Treatment with phosphodiester CpG-ODN ameliorates atopic dermatitis by enhancing TGF-β signaling

  • Synthetic oligodeoxynucleotides (ODNs) containing unmethylated CpG phosphorothioate (PS CpG-ODN) are known to decrease IgE synthesis in Th2 allergy responses. Nonetheless, the therapeutic role of PS CpG-ODN is limited due to cytotoxicity. Therefore, we developed a phosphodiester (PO) form of CpG-ODN (46O) with reduced toxicity but effective against allergies. In this study, we first compared the toxicity of 46O with CpG-ODNs containing a PS backbone (1826S). We also investigated the therapeutic efficacy and mechanism of 46O injected intravenously in a mouse model of ovalbumin (OVA)-induced atopic dermatitis (AD). To elucidate the mechanism of 46O underlying the inhibition of IgE production, IgE- and TGF-β-associated molecules were evaluated in CD40/IL-4- or LPS/IL-4-stimulated B cells. Our data showed that the treatment with 46O was associated with a lower hematological toxicity compared with 1826S. In addition, injection with 46O reduced erythema, epidermal thickness, and suppressed IgE and IL-4 synthesis in mice with OVA-induced AD. Additionally, 46O induced TGF-β production in LPS/IL-4-stimulated B cells via inhibition of Smad7, which suppressed IgE synthesis via interaction between Id2 and E2A. These findings suggest that enhanced TGF-β signaling is an effective treatment for IgE-mediated allergic conditions, and 46O may be safe and effective for treating allergic diseases such as AD and asthma.

The Effect of Level of Crude Protein and Available Lysine on Finishing Pig Performance, Nitrogen Balance and Nutrient Digestibility

  • Two trials were conducted to investigate the effect of decreasing the crude protein (CP) content of diets for finishing pigs containing two levels of available lysine on nutrient digestibility, nitrogen (N) balance and production performance. Ten finishing diets containing five levels of CP (on average 144, 155, 168, 182 and 193 g/kg fresh basis) and two levels of available lysine (6.9 and 8.2 g/kg fresh basis) were formulated. The diets were offered to pigs on a performance trial (n = 800 Large White (LW)${\times}$Landrace (LR) pigs) from 10 wk of age until finish at 21 wks+5 d of age. Average daily gain (ADG), average daily feed intake (ADFI) and feed conversion ratio (FCR) were calculated. In addition, a digestibility/N balance trial was conducted using pigs (n = 80 $LW{\times}LR$) housed in metabolism crates. Digestibility of dry matter (DM), CP, oil, fibre and energy was determined. N balance values were determined through analysis of N content of urine and faeces ('as determined'). N balance values were also calculated using ADG values and assuming that 16% of growth is protein deposition ("as calculated"). Pig performance was poor between 10 and 13 wk of age which indicated that the dietary treatments were nutritionally inadequate for pigs less than 40 kg. There was a significant (p<0.01) quadratic effect of increasing CP level on feed intake, ADG and FCR from 10 to 13 wk which indicated that the lower CP levels did not supply adequate levels of essential or non-essential amino acids. There was no effect of increasing available lysine level throughout the early period, which in conjunction with the response in older pigs, suggested that both 8.2 and 6.9 g/kg available lysine were insufficient to drive optimum growth. There was a positive response (p<0.05) to increasing available lysine level from 13 wk to finish which indicated that 6.9 g/kg available lysine was not adequate for finishing pigs. Energy digestibility decreased with decreasing CP level of diets containing 6.9 g/kg available lysine which may be attributed to the higher fibre content of the lower CP diets. Nitrogen excretion (g/d) was lowered when dietary CP was reduced regardless of whether the values were determined through balance or calculated using ADG. Calculated N excretion decreased linearly (p<0.001) and quadratically (p<0.001) with decreasing dietary CP content. When the N balance figures calculated in this study were compared with those quoted in the Northern Ireland and English Nitrates Directive Action Programmes, N excretion was less per pig (wean to finish) offered a 169 g/kg CP, 8.2 g/kg available lysine diet (2.39 kg vs 3.41 kg (Northern Ireland) and 2.93 kg (England)).

  • 8-Fluorociprofloxacin(8-FCP) is an investigational quinolone derivative that is substituted with fluorine at the C-8 position of ciprofloxacin(CP). It was found that the in vitro activity of 8-FCP against Gram(+) bacteria was more potent that of CP, but the opposite against Gram(-) bacteria was true. However, 8-FCP showed better in vivo efficacy than CP against representative Gram(-) organisms, E. coli and K pneumoniae. In an attempt to seek for factors causing this discrepancy in the antibacterial activities, a comparative pharmacokinetic study of 8-FCP and CP was conducted in mice and rats treated either intravenously or orally at a single dose of 30 mg/kg. The pharmacokinetic parameters in mice were as follows; the mean peak serum concentrations(C$_{max}$) following i.v. and oral doses were 12.4 and 5.3 $\mu\textrm{g}$/ml for 8-FCP, and 9.5 and 2.5 $\mu\textrm{g}$/ml for CP, respectively. The terminal half-life(t$_{1/2\beta}$) was 72.9 min for 8-FCP, and 98.2 min for CP, and the oral bioavailability(F) was 89.9% for 8-FCP, and 50.5% for CP. In rats, the mean ($\pm$SD) $C_{max}$ after i.v. administration were 11.6$\pm$1.6 $\mu\textrm{g}$/ml for 8-FCP, and 10.2$\pm$1.3 $\mu\textrm{g}$/ml for CP, whereas oral administration produced $C_{max}$ of 5.9$\pm$1.8 $\mu\textrm{g}$/ml for 8-FCP and 1.1$\pm$0.9 $\mu\textrm{g}$/ml for CP, respectively. The t$_{1/2\beta}$ was 67.9$\pm$8.4 min for 8-FCP, and 76.4$\pm$7.2 min for CP. The F was 88.6$\pm$6.3% for 8-FCP, and 40.7$\pm$6.5% for CP. Marked differences were observed between the two quinolones in the $C_{max}$ and the area under the concentration-time curve obtained after oral administration in mice and rats. The extent of 8-FCP absorption in both mice and rats was approximately 2-fold higher than that of CP, suggesting that the fluorine atom attached to C-8 plays an important role in facilitating oral absorption from the gastrointestinal tract.

Three-step in vitro digestion model for evaluating and predicting fecal odor emission from growing pigs with different dietary protein intakes

  • Objective: The objective of this study was to select an effective in vitro digestion-fermentation model to estimate the effect of decreasing dietary crude protein (CP) on odor emission during pig production and to suggest potential prediction markers through in vitro and in vivo experiments. Methods: In the in vitro experiment, three diet formulations with different CP contents (170 g/kg, 150 g/kg, and 130 g/kg) but containing the same standardized ileal digestible essential amino acids (SID-EAA) were assessed. Each diet was evaluated by two different in vitro gastric-intestinal phase digestion methods (flask and dialysis), combined with fresh pig feces-ferment inoculation. Eighteen growing barrows (31.9±1.6 kg) were divided into three groups: control diet (180 g CP/kg, without SID-EAA adjustment), 170 g CP/kg diet, and 150 g CP/kg diet for 4 weeks. Results: The in vitro digestion results indicated that in vitro digestibility was affected by the gastric-intestinal phase digestion method and dietary CP level. According to the gas kinetic and digestibility results, the dialysis method showed greater distinguishability for dietary CP level adjustment. Nitrogen-related odor compounds (NH3-N, indole, p-cresol, and skatole) were highly correlated with urease and protease activity. The feeding study indicated that both EAA-adjusted diets resulted in a lower odor emission especially in p-cresol and skatole. Both protease and urease activity in feces were also closely related to odor emissions from nitrogen metabolism compounds. Conclusion: Dialysis digestion in the gastric-intestinal phase followed by fresh fecal inoculation fermentation is suitable for in vitro diet evaluation. The enzyme activity in the fermentation and the fecal samples might provide a simple and effective estimation tool for nitrogen-related odor emission prediction in both in vitro and in vivo experiments.

  • 사람의 Disabled-2 (Dab2)는 정상세포에서 c-Fos의 발현을 억제하여 세포 성장을 조절하는 암억제 유전자로 추정되어 지고 있다. 많은 암세포에서 Dab2는 유방과 난소의 정상세포에서는 발현이 되지만, 약 $85\%$의 유방과 난소의 종양세포에서는 발현이 줄어들거나, 발현이 억제되는 것으로 알려져 있다. 본 연구에서는 bisulfite 반응에 의한 염기서열 분석법과 MSP방법 등을 이용하여 유방암 세포주인 MDA MB-231세포에서 Dab2 유전자의 promoter상에 존재하는 Cpg island의 methylation된 상태를 분석하였다. 그 결과, 사람의 정상 자궁내막세포에서는 Dab2 promoter 부위가 완전하게 methylation되어 있지 않았다. 그러나 MDA MB-231세포에서는 TATA box 근처 의 CpG dinucleotide에서 비정상적으로 methylation되어 있었다. 이런 비정상적인 CpG dinucleotide의 methylation은 MDA MB-231세포를 5-azacytidine으로 처리하였을 때 methylation이 풀리고, Dab2의 발현이 회복되는 것으로 나타났다. 따라서 인간 유방암 세포주인 MBA MB-231세포에서 Dab2의 발현억제는 Dab2 유전자의 promoter부위의 CpG island의 비정상적인 methylation과 관련이 있는 것으로 여겨진다.

Effect of Choice Feeding on the Nutrient Intake and Performance of Broiler Ducks

  • An experiment was conducted at the experimental duck farm of Cantho University in the Mekong Delta of Vietnam to evaluate the effects of offering pairwise feeds with different crude protein (CP) levels to Cherry Valley broiler ducks from 28-63 days of age on feed intake and performance. Five diets consisting of mixtures of soya bean meal, fish meal, maize meal and fine rice bran, were formulated to give isocaloric diets with 30, 24, 18, 16 and 12% CP. These were offered separately as pellets to growing ducklings in the following combinations: 30+18, 30+12, 24+16 and 18+12. A diet with 20% CP (CP20) was used as control. All the diets were offered ad libitum. Daily feed intakes (g dry matter, DM) were 161, 168, 150, 101 and 143 g for the 30+18, 30+12, 24+16, 18+12 and CP20 treatments, respectively. CP intakes were 29.6, 27.0, 24.8, 17.1 and 22.9% of total DM intake, and the ratio of methionine+cystine to lysine was 0.76, 0.75, 0.75, 0.76 and 0.75 for the 30+18, 30+12, 24+16, 18+12 and CP20 treatments, respectively. Live weights at 63 days of age were 2,937, 3,054, 2,898, 2,200, and 2,811 g (p<0.001), and daily live weight gains 57.8, 61.7, 57.0, 38.5 and 55.7 g (p<0.001) for the 30+18, 30+12, 24+16, 18+12 and CP20 treatments, respectively. The weights and yields of the carcass, breast and thigh muscles, and heart decreased in the order 30+18, 30+12, 24+16, CP20 and 18+12 (p<0.001). The present study suggests that growing broiler offered a combination of high and low protein feeds pairwise will eat excessive amounts of the high protein feed, thus resulting in protein intakes above requirements.

비육기 흑염소의 유지를 위한 조단백질 요구량 결정에 관한 연구 (Study on the Determination of Crude Protein Requirement for Maintenance of fattening Black Goat(Capra hircus coreanae))

  • 본 연구는 단백질 균형시험에 의한 흑염소의 1일 유지 단백질 요구량을 구하기 위해 수행되었다. 평균체중 $31.78{\pm}4.54$의 8개월 령의 비육기 수컷 흑염소 6두를 공시하였고, $3{\times}3$ Latin square design 방법으로 처리구별 사료 내 조단백질 수준을 달리하여 흑염소에 미치는 영향을 조사하였다. 시험 기간은 2014년 10월 12일부터 2014년 11월 21일까지 40일간 이천의 흑염소 농장에서 진행하였다. 사료의 조단백질 수준은 처리구별로 13%(T1), 16%(T2), 19%(T3) 이며 TDN은 66%로 하여 배합한 TMR 사료를 사용하였으며 1일 사료급여량은 체중의 1.5%를 오후 5시경에 급여하였다. 처리구별 건물섭취량은 T1구가 925.14g/d, T2구가 966.67g/d, T3구가 936.08g/d으로 T2구에서 가장 높았고, 일당증체량은 T2구가 167.13g/d으로 T1과 T3구의 각각 109.26g/d, 118.98g/d 보다 높게 나타났으나 유의적 차이는 보이지 않았다. 건물 소화율은 T1과 T2과 T3구 각각 73.68%, 71.81%, 73.77%로 처리구간에 따른 차이는 나타나지 않았다. 조단백질 소화율은 T3구가 82.42%로 T1과 T2구 각각 72.73%, 65.72% 보다 유의하게 높았다(p<0.05). 조지방 소화율은 처리구별 유의성은 인정되지 않았다. ADF 소화율은 69.13%로 T1구에서 가장 높았다(p<0.05). CF와 NDF 소화율은 46.88~59.76%, 63.29~73.73%의 범위로 처리구간 유의성은 인정되지 않았다. 조단백질 섭취량은 T1과 T2과 T3구에서 각각 128.78g/d, 154.57g/d, 181.23g/d로 사료 내 단백질 수준이 높아질수록 유의적으로 증가하였다(p<0.05). 대사 체중 당 CP 섭취량과 CP 축적량과의 관계식에서 CP 균형이 0이 되는 X축 절편은 $1.63g/BW^{0.75}$이었다. 본 연구결과 8개월 령 수컷 흑염소에게 필요한 유지 조단백질 요구량은 $1.63g/BW^{0.75}$이며 흑염소의 사양표준 설정에 기초자료가 될 것으로 사료된다. 그러나 유지 단백질 요구량에는 성장단계 및 사양방법, 성별에 따라 차이가 발생할 수 있기 때문에 향후 그러한 요인들을 기준으로 한 흑염소 단백질 연구가 단계적으로 필요할 것으로 사료된다.

Adjuvant effect of liposome-encapsulated natural phosphodiester CpG-DNA

  • Immunostimulatory CpG-DNA targeting TLR9 is one of the most extensively evaluated vaccine adjuvants. Previously, we found that a particular form of natural phosphodiester bond CpG-DNA (PO-ODN) encapsulated in a phosphatidyl-${\beta}$-oleoyl-${\gamma}$-palmitoyl ethanolamine (DOPE) : cholesterol hemisuccinate (CHEMS) (1 : 1 ratio) complex (Lipoplex(O)) is a potent adjuvant. Complexes containing peptide and Lipoplex(O) are extremely useful for B cell epitope screening and antibody production without carriers. Here, we showed that IL-12 production was increased in bone marrow derived dendritic cells in a CpG sequence-dependent manner when PO-ODN was encapsulated in Lipoplex(O), DOTAP or lipofectamine. However, the effects of Lipoplex(O) surpassed those of PO-ODN encapsulated in DOTAP or lipofectamine and also other various forms of liposome-encapsulated CpG-DNA in terms of potency for protein antigen-specific IgG production and Th1- associated IgG2a production. Therefore, Lipoplex(O) may have a unique potent immunoadjuvant activity which can be useful for various applications involving protein antigens as well as peptides.

냉장저장 중 사과 슬라이스의 갈변에 미치는 갈변저해제의 효과 (Effects of Antibrowning agents on browning of apple slices during cold storage)

  • 냉장 저장 시 사과슬라이스의 갈변정도 및 조직감과 같은 물리적인 변화와 PPO활성, 총페놀 함량등의 화학적 변화를 측정하였으며 관능검사를 행하여 비교하였으며 또한 이들 변화들 간의 상관관계를 알아본 결과는 다음과 같다. 갈변 정도는 fresh apple slice, 물에 침지한 시료군, CP 시료군, ascorbic acid 시료군 모두 증가하는 경향을 나타냈으며 그 크기는 fresh apple slice>물에 침지한 시료군>ascorbic acid 시료군>CP 시료군 순이었으며, PPO활성은 시료군 모두 증가하는 경향을 나타냈으며 그 크기는 fresh apple slice>물에 침지한 시료군>ascorbic acid 시료군>CP 시료군 순이었으며 CP 시료군의 증가가 가장 낮았다. 총페놀 함량은 fresh apple slice군은 60 mg/100 g에서 56.2 mg/100 g로 감소하는 경향을 보였으나 물에 침지한 시료군은 59.6 mg/100 g에서 56.0 mg/100 g으로, CP 시료군, ascorbic acid 처리군은 51.9 mg/100 g에서 52.8mg/100 g로, 54.1 mg/100 g에서 54.4 mg/100 g로 나타났다. 조직감은 시료군 모두 감소하는 경향을 보였으며 그 크기는 fresh apple slice>물에 침지한 시료군>ascorbic acid 시료군>CP 시료군 순으로 감소하였다. 또한 관능 검사에서 CP 시료군이 다른 처리군들 보다 더 효과적인 것으로 나타났다. 상관관계 분석에서 갈변 정도와 PPO활성, 총페놀 함량 등이 높은 상관성을 나타내었다. 이러한 연구결과로 사과 슬라이스의 냉장 저장중 효소적 갈변 변화에 대한 영향에서 카라멜 생성물에 침지한 CP 시료군이 효소적 갈변에 대한 저해 효과가 있는 것으로 나타났으며 기타 다른 요인에서도 효과적인 것으로 나타나 sucrose의 카라멜 생성물을 채소 및 과일의 저장 과정에 갈변 억제물질로 이용할 수 있을 것으로 생각된다.

단백질 균형시험에 의한 한우 거세우의 유지 조단백질 요구량 (Crude Protein Requirements for Maintenance of Hanwoo Steers by Protein Balance Trial)

  • 본 연구는 거세우의 1일 유지 단백질 요구량을 구하기 위해 실시하였다. 평균체중 173.7±26.3kg의 한우 거세우 18두를 공시하여 3수준의 단백질을 급여하면서 질소균형법을 통해 유지 단백질 요구량을 결정하였다. 급여사료의 조단백질(CP) 수준을 조절하기 위하여 분쇄옥수수를 corn gluten meal(CGM)로 0%(LCP), 7.2% (MCP), 14.2%(HCP) 대체하였고, 각 배합사료의 CP 함량은 9.8, 14.0, 18.9% DM 이었다. 1일 사료급여량은 체중의 2.0%로 하면서 배합사료와 볏짚의 비율은 80:20로 동일하게 하였다.CP 섭취량은 LCP, MCP, HCP 구에서 각각 5.1, 7.8, 10.3g/BW0.75 이었고, 단백질 섭취량이 높아지면서 DM 소화율도 LCP 구와 비교하여, MCP 구, HCP 구에서 각각 14%, 20%의 유의성 있는 증가를 보였으며, CP 소화율은 LCP 구와 비교하여 MCP 구 및 HCP 구에서 각각 약 50%, 80% 증가하였다. LCP, MCP, HCP 구에서의 CP 균형은 각각 -0.05, 2.36, 4.07g/ BW0.75 이었고, 대사체중당 CP 섭취량과 CP 축적량과의 관계식에서 CP 균형이 0이 되는 X축 절편, 즉, 유지 단백질 요구량 5.56g/BW0.75 이었다. 본 결과는 일본의 육용종 거세우보다 평균 20% 이상은 높기 때문에 본 연구결과를 적용한 한우 사양표준(농림부, 2002)의 유지 단백질 요구량이 좀 더 낮아져야 할지에 대해서는 증체 단백질 요구량을 포함한 총 단백질 요구량과 그동안의 사양시험 결과들을 수집 분석하는 종합적인 고찰이 필요하다.
