Effects of $\beta$-glucans from Coriolus versicolor on Macrophage Functions in Young and Aged mice

  • The immune system may play an important role in aging and the changes in the immune status are associated with treatment of various immunomodulators. This study examined the effects of $\beta$-glucans isolated from mushroom fungi, Coriolus versicolor on macrophages functions in young (8-weeks-old) and aged (82-weeks-old) male C57BL/6 mice. When peritoneal macrophages were treated with various concentrations of $\beta$-glucan ($1-100\;{\mu}g/ml$) for 24 hrs, tumoricidal activity, NO production and phagocytic activity were significantly increased in the young mice, whereas there are no effects in the aged mice. These results suggest that $\beta$-glucans has differential effects on the macrophage functions in young and aged mice and age nutrition might need to be considered to select proper immunomodulator. In addition, $\beta$-glucan could be used clinically for the treatment of diseases such as cancer therapy in the young.

The Comparative Immunomodulatory Effects of β-Glucans from Yeast, Bacteria, and Mushroom on the Function of Macrophages

  • The comparative immunomodulatory effects of ${\beta}$-glucans isolated from mushroom fungi (Coriolus versicol), yeast (Saccharomyces cerevisiae) and bacteria (Agrobacterium) on the major functions of macrophages were evaluated. As parameters of macrophage functions, we examined tumoricidal activity, phagocytosis, nitric oxide (NO) production, and the induction of inducible NO synthetase (iNOS) in RAW264.7 cells, following treatments with ${\beta}$-glucans from the three different sources. The results indicated that all ${\beta}$-glucan treatments significantly induced tumoricidal activity in the RAW264.7 cells, with a remarkable effect shown by the beta-glucan from Agrobacterium at a concentration of $10{\mu}g/mL$. There was also a significant increase in iNOS-NO system activity in macrophages treated with ${\beta}$-glucans extracted from yeast; however, iNOS-NO system activity was not markedly changed by the treatment of ${\beta}$-glucans from C. versicolor mushroom fungi or Agrobacterium. Furthermore, the ${\beta}$-glucans from C. versicolor had a significant phagocytotic effect at concentrations of 1, 10, and $100{\mu}g/mL$. Taken together, the present data suggest that these ${\beta}$-glucans, isolated from three different sources, have different effects on macrophage function, and therefore, may have different clinical uses in different for various types of diseases.

Anti-metastatic and Immunomodulating Activity of Water-Soluble Components from Five Mushroom Extracts

  • Objectives : This experiment was conducted to evaluate the inhibitory effects against lung metastasis and promotion of splenocytes by water-soluble components from five mushrooms extracts (WEFM): Garnoderma frondosa, Corious versicolor, Codyceps militaris, Hericium erinaceus and Lentinula edodes. Methods : Colon 26-L5 carcinoma cells were injected through the tail vein to induce lung metastatic cancer. Changes in weight of lung were observed and cytokine level was analyzed to evaluate immunological changes. Results : Oral administration of WEFM resulted in a significant inhibition of lung metastasis after intravenous injection of colon 26-L5 cells in a dose-dependent manner. There was also a significant increase in T cell and B cell mitogenic stimuli and production of IFN-g by splenocytes stimulated with Con A compared to untreated controls. Conclusion : WEFM may have anti-tumor activities via Th1-type dominant immune responses.

Decay of Populus cathay Treated with Paraffin Wax Emulsion and Copper Azole Compound

  • In order to investigate the decay process of wood treated with preservative, waterproofing agent and their compound systems, a full-cell process was applied to impregnate the sapwood of poplar (Populus cathay) at paraffin wax emulsion concentrations of 0.5% and 2.0%, Copper Azole (CA) concentrations of 0.3% and 0.5%, and their four compound systems, respectively. Leaching tests and laboratory decay resistance against the white-rot fungus Corious versicolor (L.) Murrill for treated wood were carried out according to the America Standard E11-06 and China Standard GB/T 13942.1-2009. At certain time intervals during the decay test, samples were characterized by Fourier transform infrared spectroscopy (FTIR) and X-ray diffraction technique (XRD) to investigate the time-dependent changes of chemical components and crystalline structure, thus clarifying the decay mechanisms. The results suggested that white-rot fungi degrade hemicellulose and lignin in the wood cell wall first, followed by a simultaneous degradation of polysaccharides and lignin. Besides, CA could not only slower the decomposition of both hemicellulose and lignin, but also reduce the degradation amount of hemicellulose. However, paraffin wax emulsion at high concentration had a negative effect on the impregnation of CA for the compound system treated wood.
