  • Attempts using in vitro and in vivo selection procedures have been made to search for hammerhead ribozymes that have higher activities than the wild-type ribozyme and also to determine whether other sequences might be possible in the catalytic core of the hammerhead ribozyme. Active sequences selected in the past conformed broadly to the consensus core sequence except at A9, and no sequences were associated with higher activity than that of the hammerhead with the consensus core, an indication that the consensus sequence derived from viruses and virusoids is probably the optimal sequence [Vaish et al. (1997) Biochemistry 36, 6495-6501]. Recently, during construction of ribozyme expression vectors, we isolated a mutant hammerhead ribozyme, with an insertion of G between A9 and G10.1, that appeared to show significant activity [Kawasaki et al. (1996) Nucleic Acids Res. 24, 3010-3016; Kawasaki et al. (1998) Nature 393, 284-289]. We, therefore, characterized kinetic properties of the G-inserted mutant ribozymes in terms of the NUX rule. We demonstrate that the NUX rule is basically applicable to the G-inserted ribozymes and, more importantly, one type of G-inserted ribozyme was very active with $k_{cat}$, value of $6.4\;min^{-1}$ in 50 mM Tris-HCl (pH 8.0) and 10 mM $MgCl_2$ at $37^{\circ}C$.

  • As computing paradigm of the next generation, Ubiquitous Computing has emerged owing to growing of the Internet and computer networking technologies, and RFID (Radio Frequency Identification) which is the core technology to achieve(realize, actualize) the Ubiquitous Computing environment, is being noticed. MIT's Auto-ID Center has proposed PML(Physical Markup Language) which is based on XML (Extensible Markup Language), is a standard language for describing physical objects, in order to interchange of data between each of these RFID application systems. The PML defines only core parts to describe physical objects, but on the other hand the other parts to be needed in practical application have to be defined with extended definitions separately. In this paper, therefore, the object information data model was designed, which defines the type of the object in order. to process electronic tag information in the RFID application service based on PML Core of Auto-ID Center and is applicable to the distribution service parts.

  • Electrical discharge machining (EDM) is an attractive machining technique but it requires electrically conductive ceramic materials. In this study, Alumina matrix composites reinforced with CNTs were fabricated through CNT purification, mixing, compaction and spark plasma sintering (SPS) processes. $Al_2O_3$ nanocomposites with the different CNT concentrations were synthesized. The mechanical and electrical characteristics of $Al_2O_3$/CNTs composites were examined in order to apply the materials to the EDM process. In addition, micro-EDM using wire electrical discharge grinding (WEDG) was conducted under the various EDM parameters to investigate the machining characteristics of machined hole by Field Emission Scanning Electron Microscope (FE-SEM). The results show that $Al_2O_3$/CNTs 10%Vol. was more suitable than the other materials because high conductivity and large discharge energy caused violent sparks resulting in bad machining accuracy and surface quality.

  • For a general office building having a center core, the distinction between the outer wall and the compartment should be clear, and it should be set at a stack effect pressure distribution acting on the respective compartment by a relative area ratio of the respective leak compartment. In the case of office buildings with an independent core for core there, some blocks of shares and exterior pressure distribution in buildings and office buildings typically have different characteristics. Therefore, if the stack effect reduction measures, designed on the basis of a general office building are applied to a stand-alone building, the core of the building should reflect the unique pressure distribution characteristics. This study, performed as part of the object corresponding to the flat plan of the building in the diversification trend, analyzed the stack effect that actually occurs in an office building having the properties intended for stand-core construction, and thus on the basis of and tested by the method of using a conventional stack effect reduction measures. Reviewed in the study, an independent cored office building that does not have the air flow path through the specific space with respect to the center core type office building has a feature, and the variation in characteristics of the pressure distribution inside the building according to this air flow path stack effect was reduced by a variety of measures that should be applied to determine the application.

  • Objectives: To determine and compare the fracture resistance of endodontically treated maxillary central incisors restored with different posts and cores. Materials and Methods: Forty-eight upper central incisors were randomly divided into four groups: cast post and core (group 1), fiber-reinforced composite (FRC) post and composite core (group 2), composite post and core (group 3), and controls (group 4). Mesio-distal and bucco-lingual dimensions at 7 and 14 mm from the apex were compared to ensure standardization among the groups. Twelve teeth were prepared for crown restoration (group 4). Teeth in other groups were endodontically treated, decoronated at 14 mm from the apex, and prepared for posts and cores. Resin-based materials were used for cementation in groups 1 and 2. In group 3, composite was used directly to fill the post space and for core build-up. All samples were restored by standard metal crowns using glass ionomer cement, mounted at $135^{\circ}$ vertical angle, subjected to thermo-mechanical aging, and then fractured using a universal testing machine. Kruskal-Wallis and Mann-Whitney U tests were used to analyze the data. Results: Fracture resistance of the groups was as follows: Control (group 4) > cast post and core (group 1) > fiber post and composite core (group 2) > composite post and core (group 3). All samples in groups 2 and 3 fractured in restorable patterns, whereas most (58%) in group 1 were non-restorable. Conclusions: Within the limitations of this study, FRC posts showed acceptable fracture resistance with favorable fracture patterns for reconstruction of upper central incisors.

  • In this paper, experimental studies of the regrinding of tungsten carbide (WC-Co) tools for high-speed machining were conducted. Regrinding and a subsequent evaluation test were carried out for a flat endmill tool with diameters of 10 mm and 3 mm using a CNC five-axis tool grinder and a CNC three-axis machining center. Tool wear on the two types of endmill tools increased as the cutting length increased, and the tool wear was not influenced by the regrinding state. In case of the micro endmill with a tool diameter of 3 mm, the effective regrinding time was determined for a flank wear threshold of 0.3 mm considering the tool life according to cutting length. The tool lives of the 10 mm and 3 mm endmill tools were increased by 80% and 72%, respectively. This conclusion proves the Feasibility of the recycling of tungsten carbide materials in the high-speed machining of high-hardened materials for industrial applications.

  • The importance of ventilation system is being emphasized by interest of indoor air quality. Especially, heat recovery ventilation system has attracted attention as most effective ventilation plan. Because it can reduce hazardous construction materials, indoor air pollutions, and also can reduce air conditioning energy cost. In heat recovery ventilator, the element core is the most important part. The element core is composed of liner and spacer. And liner and spacer are stacked alternately. On the Liner, heat and humidity transfer are made between supply and exhaust air. And spacer plays a role as a tunnel of exhaust and supply. In this study, we investigated and analyzed the efficiency of a heat recovery ventilator, when the spacer's properties are changed. As a result, difference spacer's properties affect an efficiency of heat recovery ventilator.

  • To elucidate genetic diversity of common reed in Korea, we collected a total of 674 common reed plants from 27 regions in South Korea. Hierarchical clustering using 7 morphological traits divided the 27 common reed populations into 7 groups. Random amplified polymorphic DNA (RAPD) results identified three distinct groups of common reed. Common reed accessions in group I mostly inhabit coastal areas. Group II includes reeds mostly collected from inland areas. Group III consists of common reed accessions collected from inland and coastal areas, suggesting that this group might contain hybrids. In summary, we suggest that parapatric speciation might be an important factor in the genetic diversity of common reed and geographical speciation of common reed that might be also affected by environmental gradients.
