Ginseng saponin was isolated from panax ginseng root using high-speed countercurrent chromatography (HSCCC). Preliminary studies were performed to optimize physical properties of two-phase solvent system and operating parameters including rotation speed of column, elution mode of mobile phase, and flow rate. Two-phase solvent system for isolation of ginseng saponins was composed of chloroform, water, and methanol as blending solvent. Chloroform-aqueous methanol (4:6) systems with various concentration of methanol in water were evaluated for retention of stationary phase in column. Retention of stationary phase decreased with increasing flow rate in tail-to-head elution mode using upper phase as mobile phase and head-to-tail elution mode using lower phase as mobile phase. Latter mode produced high retention at flow rate of 5 mL/min. Optimum conditions for isolation of saponin were chloroform/methanol/water (40/39/21) solvent system; mobile phase, of lower organic layer, flow rate, of 5 mL/min, head to tail elution mode, rotation speed, of 800 rpm, and sample injection, of $200{\mu}L$, Recovery yield of ginseng saponin from panax ginseng root extract by HSCCC was 63.6%, and the purity of HSCCC fractions was verified by TLC.

To predict the average liquid level under the condition of the countercurrent stratified two-phase flow in a pipe, an analytical model has been suggested. This is made by introducing the interfacial level gradient into the liquid-phase and the gas-phase momentum equations. The analytical method for the gas-phase pressure drop calculation with f$_i$$\neq$ f$_G$ has also been described using the liquid level prediction model developed in the present study.

An experimental study on the countercurrent two-phase flow in narrow rectangular channels has been performed. The void fraction and the pressure gradient were investigated using air and water in 760 mm long, 100 mm wide. vertical test sections with 2, 3 and 5 mm channel gaps. Tests were systematically performed with downward liquid superficial velocities and upward gas velocities covering 0 to 0.08 and 0 to 2.5 m/s ranges. respectively. the experimental results were compared with the previous correlations, which were mainly for round tubes, and the qualitative trends were found to be in good agreements. However the quantitative discrepancies were hardly neglected. as the superficial gas velocities increased, the void fraction increased and the pressure gradient decreased, where the effects of the liquid superficial velocities were infinitesimal. as the gap width of the rectangular channel increased the void fraction and the 2-phase frictional pressure gradient approached those values for the round tubes. Equi-periphery diameter, rather than the hydraulic diameter, seemed to be more effective in the analysis of two-phase flow behavior.

An experimental study on the countercurrent two-phase flow in narrow rectangular channels has been peformed. Countercurrent flow limitation (CCFL) was investigated using air and water in 760mm long, 100mm wide, vertical test sections with 1 and 3mm channel gaps. Tests were systematically performed with downward liquid superficial velocities and upward gas velocities covering 0 to 0.125 and 0 to 3.5m/s ranges, respectively. As the gap width of rectangular channel increased the CCFL water superficial velocity decreased for the given air superficial velocity. Slight increase of the air superficial velocity resulted in the abrupt decrease of water velocity when $j_g=2{\sim}4m/s$. The critical superficial velocity of air, at which the downward flow of water was no longer allowed, also decreased with the increase of gap width. The experimental results were compared with the previous correlations, which were mainly for round tubes, and the qualitative trends were found to be partially acceptable. However the quantitative discrepancies were hardly neglected. New correlation of CCFL was developed and showed good agreement with the experimental data.
An analytical model that includes the steam condensation effect has been derived and a parametric study has been performed. In addition, a series of experiments were performed and a total of 34 experimental data for the onset of CCFL in nearly horizontal countercurrent two-phase How have been obtained for various flow rates of water. Comparisons of the present CCFL data with slug formation models show that the agreement between the present as well as the existing model and the data is about the same. However, the deviation between the Taitel and Dukler's model predictions and the data is the largest when if j$_{f}$<0.04 m/s. A parametric study of the effect of the steam condensation using the present model shows that, when all local conditions are similar, the model predicted local gas velocities that cause the onset of flooding are slightly lower when condensation occurred. Based on the visual observation and the evaluation of the present work, it has been concluded that the criterion derived for the onset of slug flow can be directly used to predict the onset of inner flooding in nearly horizontal two-phase flow within the experimental ranges of the present work.

Structures of interfacial waves in nearly-horizontal countercurrent stratified air-water flow have been measured by means of a needle contact method. Based upon a statistical analysis for the liquid film distribution, statistical properties of the waves such as mean film thickness, mean wave amplitude and rms value of the wave fluctuation have been calculated. The results show that the film distribution can be described by a Gaussian probability density function for the three-dimensional wave regime. It is also indicated that the mean film thick ness and the rms value of the wave fluctuation increase as gas and liquid flow rates are increased in countercurrent two-phase flow. The dimensionless intensity of the wave fluctuation may be regarded as a function of the Froude number and the dimensionless mean film thickness.

근사수평 이상반류유동에서의 플러딩천이에 대한 실험을 수행하였으며 이것을 바탕으로 반류유 동도(flow-regime map)를 완성하였다. 또 플러딩천이에 대한 응축의 영향을 고찰하였는데 플러 딩이 액체입구에서 야기될 때 플러딩 속도는 응축량을 고려한 유효증기량으로 표시되며 이 경우 반드시 히스테리시스효과를 동반하게 된다. 이 효과는 응축에 기인하는 것으로 그 메카니즘을 구명하였다. 또 전달액체유량이 영이 될 때의 임계증기속도는 액체분출유량이나 액체서브쿠울 링의 정도에 무관하며 본 연구에서 사용한 관의 경우, 수정 Wallis 변수로 1.74로 나타났다.
Anthocyanins are water soluble plant pigments which are responsible for the blue, red, pink, violet colors in several plant organs such as flowers, fruits, leaves and roots. In recent years, anthocyanin-rich foods have been favored as dietary supplements and health care products due to diverse biological activities of anthocyanins including antioxidant, anti-allergic, anti-diabetic, anti-microbial, anti-cancer and preventing cardiovascular disease. High-performance countercurrent chromatography (HPCCC) coupled with reversed-phase medium pressure liquid chromatography (RP MPLC) method was applied for the rapid and efficient isolation of cyanidin 3-glucoside (C3G) and peonidin 3-glucoside (P3G) from black rice (Oryza sativa L., Poaceae). The crude black rice extract (500 mg) was subjected to HPCCC using two-phase solvent system composed of tert-butyl methyl ether/n-butanol/ acetonitrile/0.01% trifluoroacetic acid (TBME/B/A/0.01% TFA, 1 : 3 : 1 : 5, v/v, flow rate - 4.5 mL/min, reversed phase mode) to give enriched anthocyanin extract (37.4 mg), and enriched anthocyanin extract was sequentially chromatographed on RP-MPLC to yield C3G (16.5 mg) and P3G (8.7 mg). The recovery rate and purity of isolated C3G were 76.0% and 98.2%, respectively, and those of P3G were 58.3% and 96.3%, respectively. The present study indicates that HPCCC coupled with RP-MPLC method is more rapid and efficient than multi-step conventional column chromatography for the separation of anthocyanins.

Two-phase transient phenomena in the noncondensable gas-filled closed loop was investigated numerically using the RELAP5/MOD3 version 3.1 computer code. The condensation heat transfer correlation for noncondensable gases was studied in detail. Two modes of the reflux condensation which can be characterized by countercurrent flow of steam and its condensed water and the oscillatory between reflux condensation and natural circulation were predicted well. However, the natural circulation mode which the condensed water carried over the U-bend concurrently with steam was failed to predict.

A unified correlation of the interfacial friction factor for air-water and steam-water flows in inclined rectangular channels has been developed. The correlation was expressed in the form of a power law of the liquid and the gas Reynolds number, and the liquid-to-gas viscosity ratio. In addition, a relation between the equivalent roughness and the intensity of wave height fluctuation of the interface has been investigated. A new dimensionless intensity of fluctuation including a liquid film Reynolds number is proposed. It has been shown that the dimensionless equivalent roughness, which is calculated from the Nikuradse equation, can be uniquely related to this dimensionless intensity of fluctuation for both air-water and steam-water flows.
