Glucoamylase is an important industrial enzyme. Glucoamylase production by industrial Aspergillus niger strain featured with two major problems: (i) empirical substrate feeding methods deteriorating the fermentation performance; and (ii) the high raw materials cost limiting the economics of the glucoamylase product with delegated specification. In this study, we first proposed a novel three-stage varied-rate substrate feeding strategy for efficient glucoamylase production in a 5 L bioreactor using the standard feeding medium, by comparing the changing patterns of the important physiological parameters such as DO, OUR, RQ, etc., when using different substrate feeding strategies. With this strategy, the glucoamylase activity and productivity reached higher levels of 11,000 U/ml and 84.6 U/ml/h, respectively. The performance enhancement in this case was beneficial from the following results: DO and OUR could be controlled at the higher levels (30%, 43.83 mmol/l/h), while RQ was maintained at a stable/lower level of 0.60 simultaneously throughout the fed-batch phase. Based on this three-stage varied-rate substrate feeding strategy, we further evaluated the economics of using alternative carbon sources, attempting to reduce the raw materials cost. The results revealed that cornstarch hydrolysate could be considered as the best carbon source to replace the standard and expensive feeding medium. In this case, the production cost of the glucoamylase with delegated specification (5,000 U/ml) could be saved by more than 61% while the product quality be ensured simultaneously. The proposed strategy showed application potential in improving the economics of industrial glucoamylase production.
본 연구에서 glycerol에서 ethanol을 생산할 수 있는 P. tannophilus ATCC 32691를 선별하여, ethanol 생산조건에 대하여 조사하였다. Ethanol 생산 시, 미량의 공기 공급이 매우 중요함을 확인하였고, 이를 위하여 발효조에서 surface aeration 방법을 통하여 ethanol 생산을 시도하였다. Glycerol이 포함된 YPG 배지(1% yeast extract, 2% peptone, 2% glycerol)에서 ethanol을 생산하는 최적 조건은 880 ml의 배양액에 500 ml/min의 공기를 surface aeration 방법으로 공급하면서, 300 rpm의 agitation speed로 운전할 경우였다. 이 조건을 이용하여 fed-batch 배양을 실시한 결과 배양시작 후, 90시간에 이르러 ethanol이 최고 5.74 g/l 생산되었고, glycerol에 대한 ethanol 수율(Ye/g)은 0.166 이었다.
Cultivation of Spirulina platensis using ammonium salts or wastewater containing ammonium as alternative nitrogen sources is considered as a commercial way to reduce the production cost. In this research, by analyzing the relationship between biomass production and ammonium-N consumption in the fed-batch culture of Spirulina platensis using ammonium bicarbonate as a nitrogen nutrient source, an online adaptive control strategy based on optical density (OD) measurements for controlling ammonium feeding was presented. The ammonium concentration was successfully controlled between the cell growth inhibitory and limiting concentrations using this OD-based feedback feeding method. As a result, the maximum biomass concentration (2.98 g/l), productivity (0.237 g/l d), nitrogen-to-cell conversion factor (7.32 gX/gN), and contents of protein (64.1%) and chlorophyll (13.4mg/g) obtained by using the OD-based feedback feeding method were higher than those using the constant and variable feeding methods. The OD-based feedback feeding method could be recognized as an applicable way to control ammonium feeding and a benefit for Spirulina platensis cultivations.
The effect of acetic acid formation deficiency on recombinant E. coli fermentation was investigated using a mutant strain deficient in acetic acid formation. A mutant strain which does not grow under anaerobic conditions was isolated. The acetic acid production in this strain was negligible in aerobic batch fermentation. The cloned-gene expression in the mutant strain was higher than the wild-type strain. Fed-batch fermentations with controlled specific growth rates were carried out in order to compare the cloned-gene expression between the wild-type and the mutant strains. The expression decreased along with the specific growth rate in both strains. The cloned-gene expression in the mutant strain was 60% higher than in the wild-type strain at the same specific growth rate.
산소를 많이 소비하는 발효공정일수록 배양액중의 용존산소의 농도가 목적생산물의 생산성에 많은 영향을 주는 경우가 많다. 때문에 고농도 발효에 앞서, 발효조의 sparging hole로부터 임펠러 높이에 따른 산소전달 능력을 알아본 결과 공기공급이 1 vvm, 교반속도가 600 rpm에서 산소전달계수($K_La$)는 2.67($min^{-1}$)으로 가장 높았다. 배양 시 용존산소 농도를 20% 이상 유지시켰을 때 온도에 따른 k6ub/IFN-${\alpha}1$ 생성은 $30^{\circ}C$에서 세포증식을 하고 $25^{\circ}C$에서 IPTG로 Induction 하였을 때 발현율이 6.43mg/ml로 total protein의 37%로 가장 많은 양이 발현되는 것을 알 수 있었다. 용존산소 농도에 따른 k6ub/IFN-${\alpha}1$의 발현양은 용존산소 농도가 35%일 때 가장 높은 수율을 나타냈다. 용존산소량은 산소소비 속도를 측정함으로써 정확한 임계점을 찾을 수 있었는데 용존산소량이 35% 유지될 때 산소 전달 속도와 비교하여 가장 적당한 산소공급량임을 확인할 수 있었다.
유전자의 재조합 균주의 생산성을 향상시키기 위한 발효시스템 개발을 목적으로 regulated promoter 인 SUC2 gene 갖는 유전자 재조합 S. cerebisiae를 모델로하여 유전자 산물인 Invertase 발현에 미치는 글루코오스 농도의 영향, 발효 중 플라스미드의 불안정성, 생육특성 및 continuous fed batch system을 연구하였다. 유전자 재조합 균주는 biphasic growth 현상을 보였으며 글루코오스 농도가 0.9g/$\ell$에서 2.2g/L로 증가함에 따라 비증식 속도는 0.224 h$^{-1}$에서 0.226 h$^{-1}$로 증가했으며 숙주효모보다 낮은 값을 나타내었다. 유전자 재조합 균주의 invertase 생산은 발효조내의 글루코오스 농도에 크게 영향을 받아 글루코오스 농도가 0.25~0.4g/L로 감소될 때 invertase 생산이 시작되었으며 회분발효중 플라스미드의 분리는 글루코오스 자화기간 동안에 빈번히 일어났으나 에탄을 자화기간에는 완만해지는 경향을 보였다. 또한 통기를 해주지 않고 배지의 용존산소만으로 배양시킨 결과 통기를 한 경우에 비하여 invertase의 비활성과 총활성이 각각 1.5 및 1.3 배 증가되었다. 균체의 증식단계와 유전자의 발현단계로 구분하여 발효시키기 위하여 영양분을 함유한 글루코오스 용액을 48m1/h(0.096g 글루코오스/48L의 유량으로 12시간 연속적으로 공급하여 fed batch 배양한 결과 invertase의 비활성과 총활성이 비통기적 회분배양 보다 각각 1.74, 2.74배 증가되었다.
A simple control strategy of DO-stat was introduced to the recombinant rGuamerin production process in Pichia pastoris. This induction strategy consisted of two interrelated control loops ‘by which oxygen ratio of inlet gas and methanol feeding rate was controlled. Using this control strategy, over-feeding or under-feeding of methanol could be avoided in concomitance with the efficient control of dissolved oxygen level. As a result, the cell concentration reached 130 g/L and rGuamerin expression level was 450 iu/L, which was more than 40% increased result comparing with the fed-batch process using manual control of methanol feeding rate.
The timing of poly(3-Hydroxybutyrate) (PHB) biosynthesis was controlled by varying the agitation speed of a stirred tank fermentor during the pH-stat fed-batch culture of recombinant Escherichia coli strain GCSC 6576 harboring pSYL107. Using a concentrated whey solution containing ca. 200 g/l lactose as the nutrient feed, the PHB content was only 57% after 35h due to volumetric limitation of the fermentor. However, by limiting the oxygen by maintaining the agitation speed at 300 rpm, the final PHB content increased to 70% after 70h with a cell concentration of 15 g/l. When the agitation speed was increased up to 500 rpm, a cell concentration of 31 g/l with 80% PHB was obtained after 52h. A further increase in the maximum agitation speed increased the cell concentration, PHB concentration, and PHB productivity, however, the PHB content decreased to 56-58%.
본 연구에서는 Bacillus 유래 esterase를 생산할 수 있는 재조합 대장균을 사용하여 유가식 배양을 이용한 고농도 균체 배양을 통해 esterase 생산성을 극대화하고자 하였다. 유가식 배양 중 순수 산소의 공급을 통해 용존산소를 30% 이상 유지한 경우와 포도당농도를 1 g/l 이상 유지한 경우 각각 $OD_{600}$ 76 (35.8 g/l DCW)과 $OD_{600}$ 90 (42.4 g/l DCW)까지 균체량을 증가시킬 수 있었다. 포도당의 공급에도 불구하고 배양 후반에 세포의 성장이 정체되는 현상을 극복하기 위해 yeast extract가 강화된 추가 배지의 공급을 시도하였으며, 그 결과 $OD_{600}$ 185 (87.3 g/l DCW)까지 고농도 균체 배양이 가능함을 확인하였다. 단백질 생산 수율의 향상을 위해 성장 시기에 따라 induction에 의한 세포 성장과 esterase 생산성을 평가하였고, 그 결과 대수 성장기 후반에 induction을 유도한 경우 세포 성장 측면에서는 최대 $OD_{600}$ 190(89 g/l DCW)까지 고농도 균체 배양이 가능함을 확인하였다. Esterase 생산성 측면에서는 대수 성장기 초반에 induction 을 유도한 경우에 비해 최대 5.8배 생산성이 증가됨을 확인할 수 있었다. 따라서 본 연구를 통해 순수산소와 질소원의 공급을 통해 확립된 대장균 고밀도 배양방법을 기초로 IPTG 유도시간을 최적화 함으로써 Bacillus 유래 esterase의 최대 생산성을 확보할 수 있는 배양방법을 확립하였다.

LSW는 유기질소원으로 가격이 가장 저렴하면서도 43.3 mL/L를 이용할 경우에 250 g/L의 glucose로부터 erythritol을 53%의 수율과 0.52 g/L-hr의 생산성 및 132 g/L의 농도로 생산이 가능하였다. 따라서 산업용 쓰이기에 충분한 가능성을 보였다. Erythritol의 생산성 향상을 위한 발효 조건의 최적화에 대한 실험을 수행한 결과 기본 발효배지에 65 mL/L의 농도로 LSW를 이용할 경우에 250 g/L의 glucose로부터 erythritol의 수율은 44% 약간 낮아졌지만, 생산성은 0.66 g/L-hr로 1.3배 증가하였다. Growth stage에서 고농도 균체를 얻기 위하여 glucose를 feeding 방법에 관한 연구에서는 glucose 100 g/L와 LSW 500 mL/L를 같이 넣고 배양하는 회분식 배양 방법을 통해 23시간만에 77.6 g/L의 균체를 얻을 수 있었다. Erythritol의 수율 향상을 위하여 production stage에서 최적의 glucose 농도에 대한 연구를 수행한 결과 발효조 내에서 glucose의 농도가 450 g/L 가 되도록 할 경우에 185.5 g/L의 erythritol을 37.3%의 생성 수율과 1.66 g/L-hr의 생산성으로 얻을 수 있었다. 따라서 산업용 질소원으로 선정된 LSW를 가지고 2단계 유가식 배양을 통하여 회분식 배양과 비교해 볼 때 수율은 1.1배정도 감소되었지만 erythritol의 생산성은 2.5 배 정도 향상시킬 수 있었다.
