Studies on Antitumor Activity and Antimicrobial Activity of Coriolus Versicolor(Fr.)Quel and Ganoderma Lucidum (Fr.)Karst (구름버섯과 영지버섯의 항암효과에 관한 연구)

  • The purpose of the study was to verify if or not as funtional food by estimating antitumor activity and antimicrobial activity of Coriolus versicolor(Fr.)Quel and Ganoderma Lucidum (Fr.)Karst. 1. The contents of moisture. crude fiber and crude protein of GL were higher $(18.28\%,\;10.3\%,\;78.4\%)$ than that of CV, but the content of crude lipid GL was higher than that of CV. 2. Inhibitory effects of on the growth of AGS human gastric adenocarcinoma cells and HT-29 human colon adenocarcinoma cell were increased by increasing of concentration in added methanol extracts of CV and GL. 3. Antimicrobial activity on Escherichia col1; Staphylococcus aureus, Salmonella typhimurium existed methanol extracts of CV. Therefore. it is suggested that CV is worth as functional food.

  • The purpose of this study was to investigate the physiological functionality of Coriolus versicolor Quel (CV) and Ganoderma IUCIdum (Fr.)Karst (GL) by estimate the proximate composition and content of antioxidant components. 1. In the physicochemical property. the content of proximate composition of GL was higher 08.28% of moisture, 10.3% of crude protein, 78.4% of crude fiber) than that of CV, but the content of crude lipid and ashes of GL was higher than that of CV. 2. CV had relatively higher content of antioxidant components such as total phenol, carotenoids and v itamin C. than GL did. Antioxidant mineral components such as magnesium and zinc also had relatively higher in CV than in GL. 3. TBA value, conjugated diene production, LDL oxidation, DPPH of methanol extracts in CV and GL were as high as a-Tocopherol.

  • Present expriments were conducted to determine the possibility of cultivation of Coriolus versicolor selected among the higher fungi growing in Korea. Dried apple sawdust used in this study consisted of 31.5% C, 0.54% N, 2.41 % CaO, 0.05% $P_2O_5$, 0.48% MgO and 0.81% $K_2O$(pH 5.6). Coriolus versicolor mushrooms were cultivated on the sawdust media; apple sawdust; rice bran=80:20 in 850cc polypropylene bottles. The isolate of Coriolus versicolor used was YCV collected from an Apple farm in Youngchun district. It took 34 days to make fruitbody from spawning. The fruit bodies produced the total fresh weight 28 g in a bottles, and converted to 64% of fresh weight.

  • Attempts were made to investigate the sterol components of Coriolus versicolor (Fr.) Quel, which is one of medicinal fungi and which grows wildly in Korea.Its carpophores were collected in the Gyeong Gi Province and extracted with chloroform and methanol. Four Compounds were isolated and one of these compounds was identified as ergosterol by T.L.C., G.L.C. and chemical tests. ${\beta}-Sitosterol$ appears to exist also in the carpophore.

  • The extraction and separation methods of protein-bound polysaccharides from the mycelium and culture broth of Coriolus versicolor (Fr) Quel were investigated. The use of 2% solution of surface active agent Triton X-100, was effective for extraction of the protein-bound polysaccharides from the mycelium. For the separation and partial purification of the protein-bound polysaccharides, the column chromatography using DEAE-Cellulose and DEAE-Sephadex proved to be effective.

  • To investigate mechanism of antineoplastic activities of the protein-polysaccharide fraction of Coriolus versicolor (Fr.) Quel., the mycelium of the fungus was grown in a liquid medium and was extracted with hot water. The extract was purified and used for examining its effects upon immune response in mice. The fraction was administered at a dose of 20mg/kg/day to ICR mice for five days. After ten days the mice were immunized by sheep red blood cells. The plaque-forming cells (PFC) in their spleen were increased in the treated mice. The number of PFC in case of the protein-removed polysaccharide was lower than that of the entire protein-­polysaccharide fraction of the fungus.

  • The objectives of this investigation were to produce artificially an antitumor constituent by submerged culture of the mycelium of Coriolus versicolor (Fr.) Quel., to characterize the influence of various modifications of the nutrient and culture conditions with respect to the pro­duction, to determine chemical composition of the antitumor constituent, and to examine effects of the constituent on the immune response of mice. Submerged agitation of the mycelium in flasks containing a nutrient solution showed its adequate growth. Especially the mycelial growth in the medium containing glucose and yeast extract was abundant. The addition of cotton seed flour or ginseng waste to the medium increased the yield of mycelial growth and the production of the antitumor constituent. The replacement of glucose with starch also yielded the adequate growth. The antitumor constituent extracted from the mycelium and isolated from the culture filtrate was a protein-bound polysaccharide. The analyses of this constituent by GLC and amino acid autoanalysis showed that it contained four monosaccharides and fifteen amino acids. The protein-free polysaccharide of the constituent was also found to exert greater antitumor activity against sarcoma-180 in mice than the entire constituent. The antitumor constituent was found to potentiate the immune response of mice against sheep red blood cell. The protein-bound polysaccharide exerted more favorable influence on the immunity than the protein-free moiety.
