Cell Distribution and Shear Effect on Mammalian Cell Growth in Macroporous Gelatin Microcarriers (다공성 젤라틴 미립담체를 이용한 동물세포의 배양시 담체 내외부의 세포 분포 및 전단응력의 영향)

  • Immobilization of anchorage-dependent animal cells was investigated using macroporous gelatin microcarriers developed in our laboratory. For the observation of the distribution of cells in macroporous beads, Vero-6 cells and CHO cells were cuttured and their distribution in macroporous beads was observed using a confocal microscope. In results, the final concentration of Vero-6 cells and CHO cells on macroporous beads was 2-3 times higher than that on commercial solid microcarriers (Cytodex-3). Also, macroporous microcarriers could hold cells in their macropores. Consequently, the pores protected cells against hydrodynamic shear. Based on Kolmogorov eddy length scale, the smaller eddies (80 $\mu $m) showed the detrimental effect on cells in macroporous beads as compared with 160 $\mu $m of eddies in conventional solid microcarriers.

A Feasibility Study on the Use of Autogeneous GTAW for Correction of Distortions in Welded Aluminum Alloy Structures (알루미늄 熔接構造物의 變形橋正을 위한 Autogeneous GTAW의 適用 可能性 硏究)

  • Characteristics of two correcting methods, a new Autogeneous GTAW heating (TIG) method and the conventional GMAW bead-on plate welding(MIG) method, for distorted aluminum fabrication structures were studied. As a result of microscopic study of Autogeneous GTAW heating and GMAW bead-on plate welding areas, porosities in weld metal and surface cracks in local heating zone were found. Through the mechanical tests, it was verified that porosities decrease tensile strength and surface of distortion, angular displacement and transeverse shrinkage were measures and compared. In order to investigate changes of material properties in heating area and cause of defects such thermal stresses were calculated by ADINA. Through the computations of transient thermal stresses and microscopic observation of fracture surface, thermal stress was found to be the cause of crack during Autogeneous GTAW heating.

  • Recrystallization textures of ferritic stainless steel sheets of STS 430 were varied by means of different cold rolling procedures. The conventional normal rolling led to the evolution of strong through-thickness texture gradients in the final recrystallization texture, while the cross-rolling led to a decrease in texture gradients. Micro-texture observation by EBSD revealed that the formation of band-like orientation colonies formed close to the center layer was responsible for ridging. Modification of the recrystallization texture and microstructure by cross-rolling destroyed band-like orientation colonies and consequently reduced the ridging height.

  • A 110mm diameter aspheric metal secondary mirror for a test model of an earth observation satellite camera was fabricated by ultra-precision single point diamond turning(SPDT). Aluminum alloy for mirror substrates is known to be easily machinable, but not polishable due to its ductility. A harder material, Ni, is usually electrolessly coated on an Al substrate to increase the surface hardness for optical polishing. Aspheric metal secondary mirror without a conventional polishing process, the surface roughness of Ra=10nm, and the form error of Ra=λ/12(λ=632nm) has been required. The purpose of this research is to find the optimum machining conditions for reflector cutting of electroless-Ni coated Al alloy and apply the SPDT technique to the manufacturing of ultra precision optical components of metal aspheric reflector.

  • Development and practical emerging construction technologies has been studied as one of key solution for overcoming these barriers. Many literatures indicated that effective performance evaluation methodologies are demanding for efficient applications of new technologies. This paper presents a performance evaluation methodology using construction simulation technique and introduces studies of two cases where the steel staircase system, as an new technologies was applied and where the conventional method of staircase concrete works. Productivity was measured and calculated by site observation and construction simulations. This results show the quantified influence of new technology to site personnels and assist them to make various decisions for using or not using the technology depending on their own situations.

  • An application of contra-rotating rotors, in which a rear rotor is in tandem with a front one and these rotors rotate in the opposite direction each other, has been proposed against a demand for developing higher specific speed axial flow pump. One prototype rotors, which we have designed with a conventional method, has given the positive slope of head characteristic curve especially in the rear rotor. It is necessary to understand the internal flow behavior in the rear rotor to establish the design guideline for achieving higher and more reliable performance. In the present study, we carried out the experimental investigations of the internal flow field of the rear rotor, especially at the partial flow rate, by Laser Doppler Velocimetry (LDV) for the main flow and the limiting streamlines observation on rotor surfaces for the boundary layer flows.

  • In this paper, were introduce two types of equalizers, called equalizer-a and equalizer-b, applying to wireless communications having unknown channel characteristics. The equalizer-a, which has the single sample detector with equalizer system, is developed while the equalizer-b has the partition detectors with the same system used in equalizer-a. The methodologiy we adopt for designing the equalizers is that the sample space is partitioned into finite number of regions by using quantiles, which are estimated by robbins-monro stochastic approximation (RMSA) algorithm, and the coefficients of equalizers are calculated based on nonlinear minimum mean, square error (MMSE) algorithm. Through the computer simulation, the equalizers show much better performance in equiprobably partitioned sample subspaces of observations than the single sample detector and the detector, which has the conventional equalizer, in unquantized observation space under various noise environments.

  • It is well-known that the performance of turbo codes can be improved by allocating different energies per code symbol. In this paper, based on this observation, we propose a joint encoding and modulation scheme for quadrature amplitude modulated turbo code systems. In the proposed scheme, the amount of energy difference between the turbo coded symbols is optimized by optimizing the constellation of quadrature amplitude modulation(QAM). The proposed scheme offers better coding gain compared to the conventional combination of binary turbo code and QAM at the bit error rate of $10^{-5}$.

  • The fine blanking is a process of press-working which makes possible to produce thick sheet metal parts of finished sheared surface and close dimensional accuracy over whole material thickness in single blanking operation. Fineblanked parts in general are characterized by their cleanly sheared surface over the whole stock thickness and accurate size for the specific functions. The fine blanking technology is urgently needed to remove secondary operations which are necessary in conventional blanking operation and cost reduction. In this study, the effect of material texture and vee-ring on parts for automobile is investigated by experimental observation and analysis.
