Effects of Liquid Culture of Coriolus Versicolor on Lipid Metabolism and Enzyme Activities in Rats Fed High Fat Diet (운지버섯 균사체 배양액이 고지방 식이를 급여한 흰쥐의 지질대사 및 효소활성에 미치는 영향)

  • The effects of liquid culture of Coriolus versicolor on weight gain, food intakes, food efficiency ratios, serum and hepatic lipid concentrations, serum protein levels and serum enzyme activities, were studied in growing male rats. Sprague-Dawley rats were given four different types of diets for a succeeding period of 5 weeks, respectively: a normal diet group (7% corn oil), a high fat diet group (7% corn oil+15% lard), a 20% or 30% C. versicolor diet groups (high fat diet+20% or 30% C. versicolor in water) according to the levels of C. versicolor supplementation. The body weight gains of the rats fed the 30% C. versicolor diets were lower than those in the rats fed high fat diet. The epididymal fat pad weight of the rats fed high fat diet and 20% or 30% C. versicolor diets were significantly higher than that of the rats fed normal diet. The concentrations of triglyceride in the serum and the liver of the rats fed the 30% C. versicolor diets were more significantly decreased compared to rats on the high fat diet. The concentrations of total cholesterol in the serum and the liver of rats fed the high fat diet, 20% and 30% C. versicolor diets were similar to those of rats fed the normal diet. The HDL-cholesterol concentration and the HDL-cholesterol/total-cholesterol ratio of the rats fed 20% and 30% C. versicolor diets were significantly lower than those of the rats fed high fat diet. But the antherogenic index of the rats fed 20% or 30% C. versicolor diets were significantly higher than those of the rats fed high fat diet. There were no differences in the activities of glutamic oxaloacetic transaminase, glutamic pyruvic transaminase and alkaline phosphatase in the serum among the experimental groups. These results showed that the 30% C. versicolor diet feeding decreased the triglyceride in serum and liver of the rats.

  • Seven white rot fungi (Irpex lactenis, Coriolus hirsutus, Lopharia mirabilis, Schizopora paradoxa, Ganoderma lucidum, Pleurotus ostreatus and Pycnoporus coccineus) native to Korea and two famous exotic lignin degradable white rot fungi (Coriolus versicolor and Phanerochaete chrysosporium) were investigated to clarify their physiological and physicochemical characteristics on white-rotted wood blocks. G. lucidum degraded wood blocks more seriously than those by exotic lignin-degrading fungi, C. versicolor and P. chrysosporium, but only slightly decreasecl the strength of wood which was compared to the weight loss, persumably on the account of its small use of cellulose when attacking wood. It is quite interesting to note that the holocellulose degradation rate of G. lucidum was also higher than any of the other tested fungi. The order of fungi, according to the lignin-decomposing rates, was G. lucidum>P. coccineus>C. versicolor>S. paradoxa>P. chrysosporium>L. mirabilis>P. ostreatus>C. hirsutus>I. lactenis. The lignin degradation of G. lucidum and P. coccineus which were collected in Korea was greater than that of C. versicolor and P. chrysosporium. If holocellulose degradation is not considered. G. lcidum has the merit of actual application in biomass conversion due to linin removal.

  • The effects of liquid culture of Coriolus (C) versicolor on weight gain, food intakes, food efficiency ratios, serum and hepatic lipid concentrations were investigated in male rats fed the high cholesterol diets. Eight weeks old Sprague-Dawley rats were given three different types of diet for six weeks, respectively: a control diet ($ 20\% fat +0.5 \% $ cholesterol), two kinds of C versicolor diet (control diet + $ 30\% or 40\%$C. versicozor in water) according to the levels of C. versicolor supplementation. The body weight gains of the rats fed $ 30\% or 40\% $ C. versicolor diets were lower than those in the rats fed the control diet. The food intake, food efficiency ratios, and liver, kidney, epididymal fat pad weights of the rats fed $ 30\% or 40\%$ C. versicoEer diets were similar to those of the rats fed the control diet. The concentrations of hepatic cholesterol and triglyceride in the rats fed $ 30\% or 40\% $ C. versicolor diets were significantly lower than those in the rats fed the control diet. The concentrations in serum total cholesterol, LDL-cholesterol, and atherogenic index ratios were significantly lower in the rats fed $ 30\% or 40\%$ C. versicolor diets compared to those fed the control diet. The HDL-cholesterol/total-cholesterol ratios was significantly higher in the rats fed $ 30\% or 40\% $ C. versicolor diets compared to those fed the control diet. The fecal excretion of total lipid and triglyceride in the rats fed $ 40\% $ C. versicolor diet was significantly higher than that of the rats fed the control diet. There were no significant difference found in the serum trigly-ceride, phospholipid and HDL-cholesterol concentrations among the experimental groups. These results showed that the C. versicolor feeding decreased the hepatic cholesterol, triglyceride, and the serum total cholesterol, LDL-cholesterol and atherogenic index, and increased the serum HDL-cholesterol/ total-cholesterol ratio of the rats.

Effects of Beta-glucan from Coriolus versicolor on Scavenger Receptor B1 Expression and their Involvement of Dectin-1 and Casein Kinase 2

  • The mushroom Coriolusversicolor contains biologically active polysaccharides, most of which belong to the ${\beta}$ glucan group. Diverse physicochemical properties, due to different sources and isolated types of ${\beta}$-glucans, may induce distinct biological activities. Here, we examined the effects of ${\beta}$-glucan from Coriolusversicolor (CVG) on the scavenger receptor B1 (SR-B1) expression and the role of SR-B1 in CVG-induced phagocytosis regulation by using SR-B1-specific shRNA transfected cells. We also examined whether Dectin-1 and CK2 are involved in SR-B1 expression in CVG-treated cells. Our study results showed that CVG increased the SR-B1 expression via Dectin-1 and CK2 in macrophages. However, the inhibition of SR-B1 expression by shRNA did not completely eliminate the effect of CVG on the increase of phagocytosis suggesting that SR-B1 is not essential for CVG-stimulated phagocytosis. This study will contribute to identify CVG's mechanism of action and its use in the development of functional foods.

Ultrastructural Process of Protoplast Fusion Between Lentinula edodes and Coriolus versicolor

  • Protoplast fusion is a useful technique for establishing fungal hybrids to overcome the natural barriers. The ultrastructure of protoplast and its fusion process were observed using a scanning electron microscopy(SEM) and a transmission electron microscopy(TEM). The protoplasts were variable in size from $0.5{\sim}15{\mu}m$ in diameter, and the mean diameter was about $3{\sim}5{\mu}m$. It was impossible to discriminate protoplasts of Lentinula edodes from protoplasts of Coriolus versicolor by size and surface structure. Big aggregates of the dehydrated protoplasts were observed, after polyethylene glycol 4000 treatment. Nucleus, mitochondria, lipid granules and various vesicles having granules were scattered in the cytoplasm. The vesicles were heterogeneous in size and vary from one protoplast to another. The fused membrane layer of the two protoplasts was observed. Time protoplast membrane contact and reorganization of membrane components were essential condition for protoplast fusion. Transmission electron micrograph showed fused protoplasts and flattening of the cells in the area of the membrane contact. We hope that our electron microscopic observations provide some insights into the understanding of the fusion process of protoplast in fungi.

Anticancer and Antioxidant Activities of Coriolus versicolor Culture Extracts Cultivated in the Citrus Extracts. (감귤 농축액에서 배양한 운지버섯 배양추출물의 항산화 및 항암활성)

  • Coriolus versicolor was grown in a defined synthetic liquid medium and citrus extracts, and the culture extracts were examined for antioxidant activity, nitrite scavenging activity, and in vitro anticancer activity against HeLa, PC-3, HepG2, and A-549 cells. Whereas the culture extracts obtained from the synthetic medium and the un-inoculated citrus extract showed 60 and 22% of the 1,1-diphenyl-2-picrylhydrazyl radical scavenger activity, the culture extracts obtained from the citrus extracts medium exhibited antioxidant activity up to 89%. The nitrite scavenging activity of the culture extracts obtained from the citrus extracts medium and the synthetic liquid medium, and the un-inoculated citrus extract at pH 1.2 were up to 67, 55, and 34%, respectively. The culture extract obtained from the synthetic liquid medium inhibited the growth of HeLa, PC-3, HepG2, and A-549 up to 66, 23, 18, 10% at 48 h of incubation, respectively; however, the culture extract obtained from the citrus extracts medium inhibited the growth of HeLa, PC-3, HepG2, and A-549 up to 75, 82, 55, and 82%, respectively. As a negative control, the un-inoculated citrus extract was examined in the same way and inhibited the growth of HeLa, PC-3, and HepG2 cells 20, 6, and 15% at 48 h incubation, respectively; the inhibition of A-549 cell growth was negligible. These results clearly showed that the fermentation of C. versicolor in the citrus extracts rather than in the defined synthetic medium significantly enhanced the anticancer activity, antioxidant activity, and nitrite scavenging activity.

  • The extraction and separation methods of protein-bound polysaccharides from the mycelium and culture broth of Coriolus versicolor (Fr) Quel were investigated. The use of 2% solution of surface active agent Triton X-100, was effective for extraction of the protein-bound polysaccharides from the mycelium. For the separation and partial purification of the protein-bound polysaccharides, the column chromatography using DEAE-Cellulose and DEAE-Sephadex proved to be effective.
