Phytochemicals have been considered as alternatives for synthetic fungicides because of their biodegradability and low toxicity. In this study, we found that the methanolic extract of Corydalis ternata suppressed the development of plant diseases caused by Puccinia triticina and Colletotrichum coccodes. As the antifungal substance, three isoquinoline alkaloids (dehydrocorydaline, stylopine, and corydaline) were isolated from C. ternata. These active compounds also exhibited in vivo antifungal activity against P. triticina and C. coccodes. Taken together, our results suggest that C. ternata and its active compounds can be used to control plant diseases.
Corydalis Tuber has been used in traditional medicine for an analgesic, antispasmodic and gastric ulcers. For the quality control on this drug, isolation and quantitative determination of berberine and coptisine from Corydalis ternata Nakai (Papaveraceae) has been conducted by using HPLC method. Berberine and coptisine in quarternary alkaloidal fraction from the crude drug were separated on silicagel column using a $CHCl_3:MeOH\;(85:15)$ and $CHCl_3:MeOH:H_2O\;(70:30:4)$ as an eluent, and the average contents were about 0.93 and 0.36%.
A methanolic extract of Corydalis ternata having aldose reductase inhibitory activity was examined as a possible aldose reductase (ALR2) inhibitor, a key enzyme involved in diabetic complications. Seven alkaloids, tetrahydrocoptisine (1), corydaline (2), tetrahydropalmatine (3), isocorybulbine (4), corybulbine (5), dehydrocorydaline (6), and N-methyltetrahydroberbinium (7) were isolated from $CHCl_3$ fraction of C. ternata methanol extract. Among them, compounds 1, 5, and 7 exhibited $5.04{\pm}1.97%$, $5.00{\pm}1.26%$, and $1.80{\pm}2.33%$ inhibitions, respectively at $40{\mu}M$. The activities of the single compounds were not comparable to that of the whole extract, suggesting that the whole combination of each single compound was responsible for the activity of the extract as shown in many cases of natural medicines. Even though this is the second report on aldose reductase inhibition activity of C. ternata, recombinant human aldose reductase was employed in this study unlike in the previous report. Furthermore, the aldose reductase inhibitory activities of isocorybulbine, corybulbine, and N-methyltetrahydroberbinium, to the best of our knowledge, were evaluated for the first time in this study. These results suggest a use of the extract of C. ternata for ameliorating diabetic complications.
Dehydrocorydaline was isolated from the roots of Corydalis ternata (Papaveraceae) and identified by the comparison the $^1H-$ and $^{13}C$-NMR spectral data with those of authentic sample. The content of dehydrocorydaline was determined by the HPLC analysis based on extraction of ground plant material. Quantitative analysis of dehydrocorydaline in MeOH extract of C. ternata by HPLC showed $1.31{\pm}0.95%$ in 20 samples collected throughout regions of Korea.

Chemical carcinogenesis is associated with the increase of intracellular polyamine levels, and 12-O-tetradecanoylphorbol-13-acetate (TPA)-induced mouse epidermal ODC activity are essential to skin tumor promotion by TPA. Therefore, for the discovery of new cancer chemopreventive agents, we have evaluated about 73 kinds of natural products to study inhibitory effects against ODC activity induced by TPA in T24 cell culture system. The total methanol extracts of plants fractionated into three layers (hexane, ethyl acetate and water layer) were tested and the hexane fraction of Angelica gigas $(root\;bark,\;IC_{50}:\;7.4\;{\mu}g/ml)$ and the ethyl acetate fraction of Corydalis ternata $(root,\;IC_{50}:\;7.5\;{\mu}g/ml)$ were the most effective on the inhibition of TPA-induced ODC activity, These active fractions are under investigation with further sequential fractionation using column chromatography.
본 연구에서는 현호색의 methanol추출물에 대하여 사염화탄소에 의한 간 손상 보호효과를 확인한 후 이를 hexane, chloroform, butanol 및 물로 계통분획하여 사염화탄소에 의한 간 손상 보호효과를 검색하였다. 이 결과 현호색 물분획물이 유의성 있는 간 보호효과를 보임으로써 간손상 보호효과를 확인하고 그 기전을 밝히고자 하였다. 그 결과 혈액중의 ALT활성 및 cholesterol함량은 사염화탄소 단독투여군에 비하여 감소하였으며 간조직의 TG와 지질과산화물 함량은 사염화탄소 단독투여군에 비하여 감소하여 사염화탄소에 의한 지방의 축적이나 세포막의 손상은 억제된 것으로 보인다. 기전연구를 위하여 측정한 CYP함량과 calcium함량은 사염화탄소 단독투여군에 비하여 감소하였으며 GST활성도는 사염화탄소 단독투여군에 비하여 증가하였다. 항산화 효소의 유도로 인하여 증가할 것으로 기대한 SOD, GPX, GST활성도는 모든 처치군에서 감소하는 경향을 보여 간손상 보호 효과는 free radical scavenging effect보다는 CYP함량의 감소와 CCB로서의 작용 및 GST활성도의 증가에 기인한 것으로 생각된다.
Depression is a very difficult disease to be cured because several nervous systems are involved. In the present study, we evaluated the effects of Keum-Ryung-Ja-San (KRJS), a traditional herbal prescription, on depressive behaviors in mice using the forced swimming test. KRJS was given 1 h prior to the forced swimming test (50, 100, 200, an400 mg/kg, p.o.). The duration of immobility time in the forced swimming test was significantly reduced by KRJS treatment (200 mg/kg, P<0.05) and similar effects were observed with a classical antidepressant, imipramine (15 mg/kg, i.p.). With subchronic administrations of KRJS and its constituents at several doses for 1 week, a decreased duration of immobility time was observed with KRJS and Corydalis ternata (200 mg/kg, p.o. P<0.05). These results suggest that KRJS may have antidedpressive activities and CT may contribute to the antidepressive activity of KRJS.

A simple and rapid analytical method was developed for the determination of eight isoquinoline alkaloids in Corydalis species. Eight isoquinoline alkaloids, including 2 aporphine alkaloids (isocorydine and glaucine) and 6 protoberberine alkaloids (coptisine, palmatine, berberine, canadine, corydaline, and tetrahydrocoptisine) were used as chemical markers which have a various biological activity and determined for quality control of Corydalis (C.) species (C. ternata, C. yanhusuo, and C. decumbens). To evaluate the quality of these herbal medicines, LC chromatographic separation of alkaloids were preferentially investigated on reversed-phase C18 column with pH variation and composition of mobile phase. In addition, the separation of these alkaloids in herbal extracts was found to be significantly affected on mobile phase composition using gradient elution. Especially for C. yanhusuo extract, berberine was seriously interfered with other alkaloid extracted from sample matrix when mobile phase composition was not optimized. As results, these compounds were successfully separated within 28 min using 10 mM ammonium acetate containing 0.2% triethylamine (adjusted at pH 5.0) as a mobile phase with gradient elution. On the basis of optimized HPLC conditions, 23 different Corydalis species samples were analyzed for the determination of alkaloid levels. In addition, principal component analysis (PCA) combined with the chromatographic data could be successfully classified the different geographic origin samples.
The MeOH ext., $CH_2Cl_2$ Frac., EtOAc Frac., n-BuOH Frac., and $H_2O$ Frac. of 23 Korean medicinal herbs were prepared and were tested the inhibitory effects on Cytochrome P450 (Cyp) 1A1/2, 2B1/2, 2E1. Among the tested samples, the extracts of Selaginella tamariscina, Euonymus alatus, Salvia miltiorhiza, Angelica acutiloba, Rheum palmatum, Paeonia moutan, Scutellaria barbata, Tribulus terrestris, Hedyotis diffusa, Curcuma zedoaria, Rehmania glutinosa, Trogopterus xanthipes, Melandryum firmum, Achyranthes bidentata, Leonurus sibricus, Panax ginseng, Paeonia lactiflora, Poncirus trifoliata, Cnidium officinale, Cyperus rotundus, Corydalis ternata showed significant inhibitory effects on Cyp 1A1/2, 2B1/2, 2E1. The $IC_{50}$ values of those extracts were found to be below $50\;{\mu}g/ml$.
혈액 항응고 활성물질을 검색하기 위하여 73종의 한약재, 41종의 산채류 및 일반 채소류, 5종의 해조류들의 열수추출물들에 대해 혈액응고 지연시간(Tr)을 시험하였다 1차검색에서 비교적 활성이 높았던 택사, 삼칠근, 현호색, 운지, 마늘, 청각의 산성 수추출물에서 가장 높은 항응고 활성(대조군에 비해 4.3배)을 보였다. 청각(1.2KG)을 0.8% 염산 수용액(24l)으로 추출한 후 메탄을 환류, 에탄올 환류, 침전 및 투석하여 활성이 약 2배 증가된 조다당 CF-1을 얻었다. CF-1은 80.8%의 다당과, 8.7%의 단백질 및 13.3%의 황산기로 구성되어 있었으며, 다당류는 arabinose, galactose, 및 glucose가 주구성당으로 확인되었다. 이 조다당 CF-1의 항응고 활성 본체를 확인하기 위해 pronase처리와 periodate 산화를 행한 결과 pronase 처리시에는 활성의 변화가 없었으나 periodate 산화시에는 약 70%로 활성이 감소한다는 사실로부터 CF-1의 항응고 활성은 다당에 의해 기인됨을 알 수 있었다.
