To evaluate the sanitary conditions of four species of commercial shellfish (Crassostrea gigas, Tapes japonica, Mytilus coruscus and Anadara granosa), the psychrotrophic coliforms and the bacterial flora were monthly examined from January to December, 1978. Viable cell counts of the samples submitted were $1.3\times10^3\~2.5\times10^6/g$ and the numbers of coliform and E. coli were above $0\~5.5\times10^2/g\;and\;2\~9.2\times10^2/100g$ respectively. Among the 373 coliform bacteria isolated from the samples submitted, 298 strains were identified as coliform by IMViC test, $44^{\circ}C$ culture and gelatin liquefaction test. E. coli type I and K. aerogenes type I were the most predominant ones among the identified coliform as 138 strains and 71 strains respectively. The psychrotrophic coliform growing at $5^{\circ}C$ were 186 $strains(50\%)$ among 373 strains of isolated coliform. Vibrio and Pseudomonas were the most predominant groups. Among 453 strains of isolated bacteria, Vibrio was 240 $strains(53\%)$ and Pseudomonas was 91 $strains(20\%)$. Moraxella, Acinetobacter, Enterobacteriaceae and Flavobacterium-Cytophage were scantly detected. In seasonal variation of general bacterial flora, a lot of Vibrio appeared from May to October, did Moraxella during the winter season and the other genuses showed no seasonal change.
In coastal areas that are affected by continuous, seasonal or occasional pollution sources, bivalves accumulate biological contaminants such as pathogenic bacteria. We investigated the effect of natural seawater relaying and electrolyzed seawater depuration on the bacteriological quality of artificially contaminated oysters Crassostrea gigas and short-necked clams Ruditapes philippinarum to suggest an alternative method of shellfish sanitation control.When artificially contaminated oysters and short-necked clams (fecal coliform level 1,700 MPN/100 g) were relayed into a sea area of safe bacteriological water quality, the fecal coliform level dropped to below 110 MPN/100 g after 1 day. The bacteriological quality of oysters and short-necked clams that are contaminated at a fecal coliform level of 1,700 MPN/100 g could be improved, and become appropriate for raw consumption by a single day relay under proper environmental conditions. When artificially contaminated oysters (fecal coliform level 330 MPN/100 g) were depurated with electrolyzed seawater, 94% of fecal coliform was eliminated after 12 h and fecal coliform was undetectable after 24 h. After 24 h depuration with electrolyzed seawater, the fecal coliform level of short-necked clams with initial fecal coliform of 2,400 MPN/100 g was below 20 MPN/100 g. However, the fecal coliform level of short-necked clams with initial fecal coliform of 17,000 MPN/100 g was relatively high, at 790 MPN/100 g, even after 24 h of depuration with electrolyzed seawater, because of the repeated cycle of excretion and accumulation of fecal coliform in shellfish tissue under the closed depuration environment. Such natural seawater relaying and electrolyzed seawater depuration can be restrictively applied to improve or secure the bacteriological quality of oysters and short-necked clams in accordance with safety levels for bivalves for raw consumption.
The purpose of this study is to develop a sanitary aquarium for the safety slices of raw fish by using U.V. light. Water re-circulating system was composed of two tanks. One of the tanks $(90\times45\times45cm\;in\;size),$ was used for rearing fish and the other $(90\times45\times45cm\;in\;size),$ with 37 pieces of corrugated plastic plates was used for the growth of Nitrosomonas and Nitrobacter to remove ammonia from the water. Consequently, bactericidal effects of U.V. light were examined under the controlled condition of water with flow rate 730m1/sec (water flow thickness: 10mm), the width 41cm of water flow route, and the distance 4.75cm from the lamp to its water bottom, and U.V. light 75W (5 lamps). The water of the aquarium tank will be theroetically circulated 1 cycle per 18 min. In these conditions the bactericidal effect was $85\%$ just after passing through U,V. light and 3 log cycle in aquarium tank water. The count of Vibrio parahaemolyticus just after irradiation was decreased by about over than 3 log cycle. Under the irradiation for 72 hours, viable cell counts in both skin and gill of fish reared were decreased into about 2 log cycle, but there was no significant decrease in viscera. When the temperature of the tank was controlled at about $20-23^{\circ}C$ under the same condition, viable cell counts were reduced about 2 log cycle, and fecal coliforms were reduced about 1 log rycle and 3 log cycle in Crassostrea gigas and Mytilus edulis, respectively.
The change of cell counts of Vibrio vulnificus in meat homogenates of fish and shellfish by the storage time and temperature was examined to get basic information for precautionary steps against septicemia from slices of raw fish (sashimi). Therefore, we inoculated raw and cooked meat homogenates of fish and shellfish with Vibrio vulificus M-8 (isolated from shellfish ) and stored them at $-20^{\circ}C,\;4^{\circ}C\;and\;30^{\circ}C$ for 72 hours. Vibrio vulnificus M-8 was not detected in 32 hours when it was frozen and stored at $-20^{\circ}C$ after inoculating them into phosphate buffer solution at concentration of $10^5\;cell/ml$, while the existance of Vibrio vulnificus was identified after 72 hours of storage at the same temperature in case of inoculation into the meat homogenate of yellow tail. The cell count of Vibrio vulnificus was decreased as about $20\%$ of initial count after 2 hours storage at $4^{\circ}C$ in phosphate buffer solution with fish and shellfish homgenates. From the experimental results it was recognized that Vibrio vulnificus was labile to the cold stress. In comparison to the growth of growth of Vibrio M-8 at $30^{\circ}C$ in the raw and cooked meat of the yellow tail(Seriola guingueradita), snapper(Chrysophrys major), ark shell(Anadra brouhgtonii), and oyster(Crassostrea gigas), the raw meat homogenates were more excellent than the cooked ones though all fish and shellfish meat homogenates were proves to be good for the growth of the microbe.
The objectives of this study were to investigate color patterns of shell and mantle edge pigmentation of a Pacific oyster, C. gigas, and to estimate variance components of the two colors. A sample of 240 F0 oysters was collected from six aquaculture farms in Tongyeong, Korea to measure shell color and mantle edge pigmentation. Among the F0s, male and female individuals with black (white) shell and black (white) mantle edge were selected and mated to generate three F1 full-sib black (white) cross families (N = 265). Two and four F2 cross families (N = 286) were also produced from black and white F1 selected individuals, respectively. Variance component estimates due to residuals and families within color were obtained using SAS PROC VARCOMP procedures to estimate heritability of shell and mantle edge pigmentation. In the F0 generation, about 29% (11%) had black (white) color for both shell and mantle edge. However, in the F1 and F2 black (white) cross families, 75% (67%) and 100% (100%) of oysters had black (white) shell colors, and 59% (23%) and 79% (55%) had black (white) mantle edge, respectively. Spearman correlation coefficients between shell and mantle edge color were 0.25, 0.74, and 0.92 in F0, F1, and F2 generations, respectively, indicating that, with generations of selection process, an individual with black (white) shell color is more likely to have black (white) mantle edge pigmentation. This suggests that shell color could be a good indicator trait for mantle edge pigmentation if selection of both the colors is implemented for a couple of generations. Estimates of heritability were 0.41 and 0.77 for shell color and 0.27 and 0.08 for mantle edge pigmentation in the F1 and F2 generations, respectively, indicating that, in general, significant proportions of phenotypic variations for the shell and mantle edge colors are explained by genetic variations between individuals. These results suggest that the two color traits are inheritable and correlated, enabling effective selection on shell and mantle edge color.
A field survey and experiment on aquaculture in Costa Rica including Nicoya Gulf was carried out for a month, December $2{\sim}28$, 1981. 1) Air temperature was $27{\sim}32^{\circ}C$, water temperature, $28{\sim}29.5^{\circ}C$ and salinity, $20{\sim}28.5\%$ both in the surface and 5 m layer on December 22, 1981. 2) Dissolved oxygen plentiful, $7.3{\sim}9.95\;ml/l$. 3) Mean precipitation for the past ten years in Nicoya Gulf area was $1298.6{\sim}2196.3$ mm near Puntarenas area and rainy days a year was 137.7, Most rainfalls was concentrated from May to November, the rainy season. 4) Precipitation in main oyster farms near Chungmu, Korea was 1836.0mm a year and it was similar to that of Costa Rica. 5) Diatoms were 16 species belonged to 13 genera and dinoflagellates, 3 species belonged to 2 genera. Standing crops was $639{\sim}130,211\;cells/l$ and dominant species were Chaeteocros curvisetus, Chaetoceros decipiens, Nitzschia serata. Thalassiothrix mediterranca, a tropical esturary species, was found. 6) Nicoya Gulf has the good environmental factors in the water temperature, salinity, dissolved oxygen and planktons, and it is desirable to experiment and culture the oyster (Crassostrea rhrizophorae).
To compare the accumulation of paralytic shellfish poison (PSP) in different marine organisms, the occurrence and variation of PSP were surveyed in blue mussel Mytilus edulis, oyster Crassostrea gigas, short neck clam Ruditapes philippinarum, bay scallop Argopecten irradians, and warty sea squirt Styela clava collected from Jinhae Bay, Korea, in 2005 and 2006 year. We also investigated the ability of the blue mussel to detoxify PSP by relaying and depuration (via the water flow or water circulation system). In the marine organisms examined, PSP levels were the highest in blue mussel, followed in order by bay scallop, oyster, short neck clam, and warty sea squirt. Comparing the maximum PSP levels in the bivalve species examined in 2005 and 2006, PSP in blue mussel was 1.6-2.0, 4.0-5.9, and 5.1-6.0 times higher than in bay scallop, oyster, and short neck clam, respectively. Therefore, blue mussel could be useful as a bioindicator for PSP monitoring. With the increasing PSP levels in blue mussel in 2006, the proportion of PSP in its digestive gland increased to 95.1% when the maximum level was detected from the whole tissues of blue mussel on May 29. Subsequently, the PSP proportion in the digestive gland decreased as the PSP level in whole tissue decreased. The detoxification of PSP in blue mussel was greatest with relaying, followed by the water flow, and water circulation systems. Relaying decreased the PSP level below the regulatory limit of $80{\mu}g$/100 g after 2 days in low toxic sample with $124{\mu}g$/100 g, and after 7 days in high toxic sample with $401{\mu}g$/100 g. During depuration in the blue mussel with $401{\mu}g$/100 g via the water flow system, the PSP amounts in the digestive gland decreased by about 50% after 1 day, and about 77% after 7 days. In contrast, the PSP amounts in the soft body, gill, and mantle did not change significantly with depuration.
For the treatments and protection of bacterial fish disease, many requirements are needed for aquatic probiotics so that they are effective in aquaculture animals but are also harmless to humans. In the present study, among 17 candidate probiotic bacteria (CPB) obtained from the edible part of the shellfish, Bacillus sp. CPB-St (CPB-St) were selected and in vitro evaluated for the possibility as a probiotic strain for the control of fish streptococcosis which frequently occurs in the olive flounder farms. CPB-St showed inhibitory effects on the growth of various fish pathogenic bacteria, Streptococcus sp., S. parauberis, S. iniae, Lactococcus garvieae and L. piscium by the double layer test ranging about 18~26 mm of clear zone. Inhibitory activity of CPB-St to Streptococcus sp. was observed 6 hours after and the growth of Streptococcus sp. was decreased to 8~55 folds in the co-culture of CPB-St with Streptococcus sp.. The safety of CPB-St to fish and survival of CPB-St in the intestine were assessed in the olive flounder, Paralichthys olivaceus. Fish mortality was not observed in artificial infection with CPB-St for 2 weeks. CPB-St was entirely excreted from the stomach and intestine 24 hours after oral injection. This results indicate that CPB-St has potential applications as a probiotic for the control of fish streptococcosis in aquaculture.
Hemacolor stain, histopathological observation and FTM incubation assay were applied to detect Perkinsus sp. infection in Manila clams (Ruditapes philippinarumi taken from culture beds at Tean and Gochang from March 2002 to August 2003. The prevalency was 100% in the clams from Gochang and 20-70% from Tean. Of the three methods, histopathological observation was the most effective to detect the infection. And the parasites was most abundant in gills. When PCR assay was applied to detect Perkinsus sp. for four species of Mollusc such as manila clam, surf clam (Mactra veneriformis), oyster (Crassostrea gigas) and Thomas' rapa whelk (Rapana venosa) taken from Tean and Gochang from April to July 2004, the parasites were detected from all the species at the infection rates of 95%, 62%, 46.9% and 10% in that order. The infection rate was much higher in the species burrowing in the bottom sediments than those attaching on the tidal rock. The results suggest that Perkinsus sp. may affect almost all the molluscs inhabiting western coastal areas of Korea.

The levels of heavy metals (total-mercury, cadmium, lead and chromium) in shellfishes, such as oyster (Carassostrea gigas), blue mussel (Mytilus edulis), arkshll (Scapharca broughtonii) and little neck clam (Ruditapes philippinarum), were determined. The samples were collected from 11 stations for oyster and 12 stations for blue mussel in the South Coast of Korea, and little neck clam and arkshell were collected from Koheung and the Kangjin Bay, respectively, during 1999 to 2000. The average concentrations of heavy metal in 4 different species of shellfishes analyzed were ranged from 0.007 to 0.009 ${\mu}g/g$ for total-mercury; 0.232 to 0.559 ${\mu}g/g$ for cadmium; 0.107 to 0.220 ${\mu}g/g$ for lead; and 0.116 to 0.364 ${\mu}g/g$ for chromium on wet basis. The highest level of total-mercury, lead, cadmium, and chromium in the samples analyzed were all below the quarantine limit of the Korean regulation and guideline established by the U.S.Food and Drug Administration for human consumption. The levels of heavy metals in the samples were negligible, which could be endogenous. It was elucidated that oyster was able to accumulate 2-times more cadmium than blu mussel in this study.
