Dynamic PIV Measurements of Wake behind a Rotationally Oscillating Circular Cylinder (주기적으로 회전진동하는 원주 후류의 Dynamic PIV 속도장 측정)

  • The temporal evolution of wake behind a circular cylinder oscillating rotationally with a relatively high forcing frequency has been investigated experimentally using a dynamic PIV technique. Experiments were carried out with varying the frequency ratio $F_R\;(=f_f/f_n)$ in the range from 0.0 (stationary) to 1.6 at oscillation amplitude of ${\theta}_A=30^{\circ}$ and Reynolds number of $Re=4.14{\times}10^3$. Depending on the forcing condition ($F_R$), the flow was divided into three regimes; non-lock-on ($F_R=0.4$), transition ($F_R=0.8$, 1.6) and lock-on regimes ($F_R=1.0$) with markedly different flow structure in the near-wake region behind the cylinder. When the frequency ratio was less than 1.0 ($F_R{\le}1.0$), the rotational oscillatory motion of the cylinder decreased the length of the vortex formation region and enhanced the mutual interaction between large-scale vortices across the wake centerline. The entrainment of ambient fluid seemed to play an important role in controlling the near-wake flow and shear-layer instability. However, the flow characteristics changed markedly beyond the lock-on flow regime ($F_R=1.0$) due to high-frequency forcing. At $F_R=1.6$, the mutual interactions between the vortices shed from both sides of the cylinder were not so strong. Thereby, the flow entrainment and momentum transfer into the wake center region were reduced. In addition, the size of the large-scale vortices decreased since the lateral extent of the wake was suppressed.

  • Most of ferrous b.c.c weld materials have martensitic transformation during rapid cooling after welding. It is well known that volume expansion due to the phase transformation could influence on the relaxation of welding residual stress. To apply this effect practically, it is necessary to establish a numerical model which is able to estimate the effect of phase transformation on residual stress relaxation quantitatively. For this purpose, the analysis is carried out in two regions, i.e., heating and cooling, because the variation of material properties following a phase transformation in cooling is different in comparison with the case in heating, even at the same temperature. The variation of material properties following phase transformation is considered by the adjustment of specific heat and thermal expansion coefficient, and the distribution of residual stress in analysis is compared with that of experiment by previous study. In this study, simplified numerical procedures considering phase transformation, which based on a commercial finite element package was established through comparing with the experimental data of residual stress distribution by other researcher. To consider the phase transformation effect on residual stress relaxation, the transition of mechanical and thermal property such as thermal expansion coefficient and specific heat capacity was found by try and error method in this analysis. In addition to, since the transformation temperature changes by the kind and control of alloying elements, the steel with many kinds of transformation temperature were selected and the effect of transformation on stress releasement was investigated by the numerical procedures considering phase transformation.

Temperature-sensitive Liposomes Modified with Poly(N-isopropylacrylamide-co-acrylamide) (Poly(N-isopropylacrylamide-co-acrylamide)로 변형된 온도민감성 리포좀)

  • This study has investigated the temperature-sensitive liposomes, which release anticancer drug(doxorubicin) at the hyperthermia temperature$(~40^{\circ}C)$. The temperature-sensitive liposomes were modified with a copolymers of N-isopropylacrylamide(NIPAAm) and acrylamide(AAm), which exhibit a lower critical solution temperature (LCST) at the hyperthermia temperature. The release of doxorubicin from the modified liposomes was determined by measuring the fluorescence intensity with changing temperature and time. The release of doxorubicin from liposomes modified with poly(NIPAAm-co-AAm) copolymer was increased significantly, because poly(NIPAAm-co-AAm) could undergo the conformational transition in the narrow hyperthermia temperature region$(~40{\pm}2^{\circ}C)$. Moreover, we observed that doxorubicin released from liposomes within 5 minutes, and the size of modified liposomes was 120~170 nm. In this study, we have prepared temperature-sensitive liposomes which could be controlled by temperature. They can be applied in the field of a drug delivery system for tumor targeting by temperature control.

A Study on a grounding system of electric railway (전기철도 접지시스템에 대한 연구)

  • An electrical railway system uses high voltage system for train traction and low voltage system for train control. Railway systems are broadly distributed across mountains, sea sides and cities. Electrical accidents provoke the death and injury of a human being and the damage of the equipment by the overcurrent due to the catenary dropping and by the overvoltage due to lightning. Grounding systems are adopted for the protections of the system from the overcurrent and the overvoltage. Isolation grounding for each system can be easily installed. However, the closed circuits between grounding systems can be occurred. The currents flow through the closed circuits cause the abnormal operation of the system. To overcome the problem of the isolation grounding, the equipotential bonding is usually adopted. The equipotential bonding should have very small grounding resistance. In this paper, we showed the transition from the isolation grounding system to the common grounding system and presented the comparison and the analysis of two grounding systems by simulation. In addition, we proposed the direction for a new grounding system of electric railway.

Rad51 Regulates Reprogramming Efficiency through DNA Repair Pathway

  • Rad51 is a key component of homologous recombination (HR) to repair DNA double-strand breaks and it forms Rad51 recombinase filaments of broken single-stranded DNA to promote HR. In addition to its role in DNA repair and cell cycle progression, Rad51 contributes to the reprogramming process during the generation of induced pluripotent stem cells. In light of this, we performed reprogramming experiments to examine the effect of co-expression of Rad51 and four reprogramming factors, Oct4, Sox2, Klf4, and c-Myc, on the reprogramming efficiency. Co-expression of Rad51 significantly increased the numbers of alkaline phosphatase-positive colonies and embryonic stem cell-like colonies during the process of reprogramming. Co-expression ofRad51 significantly increased the expression of epithelial markers at an early stage of reprogramming compared with control cells. Phosphorylated histone H2AX (${\gamma}H2AX$), which initiates the DNA double-strand break repair system, was highly accumulated in reprogramming intermediates upon co-expression of Rad51. This study identified a novel role of Rad51 in enhancing the reprogramming efficiency, possibly by facilitating mesenchymal-to-epithelial transition and by regulating a DNA damage repair pathway during the early phase of the reprogramming process.

Effects of B and Cu Additions on the Microstructure and Mechanical Properties of High-Strength Bainitic Steels (베이나이트계 고강도강의 미세조직과 기계적 특성에 미치는 B 및 Cu 첨가의 영향)

  • Effects of B and Cu additions on the microstructure and mechanical properties of high-strength bainitic steels were investigated in this study. Six kinds of high-strength bainitic steels with different B and Cu contents were fabricated by thermo-mechanical control process composed of controlled rolling and accelerated cooling. The microstructures of the steels were analyzed using optical and transmission microscopy, and the tensile and impact tests were conducted on them in order to investigate the correlation of microstructure with mechanical properties. Depending on the addition of B and Cu, various low-temperature transformation products such as GB (granular bainite), DUB (degenerated upper bainite), LB (lower bainite), and LM (lath martensite) were formed in the steels. The addition of B and Cu increased the yield and tensile strengths because of improved hardenability and solid solution strengthening, but decreased the ductility and low-temperature toughness. The steels containing both B and Cu had a very high strength above 1.0 GPa, but showed a worse low-temperature toughness of higher DBTT (ductile-to-brittle transition temperature) and lower absorbed energy. On the other hand, the steels having GB and DUB showed a good combination of tensile and impact properties in terms of strength, ductility, yield ratio, absorbed energy, and DBTT.

  • During laser spot welding of the braun tube electron gun, phenomena such as serious spattering and oxidative reaction, etc. were occurred. The spatter occurred from weld pool affects the braun tube, namely it blocks up a very small hole on the shadow mask and causes short circuit between two roles of the electron gun. We guessed that high power density and oxidative reaction are main sources of these problems. So, we studied to prevent and to reduce spatter occurring in spot welding of the braun tube electron gun using pulsed Nd:YAG laser. The characteristics of laser output power was estimated, and the loss of laser energy by optical parameter and spatter was measured by powermeter. The effects of welding parameters, laser defocused distance and incident angle, were investigated on the shape and penetration depth of the laser welded bead in flare and flange joints. From these results, the laser peak power was a major factor to control penetration depth and to occur spatter. It was found that the losses of laser energy by optic parameter and sticked spatter affect seriously laser weldability of thin sheets. The deepest penetration depth is gotten on focal position, and a "bead transition" occurred with a slight displacement of focal position relative to the workpiece surface and the absorption rate of the laser energy is affected by the shape factor of the workpiece. When we changed the incident angle of laser beam, the penetration depth was decreased a little with increasing of the incident angle, and the bead width was increased. The spattering was prevented by considering laser beam energy and incident angle.ent angle.

Effects of the Cyclin D1 Polymorphism on Lung Cancer Risk - a Meta-analysis

  • Background: Cyclin D1 (CCND1) is critical in the transition of the cell cycle from G1 to S phases and unbalanced cell cycle regulation is a hallmark of carcinogenesis. A number of studies conducted to assess the association between CCND1 G870A polymorphism and susceptibility to lung cancer have yielded inconsistent and inconclusive results. In the present study, the possible association above was assessed by a meta-analysis. Methods: Eligible articles were identified for the period up to November 2011. Pooled odds ratios (OR) with 95% confidence intervals (95%CI) were appropriately derived from fixed effects or random-effects models. Sensitivity analysis excluding studies whose genotype frequencies in controls significantly deviated from the Hardy-Weinberg equilibrium (HWE) was performed. Results: Ten case-control studies with a total of 10,548 subjects were eligible. At the overall analysis the CCND1 870A allele appeared to be associated with elevated lung cancer risk (for allele model, pooled OR = 1.24, 95% CI: 1.08-1.44, P = 0.004; for homozygous model, pooled OR = 1.45, 95% CI: 1.14-1.84, P = 0.003; for recessive model, pooled OR = 1.29, 95% CI: 1.06-1.58, P = 0.013; for dominant model, pooled OR = 1.33, 95% CI: 1.08-1.65, P = 0.009). Subgroup analyses by ethnicity and sensitivity analysis further pointed to associations, particularly in Asians. Conclusion: This meta-analysis suggests that the A allele of CCND1 G870A polymorphism confers additional lung cancer risk.

Lentivirus-mediated shRNA Interference Targeting SLUG Inhibits Lung Cancer Growth and Metastasis

  • Objective: Lung cancer is a deadly cancer, whose kills more people worldwide than any other malignancy. SLUG (SNAI2, Snail2) is involved in the epithelial mesenchymal transition in physiological and in pathological contexts and is implicated in the development and progression of lung cancer. Methods: We constructed a lentivirus vector with SLUG shRNA (LV-shSLUG). LV-shSLUG and a control lentivirus were infected into the non-small cell lung cancer cell A549 and real-time PCR, Western blot and IHC were applied to assess expression of the SLUG gene. Cell proliferation and migration were detected using MTT and clony formation methods. Results: Real-time PCR, Western Blot and IHC results confirmed down-regulation of SLUG expression by its shRNA by about 80%~90% at both the mRNA and protein levels. Knockdown of SLUG significantly suppressed lung cancer cell proliferation. Furthermore, knockdown of SLUG significantly inhibited lung cancer cell invasion and metastasis. Finally, knockdown of SLUG induced the down-regulation of Bcl-2 and up-regulation of E-cadherin. Conclusion: These results indicate that SLUG is a newly identified gene associated with lung cancer growth and metastasis. SLUG may serve as a new therapeutic target for the treatment of lung cancer in the future.

Development of Verification and Conformance Testing Tools for Communication Protocol (통신 프로토콜 검정기 및 적합성시험 도구 개발)

  • As a very important part in development of the protocol, verification and conformance test for protocol specification are complementary techniques that are used to increase the level of confidence in the system functions as prescribed by their specifications. In this paper, we verify the safety and liveness properties of rail signal control protocol type 1 specified in LTS with model checking method, and experimentally prove that it is possible to check for the deadlock, livelock and rechability of the states and actions on LTS. The implemented formal checker is able to verify whether properties expressed in modal logic are true in specifications using modal mu-calculus. We also propose a formal method on generation of conformance test cases using the concept of UIO sequences from verified protocol specification. The suggested tools are implemented by C++ language under Windows NT.
