Optimization of Culture Conditions and Continuous Production of Chitosan by the Fungi, Absidia coerulea

  • The production of chitosan from the mycelia of Absidia coerulea was studied to improve cell growth and chitosan productivity. Culture conditions were optimized in batch cultivation (pH 4.5, agitator speed of 250 rpm, and aeration rate of 2 vvm) and the maximum chitosan concentration achieved was 2.3g/L under optimized conditions. Continuous culture was carried out successfully by the formation of new growth spots under optimized conditions, with a chitosan productivity of 0.052g/L(sup)-1 h(sup)-1, which is the highest value to date, and was obtained at a dulution rate of 0.05h(sup)-1. Cell chitosan concentrations reached about 14% in the steady state, which is similar to that achieved in batch culture. This study shows that for the continuous culture of Absidia coerulea it is vital to control the medium composition.

먹이가용성에 의한 고성만의 굴 양식장 수용력 (Estimation of Carrying Capacity by Food Availability for Farming Oysters in Goseong Bay, Korea)

  • For the continuous stable production of oyster, estimation of food availability (F) was carried out in Goseong Bay, south of coast Korea. Primary productivity ranged from 0.07 to $0.44gC/m^2/day$ (average $0.25gC/m^2/day$), lowest in July and highest in January. The distribution of primary productivity at Goseong Bay showed the pattern of "high in the south and low in the north." Food availability (F) was $F{\leq}0$, indicating insufficient food supply, from August to November and F > 0 from January to April. Continuous insufficient food supply was observed at 18 oyster farms in the southern part of the bay and 4 in its northern part. Mortality at the oyster farms was 56% on the average, and around 58% of death occurred during November when food supply was insufficient. The optimal population of cultured oyster per unit flow area was calculated to be $110-115indiv./m^2$ (198-201 indiv./string). When the sea area was divided into 3 regions (A, B, C) according to carrying capacity, the carrying capacity of (A) regions was $52-53indiv./m^2$ (93-95 indiv./string), (B) regions was $142-144indiv./m^2$ (255-259 indiv./string), and (C) regions was $198-202indiv./m^2$ (356-363 indiv./string). In particular, (A) regions showed extremely low productivity. For continuous stable oyster farming at Goseong Bay, it is necessary to control point and non-point source pollution through continuous environmental monitoring and to adjust harvest according to the base carrying capacity during the season of high water temperature.

Methylomonas mnosa에 의한 Exopolysaccharide의 유가식 및 연속 생산 (Exopolysaccharide Production in Fed-batch and Continuous Culture by Methylomonas mucosa)

  • Methylomonas mucosa를 이용하여 세포외 다당류의 생산을 알아보았다. 본 미생물은 성장을 위해 메탄올을 탄소원으로 이용하여 세포외 다당류를 생산한다. 효과적인 발효 공정 시스템의 개발올 위해 회 분식, 유가식, 연속식 배양을 수행하여 각각의 당류 에 대한 생산성을 알아보았다. 플라스크 배양에서 메탄올이 약 1 % (v/v) 이상에서 다당류의 생산과 세포의 성장은 저해되었다. 1% (v/v)에서 최대 비성장속도를 나타내었고 C/N 비율이 높을수록 다당류의 생산은 많았다. 다당류의 생산에 $Mg^{2+}$ 이온 농도가 크게 영향을 줌을 확인할 수있었다. 유가식 배양에서는 다당류의 농도가 회분식에 비 해 4배 이상 높았으나 수율은 오히려 감소하였다. 연속주입을 통한 유가식 배양은 간헐적 주업방법보다 생산성이 높았다. 이는 간헐적 주입에서 나타날 수 있는 메탄올에 의한 제한이나 저해가 없었기 때 문으로 보여진다. 연속조엽은 산소 제한을 피하기 위해 순수산소를 공급하였다. 희석률이 O.21h-1까지 증가하여도 수융 과 생산성은 증가하였다. 생산된 다당류 용액의 점도는 다당류의 증가에 따라 지수적으로 증가하였다.

Continuous Production of Succinic Acid Using an External Membrane Cell Recycle System

  • Succinic acid was produced by continuous fermentation of Actinobacillus succinogenes sp. 130Z in an external membrane cell recycle reactor to improve viable cell concentration and productivity. Using this system, cell concentration increased to 16.4 g/l at the dilution rate $0.2\;h^{-1}$, up to 3 times higher than that of batch culture, and the volumetric productivity of succinic acid increased up to 6.63 g/l/h at the dilution rate $0.5\;h^{-1}$, 5 times higher than that of batch fermentation. However, in the continuous culture using a high dilution rate, operational problems including severe membrane fouling and contamination by lactic acid producer were observed. Another succinic acid producer, Mannheimia succiniciproducens MBEL55E, was also utilized in this system, and the cell concentration and productivity of succinic acid at the dilution rate of $0.3\;h^{-1}$ were found to be above 3 and 2.3 times higher, respectively, compared with those obtained at the dilution rate of $0.1\;h^{-1}$. These observations give a deep insight into the process design for a continuous succinic acid production by microorganisms.

  • In this study, it was observed that hydrogen Productivity varied with stirrer speed, bead radius, input glucose concentration and dilution rate in a continuous stirred tank reactor in which immobilized R. rubrum KS-301 was used as a hydrogen-producing bacterium The mass transfer resistance due to cell immobilization was also studied. In order to estimate an effectiveness factor, Des of glucose was first obtained, which was subsequently represented by the correlation equation between Dos and Xb, As a result external mass transfer resistance could be neglected for stirrer speeds greater than 400rpn With bead radius increasing, the hydrogen productivity and internal effectiveness factor decreased. With input 91ucose concentration increasing, the hydrogen productivity and interval and external effectiveness factor increased. Although an Internal effectiveness factor was not affected, hydrogen productivity Increased with dilution rate increasing. An overall effectiveness factor remained nearly constant for the dilution rates investigate4 but increased with input 91ucose concentration increasing.

  • Welding processing is used in the various industrial fields such as shipbuilding, car, airplane and steel structure, etc.. But the welding process has a bad working condition and lack of skillful worker. The welding depended on man power causes low productivity and difficulty in keeping continuous and stable quality control. This paper shows the development results of welding mobile robot with the several functions such as continuous/intermittent welding, initial welding speed control, acceleration control, crater and deceleration speed control in welding end. The robot is developed based on microprocess which is intel 80c196kc.

  • For the continuous development of the manufacturing company, selp-helpㆍdiligence of the individual members of the organization and cooperation system of the sub-department process of the manufacture must operate under rationality. Moreover, perpetual and reasonable improvement on productivity should be focused on. Improvement on productivity implies raising $\ulcorner$output / input$\lrcorner$ ratio, and this does not only implies simple increase of the level of output. Therefore under the theoretical principle, Which is designed to increase the ratio of $\ulcorner$output / input$\lrcorner$ -economically select the relevant $\ulcorner$methodology$\lrcorner$. When rational improvement on productivity $\ulcorner$techniques$\lrcorner$ are properly applied with this methodology, calculated result will come out. The ultimate aim of improving quality does not mean simply reducing the percentage of the defects, but means improving the quality of products, for customer satisfaction. Thus recognize this, we have to contribute to the national economy growth through the enhancement of added value of individual company by $\ulcorner$improvement on quality and productivity, and reduction of the cost$\lrcorner$, $\ulcorner$improvement program on quality and productivity$\lrcorner$ shouldn't be just the parading formality. We can expect the economy result, these are applied to the learning essence and the principle of the improvement.

Zymomonas mobilis에 의한 Packed Bed Reactor를 이용한 연속적인 sorbitol의 형성 (Continuous Production of Sorbitol with Zymomonas mobilis in a Packed Bed Reactor)

  • K-carrageenan에 고정화시킨 Zymomonas mobilis를 이용한 연속적인 sorbitol 생산에 대하여 연구하였다. Toluene과 glutaraldehyde로 처리하여 투과성을 증대시킨 세포를 alginate나 chitin 고정화 공정에서 어느 정도의 효소활성을 보였으나 210시간 이상의 공정반응에서는 sorbitol 생성량이 감소되었다. 독성이 적고 투과성을 증가시키는 물질로서 toluene 대신 CTAB(Cetyltrimethylammoniumbromide)를 사용하였다. CTAB로 투과성을 증가시킨 세포를 k-carrageenan에 고정화하여 CSTR과 packed bed reactor를 이용하여 sorbitol 생산을 시도하였으며 CSTR보다도 (25일) packed bed reactor에서 (30일) 더 긴 시간 효소활성도가 유지되었다. Two-stage 연속공정에서는 희석비율이 (dilution rate, $h^{-1}$) 증가함에 따라서 sorbitol 생성률이 증가되었으며 희석비율 0.32 $h^{-1}$에서 첫번째와 두번째 반응조에서 각각 $15g/\ell-h,$ $22g/\ell-h의$ sorbitol 생성률을 얻었다. 0.32$h^{-1}$보다 높은 희석비율에서는 생성를이 감소되었다.

  • Recently batch type cold rolling processes have been replaced by continuous annealing type processes for cold rolled sheets of mild steel and high strength steel in order to obtain higher productivity, labor saving. In the continuous annealing line, it is very important to maintain the target steel strip temperature at the exit side of each furnace. The automation system of continuous annealing line is based on a hierachical composition. This paper shows how to preset the set value of furnace temperature control for the heating section in a continuous annealing line. Saying in other words, this paper presents the development of an adaptive control approach to control the exit strip temperature in the continuous annealing line. There are three parts in this approach; one is a process modelling and another is recursive parameter estimation and the other is a design of temperature controller.

혼합영양생물인 Scenedesmus acuminatus의 반연속 배양 시 성장특성 연구 (Growth Characteristics of Mixotrophic Scenedesmus acuminatus under Semi-Continuous Culture System)

  • 본 연구는 혼합영양생물인 Scenedesmus acuminatus의 최적배양조건을 찾기 위한 연구로서 최적 공기주입량, 아세테이트 주입량, $CO_2$ 주입에 대해 실험을 진행하였다. 공기주입량은 0.72 vvm에서 최대 비성장속도와 최대바이오매스 생산량을 얻을 수 있었으며, 0.3 M의 아세테이트를 주입하였을 때 최대 비성장속도와 최대바이오매스 생산량을 얻었다. 반연속배양에서는 추가로 $CO_2$ 주입이 배양에 미치는 영향을 파악하였다. $CO_2$ 주입 실험에서는 아세테이트로 50% 치환한 후에 최대비성장속도($0.460d^{-1}$)와 최대바이오매스 생산성($0.936gL^{-1}d^{-1}$)을 얻을 수 있었으며 이후 치환이 계속될수록 최대비성장속도와 최대바이오매스 생산성은 지속적으로 감소하였다. 공기주입 실험에서는 JM 배지로 배양할 때 가장 높은 비성장속도($0.381d^{-1}$)와 최대바이오매스 생산성($0.253gL^{-1}d^{-1}$)을 보여주었지만 이후 아세테이트를 함유한 배지로 50% 치환하였을 때 오히려 초기값보다 감소하는 것을 관찰하였다.
