumatic remedy, an anti-pyretic, an anti-ulcer treatment, and for the alleviation of local pain and fever in Korea and Malaysia. In order to investigate the possibility of Crinum asiaticum Linne var. japonicum extract as a cosmetic ingredient, we measured its anti-inflammatory effect by inhibition of iNOS (inducible nitric oxide synthase), and the release of PGE2, IL-6, and IL-8. HPLC experiment after extraction with 95% ethanol at pH 3.5 showed that Crinum asiaticum Linne var. japonicum was mainly composed of lycorine (up to 1%), a well-known immunosuppressant. The content of lycorine varied depending on the type of tissue analyzed and the extraction method. In anti-inflammatory assay for inhibition of nitric oxide formation on lipopolysaccharide (LPS)- activated mouse macrophage RAW 264.7 cells, the ethanolic extract of Crinum asiaticum showed inhibitory activity of NO production in dose-dependent manner ($IC_{50} = 83.5 {\mu}g/mL$). Additional study by RT-PCR demonstrated that the extract of Crinum asiaticum significantly suppressed the expression of the iNOS gene. Moreover, the extract of Crinum asiaticum did not show my cytotoxicity, but did show cell proliferation effect against LPS ($10{\sim}60%$ increase of tell viability). In an assay to determine inhibition of the $H_2O_2$-activated release of PGE2, IL-6, and IL-8 in human normal fibroblast cell lines, the release of PGE2 and IL-6 was almost completely inhibited above concentrations of 0.05% and 1%, respectively. Moreover, the release of IL-8 was completely inhibited over the entire range of concentrations (> 0.0025%). The result showed that the extract of Crinum asiaticum Linne var. japonicum has sufficient anti-inflammatory effect. There-fore, Crinum asiaticum Linne var. japonicum extract may be useful as an ingredient of cosmetic products.

Analysis of O-J-I-P Transients from Four Subtropical Plant Species for Screening of Stress Indicators under Low Temperature (저온스트레스 지표 선발을 위한 아열대성 식물 4종의 O-J-I-P곡선 분석)

  • The changes of O-J-I-P transients were investigated using leaves of four subtropical plant species (Crinum asiaticum var. japonicum, Osmanthus insularis, Chloranthus glaber and Asplenium antiquum) under the natural conditions in winter, in order to select the stress indicators for diagnosing physiological states of plants under low temperature. In the O-J-I-P transients of these species, the fluorescence intensity was found to be higher in O-step and lower in P-step in winter than in summer. Particularly, the fluorescence intensity of the P-step in Crinum asiaticum var. japonicum was lower than those of other three plant species, indicating that Crinum asiaticum var. japonicum is the most sensitive to low temperature. Of the chlorophyll fluorescence parameters derived from O-J-I-P transients of four subtropical plants, Fm, Fv/Fo, ABS/CS, TRo/CS, $\phi_{po}$ and $\phi_{po}/(1-\phi_{po})$ decreased significantly with the increase of Fo, Sm, N, EToICS, ETo/RC and $\psi_o/(1-\psi_o)$ depending on temperature drop in winter. Therefore, these parameters could be used as indicators for estimating low temperature stress and diagnosing physiological states of plants under the natural conditions in winter.

Chlorophyll Fluorescence and Antioxidative Enzyme Activity of Crinum Leaves Exposed to Natural Environmental Stress in Winter (겨울철 자연환경에 노출된 문주란 잎의 엽록소형광과 항산화효소 활성에 관한 연구)

  • Chlorophyll fluorescence and antioxidative enzyme activity were investigated from leaves of Crinum asiaticum var. japonicum under the natural condition in winter, in order to monitor plant response and physiological states such as vitality, productivity and so on. In the O-J-I-P transients, the fluorescence intensity of J, I, P-step decreased remarkably depending on temperature drop in winter. The photochemical efficiencies of PSII, Fv/Fm, were significantly low in late winter with decrease of Fm. These results indicate that Crinum plants were affected by seasonal drop of temperature. The catalase activity significantly decreased depending on temperature drop in winter. However, the activity of superoxide dismutase ascorbate peroxidase and peroxidase slightly increased in winter while some isoenzymes appeared in winter. These results, with the remarkable decrease of Ev/Fm in winter, represent that Crinum plants were exposed to oxidative stress and subsequently damaged leading to cell death.

  • The diurnal and seasonal variations of antioxidative enzyme activity and the O-J-I-P transients were investigated from the leaves of Crinum asiaticum var. japonicum under winter stress in natural habitat, in order to diagnose quantitatively physiological states of plants under stresses. The activities of superoxide dismutase and peroxidase increased slightly in winter. Especially, peroxidase acitivity was higher at dawn and night in winter and some isoforms were detected only in early winter. In the O-J-I-P transients, the fluorescence intensity of J, I, P steps decreased significantly in winter season, contrary to its high value in summer season. Of the chlorophyll fluorescence parameters derived from the O-J-I-P transients, Fm and $\Phi_{po}$ decreased with the increase of ABS/RC depending on temperature drop in winter.

Photochemical Efficiency and psbA Gene Expression of Crinum Leaves under Natural Environmental Stress in Winter (겨울철 저온스트레스에 의한 문주란의 광합성효율과 psbA 유전자의 발현양상)

  • The change of chlorophyll fluorescence parameters, O-J-I-P transients and psbA gene expression were investigated in the leaves of Crinum asiaticum var. japonicum on the natural condition in winter, in order to elucidate physiological responses of photosystem II (PS II) activity to winter stresses. The photochemical efficiencies of PS II, Fv/Fm, were significantly low in winter, contrary to its high value in summer. The values of I -qN and I-qP were lower in midday than at dawn or night both in summer and winter, although their decrease in midday was less in winter than in summer. In the O-J-I-P transients, the fluorescence intensity of J, I, P-step decreased remarkably depending on temperature drop in winter. And the D I reaction center protein of PS II decreased in late winter more than in early winter, concomitantly with relatively high content of description products of psbA gene in midday. These results indicate that low temperature in winter causes irreversible damage to PS II and subsequently leads to cell death.

  • The diurnal changes of chlorophyll fluorescence and antioxidative enzyme activity were investigated in the leaves from four subtropical plant species (Crinum asiaticum var. japonicum Bak., Osmanthus insularis Koidz., Asplenium antiquum Makino and Chloranthus glaber Makino) under the natural habitats in summer and winter. The intensity of chlorophyll fluorescence was lower in O-, I-, J-, P-steps of O-J-I-P transient in winter than summer, and prominent diurnal change was not found in the fluorescence intensity of four subtropical plant species in winter. The activity and isoenzyme pattern of SOD and catalase did irregularly change seasonally and diurnally in four subtropical plant species. In contrast, the peroxidase activity and isoenzyme pattern was different depending on plant species and growth seasons; The activity increased slightly more in winter than in summer in four subtropical plant species, and several isoenzymes appeared in the leaves from C. asiaticum var japonicum, O. insularis and A. antiquum in winter.

Apoptosis Induction of HL-60 Leukemia Cells by Extract of Crinum asiaticum (문주란 추출물의 HL-60 백혈병 세포 Apoptosis 유도 효과)

  • The present study investigated the antiproliferative effects of Crnum asiaticum var. japonicum against HL-60 human leukemia cells. The 80% MeOH extract or several solvent fractions from the C. asiaticum inhibited the growth of HL-60 cells, whereas the growth of HEL-299 cells, human embryonic lung fibroblast, was scarcely inhibited. When the HL-60 cells were treated with the $CHCl_3$ fraction, the BuOH fraction, the EtOAc fraction and the $H_2O$ fraction, DNA ladder, chromatin condensation and increase of sub-G1 hypodiploid cells were observed. Furthermore, the $CHCl_3$ fraction and the BuOH fraction reduced Bc1-2 mRNA level, whereas Bax mRNA level was increased. These results suggest that the inhibitory effect of C. asiaticum on the growth of the HL-60 cell might be mediated through the induction of apoptosis via the down-regulation of Bc1-2. Taken together, components of C. asiaticum might have a therapeutic potential for the treatment of human leukemia.
