몇가지 생리활성 저해제가 금어초 절화의 굴지성 반응에 미치는 효과 (Chemical Suppression of Gravitropic Bending Response in Flower Stalks of Snapdragon (Antirrhinum majus L.))

  • 절화의 수확 후 저장 및 유통과정 중에서 야기될 수 있는 문제 중의 하나인 굴지성 반응에 의한 화경의 구부러짐을 막아내기 위하여 CDTA, $CoCl_2$ 및 ortho-vanadate의 절화 굴지성 반응 억제 효과를 검정하였다. 절화의 굴지성 반응억제제 처리는 절화줄기 기부 혹은 굴지성 반응 부위를 시험용액에 담구는 두가지 방법에 의거 실시하였다. 절화기부를 저해제 용액에 1시간 침지시키는 방법으로 처리하였을 경우, P-type ATPase 저해제인 vanadate만 굴지성 반응 억제효과를 보였으나, 절화의 굴지성반응 활성부위를 직접 계면활성제 (Triton X-100, 0.01%)가 녹아있는 저해제 용액에 침지하였을 경우 CDTA 및 vanadate처리구에서도 상당한 정도의 굴지성 억제를 관찰 할 수 있었을 뿐만 아니라 특히 $CoCl_2$ 처리구에서 그 효과가 현저하였다. $CoCl_2$ 농도에 따른 굴지성 반응 억제효과의 변화를 조사한 결과, 20 mM 전후의 농도에서 거의 완벽한 효과를 볼 수 있었다. 이상의 결과를 고려해 볼 때, 적절한 계면활성제와 $CoCl_2$를 이용한 처리액을 준비하여 활성부위에 직접처리하는 방법을 개선한다면 산업적 응용성이 높은 절화의 굴지성 억제 수단을 마련할 수 있다는 결론을 얻었다.

키토산이 피복된 나노 크기의 자성체 분말 제조 (Preparation of Chitosan-coated Magnetite Nanoparticles)

  • 이 연구의 목적은 약물의 분리나 약물전달 시스템에 이용 할 수 있도록 마그네타이트 나노 입자의 표면을 개질, 즉 생체적합성, 저독성의 특성을 갖는 키토산을 이용하여 키토산이 피복된 나노 크기의 자성체 입자를 제조하는 것이다. 본 연구에서는 음향화학법을 적용한 공침 기술을 이용, 균일한 마그네타이트 나노 입자를 합성하였다. 계면활성제인 올레인산을 가하여 입자간의 응집을 막았다. 물과 계면활성제의 농도비 R=[물]/[계면활성제]를 조절하여 평균크기가 2nm에서 9nm인 마그네타이트 입자를 선택적으로 합성하였다. 이 방법을 통하여 합성된 마그네타이트 나노 인자를 염기성 수용액에 분산시켰다. 초음파를 조사하면서 키토산 용액을 서서히 가하여 마그네타이트 나노 입자의 표면을 키토산으로 피복하였다. 이때 키토산의 농도가 증가할수록 입자가 수 나노미터 크기씩 증가하는 것을 입도 분석기와 원자 현미경 관찰을 통해서 확인 할 수 있었다. 합성된 마그네타이트 분말과 키토산이 피복된 마그네타이트 나노 입자 모두 실온에서 초상자성을 보이는 것으로 나타났다.

Antimicrobial Active Substances from Entomopathogenic Fungi (Various Applications of Entomopathogenic Fungi)

  • Insects constitute the largest and most diverse group of animals in the world. They also serve as the hosts or nutrient sources for an immense assemblage of pathogens, parasites, and predators. More than 700 fungal species from 100 genera have adopted an entomopathogenic lifestyle. Although entomopathogenic fungi were studied as only biocontrol agents against a variety of pests in various countries, it has been recently focused their additional roles in nature. They are antagonists to/against plant pathogens, endophytes, and possibly even plant growth promoting agents. The potential antimicrobial effect against fungal plant pathogens by an isolate of entomopathogenic fungi including Beauveria bassiana, Lecanicillium spp., and Isaria fumosorosea have been reported since late 1990s, but wasn't reported pathogenicity of the isolate against pests. Later, a Canadian Lecanicillium sp. isolate and L. longisporium isolated from Vertalec$^{(R)}$ showed simultaneous control effect against both aphid and cucumber powder mildew. Therefore, the antimicrobial activities of 342 fungi isolates collected from various regions and conditions in Korea were evaluated against plant pathogenic fungus Botrytis cinerea using dual culture technique on agar plate. As a result, 186 isolates (54.4%) shown the antifungal activity against B. cinerea. The culture filtrates of selected fungi completely suppressed the growth of the microorganisms, indicating that suppression was due to the presence of antimicrobial substances in the culture filtrate. Mode of action of these fungi against insect involves the attachment of conidia to the insect cuticle, followed by germination, cuticle penetration, and internal dissemination throughout the insect. During infection process, secreted enzymes, proteinous toxins, and/or secondary metabolites secreted by entomopathogenic fungi can be used to overcome the host immune system, modify host behavior, and defend host resources. Recently, secondary metabolites isolated from entomopathogenic fungi have been reported as potential bioactive substances. Generally, most of bioactive substances produced by entomopathogenic fungi have reported low molecular weight (lower than 1,000 g/mol) as peptide and, in contrast the high molecular weight fungal bioactive substances are rare. Most substances based on entomopathogenic fungi were shown antimicrobial activity with narrow control ranges. In our study we analyzed the antimicrobial substances having antagonistic effects to B. cinerea. Antimicrobial substances in our fungal culture filtrates showed high thermostability, high stability to proteolytic enzymes, and hydrophilicity and their molecular weights were differed from substance. In conclusion, entomopathogenic fungi showed pathogenicity against insect pests and culture filtrate of the fungi also shown to antimicrobial activity. In the future, we can use the entomopathogenic fungi and its secondary metabolites to control both insect pest control and plant pathogenic fungi simultaneously.

Preparation of silica-coated gadolinium compound particle colloid solution and its application in imaging

  • A preparation method for gadolinium compound (GdC) nanoparticles coated with silica ($GdC/SiO_2$) is proposed. GdC nanoparticles were prepared with a homogeneous precipitation method at $80^{\circ}C$ using $1.0{\times}10^{-3}$ M $Gd(NO_3)_3$, 0.5 M urea and $0-3.0{\times}10^{-4}$ M ethylenediarinnetetraacetic acid disodium salt dihydrate (ETDA) in water. As a result of preparation at various EDTA concentrations, GdC nanoparticles with a size as small as $40.5{\pm}6.2$ nm, which were colloidally stable, were prepared at an EDTA concentration of $2.0{\times}10^{-4}$ M. Silica-coating of the GdC nanoparticles was performed by a St$\ddot{o}$ber method at $35^{\circ}C$ using $1.0-10.0{\times}10^{-3}$ M tetraethylorthosilicate (TEOS), 11 M $H_2O$ and $1.5{\times}10^{-3}$ M NaOH in ethanol in the presence of $1.0{\times}10^{-3}$ M GdC nanoparticles. Performance of preparation at various TEOS concentrations resulted in production of $GdC/SiO_2$ particles with an average size of $106.1{\pm}11.2$ nm at a TEOS concentration of $5.0{\times}10^{-3}$ M. The gadolinium (Gd) concentration of $1.0{\times}10^{-3}$ M in the as-prepared $GdC/SiO_2$ particle colloid solution was increased up to a Gd concentration of 0.2 M by concentrating with centrifugation. The core-shell structure of $GdC/SiO_2$ particles was undamaged, and the colloid solution was still colloidally stable, even after the concentrating process. The concentrated $GdC/SiO_2$ colloid solution showed images of X-ray and magnetic resonance with contrast as high as commercial Gd complex contrast agents.

Klebsiella pneumoniae균의 바이오 필름 형성과 활성에 대한 병두황진 추출물의 효과 (Effects of Scutellaria scordifolia Fisch. ex Schrank Extracts on Biofilm Formation and the Activities of Klebsiella pneumoniae)

  • 주술적 의료로 이용되는 식물에서 새로운 항미생물질을 찾는 있는 중에 바이오필름의 출현은 임상, 의학 제조업 분야에서 다급한 신호를 보내주고 있는 것이다. 국립보건원(NIH)은 세계 모든 미생물 감염의 70%가 바이오필름과 연관되어 있다고 추정하고 있으며, 기존의 항생제에 내성이 있는 변종의 출현은 전세계 공중 보건에 심각한 위협이 되고 있다. 따라서 대체의학 물질을 찾아 내는 것은 통합(동서양) 의학의 중요한 화두이다. 이 연구에서 Klebsiella pneumoniae의 바이오 필름 형성과 활성화에 대한 네 가지 허브 추출물을 스크린 하였다. 본 실험에 사용된 식물 추출액 중에 Scutellaria scordifolia Fisch. ex Schrank 추출물이 박테리아의 성장과 바이오 필름 형성에 억제 효과를 보여 주었다. Scutellaria scordifolia Fisch. ex Schrank 추출물이 L929세포에 대한 세포독성을 띄지 않음을 확인 하였다. Scutellaria을 첨가한 실험군에서 다른 대조군에 비해 K. pneumoniae의 성장이 억제되었다. Scutellaria scordifolia Fisch. ex Schrank 추출액이 첨가되지 않은 대조군과 비교하면, 식물 추출액이 첨가된 실험군에서는 생균수가 최대 약 60배 억제되었음을 확인 하였다. Scutellaria scordifolia Fisch. ex Schrank 추출물이 포함된 실험 군에서 K. pneumoniae 균의 개체수가 줄어들어 있음을 differential interference contrast (DIC)의 이미지를 통해서도 확인하였으며, 형광세기의 비교분석을 통해 음성대조군에 비해 추출물이 포함된 실험군에서 형광세기가 통계학적으로 유의하게 낮아져 있는 것을 확인하였다. 이러한 결과는 이 식물 추출물이 K. pneumoniae의, 특히 바이오필름 형성에서 항균제로서 적용될 수 있음을 암시해 주는 것이다.

Distinct Effect of Neurotrophins Delivered Simultaneously by an Adenoviral Vector on Neurite Outgrowth of Neural Precursor Cells from Different Regions of the Brain

  • For many years, it has been demonstrated that neurotrophins regulate the adult nervous system, implicating their potential as therapeutic agents for the treatment of neurodegenerative diseases. We generated adenoviral vectors encoding brain-derived neutotrophin factor (BDNF) and neurotrophin-3 (NT3) and tested either separately or together for the ability to induce differentiation of neuronal precursor cells with two different origins. Separate transduction of adenovirus delivering BDNF (BDNF-Ad) or NT3 (NT3-Ad) induced the neuronal differentiation in hippocampal and cortical precursor cells. NT3-Ad infected cells extended short neurites, whereas BDNF-Ad infected cells had longer neurites. In the early differentiation of hippocampal precursor cells, simultaneous infection of BDNF-Ad and NT3-Ad promoted further differentiation and neurite elongation compared with the separate infection of each virus. In contrast, simultaneous infection did not show the synergistic effect in the cortical precursor cells, suggesting that the neurotrophins play distinct roles in different regions of the brain. However, the numbers of neurites and spines per differentiated cells were markedly increased in cortical as well as hippocampal precursor cells, indicating the promotion of efficient neurite elongation and formation of dendritic spine, when BDNF-Ad and NT3-Ad were co-infected. These results suggest more studies in the effect of a combinatorial use of neurotrophins on different sites of brain need to be carried out to develop gene therapy protocols for neurodegenerative diseases.

Protective Role of Selenium and High Dose Vitamin E against Cisplatin - Induced Nephrotoxicty in Rats

  • Background: Cisplatin (CDDP) is one of the most active cytotoxic agents in the treatment of cancer. We investigated the effect of selenium (Se) with high dose vitamin E (VE) administration to prevent CDDP-induced nephrotoxicity in rats. Materials and Methods: In this study, 40 female Wistar rats were randomly divided into five equal groups. The first group, which served as the control, was administered physiological saline (2.5 cc/day, 5 days) intraperitoneally (IP), while group A was administered cisplatin (6 mg/kg BW/ single dose) plus physiological saline IP. Groups B, C, D received IP five doses of Se (1.5 mg/kg BW), and a high dose of VE (1000 mg/kg BW) (Se-VE) in combination before, simultaneously, and after CDDP, respectively. The rats were sacrificed five days after CDDP administration. Plasma malondialdehide (MDA), glutathione peroxidase (GSH-Px), reduced glutathione (GSH), catalase, urea, creatinine levels, renal histopathological changes were measured. Results: The histopathological injury score, plasma levels of MDA, urea, creatinine were found to increase in group A compared to the control (p<0.05), while plasma levels of GSH-Px, GSH and catalase decreased (p<0.05). In contrast, plasma levels of MDA decreased (p<0.05) in groups B, C, D, which were treated with Se- VE, whereas levels of GSH-Px, GSH were found to increase only for group D (p<0.05). Plasma urea, creatinine levels improved in the treatment groups compared to group A (p<0.001). Histopathological changes caused by CDDP were also significantly improved after Se-VE treatment (p<0.05). Conclusions: Oxidative stress increases with CDDP-induced nephrotoxicity in rats. Se-VE supplementation might thus play a role in the prevention of CDDP-induced nephrotoxicity in patients.

Quantification of Gadolinium Concentration Using GRE and UTE Sequences

  • Purpose: To compare different MR sequences for quantification of gadolinium concentration. Materials and Methods: Gadolinium contrast agents were diluted into 36 different concentrations. They were scanned using gradient echo (GRE) and ultrashort echo time (UTE) and R1, $R2^*$ and phase values were estimated from collected data. For analysis, ROI masks were made for each concentration and then ROI value was measured by mean and standard deviation from the estimated quantitative maps. Correlation analysis was performed and correlation coefficient was calculated. Results: Using GRE sequence, R1 showed a strong linear correlation at concentrations of 10 mM or less, and $R2^*$ showed a strong linear correlation between 10 to 100 mM. The phase of GRE generally exhibited a negative linear relationship for concentrations of 100 mM or less. In the case of UTE, the phase had a strong negative linear relationship at concentrations 100 mM or above. Conclusion: R1, which was calculated by conventional GRE, showed a high performance of quantification for lower concentrations, with a correlation coefficient of 0.966 (10 mM or less). $R2^*$ showed stronger potential for higher concentrations with a correlation coefficient of 0.984 (10 to 100 mM), and UTE phase showed potential for even higher concentrations with a correlation coefficient of 0.992 (100 mM or above).

Microbe Hunting: A Curious Case of Cryptococcus

  • C. neoformans-associated cryptococcosis is primarily a disease of immunocompromised persons, has a world-wide distribution, and is often spread by pigeons in the urban environment. In contrast, C. gattii causes infection in normal hosts, has only been described in tropical and semi-tropical areas of the world, and has a unique niche in river gum Eucalyptus trees. Cryptococcosis is acquired through inhalation of the yeast propagules from the environment. C. gattii has been identified as the cause of an emerging infectious disease centered on Vancouver Island, British Columbia, Canada. No cases of C. gattii-disease were diagnosed prior to 1999; the current incidence rate is 36 cases per million population. A search was initiated in 2001 to find the ecological niche of this basidiomycetous yeast. C. gattii was found in the environment in treed areas of Vancouver Island. The highest percentage of colonized-tree clusters were found around central Vancouver Island, with decreasing rates of colonization to the north and south. Climate, soil and vegetation cover of this area, called the Coastal Douglas fir biogeoclimatic zone, is unique to British Columbia and Canada. The concentration of airborne C. gattii was highest in the dry summer months, and lowest during late fall, winter, and early spring, months which have heavy rainfall. The study of the emerging colonization of this organism and subsequent cases of environmentally acquired disease will be informative in planning public health management of new routes of exposure to exotic agents in areas impacted by changing climate and land use patterns.

Apoptotic Effect of Co-treatment with Curcumin and Cisplatin on SCC25 Human Tongue Squamous Cell Carcinoma Cell Line

  • Curcumin is a widely used flavoring agent in food, and it has been reported to inhibit cell growth, to induce apoptosis, and to have antitumor activity in many cancers. Cisplatin is one of the most potent known anticancer agents and shows significant clinical activity against a variety of solid tumors. This study was undertaken to investigate the synergistic apoptotic effects of co-treatment with curcumin and cisplatin on human tongue SCC25 cells. To investigate whether the co-treatment efficiently reduced the viability of the SCC25 cells compared with the two treatments separately, an MTT assay was conducted. The induction and the augmentation of apoptosis were confirmed by DNA electrophoresis, Hoechst staining, and an analysis of DNA hypoploidy. Western blot, MMP and immunofluorescence tests were also performed to evaluate the expression levels and the translocation of apoptosis-related proteins following the co-treatment. In this study, following the co-treatment with curcumin and cisplatin, the SCC25 cells showed several forms of apoptotic manifestation, such as nuclear condensation, DNA fragmentation, reduction of MMP, increased levels of Bax, decreased levels of Bcl-2, and decreased DNA content. In addition, they showed a release of cytochrome c into the cytosol, translocation of AIF and DFF40 (CAD) to the nuclei, and activation of caspase-7, caspase-3, PARP, and DFF45 (ICAD). In contrast, separate treatments of $5{\mu}M$ of curcumin or $4{\mu}g/ml$ of cisplatin, for 24 hours, did not induce apoptosis. Therefore, our data suggest that combination therapy with curcumin and cisplatin could be considered as a novel therapeutic strategy for human oral squamous cell carcinoma.
