Objectives The present study aimed to investigate the change in marker compounds, anti-oxidative and anti-inflammatory effects of salt-water processed Cortex Eucommiae. Methods To evaluate the influence of processing on anti-oxidant effect of Cortex Eucommiae, changes in total phenol, total flavonoid, 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging, 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) free radical scavenging, and ferric reducing antioxidant power (FRAP) between processed and raw Cortex Eucommiae were assessed. In addition, nitrite assay was conducted to determine the influence of processing on anti-inflammatory effect of Cortex Eucommiae. Cell viability was also examined as to elucidate whether processing affects cytotoxicity of Cortex Eucommiae. Finally, high-performance liquid chromatography (HPLC) analysis was conducted to monitor changes in pinoresinol diglucoside amount of processed and raw Cortex Eucommiae. Results Salt-water processed Cortex Eucommiae showed higher total phenol and flavonoid amount, compared to raw Cortex Eucommiae. Furthermore, anti-oxidative activity of processed Cortex Eucommiae was improved as discovered in DPPH, ABTS, and FRAP assays. Anti-inflammatory effect of Cortex Eucommiae was also enhanced following salt-water processing, as evidenced in nitrite assay. HPLC analysis found that the amount of pinoresinol diglucoside, widely known as the marker compound of Cortex Eucommiae, increases through salt-water processing. All experiments were performed with non-toxic concentration of Cortex Eucommiae; processing did not affect the cytotoxicity of Cortex Eucommiae up to the currently adopted concentration. Conclusions The present results support that salt-water processing of Cortex Eucommiae is beneficial in terms of marker compound amount, anti-oxidative, and anti-inflammatory activities. Additional investigations are needed to standardize the processing method of Cortex Eucommiae.
In order to estimate the effects of Eucommiae Cortex, which is one of the most important herb medicines used for invigorating the liver and kidney and strengthening ones and muscles, fetus-soothing etc., this experiment was conducted the quantitative analysis of geniposidic acid and geniposide by HPLC, and the analgesic effects of mice, the lipid metabolism of rats, the catecholamine concentration in the brain and the plsma of rats stressed by immobilization. The results obtained are as follows. 1. The contents of geniposidic acid and geniposide were 22.3mg/g, 5.4mg/g in Eucommiae Cortex without processing, and 119.8mg, 10.4mg/g in Eucommiae Cortex fried with salts. 2. In analgesic effects of mice, there were all significant in Eucommiae Cortex without processing and Eucommiae Cortex fried with salts. 3. In the effects of lipid metabolism of rats, there were not significant on the change of organ weight and the level of serum total lipid and total cholesterol, triglyceride, phospholipid, HDL-cholesterol. But, there were significant on the level of free fatty acid. 4. In tile effects of the catecholamine concentration in the brain and the plsma of rats stressed by immobilization, there were all significant in Eucommiae Cortex without processing and Eucommiae Cortex fried with salts, especially in case of norepinephrine. From the results mentioned above, there were significant on the analgesic effects of mice, and the catecholamine concentration in the brain and the plsma of rats stressed by immobilization. And there were different from Eucommiae Cortex without processing and Eucommiae Cortex fried with salts in the contents of geniposidic acid and geniposide. These results are relative to invigorating the liver and kidney and strengthening ones and muscles, and fetus-soothing, therefore when we use Eucommiae Cortex, it is more desirable to use Eucommiae Cortex with processing.
The studies were conducted to investigate the anti-diabetic activities on the hyperglycemia induced by streptozotocin in rats, Anti-fatigue, Decrease of body weight activities in mouse and anti-gastric ulcer activities in stress-induced rats by Mixed Extracts of Acantopanacis senticosi Radicis Cortex and Eucommiae Cortex in Korea. 1. The blood glucose levels of streptozotocin-induced hyperglycemic rats were dose-dependently decreased by administrations of various doses(100, 200, 400, significantly 200+100mg/kg) of Mixed Extracts from Acantopanacis senticosi Radicis Cortex and Eucommiae Cortex. 2. The serum total cholesterol levels of streptozotocin-induced hyperglycemic rats were dose-dependently decreased by administrations of various doses (100, 200, 400, significantly 200+100mg/kg) of Mixed Extracts from Acantopanacis senticosi Radicis Cortex and Eucommiae Cortex. 3. The serum triglyceride levels of streptozotocin-induced hyperglycemic rats were dose-dependently decreased by administrations of various doses(100, 200, 400, significantly 200+100mg/kg) of Mixed Extracts from Acantopanacis senticosi Radicis Cortex and Eucommiae Cortex. 4. The swimming time levels in mouse were dose-dependently extended by administrations of various doses(100, 200, 400, significantly 200+100mg/kg) of Mixed Extracts from Acantopanacis senticosi Radicis Cortex and Eucommiae Cortex. 5. The body weight levels in mouse were dose-dependently decreased by administrations of various doses(100, 200, 400, significantly 200+100mg/kg) of Mixed Extracts from Acantopanacis senticosi Radicis Cortex and Eucommiae Cortex. 6. Stress-induced gastric ulcer were dose-dependently repaired by administrations of various doses(100, 200, 400, significantly 200+100mg/kg) of Mixed Extracts from Acantopanacis senticosi Radicis Cortex and Eucommiae Cortex.
Objectives : The present study had been undertaken to investigate the effects of Cynomorii Herba and Eucommiae Cortex on treatment of osteoporosis in ovariectomized rats. Methods : In this experiment, the rats of experimental groups were ovariectomized. Rats of medicinal groups were administered by water extracts of Cynomorii Herba or Eucommiae Cortex. The levels of bone mineral density and body weight were measured. Results : The levels of spinal bone mineral density was significantly increased in comparison with OVX group at 8 weeks after medication in Cynomorii Herba group. The levels of femoral and fibula-tibial bone mineral density were not significantly increased in comparison with OVX group after medication in Cynomorii Herba and Eucommiae Cortex group. The levels of body weight were not significantly decreased in comparison with OVX group after medication in Cynomorii Herba and Eucommiae Cortex group. Conclusions : Reviewing these experimetal results, it appears that Cynomorii Herba have efficacy on treatment of osteoporosis. And further study should be conducted to illustrate in depth the curing and prevention of osteoporosis.

Objectives : It is necessary to manage herbal medicines based on effectiveness by comparing the efficacy of herbal medicines by region. In this study, we compared anti-oxidative activity and marker compounds of Eucommiae Cortex by regional groups. Methods : Eucommiae Cortex grown and harvested in Gangjin, Sancheong, Yeongwol, Jangsu, and Jecheon were used. Eucommiae Cortex was extracted in distilled water at $100^{\circ}C$ for 3 hours, and filtered. filtered. Extract samples were freeze-dried at $-80^{\circ}C$. Comparison of anti-oxidant activity in Eucommiae Cortex samples from regional groups was measured in 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging, 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) free radical scavenging, and ferric reducing antioxidant power (FRAP) between regional groups of Eucommiae Cortex. In addition, high-performance liquid chromatography (HPLC) analysis was conducted to compare pinoresinol diglucoside concentration by regional groups. Results : HPLC analysis found that pinoresinol diglucoside concentration, widely known as the marker compound of Eucommiae Cortex, was the highest in Gangwon Yeongwol. There was a significant difference in anti-oxidative activity of Eucommiae Cortex between regional groups as discovered in DPPH, ABTS and FRAP assays. DPPH free radical scavenging was the highest in Jeonbuk Jangsu. ABTS free radical scavenging was the highest in Jeonbuk Jangsu and Chungbuck Jecheon. FRAP was the highest in Jeonbuk Jangsu. Conclusions : Although pinoresinol diglucoside concentration was high, anti-oxidative activity was not proportionately high. Pinoresinol diglucoside concentration was the highest in Gangwon Yeongwol. Anti-oxidative activity was the highest in Jeonbuk Jangsu.
The effects of 3% Cortex Eucommiae extracts on serum lipid contents were evaluated in rats. Forty-two male Sprague-Dawley rats weighing $100{\pm}10g$ were divided into seven groups and fed different fat diets for four weeks. Each group was administered with following group: Control, basal fat diet, saturated fat diet and unsaturated fat diet respectively was fed 1% cholesterol, Cortex Eucommiae extracts 3%. The contents of serum lipid contents triglyceride in the basal fat diet group were higher than those of control group. The contents of total-cholesterol in serum of the saturated fat diet group was significantly higher than other group. The contents of HDL-cholesterol in serum of the 3% Cortex Eucommiae extract group were higher than those other fat diet group. The contents of LDL-cholesterol in serum of the 3% Cortex Eucommiae ex1ract group was lower than other fat diet groups. These results suggest that 3% Cortex Eucommiae extract group may reduced elevated levels of serum lipid contents in rats different fed fat diet.

In this study, we carried out quantification analysis of the six marker components, geniposidic acid, chlorogenic acid, geniposide, pinoresinol diglucoside, liriodendrin, and genipin in the 70% ethanol extracts of non-processed Eucommiae Cortex and processed Eucommiae Cortex using a high-performance liquid chromatography coupled with photodiode array detector. The six components were separated on Gemini C18 column (5 μm, 4.6×250 mm) by the gradient elution with 1.0% (v/v) acetic acid in water and 1.0% (v/v) acetic acid in acetonitrile as mobile phase. The flow rate was 1.0 mL/min and the injection volume was 10 mL. The amount of geniposidic acid, chlorogenic acid, geniposide, pinoresinol diglucoside, liriodendrin, and genipin in non-processed Eucommiae Cortex were 1.31, 0.31, 0.66, 0.46, 0.46, and 0.03%, respectively, while the amount of the six compounds in non-processed Eucommiae Cortex were 0.04-0.78, 0.01-0.14%, 0.05-0.63%, 0.01-0.37%, 0.15-0.42%, and not detected, respectively. After processing treatment, the contents of three iridoids, two lingnan, and one phenylpropanoid decreased in Eucommiae Cortex.
Objectives : Eucommiae cortex is reported that it helps bone and skeletal muscle stronger. In case of bone, many report is presented, but reports related to skeletal muscle are rarely existed. So in order to investigate effects of Eucommiae cortex on the skeletal muscle atrophy following stroke, cerebral infarct was induced by the middle cerebral artery occlusion (MCAO) in the rats. Methods : In order to induce MCAO rats, nylon suture was advanced and then blocked middle cerebral artery(MCA). Water extract of Eucommiae cortex was treated for 15 days, once a day orally, after the MCAO. Effects were evaluated with muscle weights, muscle fiber type composition, cross-sectioned area of muscle fibers in soleus and gastrocnemius of the unaffected and affected hind limbs. And MyoD protein expression in gastrocnemius was demonstrated with immunohistochemistry and western blotting. Results : In the affected hind limb of the MCAO rats, muscle weight loss of gastrocnemius and tibialis anterior muscles were attenuated by Eucommiae cortex treatment. In soleus muscle of the affected hind limb of the MCAO rats, increase of type-I fibers and decrease of type-II fibers were induced by Eucommiae cortex treatment. In soleus muscle of the affected hind limb of the MCAO rats, decrease of cross-sectioned areas of type-I fibers was attenuated by Eucommiae cortex treatment. In gastrocnemius muscle of the affected hind limb of the MCAO rats, increase of type-I fibers and decrease of type-II fibers were induced by Eucommiae cortex treatment. In gastrocnemius muscle of the affected hind limb of the MCAO rats, decreases of cross-sectioned areas of type-I and type-II fibers were attenuated by Eucommiae cortex treatment. In gastrocnemius muscle of the affected and unaffected hind limb of the MCAO rats, MyoD expressions were increased by Eucommiae cortex treatment. Conclusions : These results suggest that Eucommiae cortex has a protective effect against muscle atrophy, through the inhibition of the muscle cell apoptosis, following the central nervous system demage.

Objectives : The purpose of this study is to examine whether Eucommiae Cortex pharmacopuncture may affect to the neuropathic pain in a rat model. Methods : To produce the model of neuropathic pain, under isoflurane 2.5 % anesthesia, underwent tight ligation by 6.0 silk thread and transection of the tibial and sural nerves, leaving the common peroneal nerve intact. After neuropathic surgery, the author examined if the exhibited the behavioral sign of allodynia. The allodynia was assessed by stimulating the plantar with Dynamic Plantar Aesthesiometer. Three days after the neuropathic surgery, Eucommiae Cortex pharmacopuncture was injected at Sinsu($BL_{23}$) once every week for 6 weeks. After that, the author examined the withdrawal response of neuropathic rats' leg by Dynamic Plantar Aesthesiometer. And also the author examined Bax, Bcl-2, Bax/Bcl-2 ratio in the spinal cord of neuropathic rats and the change of WBC, RBC, HGB, HCT count in the blood of neuropathic rats. Results : 1. The Eucommiae Cortex pharmacopuncture decreased the withdrawal response of mechanical allodynia that assessed with Dynamic Plantar Aesthesiometer in EC2-$BL_{23}$ group as compared with control group. 2. The Eucommiae Cortex pharmacopuncture decreased Bax/Bcl-2 ratio in EC1-$BL_{23}$, EC2-$BL_{23}$ group. But The Eucommiae Cortex pharmacopuncture injected at Sinsu($BL_{23}$) didn't change Bax, Bcl-2 expression level in the all group. 3. The Eucommiae Cortex pharmacopuncture decreased WBC count in EC1-$BL_{23}$, EC2-$BL_{23}$ group. Conclusions : We have noticed that Eucommiae Cortex pharmacopuncture decreased mechanical allodynia in the model of neuropathic pain compared with the control group. Bax/Bcl-2 ratio in spinal cord of that group was also decreased compared with the control group. This study can be used as a basic resource on a study and a treatment of neuropathic pain.
The dyeability of the Eucommiae Cortex were examined to establish the optimum condition for dyeing of Eucommiae Cortex. The results of this study are as follows, Wool, nylon and silk had the highest K/S value at pH 3. The dyeability was the highest in wool, nylon, and silk. In addition, the increase in K/S value corresponded to temperature in wool, nylon and silk, and the greatest K/S value at 95$^{\circ}C$. The increase in K/S value corresponded to dyeing times in wool, nylon and silk, and the greatest K/S value at 120min. Also, the K/S value increased in concomitance with the increased number of dyeing repetition. In the most cases, colorfastness of light was low but colorfastness of laundry, sweat was relatively good.
